  • The soil on the behavior of the nonlinear elastic work-hardening plasticity has a variety of stress paths due to the state of soil and the test conditions. The soil with a specific volume ${\upsilon}$ in principal stress space (${\sigma}_1$, ${\sigma}_2$, ${\sigma}_3$, and ${\upsilon}$v) displays the shape of an irregular hexagonal pyramid with an end cap. With variations of ${\upsilon}$ the size of the cap is changed but its shape remains unchanged and the movement of the cap is controlled by the increase or decrease of the plastic volumetric strain. By reflecting such a property of soil various cap models have been developed by researchers. In this thesis, a constitutive model of soil with a combination of the nonlinear elastic work-hardening plastic cap and the failure surfaces of Mohr-Coulomb (M-C cap model) has been developed. According to the the results of analyses using the work-hardening plastic cap model, the normally consolidated soil under shearing has experienced the work-hardening and plastic flow (movement of the cap). But in the shearing of the overconsolidated soil the elastic behavior is shown until the stress path has reached the failure surface and the cap does not move.

  • Consumption of high doses of ethanol can lead to amnesia, which often manifests as a blackout. This incoordination of blackout may be a major cause in various social problems in alcohol consumption. However, there is still no treatment for preventing these alcohol-induced problems. Hydrangeae dulcis folium is a drug or a tea which is made from the fermented and dried leaves of Hydrangea serrata Seringe. The present study, we tested the ethanol extract of the Hydrangeae dulcis folium (EHDF) on ethanol-induced psychological deficits. To test behavioral deficits, an object recognition test was conducted using a mouse model. To evaluate synaptic deficits, N-methyl-D-aspartate (NMDA) receptor-mediated excitatory postsynaptic potential EPSP and long-term potentiation (LTP) in the mouse hippocampal slices were tested, as they are known to be vulnerable to ethanol and are associated with ethanol-induced amnesia. In the tests, ethanol (1 g/kg, i.p.) impaired object recognition memory, but EHDF (10 or 30 mg/kg) prevented this impairment in object recognition test. Interestingly, EHDF ($30{\mu}g/ml$) significantly ameliorated ethanol-induced LTP and NMDA receptor-mediated synaptic transmission in the hippocampal slices. EHDF prevented ethanol-induced object recognition memory deficits induced by ethanol. Interestingly, EHDF significantly ameliorated ethanol-induced LTP and NMDA receptor- mediated synaptic transmission in the hippocampal slices.

  • The plasticity of nervous system is generated not only due to changes in neurons but also due to changes in neuroglial cells. Astrocyte is important for maintaining the normal brain function and controlling the neuronal functions. The amygdala receives an array of important sensory information of danger signals. This information is further transduced and integrated to produce the highly adaptive emotion, fear. In this study, morphometric changes in the cell bodies of astrocytes in the amygdala, induced by prenatal stress and restraint stress were examined. For this purpose. rats were classified into 4 groups; control group (CON), only restraint-stressed (starting on P90 for 3 days) group (CONR), prenatally-stressed group (PNS), and prenatally and restraint (on P90 for 3 days) stressed group (PNSR). Astrocytes were verified with anti-GFAP immunohistochemistry, counter stained with methylene blue/azure II and were examined using the Neurolucida. Results showed that astrocytes in the amygdala of PNS rats had significantly larger cell bodies than did CON rats and this was enhanced further by restraint stress. Thus this data showed that hypertrophy of the astrocytic cell bodies of amygdala complex is induced by prenatal and restraint stress.

