Assay of Antimutagenic Activities of Vegetable Plants (식용식물(食用植物)의 항변이원성(抗變異原性)에 관한 연구(硏究))

  • Potential antimutagenic activities of vegetable plants were investigated. 24 vegetables which are frequently consumed by Korean people were extracted with 70% ethyl alcohol to prepare the extract samples. Then those samples were added to the culture media containing mitomycin C($O.3\;{\mu}g/ml$) and the SOS-Chromotest was performed. Positive control, mitomycin C alone, showed about 330 units of ${\beta}$-galactosidase activities. Among 24 vegetable samples, Saxifraga oblougifolia Nakai (취나물, Saxifragaceae) and Platycodon grandiflorum A. De Candolle(도라지, Campanulaceae) showed 136 and 155 units, respectively when mitomycin C was treated. These results indicate that Saxifragae Herba and Platycodi Radix possess protecting action from mutagenic activity produced by mitomycin C.

  • The protective effects of a Platycodi Radix (Changkil: CK), the root of Platycodon grandiflorum A. DC (Campanulaceae). on carbon tetrachloride-induced hepatotoxicity and the possible mechanisms involved in this protection were investigated in mice. Pretreatment with CK prior to the administration of carbon tetrachloride significantly prevented the increased serum enzymatic activities of alanine and aspartate aminotransferase in a dose-dependent manner. Studies on Pest Insects of Crude Drugs (생약 및 한약재의 저장 중 발생하는 곤충류)

  • In order to estimate accurate quality of crude drugs in which pest was found during the storage, damaged 15 items were collected on current markets. We identified insect name by investigation of insect morphology, bug cluster, and morphology of damaged crude drugs. Three kind of pests were identified in the above 15 items as follows: A: Pyralis sp. (Pyralidae) was observed in damaged Platycodi Radix. B: Anthreus verbasci (Anobiidae) was found out in damaged Angelicae gigantis Radix, and Dioscoreae Rhizoma. C: Stegobium pamceum (Anobiidae) was discovered in the 12 crude drugs as follows: Puerariae Radix, Angelicae koreanae Radix, Angehcae tenuissimae Radix, Codonopsis Radix, Ledebouriellae Radix, Ginseng Radix alba, Angelicae dahuricae Radix, Belamcandae Rhizoma, Paeoniae Radix, Cnidii Rhizoma, Alismatis Rhizoma, and Fritillariae Bulbus.

  • A rapid and practical HPLC assay was developed for quantitative analysis of saponins in Platycodi Radix. Seven saponin components in Platycodi Radix, i.e., deapioplatycoside E, platycoside E, deapioplatycodin D$_3$, platycodin D$_3$, polygalacin D$_2$, platycodin D$_2$ and platycodin D were successfully resolved on C18 column and detected by ELSD. Standard curves were linear over the concentration range 1 ${\sim}$2,000 ${\mu}$g/ml (r$^2$>0.992). Intra- and inter-day coefficients for variation of seven saponin components were < 10% and limit of quantification of them were around 0.7${\sim}$1.5 ${\mu}$g/ml, respectively. Using this method, contents of seven saponins in various plant materials under different cultivating conditions were estimated.

  • Objectives: The aim of this work is the making of morphological standard in platycodi radix. Methods: The external-internal morphological standards were determined by using stereoscope and butanol series. Conclusions: The external and internal characteristics according to the shape of original plant or herbal states. entirely correspond to all the literature on this subject. According to the place of production. the difference appears. (1) In the external shapes. as the Korean products. the distinction among the first. medium. poor class products was caused by the state of processing of desiccation. But as the foreign products, it was caused by the difference of size, color and especially color of circle. (2) In the internal shapes. the Mt. Baekdu region products is not skinned and has the deep state of suberization of cork phellogen. so it makes certain distinctional point. And the Korean products has the much latex tube group relatively and the bigger crevice than the foreign one.

A Saponin Complex, KPRG-C, and Its Sapogenin Complex, KPRG-D, Reduce Nociception and Inflammation in Animals

  • To develop a clinically available saponin- or sapogenin complex from Oriental medicines, the EtOH extract (KPRG-A) was obtained by extracting from the four crude drugs, Kalopanacis Cortex, Platycodi Radix, Rubi Fructus and Glycyrrhizae Radis. The BuOH fraction (KPRG-B), a crude saponin complex, was prepared by fractionating KPRG-A, which were further completely hydrolyzed to afford the sapogenin complex (KPRG-D). In an attempt to find the antinoicpetive effects of the saponin complex and sapogenin complex, KPRG-C, and -D, were assayed by writhing-, hot plate-, and tail-flick tests using mice or rats. The three samples were also subjected to antiiflammatory tests using serotonin-induced and carrageenan-induced hind paw edema mice and rats, respectively. The three samples significantly reduced inflammations and pains of the experimental animal. The potency were found in the order of KPRG-D> KPRG-C> KPRG-B. The most active sample, KPRG-D, caused no death, no body increase or no anatomical pathlogic change even at 2,000 mg/kg dose. These results suggest that a sapogenin complex, KPRG-D, which was found to contain mainly hederagenin, platycodigenin, polygalacic acid, 23-hydroxytormentic acid, glycyrrhetic acid together with minor triterpene acids, could be a potential candidate for antiinflammatory therapeutics.

