Studies on the Biological Activity of Pleurotus ferulea (아위버섯(Pleurotus ferules) 추출물의 생리활성 탐색)

  • This study was peformed to screen the biological activities of Pleurotus ferulea (K5 and K8 strains). The cap of K5 strain is well developed than stalk, and vice versa in K8. The ethanol extract of Pleurotus ferulea exhibited significant free radical scavenging activity (35∼36%), suggesting possible effect on many degenerative diseases originated from the reaction of oxygen species. Acetylcholinesterase inhibitor is proven to be the most effective factor for Alzheimer disease induction, and ethanol extract of Pleurotus ferulea significantly inhibited acetylcholinesterase activity (25∼35%) in uitro. Moreover, ethanol extract of Pleurotus ferulea suppressed liver fibrosis by 3∼12% in uitro. However, Pleurotus ferulea feruled to inhibit glucose uptake in human intestinal cell line. Viability of gastric and colon cancer cells was also not affected by Pleurotus ferulea extract. In conclusion Pleurotus ferulea exhibited significant effect on free radical scavenging, acetylcholinesterase inhibiton and brain cell protection. However, Pleurotus ferulea failed to affect glucose uptake, and cytotoxicity of gastric and cancer cells. In general, K8 revealed more significant effects than K5.

Analysis of Nutritional Components in Pleurotus ferulea (아위버섯(Pleurotus ferulea) 영양성분 분석)

  • Nutritional components, such as approximate compositions, and amino acid, mineral, vitamin, sugar, and fatty acid contents, of artificially cultivated Pleurotus ferulea were analyzed. Contents of carbohydrates, crude lipids, dietary fibers, crude proteins, total amino acids, particularly essential amino acids, minerals, water-insoluble and-soluble vitamins, glucose, and unsaturated fatty acids such as linoleic acid of P. ferulea were higher than those of P. ostreatus and P. eryngii. Results indicate P. ferulea has abundant essential nutrients and thus is good source of functional healthy food.

Fibrinolytic, thrombin inhibitory, anti-oxidative and anti-inflammatory activities of Pleurotus ferulea (아위버섯 추출물의 혈전용해, 트롬빈저해, 항산화 및 항염증 활성)

  • Our study investigated the fibrinolytic, thrombin inhibitory, anti-oxidative and anti-inflammatory activities of the water extract and solvent fractions isolated from Pleurotus ferulea. Fibrinolytic activity was investigated using the fibrin plate method. Thrombin inhibitory activity was used to analyze thrombin inhibitor assay. The DPPH assay was used to estimate anti-oxidative activity. Inhibition of NO production was measured for anti-inflammatory activity in LPS-activated murine RAW 264.7 macrophage cells. An MTS assay was used to evaluate the effects of the water extract and solvent fractions isolated from Pleurotus ferulea on cell viability. Our results showed the fibrinolytic activity to be strong in the ethyl acetate fraction at 1.33 plasmin units. The ethyl acetate fraction also showed high thrombin inhibitory activity at 94.45%. The anti-oxidative activity of the water extract was 37.01% and the anti-inflammatory activity of the chloroform fraction was 98.13%. These findings suggest that Pleurotus ferulea's extract and fractions could be applicable in the development of functional foods for the treatment and prevention of cardiovascular diseases.

Physiological Activities of Hot Water Extract and Solvent Fractions of Pleurotus ferulea (아위버섯 열수추출물 및 유기용매 분획물의 생리활성 효과)

  • To investigate the physiological activities of hot water extract and solvent fractions isolated from Pleurotus ferulea, antioxidative, fibrinolytic, thrombin inhibitory, and ${\alpha}$-glucosidase inhibitory activities were examined. Pleurotus ferulea, hot water extract was fractionated into hexane, chloroform, ethyl acetate, butanol and water fraction. Each of these was assayed individually. The antioxidative activities of ethyl acetate and butanol fractions were 86.79% and 87.82%, respectively. Using the fibrin plate method, only the ethyl acetate fraction showed a plasmin activity of 0.08 units/ml. Thrombin inhibitory activities of chloroform and ethyl acetate fractions were 74.90% and 71.08%, respectively. In the ${\alpha}$-glucosidase inhibitory activity test, butanol fraction showed the highest activity at 49.67%. From the above results, we anticipate that solvent fractions of Pleurotus ferulea can be used as a materials for the development of biofunctional foods for cardiovascular diseases.

Screening of Fibrinolytic Activities from Cultured Mushrooms (인공재배 버섯의 혈전분해효소 활성검색)

  • Fibrinolytic activities of cultured medium and mycelium from 13 selected culturable mushrooms ranged 0.00-0.16 U and 0.00-0.48 U, respectively. Dry cell weight of Armillaria mellea was 6.50g/100mL, 1.9-5.0 fold higher than other tested mushrooms. Among artificially cultured mushroom fruiting bodies tested, fibrinolytic activity was lowest in Hericium erinaceus (0.17 U) and highest in Pleurotus ferulea (0.94 U).
