Age and Growth of Brown Sole, Pleuronectes herzensteini (Jordan et Snyder) in the East Sea of Korea (한국 동해안 참가자미, Pleuronectes herzensteini (Jordan et Snyder)의 연령과 성장)

  • The age and growth of brown sole, Pleuronectes herzensteini were investigated from samples randomly collected in the East Sea of Korea from April, 2003 to March, 2004. Ages were determined from annuli in otoliths and annuli were formed between March and May once a year. Also, main spawning period were estimated between March and April, thus rings were considered as annual marks. The von Bertalanffy growth parameters estimated from a non-linear regression method were $L_{\infty}=37.2cm$, K=0.131/year, $t_0=-2.008years$ for female and $L_{\infty}=28.3cm$, K=0.177/year, $t_0=-2.135years$ for male, and the growth between female and male was different.

  • The maturity and spawning of brown sole, Pleuronectes herzensteini were investigated using samples randomly collected in the East Sea of Korea from April, 2003 to March, 2004. We analyzed monthly changes in maturity stages, gonadosomatic index (GSI), egg diameter, fecundity (F), total length (TL) at 50% group maturity, and sex ratio. The spawning period was February to May, and the main spawning period was March to April. The fecundity was 38,702 eggs at 22 cm (TL) to 133,085 eggs at 37 cm (TL), and the relationship between TL and F was $F=11.307TL^{2.628}$. The TL at first maturity was 15 cm and at 100% maturity, 21 cm. Thus, TL at 50% group maturity was estimated to be 17.8 cm, which corrensponded to the age of 3 years. The sex ratio was 60.2 for female and 39.8 for male, where the female population was seen to be slightly predominant.

Community Structure and Distribution Pattern of the Pleuronectiform Fishes in the Uljin Marine Ranching Area, Korea (울진바다목장에서 어획된 가자미목(Pleuronectiformes) 어류의 군집구조 및 공간분포)

  • To elucidate the variation of species composition, biomass and length distribution of flatfishes by the period and region in the Uljin marine ranching area, bottom trawl survey was investigated at the six stations from Feb. 2008 to Dec. 2010. During the survey period, a total 4 families 17 species in the Pleuronectiformes, average 69,158 $ind./km^2$ and 5,625 $kg/km^2$ were identified, in 2008 appeared in 14 species, average 25,798 $ind./km^2$ and 2,333 $kg/km^2$ and in 2009 appeared in 13 species, average 102,360 $ind./km^2$ and 5,634 $kg/km^2$ and in 2010 appeared in 14 species, average 75,704 $ind./km^2$ and 8,632 $kg/km^2$. The individual dominant species, occupying over 10% of total individuals, was Pleuronectes herzensteini (20,811 ind., 30.0%), Hippoglossoides pinetorum (18,666 ind., 26.9%) and Glyptocephalus stelleri (13,499 ind., 19.4%) also the biomass dominant species, occuping over 10% of total biomass, was P. herzensteini (2,207 kg, 39.3%), Pleuronectes yokohamae (857 kg, 15.3%), H. pinetorum (761 kg, 13.5%), Kareius bicoloratus (677 kg, 12.1%). From the cluster and MDS analysis based on Bray-Curtis similarity matrix of fourth root transformed data of species number and individuals in the Uljin marine ranching area from Feb. 2008 to Dec. 2010 was divided into two different groups of the flatfishes community in 2008 and from Jan. to Apr. in 2009 and 2010 (Group A) and the pleuronectiform community in from May to Dec. in 2009 and 2010 (Group B). From the cluster and MDS analysis using the similarity of demersal organisms community among six stations, the Uljin marine ranching area was divided into two different groups of Group 1 (St. 1 and St. 3) and Group 2 (St. 2, St. 4, St. 5 and St. 6).

