We selected eight resistant cultivars including 'Heukryongsamcheok', 'Heukgeumsolsamcheok' and 'Gangryeoksamcheok' showing high suppressive effect against cucumber downy mildew (CDM) through plastic film house and field trials in spring and autumn season in 2015. Of them, these three varieties, 'Heukryongsamcheok', 'Heukgeumsolsamcheok' and 'Gangryeoksamcheok' were used to evaluated suppressive effect against CDM by comparing disease severity (area under disease development progress curve, AUDPC) with those of three susceptible varieties under plastic film house condition. AUDPC of three resistant cultivars was in the range of 10.9 to 23.6, meanwhile those of three susceptible cultivars was in the range of 286.7 to 290.3. Consequently, we confirmed that cultivation of the selected resistant cultivars can reduce CDM in vivo. When disease severity (diseased leaf area) was investigated on a resistant cultivar ('Heukryongsamcheok') sprayed without any organic materials and susceptible cultivars sprayed single-, two times- and three times with Bordeaux mixture, sulfur-loess mixture and $Kocide^R$ (copper hydroxide), respectively, it was the lowest in the resistant cultivar plot compared to susceptible cultivar plots sprayed with any organic materials. In addition, we evaluated control effect of the selected resistant cultivar by comparing that of organic materials including lime sulfur mixture, Bordeaux mixture and garlic extract. As a result, Bordeaux mixture showed the highest control effect against CDM. AUDPC of Bordeaux mixture, resistant cultivar, lime sulfur, garlic extract and untreated control was 3.9, 10.6, 95.6, 24.9, and 258.7, respectively. Based on the above-mentioned results, we think that the resistant cultivars and Bordeaux mixture can be effectively used to control CDM as one of control measures under the farmhouse condition.
Rhizopus soft rot of lily (Lilium longiflorum) caused by Rhizopus oryzae was observed in the experimental field in Taean Lily Experiment Station in Korea, 2012. The typical symptoms were water-soaked lesions on bottom stem and leaf rot. The lesion rapidly expanded and the plant was softened totally. The fungus grew vigorously at an optimum temperature ($25^{\circ}C$) and brownish colony and black sporangia were formed on potato dextrose agar medium. Sporangiophores formed on end of sporangia were sub-globose, brownish and $6-10{\mu}m$ in size. Sporangia were globose, blackish and $87-116{\mu}m$ in size. Sporangiospores were irregularly oval and sub-globose, brownish $4-8{\mu}m$ in size. On the basis of mycological characteristics, analyzing sequences of internal transcribed spacer region of ribosomal DNA, and pathogenicity test on host plants, the causal fungus was identified as R. oryzae. This is the first report of Rhizopus soft rot on lily caused by R. oryzae in Korea.

This study was performed to develop a new natural antimicrobial materials by analyzing the effect of extracts obtained from Ten Lauraceae Species on the inhibitory activity against Propionibacterium acnes. The plant materials were collected from Wando and Jeju islands, and the antimicrobial activity of the crude extracts was examined by the agar diffusion method with different part (i.e., leaf and branch), solvents (i.e., distilled water, 80% ethanol, and 100% methanol) and at different ultrasonic extracting times (i.e., 15, 30, and 45 minutes). The control agents used were synthetic antimicrobials, methylparaben and phenoxyethanol, at concentrations of 0.4, 1, 2, and 4 mg/disc. Altogether, extracts of 10 species used in the study showed inhibitory activity, which confirmed their antimicrobial action against acnes. Among these, leaves of Laurus nobilis L. which was extracted in 80% ethanol for 45 min showed the largest clear zone (19.8 mm). Leaves of L. nobilis L., showing highest antimicrobial activities among 10 species, were successively reextracted with n-hexane, chloroform, ethylacetate and n-butanol. As a results, in all fractions except butanol, clear zone above 10 mm were formed. The ethyl acetate fraction showed the highest inhibitory activity (13.3 mm) and the inhibitory activity was significantly higher than that of crude extract (10.2 mm) and phenoxyethanol as a control (12.5 mm).
