Simultaneous determination of saikosaponin derivatives in Bupleurum falcatum by HPLC-ELSD analysis using different extraction methods

  • Saikosaponin derivatives such as saikosaponins A, B1, B2, B3, B4, C, and D present in Bupleurum falcatum were analyzed by a high performance liquid chromatograph equipped with an evaporative light scattering detector, using different extraction solvents (water and 70% ethanol). The samples were injected into a YMC Pack Pro C18 column and separated using a gradient elution system with a mobile phase composed of acetonitrile and water at a flow rate of 1.1 mL/min. The content of saikosaponin derivatives was higher in 70% ethanol extract than in water extract. This study provides an efficient analytical method for determining the optimal conditions for extraction of saikosaponin derivatives, which can be used as a basis for development of functional foods and pharmaceutical products from B. falcatum.

  • For more accurate height/depth control of the agricultural implements , the soil surface as a reference position should be measured as accurate as possible. A new measurement system using microwave was developed to detect the true soil surface even under plant and/or vegetation. Two-frequency continuous-wave radar was used as the measurement system. It could estimate the distance to the target by measuring the phase difference between two different frequencies continuous-waves which reflected on the target surface. The system performance was evaluated on the barely field where the average height of barley was 91.5 cm. The experimental results showed that the system performance was not affected by the existence of barely. The maximum measurement errors were 8.91 com and 8.44cm for two different experimental plots.

  • A model reference adaptive control(MRAC) theory is very useful for controlling a plant of which the parameters are unknown or vary during operation usint only input-output signal of plant. In this study, 2' nd order discreter time MRAC controller is designed for an electrohydraulic position control system which is represented with nonlinear mathematical model and the least square method is adopted for the para-meter adjustment law. This control algorthm is applied to the position control of electrohydraulic servosystem through computer simulation and the effect of the change of load, sampling time upon the performance following reference model and upon the performance of estimating plant parameters are examined.

Comparison of the contents of total polyphenol, total flavonoid, and flavonoid derivatives in unfermented and fermented barley sprouts

  • Barley (Hordeum vulgare) belongs to the Poaceae family. This study compared the polyphenol and flavonoid levels of unfermented and fermented barley sprouts using spectrophotometric assays. The findings indicated that fermentation greatly boosted the flavonoid content but caused only a slight increase in the polyphenol content. However, this does not imply that fermentation has no effects whatsoever on the polyphenol content of barley sprouts. This was due to the fact that some flavonoids cannot be detected by the wavelength used to calculate the overall polyphenol concentration. Both samples were subjected to high-performance liquid chromatography analysis and detected the flavonoids lutonarin, saponarin, isoorientin, isovitexin, and tricin-all of which have bioactive properties-most notably known for their antioxidant activity. These results augment the ongoing phytochemical profiling research and can possibly valorize the already thriving barley industry.

  • Plant Configuration Management(CM) is an activity to maintain consistency of design requirements, physical configuration, facility configuration information throughout the life cycle of the plant by systematically managing changes that occur during the plant design and operation process. Conformity between information must be ensured not only in the design stage, but also in the case of design changes in the operation and maintenance stages, and thus a computer system capable of efficiently managing them is required. In particular, in consideration of an application to small and medium-sized domestic plants, a computer system that can support configuration management at a low cost is needed. Accordingly, in this study, a configuration management system has been developed to support the management of plant design information and change procedures in the operation stage of small and medium-sized plants. Here, a case for quantitatively evaluating the effectiveness of the developed system is described.

  • As a preceding process for developing design technology and establishing operation technology, the design procedure of the SCR(Selective Catalytic Reduction) pilot plant that can handle $1,000Nm^{3}/hr$ of flue gas was reported in this paper. And we also considered several factors that might cause abnormality of the plant in the designing process. The plant was designed and fabricated to test the $DeNO_{x}$ performances in variable operating conditions in the range of $3,000{\sim}36,000hr^{-1}/hr$ in space velocities, $1.67{\sim}6\;m/s$ in linear velocities, $200{\sim}500^{\circ}C$ temperatures, $300{\sim}1,000Nm^{3}/hr$ flow rates, and $0{\sim}1.4:1\;NH_{3}/NO$ ratios. In order to maintain the flow uniformity, the guide vanes and flow straightener were designed and constructed in the plant. The SCR pilot plant can be operated by the automatic control system, which enable to obtain performance data in real time and to set up the operating technology. The catalyst reactor consists of 4 catalyst layers and surface area of each layer can be adjusted to be of small size. Arrangement of catalysts per layer is $3{\times}6$ with the catalyst dimensions of $150{\times}150{\times}500mm(L{\times}W{\times}H)$.

  • The aim of this study is to develop an optimal analytical method for gonadorelin, progesterone, oxytocin and estradiol, the major components of hormones. A relatively simple and reproducible method using high performance liquid chromatography was developed and as a result of the measurement of specificity, linearity, repeatability, accuracy and intermediated precision, the validity of the developed method was verified with the result of meeting the verification criteria of analytical method validation. Using this newly developed method, 12 post-market veterinary products were tested and the ingredient content were 91.9~116.4%, which satisfied the 90~120% condition of the administrative measure standard. Therefore, if the newly developed method is used for the collection examination of hormone in veterinary medicine, it can be useful as an approved test method.

  • Many research efforts have made on various fields to develop a system which is able to process diverse data in terms of efficiency and accuracy as well as minimizing the error in data-processing. As known, it is obvious that procurement is crucial process for the plant projects due to its importance and impact on the performance. It is strongly required to develop an effective metadata system to assist optimal controlling data for the logistics discipline of procurement processes in plant projects. Therefore, the main purpose of this paper is to apply a metadata system for logistics of the plant projects. The suggested technical module is tested using sample data and evaluated its performance in terms of accuracy and running speed. The results of this research will be extended to the other processes of plant projects in order to achieve a ultimately process-based life-cycle plant project information management system

Identification and Growth Inhibition of Phytotoxic Substances from Tomato Plant (토마토식물의 독성물질 확인과 생장억제작용)

  • Phenolic compounds such as gallic acid, ferulic acid, p-hydroxybenzoic acid, vanillic acid, salicylic acid, tannic acid, and hydroquinone were identified from the aqueous extracts and volatile substances of tomato plant by paper chromatography, high performance liquid chromatography and gas chromatography. The seed germination and seedling growth of the experimental species, lettuce and egg plant, were severely inhibited in 5$\times$10-3M of phenolic reagents identical to those identified from tomato plant. Germination and growth rate of test species in 5$\times$10-4M and 5$\times$10-5M were higher than that of 5$\times$10-3M. Therefore, 5$\times$10-3M of phenolic compounds would be assumed to be threshold concentration for allelopathic effects.

Agronomic performance of 20 soybean recommended varieties in Korea

  • A total of 20 soybean recommended varieties which were developed until late 1980's in Korea was evaluated at Suwon. Comprehensive evaluation and correlation analysis were conducted on the agronomic characters. Great variations were found in these genotypes for branch number, pod number, and grain yield per plant. The variation in number of pods/plant ranged from 53 to 164, and in grain yield from 25.9 to 68.8 g. The coefficient of variation for most of the characters had a wide range. In correlation coefficient, grain yield per plant showed a positive phenotypic association with weight of pods, pod number of branches, and weight of stem. Multiple regression analysis was done to formulate selection criteria. It indicated that stout and medium-stature genotypes with more branches, resulting in varieties with more pods per plant but with medium-size seeds are available to obtain high-yielding varieties.
