Pueraria lobata is a perennial legume plant, widely distributed in the countries of East Asia. It is a medicinally important leguminous plant and produces various isoflavones such as puerarin, daidzein etc which have potential for preventing several chronic diseases including osteoporosis, cardiovascular disease and cancer. In this study, we tried to induce hairy roots in vitro from Korean wild arrowroot P. lobata and investigated the effects of hormones and light conditions. Initially leaf and stem segments were infected with Agrobacterium rhizogenes and incubated in different conditions. Hairy roots were induced from only stem segments and the induction was best at dark condition and the presence of IBA during incubation. Secondary roots were also significantly much more induced at the dark condition than at the 16 hours light condition. Among plant growth regulators of auxin, IBA was best for secondary root formation while 2,4-D, IAA and NAA produced callus or less hairy roots. The presence of the foreign gene rolC transferred by A. rhizogenes that plays a major role in hairy root induction was confirmed by PCR. The accumulation of isoflavones such as puerarin and daidzin was also confirmed. These results will facilitate mass production of hairy root and can be used for the production of functional substances from wild arrowroots.

Background: Crop rotation plays an important role in improving soil chemical properties, minimizing the presence of disease pathogens, and assists in neutralizing autotoxic effects associated with allelochemicals. Methods and Results: Five rotation crops of sudan grass, soybean, peanut, sweet potato, and perilla were cultivated for one year with an aim to reduce yield losses caused by repeated cropping of ginseng. In 2-year-old ginseng grown in the same soil as a previous ginseng crop, stem length and leaf area were reduced by 30%, and root weight per plant was reduced by 56%. Crop rotation resulted in a significant decrease in electrical conductivity, $NO_3$, and $P_2O_5$ content of the soil, whereas organic matter, Ca, Mg, Fe, Cu, and Zn content remained-unchanged. Soil K content was increased following crop rotation with sudan grass and peanut only. Rotation with all alternate crops increased subsequent ginseng aerial plant biomass, whereas root weight per plant significantly increased following crop rotation with perilla only. A significant positive correlation was observed between root rot ration and soil K content, and a significant negative correlation was observed between ginseng root yield and the abundance of actinomycetes. Crop rotation affected the soil microbial community by increasing gram negative microbes, the ratio of aerobic microbes, and total microbial biomass whereas decreases were observed in actinomycetes and the ration of saturated fatty acids. Conclusions: In soil exhibiting crop failure following replanting, crop rotation for one year promoted both soil microbial activity and subsequent ginseng aerial plant biomass, but did not ameliorate the occurrence of root rot disease.
Root and basal stem rot disease occurred on moth orchid (Phalaenopsis spp.), Pung-nan (Neofinetia falcata) and Nadopung-nan (Aerides japonicum) grown in the farmers' greenhouses located in Namyangju Kyonggi province, Korea during 2005 to 2006. Wilting symptoms occurred on these orchard plants at initial stage and the infected plant leaves turned yellow to red. The discolored leaves were fallen down to lead to eventual death of the entire plant. A total of 59 isolates of Fusarium spp. was obtained from roots and leaf bases of the diseased plants. The cultural and morphological characteristics of isolated Fusairum spp. were identified as Fusarium oxysporum, F. proliferatum and F. solani. F. oxysporum and F. proliferatum were isolated from all these orchard plants but F. solani was isolated only from Phalaenopsis spp. Pathogenicity of the three Fusarium spp. was confirmed by artificial inoculation. Although F. oxysporum, F. proliferatum and F. solani cusing root rot disease in Phalaenopsis spp. have been reported in Korea, the pathogens in N. falcata and A. japonicum were not reported yet. Therefore, this is the first report on the root and stem rot of N. falcata and A. japonicum caused by F. oxysporum and F. proliferatum in Korea.

The nursery test against rice blast in Korea from 2014 to 2015 was analyzed. The average of disease severity of leaf blast in 12 sites showed $3.7{\pm}2.1$ in 2014 and $4.4{\pm}2.1$ in 2015. Disease severity of leaf blast in Icheon and Cheolwon was increased ranging from $2.8{\pm}2.2$ in 2014 to $6.3{\pm}1.8$ in 2015. Using a designation system, a total of 588 isolates collected those years were categorized into 34 races in 2014 and 51 races in 2015 based on the reaction pattern of Korean differential varieties. The blast isolates of 2015 were more diverse than those in 2014. The ratios of KI race to KJ race were 54:46 in 2014 and 70:30 in 2015; however, the predominant race population was KJ-301 as 16%, and KI-101 as 15% in 2014 and 2015, respectively. These results indicate that the distribution of the blast races is getting more diverse in Korea, therefore, this research would provide the possibility to predict race distribution and change to prevent the outbreak of rice blast and will also serve as a useful information for breeding of resistant rice cultivar against blast.
