Cabbage plants were transformed with the potato proteinase inhibitor II (PINII) gene, bar gene, and hpt gene using Agrobacterium. The expression of the PINII gene was driven by its own promoter which was wound-inducible. Ten transgenic plants were obtained from medium containing hygromycin as a selection antibiotic. The integration and expression of PINII and bar genes were confirmed by Southern and Northern hybridization. Growth and development of diamondback moths (Plutella xylostella) and tobacco cutworm (Spodoptera litura) larvae were examined on $T_1$ plants. The weight of the larvae and pupae of these two insects grown on transgenic plants was not different compared to those grown on wild type plants. However, the pupation and emergence rate of diamondback moths and tobacco cutworms fed on some transgenic plants was lower than on wild type plants. These results suggest that the PINII transgene under the control of a wound-induced promoter may be used for control of insects in transgenic cabbage through reduction of insect progeny number.
Plants need essential mineral elements to favorably develop and to complete their life cycle. Despite the irreplaceable roles of microelements, they are often ignored due to the relative importance of macroelements with their influence on crop growth and development. We focused on the changes in primary metabolites in the leaves and roots of bell pepper plants under 6 microelements-deficient conditions: Copper (Cu), Zinc (Zn), Iron (Fe), Manganese (Mn), Boron (B) and Molybdenum (Mo). Bell pepper plants were grown in hydroponic containers, and individual elements were adjusted to 1/10-strength of Hoagland nutrient solution. A remarkable perturbation in the abundance of the primary metabolites was observed for the Fe and B and the Mn and B deficiencies in the leaves and roots, respectively. The metabolites with up-accumulation in the Fe-deficient leaves were glucose, fructose, xylose, glutamine, asparagine and serine. In contrast, the Mn deficiency also resulted in a higher accumulation of glucose, fructose, xylose, galactose, serine, glycine, β-alanine, alanine and valine in the roots. The B deficiency noticeably accumulated alanine, valine and phenylalanine in the roots while it showed a substantial decrease in glucose, fructose and xylose. These results show that the primary metabolism could be seriously disturbed due to a microelement deficiency, and the alteration may be either the specific or adaptive responses of bell pepper plants.
GA 생합성에 결정적 역할을 하는 GA 3$\beta$-hydroxylase를 pIG121-Hm 벡터에 GUS유전자를 빼고 antisense로 클로닝하여 이를 동진벼에 도입한 결과 17개체의 절간신장이 억제된 형질전환한 식물체를 얻을 수 있었다. 일반 재배 동진벼를 대조군으로 하여 비교하였을때 antisense GA 3$\beta$-hydroxylase 유전자가 형질 도입된 식물체의 획득형질은 평균적으로 대조군에 비해 절간 신장의 억제가 확인되었다. 절간신장의 억제가 보인 개체의 엽육조직을 co취하여 Southen blot hybridization분석 결과 3개의 line에서 모두 single copy로 도입된 것으로 나타났다. 이로써$T_o$ 식물체 내에 antisense GA 3$\beta$-hydroxylase 유전자를 내포하고 있는 것으로 확인되었다. 이것은 antisense GA 3$\beta$-hydroxylase 유전자가 생체내에서 직접 또는 간접적으로 GA 3$\beta$-hydroxylase유전자의 발현에 관여한 것이라 사료되어진다.
Invasion mechanisms of pathogens and counteracting defense mechanisms of plants are highly diverse and perpetually evolving. While most classical studies of plant defense have focused only on defense-specific factor-mediated responses, recent work is beginning to shed light on the involvement of non-stress signal components, especially growth and developmental processes. This shift in focus links plant resistance more closely with growth and development. In this review, we summarize our current understanding of how pathogens manipulate host developmental processes and, conversely, of how plants deploy their developmental processes for self-protection. We conclude by introducing our recent work on UNI, a novel R protein in Arabidopsis which mediates cross-talk between developmental processes and defense responses.
Sweetpotato fields in Korea are highly infected with virus and virus like diseases that greatly diminish both yield and quality as indicated by field observations and laboratory tests. In order to solve this problem, there is an urgent need to produce and mass propagate virus-free planting materials for distribution to the farmers. These experiments were conducted, firstly, to determine the most appropriate culture media, nutrient solution, and cutting intervals to maintain growth and vigor of tissue cultured plantleta as mother plants for propagation in insect-proof greenhouse. And as a labor saving method, the production efficiency of plug trays for rapid propagation of stem cuttings as a source of planting materials was likewise evaluated. Results showed that plants grown in medium B supplied with 0.5 and 1.0 strength of MS nutrients had high growth rate, and 20-day cutting interval was the best. 72-plug tray was better than 128-plug. Secondly, it was to develop a technique for the production of first-generation seed roots using hydroponics cultivation system. The yield of virus-free plants propagated in the non-insect proof and open-field cultivation was 2,402 kg/10a, 6% higher than those in the insect-proof cultivation, and the rate of virus re-infection was 18% higher compared to 3.3% with insect-proof cultivation. Lastly, it was to investigate the growth performance of virus free plants in farmers' field. Differences were existed in the yield depending on the variety used, but virus free plants showed an increase of $6{\sim}24%$ over virus infected plants.
