Proteomics of plant-fungal pathogen interaction: an overview (식물과 곰팡이 병원균과의 상호작용에 대한 프로테오믹스 최근 연구 동향)

  • So far it has been generally considered that proteomic approaches are very useful for studying plant-microbes interaction. In this review, recent studies based on papers published from 2010 to 2013 have investigated proteomics analysis in various interaction during plant-fungal pathogen infection by means of gel-based proteomics coupled with mass spectrometry (MS)-based analysis. In rice, three papers focused on rice-Magnaporthe oryzae interaction were mainly reviewed in this study. Interestingly, another study showed proteomic changes in rice inoculated with Puccinia triticina, which is not only an fungal pathogen in wheat and but also results to the disease resistance with non-host defense manner in rice. Additionally, proteomics analysis has been widely subjected to understand defense mechanism during other crops (wheat, tomato, strawberry and mint) and their fungal pathogen interaction. Crops inoculated are analyzed to identify differentially regulated proteins at various tissues such as leaf and apoplast using 2-DE analysis coupled with various MS approaches such as MALDI-TOF MS, nESI-LC-MS/MS and MudPIT, respectively. Taken together, this review article shows that proteomics is applicable to various organisms to understand plant-fungal pathogen interaction and will contribute to provide important information for crop disease diagnosis and crop protection.

Failure Analysis of Circulating Water Pump Shaft in Power Plant (발전 계획에서 순환 물 펌프 고장 분석)

  • This paper presents the root cause failure analysis of the circulating water pump in the 560 MW thermal power plant. A fractured austenitic stainless-steel shaft operated for 24 years was examined. Fracture morphology was investigated by micro and macro-fractographic analysis. The metallurgical analyses including chemical analysis, metallography and hardness testing were performed. The analysis reveals that the pump shaft was fractured due to the reverse bending load with combination of rotating bending load. Corrective actions for plant operator was recommended based on the analysis.

  • This paper presents the numerical methodology of ATHOS3 code for thermal hydraulic analysis of steam generators in nuclear power plant. Topics include porous media approach, governing equations, physical models and correlations for solid-to-fluid interaction and heat transfer, and numerical solution scheme. The ATHOS3 code is applied to the thermal hydraulic analysis of steam generator in the Korea Kori Unit-1 nuclear power plant and the computed results are presented

  • The purpose of this study is to suggest analysis model of RCB construction in nuclear power plant. For the objective, This study drew the risk factors of RCB construction from existing literature. The results of the study proposed analysis model made hierarchy in rebar, form, and concrete work. These will be baseline data for risk management in construction project of nuclear power plant.

  • Purpose: This study aims to analyze the fire resistance performance applied to plant facilities with high fire risk in Korea, secure suitable fire resistance performance, and ensure the fire safety of plant facilities. Method: Using the finite element analysis program Ansys, thermal transfer analysis and structural analysis were performed with fire load and fireproof coating as variables, and the fire resistance performance of plant facilities was analyzed based on the analysis results. Result: The fireproof coating applied to domestic plant facilities failed to secure fire resistance performance when the fire load of hydrocarbon fire presented in UL 1709 was applied, and it was confirmed that the deformation of steel after the fire was also significant. Conclusion: The current fire resistance performance applied to plant facilities in Korea cannot secure fire resistance performance in sudden fire growth and large fire loads like petrochemical plants, and it is necessary to secure fire safety by evaluating suitable fire resistance performance through performance evaluation of plant facilities.

Quantitative Trait Locus Mapping and Candidate Gene Analysis for Plant Architecture Traits Using Whole Genome Re-Sequencing in Rice

  • Plant breeders have focused on improving plant architecture as an effective means to increase crop yield. Here, we identify the main-effect quantitative trait loci (QTLs) for plant shape-related traits in rice (Oryza sativa) and find candidate genes by applying whole genome re-sequencing of two parental cultivars using next-generation sequencing. To identify QTLs influencing plant shape, we analyzed six traits: plant height, tiller number, panicle diameter, panicle length, flag leaf length, and flag leaf width. We performed QTL analysis with 178 $F_7$ recombinant inbred lines (RILs) from a cross of japonica rice line 'SNU-SG1' and indica rice line 'Milyang23'. Using 131 molecular markers, including 28 insertion/deletion markers, we identified 11 main- and 16 minor-effect QTLs for the six traits with a threshold LOD value > 2.8. Our sequence analysis identified fifty-four candidate genes for the main-effect QTLs. By further comparison of coding sequences and meta-expression profiles between japonica and indica rice varieties, we finally chose 15 strong candidate genes for the 11 main-effect QTLs. Our study shows that the whole-genome sequence data substantially enhanced the efficiency of polymorphic marker development for QTL fine-mapping and the identification of possible candidate genes. This yields useful genetic resources for breeding high-yielding rice cultivars with improved plant architecture.

Antifungal Activity-Guided Analysis of Actinostemma lobatum Extracts through Serial Sub-fractions

  • Plants are treasure trove of novel compounds that have potential for antifungal chemicals and drugs. In our previous study, we had screened plant extracts obtained from more than eight hundred plant materials collected in Korea, and found that butanol fraction of the Actinostemma lobatum were most potent in suppressing growth of diverse fungal pathogens of plants. Here in this study, we describe further analysis of the butanol fraction, and summarize the results of subsequent antifungal activity test for the sub-fractions against a selected set of plant pathogenic fungi. This line of analyses allowed us to identify the sub-fractions that could account for a significant proportion of observed antifungal activity of initial butanol fraction from A. lobatum. Further analysis of these sub-fractions and determination of structure would provide the shortlist for novel compounds that can be a lead to new agrochemicals.

  • This paper discusses an economic study analysis of captive power plant as a commercial plant in the cost based pool market. In this paper I assumed the conversion of a captive power plant owned by factories to a commercial plant and investigated the changes in profitability associated with this. I set the total electricity expense of a captive purpose plant as state A and the costs associated with converting to a commercial purpose plant as state B. Each state subdivided by case which is classified its plant variable cost, type of generation (combined cycle, single cycle) and type of power contract received. After set model for each case, different economic benefits by each case can be calculated.

Development of Performance Analysis Methodology for Nuclear Power Plant Turbine Cycle Using Validation Model of Performance Measurements (원전 터빈사이클 성능 데이터의 검증 모델에 의한 성능분석 기법의 개발)

  • Verification of measurements is required for precise evaluation of turbine cycle performance in nuclear power plant. We assumed that initial acceptance data and design data of the plant could provide correlation information between performance data. The data can be used as sample sets for the correct estimation model of measurement value. The modeling was done practically by using regression model based on plant design data, plant acceptance data and verified plant performance data of domestic nuclear power plant. We can construct more robust performance analysis system for an operation nuclear power plant with this validation scheme.