  • Brain-derived neurotrophic factor (BDNF) is a neurotrophic factor involved in neuronal differentiation, plasticity, survival and regeneration. BDNF draws massive attention mainly due to the potential as a therapeutic target in neurological diseases such as depression and Alzheimer's disease. In a primary screening for the natural compounds enhancing BDNF release from cultured rat primary cortical neuron, we found that compounds such as baicalein, tanshinone IIa, cinnamic acid, epiberberine, genistein and wogonin among many others increased BDNF release. All the compounds at $0.1{\mu}M$ of concentration barely showed stimulatory effect on BDNF induction, however, their combination (mixture 1; baicalein, tanshinone IIa and cinnamic acid, mixture 2; epiberberine, genistein and wogonin) showed synergistic increase in BDNF release as well as mRNA and protein expression. The level of BDNF expression was comparable to the maximum BDNF stimulation attainable by a positive control oroxylin A ($20{\mu}M$) without cell toxicity as determined by MTT analysis. Both mixtures synergistically increased the phosphorylation of extracellular signal-regulated kinase (ERK) as well as cAMP response element binding protein (CREB), an immediate and essential regulator of BDNF expression. Similar to these results, mixture of these compounds synergistically inhibited the up-regulation of inducible nitric oxide synthase (iNOS) induced by lipopolysaccharide treatments in rat primary astrocytes. These results suggest that the combinatorial treatment of natural compounds in lower concentration might be a useful strategy to obtain sufficient BDNF stimulation in neurological disease condition such as depression, while minimizing potential side effects and toxicity of higher concentration of a single compound.

  • FEM analyses which are based on modified Cam-clay theory have been generally used in such cases as analyses of stability and displacement fur embankment construction on soft clays. However, the Modified Cam Clay Model can generate some problems in anisotropic stress conditions of field because the critical state theory has been developed through many laboratory tests in isotropic conditions. Thus, the applicability on the prediction of undrained shear strength and pore water pressure which was based on the critical state theory was evaluated by triaxial tests and numerical analyses in isotropic and anisotropic conditions. Used samples often come out in domestic area, together with general low plastic clays which are showing dilatant behavior in shearing process. They were evaluated by laboratory tests and FEM based on MCCM. From the results of test and numerical analysis, the predictions of undrained strength in low plastic clays well coincided with each other in both isotropic and anisotropic conditions. However, the generation of porewater pressure was often overestimated during undrained shearing in anisotropic conditions. The results can generate the errors in the prediction of stress path of field sites during loading such as construction of embankment on soft clays because the field is subjected to anisotropic conditions during loading.

  • Mixtures of sand and rubber particles ($D_{sand}/D_{rubber}=1$) are investigated to explore their characteristics under different stain level. Mixtures are prepared with different volumetric sand fractions ($sf=V_{sand}/V_{total}$). Experimental data are gathered from a resonant column, an instrumented oedometer, and a direct shear tests. Results show that sand and rubber differently control the behavior of the whole mixture with strain level. Non-linear degradation of small strain stiffness is observed for the mixtures with $sf{\geq}0.4$, while the mixtures with low sand fraction ($sf{\leq}0.2$) show significantly high elastic threshold strain. Vertical stress-deformation increases dramatically when the rubber particle works as a member of force chain. The strength of the mixtures increases as the content of rubber particle decreases, and contractive behavior is observed in the mixtures with $sf{\leq}0.8$. Rubber particle plays different roles with strain level in the mixture: it increases a coordination number and controls a plasticity of the mixture in small strain; it prevents a buckling of force chain in intermediate strain; it leads a contractive behavior in large strain.

  • The rockbolt functions as a main support, which restricts enlargement of the plasticity area and increases stability in the original ground around tunnels, and prevents a second deformation of an excavated surface by supplementing vulnerability arising from opening of the excavated surface. System bolting is generally applied if ground conditions are bad. System bolting is generally installed perpendicular to the excavation direction in every span. If a place is narrow, or it is difficult to insert bolts due to construction conditions, it may be connected and used with short bolts, or installed obliquely. In this study, laboratory model tests were performed to analyze the effect of the ground being reinforced by inclined bolts, based on a bending theory that assumes that the reinforced ground is a simple beam. In all test cases, deflections and vertical earth pressures induced by overburden soil pressure were measured. Total of 99 model tests were carried out, by changing the installation angle of bolts, lateral and longitudinal distance of bolts, and soil height. The model test results indicated that when the installation angle of bolts was less than $75^{\circ}$, deflections of model beams tended to increase rapidly. Also, the relaxed load that was calculated by earth pressure was rapidly increased when the installation angle of bolts was less than $75^{\circ}$. However, the optimum installation angle of inclined bolts was judged to be in the range of $90^{\circ}{\sim}75^{\circ}$. Also, as might be expected, the reinforcement effect of bolts was increased when the longitudinal and lateral distance of bolts was decreased.