Analysis of Fungicide Prochloraz in Platycodi Radix by GC-ECD (GC-ECD를 이용한 한약재 길경(Platycodi Radix) 중 살균제 Prochloraz의 분석)

  • BACKGROUND: Prochloraz has been widely used as an imidazole fungicide on fruits and vegetables in Korea. Analytical approaches to evaluate prochloraz residues in herbal medicine are required for their safety management. In this study, we developed a GC-ECD method for quantitative determination of prochloraz in Platycodi Radix. The metabolite 2,4,6-trichlorophenol (2,4,6-T) was used as a target compound to evaluate total prochloraz residues as it is categorized to a representative residue definition of prochloraz. All residues containing 2,4,6-T were converted to 2,4,6-T and subjected to GC-ECD. METHODS AND RESULTS: In order to verify the applicability, the method was optimized for determining prochloraz and it metabolite 2,4,6-T in Platycodi Radix. Prochloraz and its metabolite 2,4,6-T residuals were extracted using acetone. The extract was diluted with and partitioned directly into dichloromethane to remove polar co-extractives in the aqueous phase. The extract was decomposed to 2,4,6-T, and then the partitioned ion-associate was finally purified by optimized aminopropyl solid-phase extraction (SPE). The limits of quantitation of the method (MLOQs) were 0.04 mg/kg and 0.02 mg/kg, respectively for prochloraz and 2,4,6-T, considering the maximum residue level (MRL) of prochloraz as 0.05 mg/kg in Platycodi Radix. Recovery tests were carried out at two levels of concentration (MLOQ, 10 MLOQ) and resulted in good recoveries (82.1-89.7%). Good reproducibilities were obtained (coefficient of variation < 2.8%), and the linearities of calibration curves were reasonable (r2 > 0.9986) in the range of 0.005-0.5 ㎍/mL. CONCLUSION(S): The method developed in this study was successfully validated to meet the guidelines required for quantitative determination of pesticides in herbal medicine. Thus, the method could be useful to monitor prochloraz institutionally in herbal medicine.

  • Objectives : The aim of this study was to investigate the effect of Asthma-depression and Immunoregulation with PR-HAS(Herbal-acupuncture with Platycodi Radix infusion solution) injection at Joksamni(St36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week). The experimental group(OVA-PR-HA) wase treated with concentrations(1%) of PR-HAS at Joksamni(St36) for the later 8 weeks(3times/week). The second experimental group(OVA-Needle prick) was treated with Needle-Prick at Joksamni(St36) for the later 8 weeks(3times/week). Results : 1. The weight and total cells in the mice lung treated with PR-HA decreased significantly compared with that of control group. 2. Total leukocytes and eosinophils in BALF of the mice group treated with PR-HA decreased remarkably compared with those of control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with PR-HA decreased significantly compared with those of control group. 4. The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. 5. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. 6. The numbers of $CCR3^+\;cells,\;CD4^+\;cells\;and\;CD8^+\;cells$ in the lungs, and $CD3e^+/CD69^+$ in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. 7. The mRNA expression of ${\beta}-actin,\;TNF-{\alpha}$, IL-4, IL-5,IL-13 in the mice group treated with PR-HA with RT-PCR decreased significantly compared with those of control group. Conclusion : The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. The numbers of $CCR3^+\;cells,\;CD4^+\;cells\;and\;CD8^+\;cells$ in the lungs, and $CD3e^+/CD69^+$ in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. Determination of Sulfur Dioxide in Herb Drugs

  • This study was carried out to investigate sulfur dioxide in herb drugs.(Puerariae Radix, Lycii Fructus, Platycodi Radix, Dioscoreae Rhizoma, Mori Radicis Cortex, Nelumbo Seed, Paeoniae Radix, Remotiflori Radix, Astragali Radix, Polygonatum Rhizome) Experimental subjects included 105 cases of 10 species of herb drugs collected in Kyung-dong market from December in 2001 to February in 2002. Sulfur dioxide was determined by Optimized Monier-Wiliams method. In 65 cases(61.9%) of the 105 cases, we detected $SO_2$ over 10mg/kg. (omitted)
