American ginseng (Panax quinquefolius L.) is grown in some regions of the USA and Canada and marketed for its health promoting attributes. While cultivation of this plant species has taken place in North America for over 100 years, there are many challenges that need to be addressed. In this article, the current production method used by growers is described and the challenges and opportunities for research on this valuable plant are discussed. These include studies on pharmacological activity, genetic diversity within the species, genetic improvement of currently grown plants, molecular characterization of gene expression, and management of diseases affecting plant productivity. The current research developments in these areas are reviewed and areas requiring further work are summarized. Additional research should shed light on the nature of the bioactive compounds and their clinical effects, and the molecular basis of active ingredient biosynthesis, and provide more uniform genetic material as well as improved plant growth, and potentially reduce losses due to pathogens.
Flower industry is now growing due to the development of economy in many countries. Simultaneously, needs from consumers in flower market are varied widely. To satisfy the needs from consumers and deal with a variety of diseases from a lots of pathogens as well as climate change, new elite flower cultivars should be released in flower market. For this purpose, conventional and biotechnological techniques can be employed to make good cultivar. Therefore, this review describes the general overview of flower breeding techniques including cross-hybridization, mutation breeding and genetic transformation systems. Also, breeding systems for ornamentals derived from plant tissue culture such as embryo culture, in vitro fertilization, ovary/ovule culture and haploid production were reviewed. Furthermore, in this study recent development of the generation of new flower cultivars using marker-assisted breeding, plant transformation including particle bombardment and Agrobacterium tumefaciens as well as genome-editing technology were described. This review will be contributed to the development and releasement of new flower cultivars with horticulturally useful traits in the future.
Fifty isolates of myxobacteria were isolated from soils from several areas in Korea during 1996-1997 and bioactivity against plant pathogenic fungi of these isolates was examined. A myxobacterial isolate KR025 showed good antifungal activities against Pyricularia oryzae, Cryphonectria parasitica, Colletotrichum lagenarium, and C. gloeosporioides but did not against Rhizoctonia solani, Fusarium oxysporum and Pythium ultimum. The bacterium was identified as Myxococcus fulvus based on morphological and physiological characteristics. Antifungal substances were extracted from culture broth and bacterial cell of Myxococcus fulvus KR025 by ethyl acetate. Antifungal substance of Myxothiazole (100 ${\mu}{\textrm}{m}$/ml) produced by Myxococcus fulvus KR 025 controlled 97.0% rice blast, tomato late blight, wheat leaf rust, and barley powdery mildew and showed 45.0 and 82.6% disease control of rice sheath blight and cucumber gray model, respectively.
A phage technique for detection of Xanthomonas axonopodis pv. citri, a causal bacterium of canker on Sastuma mandarin fruits was developed. Phage and ELISA techniques were compared for their sensitivity for detection of Xanthomonas axonopodis pv. citri on orange fruits. Both of techniques revealed a similar efficiency for the bacterial detection; the pathogenic bacteria were observed in pellet from the fruits with over one canker spot with below 2 mm in diameter. In field assays, the increase of phage population(120%) on surface of the fruits related to the disease development one month later indicated that the bacterial pathogens inhabit on the surface. The procedure will be effectively used for detection of only living bacterial pathogen on fruit surfaces of Satsuma mandarin and for the disease forecasting.
The monoxenic culture of the fungivorous nematode, Aphelenchus avenae, was applied for the control of soil-borne ginseng pathogens such as Fusarium solani and Rhizoctonia solani. Fungivorous nematode populations were measured in a field to examine relationships between the nematode populations and suppression of ginseng root diseases. Inoculation of A. avenae (5000 nematodes per petri-dish) reduced the colonization of the Fusarium mycelium on root discs of ginseng and carrot by 80.0% and 60.5%, respectively. A. avenae also significantly reduced the occurrence of damping-off of ginseng by R. solani pathogenic to ginseng, and no plant damage by the nematode was noted. In a 3-year-old ginseng field infested with Cylindrocarpon destructans, plant missing caused by root rot positively correlated to the density of potato rot nematode, Ditylenchus destructor, but it was reduced with the population of A. avenae, suggesting that A. avenae might inhibit the occurrence of ginseng root rot.
