Overproduction of Laccase by the White-Rot Fungus Pleurotus ostreatus Using Apple Pomace as Inducer

  • Laccase activity of Pleurotus ostreatus is significantly increased by the addition of apple pomace. Among various conditions, the best concentration of apple pomace and cultivation time for the production of laccase by P. ostreatus was 2.5% and 9 days, respectively. Reverse transcription polymerase chain reaction analyses of laccase isoenzyme genes, including pox1, pox3, pox4, poxc, poxa3, and poxa1b, revealed a clear effect of apple pomace on transcription induction. Our findings reveal that the use of apple pomace can be a model for the valuable addition of similar wastes and for the development of a solid-state fermenter and commercial production of oyster mushroom P. ostreatus.

Yield, Nutritional Content, and Antioxidant Activity of Pleurotus ostreatus on Corncobs Supplemented with Herb Residues

  • Improper disposal of herb residues in China has caused severe problems to the surrounding environment and human safety. Three herb residues, i.e., compound Kushen injection residues (CKI) and part one and part two of Qizhitongluo Capsule residues (QC1 and QC2, respectively), were used for the cultivation of Pleurotus ostreatus. The effect of the supplementation of corncobs (CC) with different herb residues on yield, nutritional composition, and antioxidant activity of P. ostreatus was investigated. Compared to the control, the higher mycelial growth rate was observed on substrates CC +30% CKI and CC +30% QC1, while the higher yield was obtained from substrates CC +30% QC2 and CC +30% CKI. Moreover, chemical analysis of fruit bodies revealed that the addition of herb residues to CC significantly increased proteins, amino acids, ashes, minerals (Na and Ca), and total phenolic contents but significantly reduced carbohydrates and $IC_{50}$ values of DPPH radicals. In addition, no heavy metals (Pb, Cd, and As) were detected in the fruiting bodies harvested from different substrate combinations. These results demonstrated that mixtures of CC with herb residues might be utilized as a novel, practical, and easily available substrate for the cultivation of P. ostreatus, which is beneficial for the effective management of herb residues.

Changes in the Activities of Enzymes Involved in the Degradation of Butylbenzyl Phthalate by Pleurotus ostreatus

  • Degradation of butylbenzyl phthalate (BBP) by the white rot fungus Pleurotus ostreatus and the activities of some degrading enzymes were examined in two different media containing 100 mg/l of the compound. P. ostreatus pre-grown for 7 days in complex YMG medium was able to completely degrade BBP within an additional 24 h but degraded only 35 mg/l of BBP in 5 days of incubation in minimal medium. Fungal cell mass in the culture in YMG medium was higher in the presence than in the absence of BBP. The esterase activity of the fungal culture in YMG medium was higher than that in minimal medium and increased with the addition of BBP. On the contrary, laccase activity was higher in minimal medium and it did not increase upon the addition of BBP. General peroxidase activity increased for a few days after the addition of BBP to both media. The degradation of BBP and its metabolites by P. ostreatus thus may be attributed mostly to esterase rather than lignin-degrading laccase. In addition, the activities of the enzymes involved in BBP degradation and their changes varied significantly in the different media and culture conditions.

Biodegradation of Endocrine-Disrupting Phthalates by Pleurotus ostreatus

  • Biodegradation of endocrine-disrupting phthalates [diethyl phthalate (DEP), dimethyl phthalate (DMP), butylbenzyl phthalate (BBP)] was investigated with 10 white rot fungi isolated in Korea. When the fungal mycelia were added together with 100 mg/l of phthalate into yeast extract-malt extract-glucose (YMG) medium, Pleurotus ostreatus, Irpex lacteus, Polyporus brumalis, Merulius tremellosus, Trametes versicolor, and T. versicolor MrP1 and MrP13 (transformant of the Mn-repressed peroxidase gene of T. versicolor) could remove almost all of the 3 kinds of phthalates within 12 days of incubation. When the phthalates were added to 5-day pregrown fungal cultures, most fungi except I. lacteus showed the increased removal of the phthalates compared with those of the non-pregrown cultures. In both culture conditions, p. ostreatus showed the highest degradation rates for the 3 phthalates tested. BBP was degraded with the highest rates among the 3 phthalates by all fungal strains. Only 14.9% of 100 mg/I BBP was degraded by the supernatant of P. ostreatus culture in YMG medium in 4 days of incubation, but the washed or homogenized mycelium of P. ostreatus could remove 100% of BBP within 2 days even in distilled water, indicating that the initial BBP biodegradation by P. ostreatus may be attributed to mycelium-associated enzymes rather than extracellular enzymes. The biodegradation rate of BBP by the immobilized cells of P. ostreatus was almost same as that in the suspended culture. The estrogenic activity of 100 mg/I DMP decreased during biodegradation by P. ostreatus.

