This study was designed to determine the effects of daidzein (DE) on hepatic lipid metabolism in chicks fed with low protein (LP) diet based on casein. In experiment 1, the male chicks were fed with one of the three levels of dietary protein containing 10.95%, 21.9% and 43.8% protein content for 2 days. In experiment 2, the chicks were fed one of the three levels of protein with or without DE at 1,000 mg/kg diet for 2 days. Experiment 3 was conducted to compare DE (LP+DE) with estradiol (LP+E2) in chicks fed with LP diet for 7 days. Plasma lipid profiles, hepatic lipid profiles, activities of hepatic malic enzyme and isocitrate dehydrogenase (ICDH) were measured. Transcriptions of hepatic fatty acid synthase, apolipoprotein-B (APO-B), and fructose bisphosphatase mRNA were measured by RT-PCR. Increasing dietary protein levels markedly decreased the concentrations of plasma triglycerides, hepatic total lipids, hepatic TG, and the mRNA transcriptions while the increased dietary protein levels increased hepatic ICDH activities in experiment 1. In experiment 2, the effects of dietary protein levels on blood and hepatic lipid content were more prominent than those of the additional DE. Interestingly, plasma TG levels were affected by DE supplementation (p<0.05). In experiment 3, DE inhibited APO-B mRNA expressions and stimulated the accumulation of lipid in the liver through mechanisms different from E2. In this study, we demonstrate that DE has beneficial effects on blood lipid profiles, but that it inhibits APO-B mRNA transcription and aggravates the fatty liver induced by LP diet in chicks.
Response surface analysis was used to study dietary ratios of n-3/n-6 fatty acid and P/S to minimize plasma triglycerides, total cholesterol and LDL ${\cdot}$ VLDL-C levels and maximize plasma HDL ${\cdot}$ C levels of rats. Because the dietary components were not statistically independent, they were studied in combinations of two variables. The two-variable combinations were the most useful in locating the desired maximum or minimum plasma triglycerides, total cholesterol and LDL ${\cdot}$ VLDL-C response in terms of the proportions of the dietary components. Response surface contours and three dimensional plots were developed for each plasma lipid response. The contours and three dimensional plots were used to help determine those combinations of the dietary fatty acid ratios that would produce the desired minimum or maximum lpid responses. The statistical analyses indicated that the minimized plasma cholesterol response levels could be attained with a diet consisting of 2.26 n-3/n-6 fatty acid and 2.15 P/S ratios.
The pharmacokinetic properties of S-amlodipine were studied using racemic amlodipine and single S-enantiomer (SK310) administration to rats. Plasma levels of the drug were determined using chiral liquid chromatography coupled with tandem mass spectrometry following solid phase extraction. The stereospecific analysis of amlodipine was performed on an ${\alpha}$-acid glycoprotein (AGP) column using a mobile phase comprising 10 mM ammonium acetate (pH 4.0) and propanol at a flow rate of 0.2 mL/min. This method was used to perform a comparative study of the pharmacokinetics of amlodipine and SK310. The results revealed that the pharmacokinetic profile of S-amlodipine after the administration of SK310 was comparable to that following the administration of the racemic mixture.
Effects of Gal geun (Puerariae Radix) EtOH ext. on lipid lowering and antioxidant were investigated in hyperlipidemic rat. Concentration of FFA and triglyceride in plasma showed a tendency to decrease in Gal geun ext. groups. Concentration of plasma total cholesterol and LDL-cholesterol decreased in Gal geun ext. groups. However the concentration of HDL-cholesterol showed no significantly different in all treatment groups. Concentration of liver total cholesterol and triglyceride showed a tendence to decrease in Gal geun ext. groups. Concentration of plasma and liver TBARS showed a low values in Gal geun ext. groups. The values of GSH-Px and SOD activity showed no significantly different among all the treatment groups. However the values of SOD and CAT activity showed a high value in the Gal geun ext. groups.
Effect of 4-nonylphenol (4-NP), endocrine disrupting compounds (EDCs), on glutamate oxaloacetate transaminase (GOT) and glutamate pyruvate transaminase (GPT) were investigated in the plasma of juvenile rockfish, Sebastes schlegeli. Fish were injected with 4NP (10, 50, 100 and 200 mg/kg body weight) in $70\%$ ethanol twice at 3-day intervals and plasma sampling were extracted at 7 days after the last injection. Controls received solvent only. 4-NP significant increased GOT in a dose-dependent manner. GPT was markedly elevated to $61\%$ (P<0.05) and $82\%$ (P<0.01) than that of the control at the 4-NP doses of 100 and 200mg, respectively. These results suggest that the estrogenic activity of 4-NP increase plasma GOT and GPT by toxic effect on hepatocyte.

The influence of TBTO (Tributyltin-Oxide) at a series of concentrations (0.52, 1.41 and 3.05 $\mu$g/L) for a period of 3 weeks on estradiol-17$\beta$, testosterone and cortisol levels in male Korean rockfish, Sebastes schlegeli was investigated. Compared to the stable concentration of estradiol-17$\beta$, plasma testosterone level was decreased significantly at the 3rd week after TBTO exposure in plasma of fish (>1.41 $\mu$g/L). The most profound physiological variation in TBTO exposed fish was a dramatic increase in plasma cortisol level at 3rd week. Conclusively, TBTO exposure caused increase level of cortisol as well as alteration of testosterone in the Korean rockfish. These results suggest that TBTO at environmentally relevant (nanomolar) concentrations disrupt endocrine secretions.

