  • Influence of fructose addition to the cultivation medium on the production of taxanes and the growth of callus culture of Taxus baccata was studied. The cultures showed an ability to adjust to the substitution of some of the sucrose in the media by fructose and the fresh biomass accumulation was higher on the media containing different concentrations of fructose during the second cultivation period.

Antifungal Activity of Thymol against Aspergillus awamori and Botrytis aclada Isolated from Stored Onion Bulbs

  • The antifungal activity of thymol against Aspergillus awamori F23 and Botrytis aclada F15 in onions was examined through direct treatment with amended media and gaseous treatment with I-plates (plastic plates containing central partitions). The protective and curative control efficacy of thymol was examined 24 h before and after the inoculation of onion bulbs with the fungal isolates. Mycelial growth, sporulation, and spore germination of the isolates were inhibited on potato dextrose agar amended with various concentrations of thymol or acetic acid (positive control). Overall, thymol produced a stronger inhibitory effect on the mycelial growth and development of the isolates than acetic acid. Following gaseous treatment in I-plates, mycelial growth, sporulation, and spore germination of the isolates were inhibited at higher concentrations of thymol or acetic acid; however, acetic acid showed a little effect on the sporulation and spore germination of the isolates. Following the treatment of onion bulbs with 1000 mg L-1 of thymol 24 h before and after fungal inoculation, lesion diameter was greatly reduced compared with that following treatment with 0.5% ethanol (solvent control). Onion bulbs sprayed with thymol 24 h before fungal inoculation generally showed reduced lesion diameters by isolate F23 but not in isolate F15 compared with those sprayed 24 h after fungal inoculation. Collectively, thymol effectively inhibited the growth and development of A. awamori and B. aclada on amended media and in I-plates. In addition, spraying or fumigation of thymol is more desirable for effectively controlling these postharvest fungal pathogens during long-term storage conditions.

Co-treatment with Origanum Oil and Thyme Oil Vapours Synergistically Limits the Growth of Soil-borne Pathogens Causing Strawberry Diseases

  • Vapours from origanum oil (O) and thyme oil (T) were applied to the four soil-borne strawberry pathogens Fusarium oxysporum f. sp. fragariae, Colletotrichum fructicola, Lasiodiplodia theobromae, and Phytophthora cactorum, causing Fusarium wilt, anthracnose, dieback, and Phytophthora rot, respectively. Increasing T vapour doses in the presence of O vapour strongly inhibited mycelial growths of the four pathogens and vice versa. When mycelia of F. oxysporum f. sp. fragariae and P. cactorum exposed to the combined O + T vapours were transferred to the fresh media, mycelial growth was restored, indicating fungistasis by vapours. However, the mycelial growth of C. fructicola and L. theobromae exposed to the combined O + T vapours have been slightly retarded in the fresh media. Prolonged exposure of strawberry pathogens to O + T vapours in soil environments may be suggested as an alternative method for eco-friendly disease management.

Effects of Temperature and Culture Media Composition on Sporulation, Mycelial Growth, and Antifungal Activity of Isaria javanica pf185

  • The fungal isolate Isaria javanica pf185 has potential as a mycopesticide because it demonstrates insecticidal activity against the green peach aphid and antifungal activity against Colletotrichum gloeosporioides. For commercialization of this isolate, determination of the optimal and least expensive culture conditions is required; however, these data are not currently available. This study describes the conditions for optimal development of conidia and production of metabolites for the biocontrol of the fungal pathogen. The optimal culture conditions were examined using cultures on solid agar and liquid media. High growth temperature enhanced spore formation but reduced antifungal activity in both solid and liquid media. The highest spore yield was obtained in a medium containing glucose as a carbon source and yeast extract as a nitrogen source. Soybean powder and wheat bran were effective nitrogen sources that promoted spore production and antifungal activity of the isolate. These results revealed the basic, cost-effective growth media for commercial production of a biopesticide with insecticidal and antifungal properties for use in integrated pest management.

감나무 둥근무늬낙엽병균(Mycosphaerella nawae)의 배양적 특성 (Cultural Characteristics of Mcyosphaerella nawae causing Spotted Leaf Casting of Persimmon)

  • Mycosphaerella nawae, the causal organism, of spotted leaf casing disease of persimmon, was isolated from infected leaves showing typical symptom. The cultural characteristics of the fungus were compared on artificial media. Among 24 different combinations of culture media and supplements, oatmeal agar+persimmon leaf extract (PLE) and PAD+ PLE+streptomycin showed the highest rate of isolating as 57.1%. The best medium for mycelial growth of the fungus was PDA+persimmon leaf powder (PLP). The colony diameter was reached 47mm for 30 days at 2$0^{\circ}C$. PDA+PLE also supported good mcyelial growth showing 46mm of diameter in same condition. The optimum growth temperature of this fungus in PDA was recognized fairly low. The mycelial growth was higher at 2$0^{\circ}C$ than 15$^{\circ}C$. The variation of pH between pH 4 to pH 8 did not affect to the mycelial growth of the pathogen.

분주 근경 크기와 배지 종류가 고추냉이 생육에 미치는 영향 (Effect of Divided Rhizome Size and Medium Type on Growth of Wasabia japonica Matsum.)