To determine as optimal sowing time of forage rape in spring time in southern areas of Korea, forage rape cv. Velox, the highest yielding variety among introduced varieties of forage rape, was grown under five different sowing times. Yield components such as plant length, number of branches and number of leaves etc. were higher at the sowing time of Mar. 15 and Mar. 25. The plants sown at Mar. 15 and Mar. 25 also showed highest fresh and dry matter yield. When plants were grown under later sowing time, they showed higher values in content of crude protein and lower values in contents of crude fiber such as Neutral Detergent Fiber (NDF), Acid Detergent Fiber (ADF), cellulose and lignin. There was no relationship between variation of In Vitro Dry matter Digestibility (IVDMD) and sowing time. The plants sown at Mar. 15 and Mar. 25 showed highest digestible dry matter yields.
Among abiotic stresses in plants, drought and chilling stresses reduce the supply of moisture to plant tissues, inhibit photosynthesis, and severely reduce plant growth and yield. Thus, the application of water stress-tolerant agents can be a useful strategy to maintain plant growth under abiotic stresses. This study assessed the effect of exogenous bio-based 2,3-butanediol (BDO) application on drought and chilling response in tomato and turfgrass, and expression levels of several plant signaling pathway-related gene transcripts. Bio-based 2,3-BDOs were formulated to levo-2,3-BDO 0.9% soluble concentrate (levo 0.9% SL) and meso-2,3-BDO 9% SL (meso 9% SL). Under drought and chilling stress conditions, the application of levo 0.9% SL in creeping bentgrass and meso 9% SL in tomato plants significantly reduced the deleterious effects of abiotic stresses. Interestingly, pretreatment with levo-2,3-BDO in creeping bentgrass and meso-2,3-BDO in tomato plants enhanced JA and SA signaling pathway-related gene transcript expression levels in different ways. In addition, all tomato plants treated with acibenzolar-S-methyl (as a positive control) withered completely under chilling stress, whereas 2,3-BDO-treated tomato plants exhibited excellent cold tolerance. According to our findings, bio-based 2,3-BDO isomers as sustainable water stress-tolerant agents, levo- and meso-2,3-BDOs, could enhance tolerance to drought and/or chilling stresses in various plants through somewhat different molecular activities without any side effects.
The Development of science and technology began with the use of fire. In addition to the invention of an artificial light, more time and activity areas were provided so that continuous technological development has continued. Also, artificial lights brought an improvement in productivity through the applications in various fields such as agriculture, livestock farming, fisheries, and so on. Especially, in the field of agriculture, LED has come to the front as an alternative for food security against extreme climatic change. Meanwhile, with great interest, many researches about the convergence of agricultural biotechnology and LED industry are ongoing over the world. This paper shows the correlation of the plants growth using the LED artificial lighting similar to sunlight, which is needed for photosynthesis, in subway station which has actually no natural lights. Also, it is expected that this paper will lay out the groundwork for improving an environmentally friendly image which gives a relief and an emotional effect to passengers by using nature friendly lighting.
To determine optimal plant density of forage rye in southern areas of Korea, forage rye cv. Paldanghomil, the highest yielding variety among experimental varieties of forage rye, was grown under five different plant density. Yield components such as plant height, and number of leaves were highest at the broad casting. Plants sown at broadcasting also showed highest fresh and dry matter yield. When plants were grown of the broad casting, they showed higher values in content of crude protein and lower values in contents of crude fiber such as neutral detergent fiber(NDF), acid detergent fiber(ADF), cellulose and lignin. There was no relationship between variation of in vitro dry matter digestibility(IVDMD) and plant density. Plants sown at the broad casting showed highest digestible dry matter yields.

Exposing plant organs to high doses of ionizing irradiation, penetrating into the plant tis-sues and cells, along the track structure of particles, lesions, and sublesions are formed on the molecules and organelles. As a result, disorders in the growth and development as well as chlorophyll-deficiency symptoms occur. The time scale of their reparation, recovery and over compensation during ontogenesis, constitutes a question of high theoretical and practical importanced, with special regard to nuclear fallout. With an aim to model the “ut supra”stated phenomena, the seeds of bean, Echo elit licensed variety, were irradiated by 300 Gy dose of X-ray-irradiation (120 kV:4.5 mA). According to the data obtained, the biosynthesis of photosynthetic pigments, will have been completed by the beginning of flowering. In consequence of the overcompensation of the repairing processes, the organs of plants developed from irradiated seeds, showed a partly differing correlative growth, compared to those of control plants. In order to characterize the vivo response of radiation-injured plants, a new method and approach were used. The changes of the electric capacitance of the plants during their ontogenesis, were continously monitored and recorede via a computer-aided and controlled measurement. In view of the data collected in such a way, the repairing plants may respond more quickly and intensively to the changes of environmental factors.