Inhibitory Effects of Chitosanascorbate on Growth of Staphylococcus aureus and Escherichia coli Contaminated in Flounder during Storage (참가자미의 저장 중 오염미생물 Staphylococcus aureus와 Escherichia coli의 생육에 미치는 Chitosanascorbate의 영향)

  • We performed an experiment of keeping the extension of raw and semi-dried flounder (Pleuronectes herzensteini). The effect of with (WG) or without gill (OG), drying degree (20% drying: 20D, 40% drying: 40D) and storage temperature($5^{\circ}C$) and 0.1% chitosan-ascorbate (CA) treatment of vacuum packaging flounder on the growth of contaminated microorganism during storage for 10 days were investigated. Total aerobacter (TA) in the OG-treated raw flounder was $0.29{\sim}0.44$ log cycle lower than that of WG-treated flounder. Also, the number of Staphylococcus aureus (SA) and E. coli (EO) in OG were lower compared with WG. The number of TA, SA and EO in 20 D among 0 D, 20 D and 40 D stored at $5^{\circ}C$ were lowest. Especially, the SA and EO was $0.13{\sim}0.53$, 0.3-0.88, and 0.13-0.74 log cycle lower compared with raw flounder. The growth of TA, SA and EO separated from raw flounder in tryptic soy broth were completely inhibited by 0.1% CA. The anti-biotical effect of CA of two microorganisms SA and EO that separated from flounder, and the growth of all of them were 90% (SA), 96% (EO) inhibited at the 0.1% CA. The inhibition times at $37^{\circ}C$ in soy broth was 36 hr. However when CA was added directly to flounder, it appeared inhibition effect to 0.88 log cycle. The effect of CA was better when gills removed and 20% drying.

Comparison of Biological Activites in Sikhae, Traditional Fermented Sea Products (전통 수산발효식품인 식해류의 생리적 기능성 효과 비교)

  • This study investigated and compared chemical properties and biological activities of four commercial sikhae products (three flat-fish, Pleuronectes herzensteini, Sikhae: product A, B, C; one Alaska pollack, Theragra chalcogramma, Sikhae: product D) and Alaska pollack Sikhae (product E). Total acidity, amino nitrogen, salinity and pH in all products were in ranges of $0.47-1.93\;g\%$, $145.94-204.81\;mg\%$, $3.40-4.00\%$ and 4.38-5.55, respectively. All products showed antimicrobial activities against Bacillus ceraus subtilis, B. cereus, Staphylococcus aureus, Listeria monocytogenes, Vibrio parahaemolyticus, except for Salmonella typhimurium. Particularly the activities were more effective with gram positive bacteria than gram negative bacteria. Antioxidative $(EDA_{50};\;14.20\;mg/mL)$ and Fibrinolytic (0.95 plasmin unit/mL) activities of product E were much stronger than commercial Sikhae products (EDA50; 18.87-34.60 mg/mL and 0.69-0.85 plasmin unit/mL).

Genetic structure of wild brown sole inferred from mitochondrial DNA analysis

  • The population structure of brown sole was examined in a total of 308 samples collected from three geographical groups: one locality (Gangneung) on the east side of the Korean Peninsula, two localities (Erimo and Tomakomai) on the southeastern side and four localities (Onishika, Teshio, Tomamae and Yoichi) on the northwestern side of Hokkaido Island, Japan, by using sequences of 484 bp from the 5' end of the control region of mtDNA. We detected 225 haplotypes, but 183 of them were unique to an individual. A total of 116 nucleotide sites were variable. Haplotype diversity (h) was very high, ranging from 0.989 to 1.000, and nucleotide diversity (${\pi}$) was detected between 0.015 and 0.022. Genetic distances (${\Phi}_{ST}$) within populations, among populations and among geographical groups were low (0.0002 to 0.0014). No significant difference was detected by the AMOVA test (P < 0.05). Pairwise $F_{ST}$ values between sampling localities were also low and not significant. Genetic differentiation was not detected among sampling localities.