Objective: This study investigated the effects of surgical castration on behavior, physiological and inflammatory indicators, and leukocyte cytokine mRNA levels in Korean cattle bull calves. Methods: Nineteen Korean cattle bull calves (average body weight, 254.5 kg; average age, 8.2 months) were divided into two treatment groups: control (n = 9) and castration (n = 10). Surgical castration was performed using Newberry knives and a Henderson castrating tool. Blood was obtained just before castration (0 h) and at 0.5 h, 6 h, 1 d, 3 d, 7 d, and 14 d after castration. Plasma cortisol (PC), saliva cortisol (SC), plasma substance P, and plasma haptoglobin concentrations, and the leucocyte mRNA levels of the interleukin-1-alpha (IL1A), interleukin-1-beta (IL1B), interleukin-1 receptor antagonist (IL1RN), and interleukin-6 (IL6) genes were analyzed. Results: Castration decreased (p<0.01) the average daily gain and gain/feed ratio. Castration reduced the time spent eating (p<0.001) and the eating frequency (p<0.01) and increased (p<0.001) the lying frequency. Castration temporarily increased (p<0.05) circulating PC and SC concentrations at 0.5 h after castration. Castration temporarily increased (p<0.05) plasma substance P concentrations at 1 d after castration. Castration increased (p<0.05) plasma haptoglobin concentrations at 1 and 3 d after castration. Castration increased (p<0.05) leukocyte mRNA levels of the IL1A, IL1B, IL1RN, and IL6 genes at 6 h after castration. Conclusion: Castration temporarily induced stress and expression of leucocyte inflammatory cytokine genes in Korean cattle bull calves.
무척추동물에서 척추동물에 이르기까지 대부분의 난생동물들의 난황 단백질의 전구체를 vitellogenin(VTG)이라 한다. 난생 척추동물에서 VTG는 간에서 합성되어 혈액을 통해 난세포로 전이된다. 암컷 어류는 정상적인 생식주기에서 난황단백전구체 형성이 시작되면 혈중 농도는 급격히 증가하게 된다. 수컷도 VTG의 유전자를 가지고 있기 때문에 낮은 수준의 내인성 에스트로겐으로 아주 적은 양의 단백질이 있을 수 있다. 그렇지만 수컷에 외인성에스트로겐 유사물질에 노출되면 그 양은 증가하게 된다. 따라서 어류에서 VTG는 내분비계 장애물질 효과를 조사하는 유용한 생물학적 추적자로 사용할 수 있다. 이 연구에서는 에스트로겐 유사물질에 오염된 지역에 서식하는 망둑어류의 혈중 VTG의 수준을 정량할 수 있는 방법을 제공하고자 한다. 이러한 목적을 위해 꾹저구(Chaenogobius annularis)의 VTG를 분리하고, 이에 대한 단클론 항체와 다클론 항체를 제작하여 혈중 VTG 수준을 정량할 수 있는 sandwich 경합 ELISA system을 만들었다. 난황 단백질에 대한 단클론 항체와 다클론 항체를 사용한 ELISA system에 대한 타당성 검토를 하였다. 순차적으로 희석한 암 컷의 혈청에 대한 흡광도 곡선은 VTG 표준농도의 곡선과 평행하였으나 수컷의 혈청에 대한 흡광도 곡선은 평행하지 않았다. 이 VTG에 대한 sandwich ELISA system으로 꾹저구(C. annularis)의 혈중 VTG의 수준을 정량뿐만 아니라 문절망둑(Acanthogobius flaviman)과 풀망둑(A. hasta)의 혈중 VTG 수준을 정량할 수 있다.
본 실험에서 암모니아 급성 노출은 대왕범바리의 혈액학적 성상 및 혈장 성분에 유의적 변화를 나타내었다. 본 실험의 결과 20mg L-1 이상의 급성 암모니아 노출은 대왕범바리의 혈액학적 성상 및 혈장 성분에 영향을 주어 생리적 변화를 일으키며, 40mg L-1의 급성 암모니아 농도는 대량 폐사를 유발할 수 있음을 나타낸다. 향후 대왕범바리 바이오플락 양식기술 적용을 위한 양식연구에 이러한 결과를 지표로 활용할 수 있을 것이다.
본 연구는 넙치 치어(평균 체장 $13.3{\pm}1.6cm$, 평균 체중 $25.6{\pm}3.7g$)를 수중 6가크롬에 10일간 0, 0.5,1.0, $2.0mg\;L^{-1}$의 농도로 노출시켜 독성영향 판단하기 위해 수행되었다. 넙치 hemoglobin과 hematocrit와 같은 혈액학적 성상은 수중 크롬노출에 의해 유의적으로 감소하였다. 혈장 무기성분인 calcium과 magnesium은 크롬노출에 의해 유의적인 변화는 나타나지 않았다. 혈장 무기성분인 glucose와 cholesterol과 같은 $1.0mg\;L^{-1}$ 이상의 크롬노출에 의해 유의적으로 증가하였지만, total protein은 유의적인 변화는 나타나지 않았다. 혈장 효소성분인 AST, ALT, ALP는 크롬노출에 의해 유의적인 증가가 나타났다. 본 실험의 결과는 수중 크롬노출은 넙치의 혈액 및 혈장성분에 유의적인 변화를 유발하며, 이러한 지표의 변화는 수중 크롬노출의 독성영향을 판단하는 주요한 지표가 될 것이다.