Attempts to search infection period, infection speed in the tissue of neck blast of rice plant, location of inoculum source and effects of several conditions about the leaf sheath of rice plants for neck blast incidence have been made. 1. The most infectious period for neck blast incidence was the booting stage just before heading date, and most of necks have been infected during the booting stage and on heading date. But $Indica{\times}Japonica$ hybrid varieties had shown always high possibility for infection after booting stage. 2. Incubation period for neck blast of rice plants under natural conditions had rather a long period ranging from 10 to 22 days. Under artificial inoculation condition incubation period in the young panicle was shorter than in the old panicle. Panicles that emerged from the sheath of flag leaf had long incubation period, with a low infection rate and they also shown slow infection speed in the tissue. 3. Considering the incubation period of neck blast of rice plant, we assumed that the most effective application periods of chemicals are 5-10 days for immediate effective chemicals and 10-15 days for slow effective chemicals before heading. 4. Infiltration of conidia into the leaf sheath of rice plant carried out by saturation effect with water through the suture of the upper three leaves. The number of conidia observed in the leaf sheath during the booting stage were higher than those in the leaf sheath during other stages. Ligule had protected to infiltrate of conidia into the leaf sheath. 5. When conidia were infiltrated into the leaf sheath, the highest number of attached conidia was observed on the panicle base and panicle axis with hairs and degenerated panicle, which seemed to promote the infection of neck blast. 6. The lowest spore concentration for neck blast incidence was variable with rice varietal groups. $Indica{\times}Japonica$ hybrid varieties were infected easily compared to the Japonica type varieties, especially. The number of spores for neck blast incidence in $Indica{\times}Japonica$ hybrid varieties was less than 100 and disease index was higher also in $Indica{\times}Japonica$ hybrid than in Japonica type varieties. 7. Nitrogen content and silicate content were related with blast incidence in necks of rice plants in the different growing stage changed during growing period. Nitrogen content increased from booting stage to heading date and then decreased gradually as time passes. Silicate content increased from booting stage after heading with time. Change of these content promoted to increase neck blast infection. 8. Conidia moved to rice plant by ascending and desending dispersal and then attached on the rice plant. Conidia transfered horizontally was found very negligible. So we presumed that infection rate of neck blast was very low after emergence of panicle base from the leaf sheath. Also ascending air current by temperature difference between upper and lower side of rice plant seemed to increase the liberation of spores. 9. Conidial number of the blast fungus collected just before and after heading date was closely related with neck blast incidence. Lesions on three leaves from the top were closely related with neck blast incidence, because they had high potential for conidia formation of rice blast fungus and they were direct inoculum sources for neck blast. 10. The condition inside the leaf sheath was very favorable for the incidence of neck blast and the neck blast incidence in the leaf sheath increased as the level of fertilizer applied increased. Therefore, the infection rate of neck blast on the all panicle parts such as panicle base, panicle branches, spikelets, nodes, and internodes inside the leaf sheath didn't show differences due to varietal resistance or fertilizers applied. 11. Except for others among dominant species of fungi in the leaf sheath, only Gerlachia oryzae appeared to promote incidence of neck blast. It was assumed that days for heading of varieties were related with neck blast incidence.

Effects of green tea, buckwheat, and grape leaf extracts on factors related to blood circulation were studied using rats fed high-cholesterol diet for 4 weeks. Rats were randomly divided into five groups, and plant extracts were orally administered. Green tea extract increased bleeding time in rat tails, suggesting it could prevent platelet aggregation. Administration of green tea, buckwheat, and grape leaf extracts decreased total cholesterol level in liver. Grape leaf extracts decreased thiobarbituric acid-reactive substances in plasma, whereas buckwheat and grape leaf extracts decreased the substances in liver. These results showed extracts of green tea, buckwheat, and grape leaf were effective for improving lipid composition in blood and liver and inhibiting lipid peroxidation in animal tissue, suggesting they may have potential to prevent cardiovascular diseases.
