A Biostimulant Preparation of Brown Seaweed Ascophyllum nodosum Suppresses Powdery Mildew of Strawberry

  • Strawberry, an important fruit crop, is susceptible to a large number of pathogens that reduce fruit quality and productivity. In this study, the effect of a biostimulant prepared from Ascophyllum nodosum extract (ANE) (0.1%, 0.2%, and 0.3%) was evaluated on powdery mildew progression under greenhouse and field conditions. In the greenhouse, application of 0.2% ANE showed maximum reduction in powdery mildew progression as compared to the control. Forty-eight hour post-inoculation, foliar spray of 0.2% ANE reduced spore germination by 75%. Strawberry leaves sprayed with ANE showed higher total phenolic and flavonoid content in response to powdery mildew infection. Furthermore, application of ANE elicited defense response in strawberry plants by induction of defense-related enzymes, such as phenylalanine ammonia lyase, polyphenol oxidase, and peroxidase activity. In field conditions, foliar spray of 0.2% ANE showed a reduction of 37.2% of natural incidence of powdery mildew infection as compared to the control. ANE sprayed plant also reduces the severity of powdery mildew infection under natural conditions. These results indicate that application of ANE induces the strawberry plant's active defense against powdery mildew infection by induction of secondary metabolism and regulating the activities of defense-related enzymes.

A Marine Bacterium with Animal-Pathogen-Like Type III Secretion Elicits the Nonhost Hypersensitive Response in a Land Plant

  • Active plant immune response involving programmed cell death called the hypersensitive response (HR) is elicited by microbial effectors delivered through the type III secretion system (T3SS). The marine bacterium Hahella chejuensis contains two T3SSs that are similar to those of animal pathogens, but it was able to elicit HR-like cell death in the land plant Nicotiana benthamiana. The cell death was comparable with the transcriptional patterns of H. chejuensis T3SS-1 genes, was mediated by SGT1, a general regulator of plant resistance, and was suppressed by AvrPto1, a type III-secreted effector of a plant pathogen that inhibits HR. Thus, type III-secreted effectors of a marine bacterium are capable of inducing the nonhost HR in a land plant it has never encountered before. This suggests that plants may have evolved to cope with a potential threat posed by alien pathogen effectors. Our work documents an exceptional case of nonhost HR and provides an expanded perspective for studying plant nonhost resistance.

Antifungal Activity-Guided Analysis of Actinostemma lobatum Extracts through Serial Sub-fractions

  • Plants are treasure trove of novel compounds that have potential for antifungal chemicals and drugs. In our previous study, we had screened plant extracts obtained from more than eight hundred plant materials collected in Korea, and found that butanol fraction of the Actinostemma lobatum were most potent in suppressing growth of diverse fungal pathogens of plants. Here in this study, we describe further analysis of the butanol fraction, and summarize the results of subsequent antifungal activity test for the sub-fractions against a selected set of plant pathogenic fungi. This line of analyses allowed us to identify the sub-fractions that could account for a significant proportion of observed antifungal activity of initial butanol fraction from A. lobatum. Further analysis of these sub-fractions and determination of structure would provide the shortlist for novel compounds that can be a lead to new agrochemicals.

The Ralstonia pseudosolanacearum Type III Effector RipL Delays Flowering and Promotes Susceptibility to Pseudomonas syringae in Arabidopsis thaliana

  • The plant defense responses to microbial infection are tightly regulated and integrated with the developmental program for optimal resources allocation. Notably, the defense-associated hormone salicylic acid (SA) acts as a promoter of flowering while several plant pathogens actively target the flowering signaling pathway to promote their virulence or dissemination. Ralstonia pseudosolanacearum inject tens of effectors in the host cells that collectively promote bacterial proliferation in plant tissues. Here, we characterized the function of the broadly conserved R. pseudosolanacearum effector RipL, through heterologous expression in Arabidopsis thaliana. RipL-expressing transgenic lines presented a delayed flowering, which correlated with a low expression of flowering regulator genes. Delayed flowering was also observed in Nicotiana benthamiana plants transiently expressing RipL. In parallel, RipL promoted plant susceptibility to virulent strains of Pseudomonas syringae in the effector-expressing lines or when delivered by the type III secretion system. Unexpectedly, SA accumulation and SA-dependent immune signaling were not significantly affected by RipL expression. Rather, the RNA-seq analysis of infected RipL-expressing lines revealed that the overall amplitude of the transcriptional response was dampened, suggesting that RipL could promote plant susceptibility in an SA-independent manner. Further elucidation of the molecular mechanisms underpinning RipL effect on flowering and immunity may reveal novel effector functions in host cells.