Evaluation of the Synergistic Effect of Mixed Cultures of White-Rot Fungus Pleurotus ostreatus and Biosurfactant-Producing Bacteria on DDT Biodegradation

  • DDT (1,1,1-trichloro-2,2-bis(4-chlorophenyl) ethane) is one of the organic synthetic pesticides that has many negative effects for human health and the environment. The purpose of this study was to investigate the synergistic effect of mixed cutures of white-rot fungus, Pleurotus ostreatus, and biosurfactant-producing bacteria, Pseudomonas aeruginosa and Bacillus subtilis, on DDT biodegradation. Bacteria were added into the P. ostreatus culture (mycelial wet weight on average by 8.53 g) in concentrations of 1, 3, 5, and 10 ml ($1ml{\approx}1.25{\times}10^9$ bacteria cells/ml culture). DDT was degraded to approximately 19% by P. ostreatus during the 7-day incubation period. The principal result of this study was that the addition of 3 ml of P. aeruginosa into P. ostreatus culture gave the highest DDT degradation rate (approximately 86%) during the 7-day incubation period. This mixed culture combination of the fungus and bacteria also gave the best ratio of optimization of 1.91. DDD (1,1-dichloro-2,2-bis(4-chlorophenyl) ethane), DDE (1,1-dichloro-2,2-bis(4-chlorophenyl) ethylene), and DDMU (1-chloro-2,2-bis(4-chlorophenyl) ethylene) were detected as metabolic products from the DDT degradation by P. ostreatus and P. aeruginosa. The results of this study indicate that P. aeruginosa has a synergistic relationship with P. ostreatus and can be used to optimize the degradation of DDT by P. ostreatus.

Viral Effects of a dsRNA Mycovirus (PoV-ASI2792) on the Vegetative Growth of the Edible Mushroom Pleurotus ostreatus

  • A double-stranded RNA (dsRNA) mycovirus was detected in malformed fruiting bodies of Pleurotus ostreatus strain ASI2792, one of bottle cultivated commercial strains of the edible oyster mushroom. The partial RNA-dependent RNA polymerase (RdRp) gene of the P. ostreatus ASI2792 mycovirus (PoV-ASI2792) was cloned, and a cDNA sequences alignment revealed that the sequence was identical to the RdRp gene of a known PoSV found in the P. ostreatus strain. To investigate the symptoms of PoV-ASI2792 infection by comparing the isogenic virus-free P. ostreatus strains with a virus-infected strain, isogenic virus-cured P. ostreatus strains were obtained by the mycelial fragmentation method for virus curing. The absence of virus was verified with gel electrophoresis after dsRNA-specific virus purification and Northern blot analysis using a partial RdRp cDNA of PoV-ASI2792. The growth rate and mycelial dry weight of virus-infected P. ostreatus strain with PoV-ASI2792 mycovirus were compared to those of three virus-free isogenic strains on 10 different media. The virus-cured strains showed distinctly higher mycelial growth rates and dry weights on all kinds of experimental culture media, with at least a 2.2-fold higher mycelial growth rate on mushroom complete media (MCM) and Hamada media, and a 2.7-fold higher mycelial dry weight on MCM and yeast-malt-glucose agar media than those of the virus-infected strain. These results suggest that the infection of PoV mycovirus has a deleterious effect on the vegetative growth of P. ostreatus.

  • 느타리(Pleurotus ostreatus)의 이핵-단핵 계통간(di-mono) 교잡주의 DNA 유전에 관한 특성을 구명하기 위하여 느타리 6계통 및 사철느타리 1계통으로 이핵-단핵 계통간 12조합 48교잡주를 얻어 교잡율, 교잡주의 핵 DNA 패턴 양상과 유연관계도, 자실체의 형태, 갓 색깔을 분석하였다. 이핵-단핵 계통간 교잡에서 느타리와 느타리간, 느타리와 사철느타리간 교잡은 모두 교잡율 100%로 나타났다. 이핵-단핵 계통간 교잡주는 공여체(donor) 이핵체의 핵이수용체(recipient) 단핵체로 전이되었다. 이핵-단핵 계통간 교잡주의 DNA 패턴은 이핵체와 유사하거나 동일한 것이 87.5%, 양친의 중간 패턴이 12.5%였다. 즉, 느타리 이핵주와 느타리 단핵 계통간 교잡주는 이핵체와 유사한 DNA 패턴이 70.9%, 양친의 핵이 공존하는 중간 패턴이 12.5%였으며, 사철느타리와 느타리간 이핵-단핵 계통간 교잡주는 16.6%로 모두 사철느타리 핵 DNA 패턴과 유사하거나 동일하였다. 교잡주의 핵 DNA 패턴은 교잡조합에 따라 차이가 나타났는데 12교잡조합 중에서 4조합에서만 단핵주와 유사하거나 중간 형태를 나타내었고 나머지는 이핵주와 동일한 양상이었다. 교잡주의 자실체 형태는 이핵주 형태가 79.2%, 양친의 중간형태 또는 단핵체 모군주의 형태가 20.8%였다. 하지만 이핵체 형태라 하더라도 자실체 색깔은 다소 달랐다. 사철느타리 이핵-느타리 단핵주간 교잡주의 자실체 갓 색깔은 모두 이핵체 사철느타리와 유사하거나 동일하였다. 느타리 이핵-사철느타리 단핵 계통간 교잡주는 양친의 중간 갓 색깔로 모두 나타났으며 다소 이핵체와 유연관계가 가까운 색깔이었다. 따라서 사철느타리가 다소 우성으로 나타나는 경향이었다. 이러한 결과로 보아 교잡주는 3종류의 핵이 모두 공존하는 세포가 많을 것으로 생각되며, 이핵-단핵 계통간 교잡 방법은 우수한 계통을 육성하는 훌륭한 방법으로 이용될 수 있을 것으로 기대된다.