The genetically modified leaffolder-resistant (Cry1Ac1) rice plant was evaluated for the changes of resistance by comparing the occurrence of major diseases with a japonica type Korean rice variety, Nakdong which was the mother plant of the transgenic rice event, in greenhouse and field conditions. There was no difference in the occurrence of sheath blight and Helminthosporium blight between the two varieties in the fields. We couldn't find any difference of resistance for fungal blast and bacterial leaf blight by artificial inoculation in greenhouse. There was also no difference in the susceptibility to sheath blight in artificial inoculation tests confirming the results in the fields. The possibility of gene transfer of Bar and Cry1Ac1 from the genetically modified rice plant to naturally infected pathogens such as Fusarium moniliforme and Pyricularia oryzae in the field conditions was tested by PCR. And the possible transfer of those genes by continuous inoculation of Xanthomonas oryzae pv. oryzae and Rhizoctonia solani was also tested. However, we couldn't find any possibility of transfer of the genes in natural and artificial conditions.
Background: Korean ginseng (Panax ginseng Meyer) is a perennial herb prone to various root diseases, with Phytophthora cactorum being considered one of the most dreaded pathogens. P. cactorum causes foliar blight and root rot. Although chemical pesticides are available for disease control, attention has been shifted to viable, eco-friendly, and cost-effective biological means such as plant growth-promoting rhizobacteria (PGPR) for control of diseases. Methods: Native Bacillus amyloliquefaciens strain HK34 was isolated from wild ginseng and assessed as a biological control agent for ginseng. Leaves from plants treated with HK34 were analyzed for induced systemic resistance (ISR) against P. cactorum in square plate assay. Treated plants were verified for differential expression of defense-related marker genes using quantitative reverse transcription polymerase chain reaction. Results: A total of 78 native rhizosphere bacilli from wild P. ginseng were isolated. One of the root-associated bacteria identified as B. amyloliquefaciens strain HK34 effectively induced resistance against P. cactorum when applied as soil drench once (99.1% disease control) and as a priming treatment two times in the early stages (83.9% disease control). A similar result was observed in the leaf samples of plants under field conditions, where the percentage of disease control was 85.6%. Significant upregulation of the genes PgPR10, PgPR5, and PgCAT in the leaves of plants treated with HK34 was observed against P. cactorum compared with untreated controls and only pathogen-treated plants. Conclusion: The results of this study indicate HK34 as a potential biocontrol agent eliciting ISR in ginseng against P. cactorum.
In April of 2002, the gray mold disease occurred severely on kalanchoe (Kalanch oe blossfeldiana) grown in a nursery in Daesan-myon, Changwon City, Korea. The lesions were started with water-soaked spot and the leaves discolored from the tip and became gray or dark and gradully expanded to whole leaf. The conidia and mycelia of pathogen appearedd on flower, flower stalk and stem. Conidia were gray, 1-celled, mostly ellipsoid or ovoid in shape and were 8~17 ${\times}$5~12${\mu}m$ in size. Conidiophores were 13~34${\mu}m$ in size. The sclerotia were formed abundantly on potato-dextrose agar. The optimum temperature for sclerotial formation was $20^{\circ}C$. Pathogenicity of the causal organism was proved on Kalanchoe blossfeldiana according to Koch,s postulate. The causal organism was identified as Botrytis cinerea based on mycological characteristics. This is the first report on gray mold of Kalanchoe(Kalanchoe blossfeldiana) caused by Botrytis cinerea in Korea.
Apple is the most economically important fruit in Korea. The suspected viroid disease of dapple apple was found in apple fruits cultivated in Kyungpook province. Symptoms begin in mid-July as small circular spots, which stand out against the background color on the young fruit. Dappling of the fruit becomes more intense and easier to detect as the fruit approaches maturity; the affected spots remain yellowish as the fruit matures. no leaf or bark syndromes have been associated with this disease. The infected fruits are downgraded considerably during quality grading. The low molecular weight RNA containing viroid RNA molecules were extracted from the peels of the apples with dapple symptoms. The RNA molecules were extracted from the apples using Qiagen column chromatography. The purified RNAs were used for the synthesis of cDNA with RT-PCR. The PCR products were then ligated into a pGEM-T Easy vector, cloned and sequenced. The sequence of the viroid RNA molecule shows 331 nucleotides with one base difference ("G" insertion between the position of 133 and 134) compared with that of the Apple scar skin viroid (ASSVd) reported by Hashimoto and Koganezawa in Japan. This is the first report on the occurrence of the ASSVd in apple trees cultivated in Korea, as well as the identification of a new Korean strain of the ASSVd.the ASSVd.