식물 병원균에 대한 항진균 활성을 갖는 Bacillus subtilis KYS-10의 분리 (The isolation of Bacillus subtilis KYS-10 with antifungal activity against plant pathogens)

  • 본 연구는 9종 식물병원균에 대한 항진균 활성을 나타내는 길항세균을 분리하여 동정한 결과 Bacillus subtilis KYS-10로 명명하였고, sucrose 4%, yeast extract 1.0%, $K_2HPO_4$ 0.2%, pH 7의 최적배지에서 150 rpm, $30^{\circ}C$, 8일간 배양 시 가장 좋은 생육을 나타냈다. B. subtilis KYS-10의 9종 식물병원균에 대한 항진균 활성을 확산법에 의해 생육저지환의 크기를 측정한 결과 G. zeae (맥류 붉은곰팡이병) 70 mm로 가장 높았고, 35~39 mm의 생육저해환을 나타낸 식물병원균은 P. KACC 40439(벼도열병), P. capsici KACC 40177(고추역병균), C. destructans KACC 41077(인삼뿌리썩음병) 40~43 mm, C. gloeosporioides KACC 43520(포도 탄저병균), C. gloesporioides KACC 40003(고추 탄저병균), S. shiraiana KACC 41065(상추 균핵병균), S. shiraiana(오디 균핵병균) 있었으며, F. Oxysporum KACC 44452(인삼 부패병균)은 28 mm로 다종의 식물병원균에 대해 높은 저해활성을 보였다. 이상의 결과들은 추후 포장실험을 통하여 친환경 미생물제제로의 개발 가능성을 시사해 주고 있다.

Seed-borne Pathogenic Bacterium Interact with Air-borne Plant Pathogenic Fungus in Rice Fields

  • Air-borne plant pathogenic fungus Fusarium graminearum and seed-borne plant pathogenic bacterium Burkholderia glumae are cause similar disease symptoms in rice heads. Here we showed that two pathogens frequently co-isolated in rice heads and F. graminearum is resistant to toxoflavin produced by B. glumae while other fungal genera are sensitive to the toxin. We have tried to clarify the resistant mechanism of F. graminearum against toxoflavin and the ecological reason of co-existence of the two pathogens in rice. We found that F. graminearum carries resistance to toxoflavin as accumulating lipid in fungal cells. Co-cultivation of two pathogens resulted in increased conidia and enhanced chemical attraction and attachment of the bacterial cells to the fungal conidia. Bacteria physically attached to fungal conidia, which protected bacterium cells from UV light and allowed disease dispersal. Chemotaxis analysis showed that bacterial cells moved toward the fungal exudation compared to a control. Even enhanced the production of phytotoxic trichothecene by the fungal under presence of toxoflavin and disease severity on rice heads was significantly increased by co-inoculation rather than single inoculation. This study suggested that the undisclosed potentiality of air-born infection of bacteria using the fungal spores for survival and dispersal.

식물유래 2차 대사물질의 병충해 및 잡초 방제효과 (Pathogen, Insect and Weed Control Effects of Secondary Metabolites from Plants)

  • 농작물이 병해충 및 잡초 등에 의하여 피해를 받게 되면 농업생산성은 상당히 저하된다. 따라서 농업생산성을 높이기 위하여 합성농약을 과용해 왔고, 이로 인하여 토양의 질과 농업환경이 파괴되는 결과를 초래하고 있어서 지속적인 농업이 점차 어려워지고 있다. 오늘날의 농업은 농업환경의 파괴를 최소화하고 농산물의 안전성을 확보함과 동시에 생산성을 증대해야하는 복합적인 문제에 직면해 있다. 한편, 식물은 일반적으로 생존하기 위하며 외부의 침입으로부터 자신을 보호하거나 자신의 영역을 확보하기 위하며 다양한 종류의 2차대사물질을 생산하고 방출한다. 병원균이 침입했을 때 식물이 생산하는 항생물질인 phytoalexin으로는 화본과 식물의 경우 flavonoid계와 diterpenoid계의 물질을 생성하고, 쌍자엽 식물은 감염된 병원균의 종류에 상관없이 Leguminosae과는 flavonoids계, Cruciferae과는 indole 유도체, Solanaceae과는 sesquiterpenoid계, Umbelliferae과는 coumarin계 물질들을 생성하여 병원균에 저항성을 가진다. 곤충의 생리작용을 저해하거나 섭생을 싫어하게 하는 기능의 물질로는 terpene계의 pyrethrin, azadirachtin, limonin, cedrelanoid, toosendanin, fraxinellone/dictamnine 등이 있으며, alkaloid계로서는 terpenoid와 alkaloid가 결합된 sesquiterpene pyridine 및 norditerpenoids alkaloids와 azepine계, amide계, loline계, stemofoline계, pyrrolizidine계 alkaloids 등이 있다. 식물은 또한 자신의 영역을 확보하기 위하여 다른 식물의 생장을 저해하는 물질을 생성하는데 여기에는 terpene계의 essential oil 및 sesquiterpene lactone과 이외에 benzoxazinoids, glucosinolate, quassinoid, cyanogenic glycoside, saponin, sorgolennone, juglone 등 다수의 2차 대사물질들이 있다. 이와 같은 기능의 2차 대사물질을 병해충 및 잡초 방제에 직, 간접으로 이용하는 것은 친환경농업의 한 가지 방법일 수 있다. 그러나 천연물질들은 자연계에서 쉽게 분해되어 효율이 떨어지는 경우가 많고 식물을 통하여 생산하는데도 한계가 있다. 따라서 보다 안전성과 효율성이 뛰어난 2차 대사물질을 찾아내는 연구와 아울러 방제기능이 있는 물질의 생합성경로를 구명하고 대사공학적으로 이용하므로 병해충에 저항성이 있고 잡초 방제효과를 갖는 형질전환 식물을 육성하는 연구가 지속적으로 이루어져야 할 것이다.