Rehmannia glutinosa is a perennial medicinal plant belonging to the family Scrophulariaceae with more than 300 species known in the world, especially in temperate regions. Its roots have been used widely in Korea for medicinal purposes. However, it is commonly infected by various pathogens during storage, causing great damage to the roots, and impedes the intensive farming of the crop. Therefore, an attempt has been made to isolate and screen a resistance gene against the pathogen Fusarium oxysporum using differential display. We treated salicylic acid(SA), and isolated a resistance gene that responds to SA. As a result, we found that SA was involved in plant defense mechanism in pathogenicity tests with SA treated and non-treted plants, and we isolated a partial PR-la gene through differential display polymerase chain reaction(DD-PCR) method.

The use of a microorganism, or its secretions, to prevent plant disease offers an attractive alternative or supplement to synthetic fungicides for the management of plant disease without the negative effects of chemical control mechanisms. During a screening for microorganisms with the potential to be used as microbial fungicides, Bacillus sp. BS061 was isolated from a plant leaf. The strain BS061 potently inhibited the mycelial growth of Botrytis cinerea, and significantly reduced disease incidence of powdery mildew in cucumber and strawberry. We also found that the culture filtrate of BS061 inhibited the mycelial growth of various plant pathogens.
In order to detect quarantine plant viruses, we developed reverse transcription-polymerase chain reaction (RT-PCR) primer pairs for five single-stranded (ss) plant RNA viruses that are not currently reported in Korea but could be potential harmful plant viral pathogens. Three viruses such as Chilli veinal mottle virus (ChiVMV), Colombian datura virus (CDV), and Tobacco etch virus (TEV) belong to the genus Potyvirus while Chrysanthemum stem necrosis virus (CSNV) and Iris yellow spot virus (IYSV) are members of the genus Tospovirus. To design RT-PCR primers, we used reported gene sequences corresponding to the capsid protein and polyprotein for ChiVMV, CDV, and TEV while using nucleocapsid protein regions for CSNV and IYSV. At least two different primer pairs were designed for each virus. Fifteen out of 16 primer pairs were successfully applied in detection of individual quarantine virus with high specificity and efficiency. Taken together, this study provides a rapid and useful protocol for detection of five quarantine viruses.
Rehmannia glutinosa is one of the most important medicinal crops in Korea. However, various plant pathogens, including Fusatium spp., cause great damage on R. glutinosa and result in enormous economic losses. This study was conducted to breed Fusarium-resistant plants by using Agrobacterium tumefaciences and AFP (anti-fungal protein) gene. The plant material used was a native accession of R. glutinosa. The PCR analysis was conducted to verify transgenicity. Based on the PCR analysis, nptII band was observed in transgenic plant genome. Southern blot and AFP protein analyses also showed the expression of this gene in transgenic plants. Expression of AFP in transgenic plants offers the possibility of developing resistance to fungal infection.
WRKY family proteins are a class of plant-specific transcription factors involved in stress response signaling pathways. In this study a gene encoding a putative WRKY protein was isolated from a pepper EST database (http://genepool.kribb.re.kr). The cDNA, named Capsicum annuum WRKY2 (CaWRKY2), encodes a putative polypeptide of 548 amino acids, containing two WRKY domains with zinc finger motifs and two potential nuclear localization signals. Northern blot analyses showed that CaWRKY2 mRNA was preferentially induced during incompatible interactions of pepper plants with PMMoV, Pseudomonas syringae pv. syringae 61, and Xanthomonas axonopodis pv. vesicatoria race 3. Furthermore, CaWRKY2 transcripts were strongly induced by wounding and ethephon treatment, whereas only moderate expression was detected following treatment with salicylic acid and jasmonic acid. CaWRKY2 was translocated to the nucleus when a CaWRKY2-smGFP fusion construct was expressed in onion epidermal cells. CaWRKY2 also had transcriptional activation activity in yeast. Taken together our data suggest that CaWRKY2 is a pathogen-inducible transcription factor that may have a role in early defense responses to biotic and abiotic stresses.