In order to develop a disease-resistant root stock for the growth of watermelon, an efficient regeneration system of the gourd(Lagenaria leucantha Duch.) inbred line GO701-2 via organogenesis was established in this experiment. Using proximal parts of cotyledon explant excised from germinated seedling in vitro, maximum adventitious shoot formation (39%) was achieved on MS medium where cytokinin (BA) and auxin (IAA) were added at a concentration of 3mg/L and 0.1mg/L, respectively. Roots of the elongated shoots were successfully formed on MS medium without adding any plant growth regulators. The cucumber CsGolS1 gene known as a resistance gene against biotic and abiotic stresses, was constructed into the binary vector pBI121 under the control of CaMV 35S promoter. When the gene was introduced into the genome of gourd by Agrobacterium-mediated transformation, putative transgenic plants were obtained with the transformation efficiency of approximately 20 percent.
This study aims to investigate if the properly use of pesticide in Ginseng farmland at Gangwon-do, Korea. Questionnaire included 36 questions such as control method for plant diseases, pests and weeds and pattern of pesticide use was answered by 271 Ginseng farmers lived in 4 cities and 8 counties at Gangwon-do. Ginseng farmers have noxious plant diseases, insects and weeds such as leaf spot, anthracnose, snail, stem-mining fly, horseweed and hairy crabgrass. To control of these, 35% of farmers relied on various type of pesticides and 31.5% of farmers used pesticides mingling with natural enemy, microorganism and organic materials. Farmers selected the pesticide based on the their own experiences or by recommendation of market dealers and neighbors, resulting in the use of inappropriate pesticides in the Ginseng farmland(78.8 % ). They followed standard dosage of the pesticide from Handbook of Pesticide Application(96.3%). They, however, used the same pesticides from 2 to 5 years(44.1 %). This consecutive use of pesticide could be induced resistance. This survey resulted that study for pesticide resistance and systematic educational program for proper use and selection of pesticide to Gingseng farmers should be conducted in farmland at Gangwon-do, Korea.

In 2006, pinewood nematode, Bursaphelenchus xylophilus, was isolated from about 50 years old trees of Pinus densiflora and P. koraiensis showing leaf-wilt and -drying symptoms in Gwangju, Gyeonggi-do and Chuncheon, Gangwon-do. Isolation of pinewood nematodes from sapwood of infected pine trees showed no difference in population density between tree species and among the sampling heights on the main stem. Migration of pinewood nematodes in the host tree were investigated by inoculation of red pine (P. densiflora, 3 years old) seedlings with B. xylophilus. The nematodes seemed to move in red pine seedlings prior to multiplication and it might have taken about 20 days to start multiplication and expression of symptoms including wilt and dieback. In initial time after inoculation, nematodes started migration through the cortical resin canal from inoculated site and further showed upward and downward movements. More nematodes were observed in cortical resin canal during early period of inoculation and later in resin canal of xylem and tracheid also while, the pith still remained free from nematode. The density of B. xylophilus was higher in seedlings of low-vigor with poor root growth than in seedlings of normal root growth. Seedlings showing high density of B. xylophilus exhibited stem discoloration and secondary infection by fungus at the inoculation site.
The new Camellia sinensis L. variety, "Hankwang" was selected from 630 clones of the tea plant clone garden in Bioenergy Crop Research Center, National Institute of Crop Science (NICS), RDA. This variety was selected to develop superior variety for high productivity and nutrient components from 2004-2008. We evaluated the clones by the characteristics of the leaf, nutrient components, and yield. "Hankwang" was finally selected from six regional trials from 2006-2008. The number of buds and weight of "Hankwang" were better than the control cultivar "Yabukata". "Hankwang" was observed to be more resistant to cold and disease compared to the control. The nutrient components of "Hankwang" such as tannin, catechin, total nitrogen, and caffeine were 16.7, 6.08, 2.32 and 2.00%, respectively.