  • The purpose of this study was carried out to investigate the effect of electroacupuncture on reactive gliosis expressing GFAP in rat with transient global cerebral ischemia. Subjects were randomly divided into two groups, a control group and a electroacupuncture group on ST36, LI11 and SP9 with 2 Hz and 1 mA. The rats were sacrificed on 1, 3 and 7 days after transient cerebral ischemia using ligation of left common carotid artery. After making brain slide sections, they were immunostained with GFAP antisera(1:2,500). The results were as follows: The numbers of astrocytes of electroacupuncture group were decreased than those of control group at every 1, 2 and 7 days. Especially, the numbers of astrocytes at 3 days(p<0.01) and 8 days(p<0.05) were different statistically. And astrocytes had resting, hypertrophic and moving types on cerebral cortex. The decrease of numbers of astrocytes expressing GFAP showed that electroacupuncture could localise and minimize the brain damage by transient cerebral ischemia and cause brain cell plasticity.

  • Purpose: Normal aging results in detectable changes in the brain structure and function. We evaluated the changes of regional cerebral glucose metabolism in the normal aging process with FDG PET. Materials and Methods: Brain PET images were obtained in 44 healthy volunteers (age range 20-69 'y'; M:F = 29:15) who had no history of neuropsychiatric disorders. On 6 representative transaxial images, ROIs were drawn in the cortical and subcortical areas. Regional FDG uptake was normalized using whole brain uptake to adjust for the injection dose and correct for nonspecific declines of glucose metabolism affecting all brain areas equally. Results: In the prefrontal, temporoparietal and primary sensorimotor cortex, the normalized FDG uptake (NFU) reached a peak in subjects in their 30s. The NFU in the prefrontal and primary sensorimotor cortex declined with age after 30s at a rate of 3.15%/decade and 1.93%/decade, respectively. However, the NFU in the temporoparietal cortex did not change significantly with age after 30s. The anterior (prefrontal) posterior (temporoparietal) gradient peaked in subjects in their 30s and declined with age thereafter at a rate of 2.35%/decade. The NFU in the caudate nucleus was decreased with age after 20s at a rate of 2.39%/decade. On the primary visual cortex, putamen, and thalamus, the NFU values did not change significantly throughout the ages covered. These patterns were not significantly different between right and left cerebral hemispheres. Of interest was that the NFU in the left cerebellar cortex was increased with age after 20s at a rate of 2.86%/decade. Conclusion: These data demonstrate regional variation of the age-related changes in the cerebral glucose metabolism, with the most prominent age-related decline of metabolism in the prefrontal cortex. The increase in the cerebellar metabolism with age might reflect a process of neuronal plasticity associated with aging.

  • At the archaeological site, there are a variety of fired soil remains with which archaeomagnetic dating is measured, and there are also different kinds of kilns equipped with favorable plasticity conditions, so quality data can be gained. Among them, roof tile kilns indicate fairly great results of archaeomagnetic measuring. This should be associated with the properties of soil consisting of the kilns. With a lot of experimental results, it has been found that the fired soil samples of roof tile kilns have formed very stable residual magnetization. This author conducted archaeomagnetic research to present archaeomagnetic results of measuring about roof tile kilns from Goryeo in Gyeonggi. With the archaeomagnetic data of measuring 21 samples, this researcher tried to estimate the archaeomagnetic dating of roof tile kilns from Goryeo in Gyeonggi and divided them into three groups, group A (A.D.900~1150), group B (A.D.1150~1250), and group C (A.D.1300~1400) to figure out their relative order and arrange them in order. Through this kind of archaeomagnetic research, it will be possible to identify the dating of 21 roof tile kilns from Goryeo in Gyeonggi and also contribute considerably to chronological research on roof tile kilns from Goryeo. In the body, the remains and kilns belonging to each group are presented in detail.