Computer-Aided Drug Discovery in Plant Pathology

  • Control of plant diseases is largely dependent on use of agrochemicals. However, there are widening gaps between our knowledge on plant diseases gained from genetic/mechanistic studies and rapid translation of the knowledge into target-oriented development of effective agrochemicals. Here we propose that the time is ripe for computer-aided drug discovery/design (CADD) in molecular plant pathology. CADD has played a pivotal role in development of medically important molecules over the last three decades. Now, explosive increase in information on genome sequences and three dimensional structures of biological molecules, in combination with advances in computational and informational technologies, opens up exciting possibilities for application of CADD in discovery and development of agrochemicals. In this review, we outline two categories of the drug discovery strategies: structure- and ligand-based CADD, and relevant computational approaches that are being employed in modern drug discovery. In order to help readers to dive into CADD, we explain concepts of homology modelling, molecular docking, virtual screening, and de novo ligand design in structure-based CADD, and pharmacophore modelling, ligand-based virtual screening, quantitative structure activity relationship modelling and de novo ligand design for ligand-based CADD. We also provide the important resources available to carry out CADD. Finally, we present a case study showing how CADD approach can be implemented in reality for identification of potent chemical compounds against the important plant pathogens, Pseudomonas syringae and Colletotrichum gloeosporioides.

Survey of bovine norovirus infections from diarrheic calves in South Korea, 2015-2017

  • This study examined complex infections with various enteropathogens and the genetic diversity of bovine norovirus (BNoV) in 932 fecal samples from diarrheic calves in South Korea. Overall, seventeen (1.8%) of the samples tested positive for BNoV following RT-PCR examination. All BNoV-positive samples were co-infected with other intestinal pathogens, including bovine Rotavirus, Giardia, Cryptosporidium, and Escherichia coli. The genetic diversity of the BNoVs shared high nucleotide identity (98.1-99.5%) and amino acid homology (93.5-98.1%) with genotype 2 BNoV (GIII.2) strains. In conclusion, BNoV infections with GIII genotypes were detected in complex infections of diarrheic calves in South Korea.

Antimicrobial Activities of 1,4-Benzoquinones and Wheat Germ Extract

  • We evaluated the antibacterial activities of selected edible Korean plant seeds against the food-borne pathogens Staphylococcus aureus KCTC1927, Escherichia coli KCTC2593, Salmonella typhimurium KCTC2054, and Bacillus cereus KCTC1014. While screening for antibacterial agents, we discovered that wheat germ extract contains 2,6-dimethoxy-1,4-benzoquinone (DMBQ) and is highly inhibitory to S. aureus and B. cereus. This is the first report of the antibacterial activity of wheat germ extract. We also investigated the antibacterial activities of the 1,4-benzoquinone standards 1,4-benzoquinone (BQ), hydroquinone (HQ), methoxybenzoquinone (MBQ), and 2,6-dimethoxy-1,4-benzoquinone (DMBQ). DMBQ and BQ were the most highly inhibitory to S. aureus and S. typhimurium, followed by MBQ and HQ. MICs for DMBQ and BQ ranged between 8 and 64 ${\mu}g/ml$ against the four foodborne pathogens tested. DMBQ and BQ showed significant antibacterial activity; the most sensitive organism was S. aureus with an MIC of 8 ${\mu}g/ml$. BQ exhibited good activity against S. typhimurium (32 ${\mu}g/ml$) and B. cereus (32 ${\mu}g/ml$). The results suggest that wheat germ extract has potential for the development of natural antimicrobials and food preservatives for controlling foodborne pathogens.