Tomato crops are highly affected by tomato diseases, and if not prevented, a disease can cause severe losses for the agricultural economy. Therefore, there is a need for a system that quickly and accurately diagnoses various tomato diseases. In this paper, we propose a system that classifies nine diseases as well as healthy tomato plants by applying various pretrained deep learning-based CNN models trained on an ImageNet dataset. The tomato leaf image dataset obtained from PlantVillage is provided as input to ResNet, Xception, and DenseNet, which have deep learning-based CNN architectures. The proposed models were constructed by adding a top-level classifier to the basic CNN model, and they were trained by applying a 5-fold cross-validation strategy. All three of the proposed models were trained in two stages: transfer learning (which freezes the layers of the basic CNN model and then trains only the top-level classifiers), and fine-tuned learning (which sets the learning rate to a very small number and trains after unfreezing basic CNN layers). SGD, RMSprop, and Adam were applied as optimization algorithms. The experimental results show that the DenseNet CNN model to which the RMSprop algorithm was applied output the best results, with 98.63% accuracy.
In June 2019, Angelica acutiloba plants showing virus-like symptoms such as chlorotic local lesion and mosaic on the leaves were found in a greenhouse in Nonsan, South Korea. To identify the causal virus, we collected 6 symptomatic A. acutiloba leaf samples and performed reverse transcription polymerase chain reaction (RT-PCR) analysis using specific detection primers for three reported viruses including tomato spotted wilt virus (TSWV). RT-PCR results showed that five symptomatic samples were positive for TSWV. Mechanical sap inoculation of one of the collected TSWV isolate (TSWV-NS-AG28) induced yellowing, chlorosis and mosaic symptoms in A. acutiloba and necrotic local lesions and mosaic in Solanaceae species. Phylogenetic analysis based on the complete genome sequences showed that TSWV-NS-AG28 had a maximum nucleotide identity with TSWVNS-BB20 isolated from butterbur in Nonsan, South Korea. To our knowledge, this is the first report of TSWV infection in A. acutiloba.
In order to search potent antifungal substances from domestic plants, 40 plants cultivated in Korea were collected. After extracting with methanol (MeOH) and concentrating to dryness, the MeOH extracts were screened for in vivo antifungal activity against six plant diseases at a concentration of $2000{\mu}g/mL$. Fourteen extracts showed disease-controlling activity more than 90% against at least one of the 6 plant diseases tested; eight, seven, and three extracts controlled more than 90% the development of rice blast, tomato late blight, and wheat leaf rust, respectively. However, none of the extracts exhibited in vivo antifungal activity more than 90% against rice sheath blight, tomato gray mold, and barley powdery mildew. From the MeOH extracts of Angelica gigas and A. dahurica showing potent controlling activity against rice blast, 1 and 2 antifungal substances, respectively, were isolated by solvent partitioning and column chromatography. The three compounds were identified to be coumarins, namely, decursin, imperatorin, and isoimperatorin, by mass spectrometry and NMR spectroscopy. They were examined for in vitro and in vivo antifungal activities together with umbelliferone (7-bydroxycournarin) and scopoletin (6-methoxy-7-hydroxycoumarin) containing a free hydroxyl group at position 7 to investigate the structure-activity relationship. In vitro, most of 50% growth inhibitory concentrations ($IC_{50}$) were over $200{\mu}g/mL$, indicating that they have relatively weak antifungal activity. The antifungal activity of decursin and scopoletin, containing cyclic alkoxy groups instead of free hydroxyl group at position 7, was stronger than umbelliferone and scopoletin. Especially, decursin and imperatorin showed potent antifungal activities against Pythium ultimum and Magnaporthe grisea, respectively, with $IC_{50}$ values less than $25{\mu}g/mL$. In vivo, decursin and imperatorin showed potent antifungal activity against rice blast, whereas other coumarins hardly controlled the development of 6 plant diseases tested. These results suggest that the antifungal activity of 7-hydroxycoumarin derivative is substantially increased when the hydroxyl group at position 7 is protected by a stable cyclic alkoxy grouping.