Seasonal Variations in the Species Composition of Fisheries Resources Caught by Trammel Net in the Uljin Marine Ranching Area, East Sea (울진바다목장에서 자망으로 어획된 수산자원의 종조성과 계절변동)

  • Variations in the species composition, biomass and size distribution of fisheries resources in the Uljin marine ranching area were investigated using trammel nets at two stations (artificial reef and natural rocky area) from 2009 to 2010. During the survey, a total of 74 species were sampled with a mean density of 132 ind./net and mean biomass of 21.56 kg/net. In the natural rocky area, a total of 45 species were sampled at a mean density of 202 ind./net and mean biomass of 28.81 kg/net, while in the artificial reef area, samples included a total of 56 species, with means of 62 ind./net and 14.30 kg/net. The dominant species, comprising over 3% of the total number of individuals, were Suberites ficus (30.8%), Ovalipes punctatus (19.2%), Paralichthys olivaceus (11.7%), Pleuronectes herzensteini (4.7%), Kareius bicoloratus (3.5%), Pseudopleuronectes yokohamae (3.5%) and Eopsetta grigorjewi (3.0%). The dominant species, in terms of biomass, comprising over 5% of the total biomass, were P. olivaceus (22.1%), S. ficus (18.7%), O. punctatus (7.2%), Hexagrammos otakii (6.6%), P. yokohamae (5.7%), K. bicoloratus and P. herzensteini (5.3%). A cluster analysis and non-metric multidimensional scaling (nMDS) analysis based on the Bray-Curtis similarity of fourth root transformed data for number of species and individuals, was divided into two groups: the artificial reef area (group A) and the natural rocky area (group B).

Contamination and Risk Assessment of Lead and Cadmium in Commonly Consumed Fishes as Affected by Habitat (서식지에 따른 다소비 어류의 납과 카드뮴의 오염 및 위해 평가)

  • This study determined the concentrations of lead and cadmium in 18 species of commonly consumed fish and assessed the risk based on provisional tolerable weekly (monthly) intakes [PTW(M)I] % as affected by behavioral characteristics, such as migration and settlement. In the 18 species, the mean concentrations of lead and cadmium were higher in the 11 species of migratory fish (llargehead hairtail Trichiurus lepturus, chub mackerel Scomber japonicus, Pacific saury Cololabis saira, skipjack tuna Katsuwonus pelamis, Pacific cod Gadus macrocephalus, anchovy Engraulis japonicus, Alaska pollack Theragra chalcogramm, brown croaker Miichthys miiuy, Japanese Spanish mackerel Scomberomorus niphonius, yellow croaker Larimichthys polyactis, and Pacific herring Clupea pallasii) than in the seven demersal species (red stingray Dasyatis akajei, brown sole Pleuronectes herzensteini, bastard halibut Paralichthys olivaceus, conger eel Conger myriaster, blackmouth angler Lophiomus setigerus, rockfish Sebastes schlegelii, and filefish Stephanolepis cirrhifer). Based on the mean concentrations, the PTWI % of lead and cadmium in commonly consumed migratory fish were 1.900 and 2.986%, respectively, which were higher than the values for lead and cadmium in the commonly consumed demersal fishes (0.257 and 0.318%, respectively). The estimation of weekly (monthly) intakes and target hazard quotients for the toxic elements lead and cadmium revealed that the commonly consumed migratory and demersal fish do not pose any health risks for consumers.

Effect of Gill, Drying and Temperature and Chitosan-Ascorbate Treatment on Growth of Contaminated Microorganism in Flounder during Storage (참가자미의 저장 중 오염미생물의 생육에 미치는 아가미의 유무, 건조도, 저장온도 및 키토산-아스코베이트의 처리효과)

Total aerobacter (TA) in the OG-treated raw flounder was $0.3{\sim}0.5$ log cycle lower than that of WG-treated flounder and also, number of coliform (CF) and E. coli (CO) in OG were lower compared with WG. Number of TA,. Especially, the TA was $0.42{\sim}1.20$ log cycle lower compared with raw flounder. The TA of the raw flounder stored at $5^{\circ}C$ compared to $10^{\circ}C$ was $0.6{\sim}1.3$ log cycle lower. The growth of total aerobacter, coliform (and E. coli separated from raw flounder in tryptic soy broth were completely inhibited by 0.1% CA. But the growth of TA in the raw and 20% dried flounder was 1.5 log cycle inhibited by 0.1% CA, and the growth of CF and CO were also slightly inhibited. We did value raw sample that treated CA 0D and 20D, and did vaccum packaging at the $10^{\circ}C$ for 10 days, Sensory quality such as appearance, freshness, and texture and overall acceptability after cooking of the 0.1% CA-treated raw and 20% dried flounder were evaluated from good to very good, while CA non-treated products were evaluated to moderate.