식용(食用)버섯의 원형질체(原形質體) 분리(分離)에 관한 연구(硏究) (Studies on Protoplast Isolation from Edible Fungi)

  • 고등균류의 유전연구와 균주개발을 위한 원형질체 조작의 기초 연구로서 몇가지 주요 버섯류의 원형질체 분리에 관하여 실험한 결과를 요약하면 다음과 같다. 1. P. ostreatus와 V. volvacea의 원형질체 분리에 알맞는 효소는 ${\beta}-D-Glucanase+Novozym234+Snail enzyme$이었으며, 삼투압 조절제로는 0.6M $MgSO_{4}$였고, 이들을 Na-Maleate buffer로 pH 5.8에 조절하여 사용했을때 가장 많은 원형질체를 생성하였다. 2. P. ostreatus, P. florida, P. sajor-caju는 MCM에서 각각 4, 5, 3일간 배양한 균사체에서, L. edodes, F. velutipes는 PDA 배지에서 각각 2일과 5일간 배양했을때 가장 효과적이었으며, A. bisporus와 V. volvacea는 분리량이 극히 적었다. 3. V. volvacea의 균사체에 2-mercaptoethanol의 전처리는 효과가 없어 원형질체 분리는 적었으며, 액체배양 보다는 agar 배지상의 cellophan방법이 원형질체 분리에 있어서 효과가 좋았다. 4. 균사체에 효소처리후 1시간 부터 원형질체가 분리되면서 점차 증가하여 9시간경에 가장 많은 원형질체를 얻었으며, 시간이 갈수록 액포화현상으로 원형질체의 크기가 커졌다.

자외선 조사에 의한 버섯의 ergocalciferol(비타민 D) 함량의 증진 (Enhancement of ergocalciferol (vitamin D) content in mushrooms by UV irradiation)

  • Ergocalciferol은 vitamin D의 활성을 가지는 것으로 알려져 있다. 본 연구에서는 팽이(Flammulina velutipes), 느타리 (Pleurotus ostreatus), 애느타리(Pleurotus ostreatus), 새송이(Pleurotus eryngii), 양송이(Agaricus bisporus), 표고(Lentinula edodes), 목이(Auricularia auricula-judae) 등 7종의 버섯을 대상으로 자외선 조사에 따른 ergocalciferol(EC, 비타민 $D_2$) 함량의 증진 효과를 조사하였다. 자외선을 조사하지 않은 버섯의 ergocalciferol 함량은 극히 낮았으나 자외선 조사에 의해 ergocalciferol 함량이 크게 증가하였으며, 자외선 A, B, C 중 특히 자외선 B가 EC 함량 증진에 효과적이었다. 버섯에서 ergocalciferol의 생성은 자외선에 직접 노출되는 부위 즉 깊이 1 mm 이내의 버섯 표면에서만 일어나는 것으로 조사되었다. 따라서 세절하지 않은 생 버섯에 자외선을 조사할 경우 팽이 버섯과 같이 체적에 비해 표면적인 넓은 종류의 버섯에서 ergocalciferol 생성이 많았고, 건조 및 세절할 경우에는 EC 생성의 기질인 ergosterol의 함량이 높은 버섯에서 ergocalciferol 생성이 많았는데, 새송이, 표고, 양송이 등이 이에 속하였다.

느타리버섯 중의 Light-Induced Mitochondrial ATPase에 관한 연구 -유기물 효과- (Studies on Light-Induced Mitochondrial ATPase in Pleurotus ostreatus -Effects of Organic Compounds-)

  • Mitochondria in Pleurotus ostreatus were isolated and purified by stepped sucrose density gradient centrifugation, to compare the effects of organic compound on the activities of mitochondrial ATPase in Basidiomycotina with those in mammalian cell. The effects of N, N'-dicycio-hexylcarbodiimide (DCCD), carbonyl cyanide m-chlorophenylhydrazone (CCCP), sodium azide and aurovertin known as compounds to be related to electron transfer system in mitochondria were studied. A activity of mitochondrial ATPase was inhibited by 64%, 57% and 53% in the presence of 0.25 mM DCCD, 0.02 mM sodium azide and 1.5 $({\mu}g/mg\;of\;protein)$ aurovertin B, respectively. It was stimulated by 22% in the presence of 0.15 ${\mu}M$ CCCP.