  • 분주 크기와 배지 종류가 고추냉이 생존율과 생육에 미치는 영향을 알아보고자 환경이 조절된 생장상에서 60일간 재배하였다. 수태로 감싼 근경 크기(직경) 5mm 이상 분주 묘의 생존율은 100%였으며, 생육(초장 12cm 내외, 엽수 $3{\sim}4$장)은 양호하였다. 생육 60일의 분주 묘는 초장 20cm 이상, 엽수가 $1{\sim}2$장 증가하였다. 루톤 분의제 처리는 생존율과 생육에 효과가 없었다. 배지 종류(마사, 난석)에 따른 직경 $5{\sim}10$mm 크기의 분주 묘를 담액 수경으로 재배하였을 때 83% 이상의 생존율을 보였으며 배지 종류에 따른 생육 차이는 없었다. 따라서 근경 크기 5mm 이상의 고추냉이 분주를 환경이 조절된 생장상에서 30일간 수경재배 방식으로 순화하여 고추냉이 건전 묘를 생산할 수 있다.

황칠나무의 揷木發根力 增進 (Promotion of Rootability of the Cuttings of Dendropanax morbifera Lev)

  • This study was conducted to examine the effects of media, crown position, clone ,a nd plant growth substances upon the rootability of cuttings for the establishment of the method of mass production by cutting of Dendropanax morbifera Lev. the artifical soil mixtureof vermiculite, perlite and peatmoss (1 : 1 : 1 v/v/v) with IBA 100mg /l treatment was most effective on the rooting rate (85%) was obtained with semihardwood cuttings, which were collected early-August after the first growing seasons. The effect of crown position on rooting of cuttings were obsreved. Average percentage rooting of cuttings on crown position showed 78% , 72% and 65% in middle , upper and lower part with IBA 100mg/l treatment , respectively. Among the four treatment levels of IBA, the highest rooting rate was observed in 100mg/l IBA treatment.

고추의 생장에 미치는 Azospirillum brasilense 및 Methylobacterium oryzae 접종 효과 (Effect of Inoculation of Azospirillum brasilense and Methylobacterium oryzae on the Growth of Red Pepper Plant)

  • BACKGROUND: Rhizosphere bacteria may improve plant growth and productivity both by supply nutrients and hormonal stimulation. Although many experiments have shown improvements in plant growth with inoculation of bacterial cultures to the rhizosphere, the main obstacle in the applications of plant growth promoting rhizobacteria in a large scale is the inconsistency of the results. We tested the growth promoting effects of Azospirillum and Methylobacterium strains on red pepper plant. METHODS AND RESULTS: Red pepper seedlings were grown for 25 days in a growth media inoculated with A. brasilense CW903 or M. oryzae CBMB20. The seedlings were transplanted and grown for 45 days in pots with soil in a greenhouse, at half the recommended level of fertilizer. Bacterial culture, $4.0{\times}10^9$ for A. brasilense CW903 and $5.8{\times}10^8$ CFU for M. oryzae CBMB20, was applied in root zone soil periodically every 10 days during the experiment. Inoculation of M. oryzae CBMB20 significantly increased the red pepper plant growth in terms of leaf number, height and mass of shoot, or root mass compared to uninoculated control plants. Although beneficial effects of A. brasilense on plant growth of many crops were observed, the growthpromoting effect of A. brasilense CW903 on red pepper plant was not found in this study. CONCLUSION(s): The factors responsible for the irregularities in plant growth promoting of rhizobacteria are difficult to elucidate. Extensive inoculation experiments in the greenhouse and in the field should enable us to define the factors critical to obtain successful application of plant growth promoting rhizobacteria.

Response of Mulberry Brown Leaf Spot Fungus Myrothecium roridum to Different Plant Extracts

  • Anti-fungal potential of 5 plant extracts viz., Eucalyptus citriodora, Allium sativum, Cassia sophera, Chromolaena odorata and Datura metel on the growth of mulberry brown leaf spot pathogen Myrothecium roridum were examined. Except fur the aqueous extract of Allium bulb, ethanolic leaf extract of all other plants more efficiently reduced the colony growth of the fungus on potato-dextrose-agar, Of which, Allium and Eucalyptus extracts were more effective. Initiation of radial growth of M. roridum on solid media was deferred maximum 6 days by ethanolic Eucalyptus extract and 4 days by aqueous Allium extract at $0.4 mg.ml^{-1}$. In the liquid media amended with Eucalyptus extract ($0.4 mg.ml^{-1}$) complete inhibition of sporulation was noticed upto 8 days, and initial inhibition of mycelial bio-mass generation was considerably diminished with time and reduction was 1.3 fold 14 days after application. While, complete inhibition of mycelial growth for 6-14 days was recorded with $\geq$0.1 mg.ml$^{-1}$ commercial eucalyptus oil. However, rejuvenation of growth appeared when fungus was re-inoculated in fresh media. Post-inoculate application of different doses Of Eucalyptus and Allium extracts significantly (p < 0.05) reduced the disease severity in pot-ted mulberry. However, persistence of the effect up to 28 days was apparent at $\geq$ 1.0 mg.ml$^{-1}$ and effectively was on par with carbendazim (1 mg.ml$^{-1}$ ). Almost equal control ability of 1.0 mg.ml$^{-1}$ Eucalyptus extracts can be achieved by ca. 10 times lowered dose of commercial eucalyptus oil. It seems, the toxic principle of E. citrodora to M. roridum is fungistatic in nature and may have essential oil based origin.
