싸리나무에 있어서 수분공급량이 생장에 미치는 영향 (Influence of Different Soil Moisture on the Growth of Lespedeza bicolor)

  • For the evaluation of drought resistance of the plant, the growth of Lespedeza bicolor Turcz. var. japonica Nakai was analyzed by the control of water content of soil: 1. The growth of leaf, stem and root showed high value in accordance with the increase of soil water content. 2. The formation of nodule was alos increased as the content of soil water became higher. 3. The highest water content of the plant was shown in the plots of 30 and 40% of water centent in soils and in the middle of the growing period (August-September). 4. The C/F ratio in the early period of the growth(July) was similar in each plot, but showed a higher value as the water centent of soil became higher at the later period(October). 5. The T/R ration increased in early period of the growth as the soil water content became higher, but it was decreased to the value of 1 in each plot.

Effects of High Voltage pulse on Seed Germination and Plant Growth

  • It is generally known that electron beam has sterilization effects and can activate plant germination and growth. Compared to electron beam, electrical pulse has not been frequently studied with respect to the biological application. In this study, we have analyzed the effects of high voltage pulse on seed germination and growth using various plant species. We have used the high voltage generator for examining seed's responses to the high voltage pulse. The operating voltage and currents of the generator are about 300 kV and 30 kA, respectively. Pulse width is 60 ns. High voltage pulse has slightly activated germination and growth of radish during early stage. Various levels of germination and growth are observed in different plant species after treated with high voltage pulse.

Altered Expression of Pyrophosphate: Fructose-6-Phosphate 1-Phosphotransferase Affects the Growth of Transgenic Arabidopsis Plants

  • Pyrophosphate:fructose-6-phosphate 1-phosphotransferase (PFP) catalyzes the reversible interconversion of fructose-6-phosphate and fructose-1,6-bisphosphate, a key step in the regulation of the metabolic flux toward glycolysis or gluconeogenesis. To examine the role of PFP in plant growth, we have generated transgenic Arabidopsis plants that either overexpress or repress Arabidopsis PFP subunit genes. The overexpressing lines displayed increased PFP activity and slightly faster growth relative to wild type plants, although their photosynthetic activities and the levels of metabolites appeared not to have significantly changed. In contrast, the RNAi lines showed significantly retarded growth in parallel with the reduced PFP activity. Analysis of photosynthetic activity revealed that the growth retardation phenotype of the RNAi lines was accompanied by the reduced rates of $CO_2$ assimilation. Microarray analysis of our transgenic plants further revealed that the altered expression of $AtPFP{\beta}$ affects the expression of several genes involved in diverse physiological processes. Our current data thus suggest that PFP is important in carbohydrate metabolism and other cellular processes.

Spore Associated Bacteria (SAB) of Arbuscular Mycorrhizal Fungi (AMF) and Plant Growth Promoting Rhizobacteria (PGPR) Increase Nutrient Uptake and Plant Growth Under Stress Conditions

  • Microorganisms present in the rhizosphere soil plays a vital role in improving the plant growth and soil fertility. Many kinds of fertilizers including chemical and organic has been approached to improve the productivity. Though some of them showed significant improvement in yield, they failed to maintain the soil properties. Rather they negatively affected soil eventually, the land became unsuitable for agricultural. To overcome these problems, microorganisms have been used as effective alternative. For past few decades, plant growth promoting rhizobacteria (PGPR) and arbuscular mycorrhizal fungi (AMF) have been used as effective inoculants to enhance the plant growth and productivity. PGPR improves the plant growth and helps the plant to withstand biotic and abiotic stresses. AM fungi are known to colonize roots of plants and they increase the plant nutrient uptake. Spore associated bacteria (SAB) are attached to spore wall or hyphae and known to increase the AMF germination and root colonization but their mechanism of interaction is poorly known. Better understanding the interactions among AMF, SAB and PGPR are necessary to enhance the quality of inoculants as a biofertilizers. In this paper, current knowledge about the interactions between fungi and bacteria are reviewed and discussed about AMF spore associated bacteria.

Effect of Nitrogen and Silicon Nutrition on Bioactive Gibberellin and Growth of Rice under Field Conditions

  • Gibberellins are growth hormones that play a pivotal role in the growth and development of plants. Present investigations were carried to check the effect of nitrogen(N) and silicon(Si) on bioactive $GA_1$ and its immediate precursor $GA_{20}$ at different growth stages of two rice cultivars with different maturity traits. It was observed that the endogenous bioactive $GA_1$ level gradually increased during vegetative stage and anthesis stage of both Junghwabyeo(early flowering cultivar) and Daesanbyeo(late flowering cultivar). However, the $GA_1$ and $GA_{20}$ content start decreasing during the seed filling stage in both rice cultivars, which indicated a possible relationship of bioactive $GA_1$ and floral development. Our results also confirmed that early 13-hydroxylation pathway was operated at all developmental stages of rice plant. Variation in the levels of the endogenous gibberellins in rice shoots were measured by GCMS-SIM using $^2H_2$-labeled gibberellins as internal standards. Combined application of N and Si enhanced growth parameters and reduced lodging index of both rice cultivars. It was thus concluded that the level of physiologically active $GA_1$ increased during vegetative and early reproductive stage, but starts declining at seed filling stage.

  • 미생물에 의한 탄산칼슘 침전(MICP)은 생물학적 기술로 지반의 공학적 특성을 향상시키는 개량공법이다. 본 논문에서는 식물 생장에 MICP 약액의 농도가 미치는 영향을 분석하였다. 큰김의털 종자를 주문진 표준사로 채워진 식물 용기에서 생장시킨 다음에 표면 처리법으로 MICP 약액을 살포하였다. 실험 결과는 MICP 약액이 처리된 식물의 줄기 생장은 무처리 식물과 비교하여 억제되었다. MICP 약액으로 인한 토양의 화학적 성분 변화를 분석하기 위해 pH와 전기전도도를 측정하였으며, pH는 MICP 약액 처리와 상관 없이 모든 경우에서 pH 7에 가까운 중성상태를 보였지만, 전기전도도는 MICP 약액의 농도가 높을수록 증가하였다. MICP 처리 과정에서 발생한 이온들이 식물 뿌리 주변 토양의 삼투압을 증가시켜 식물 성장에 필요한 물과 양분의 흡수를 저해하였고, 궁극적으로 식물 생장을 억제하였기 때문이다.

Global Transcriptome Profiling of Xanthomonas oryzae pv. oryzae under in planta Growth and in vitro Culture Conditions

  • Xanthomonas oryzae pv. oryzae (Xoo), the causative agent of bacterial blight, is a major threat to rice productivity. Here, we performed RNA-Seq based transcriptomic analysis of Xoo transcripts isolated under in planta growth (on both susceptible and resistant hosts) and in vitro culture conditions. Our in planta extraction method resulted in successful enrichment of Xoo cells and provided RNA samples of high quality. A total of 4,619 differentially expressed genes were identified between in planta and in vitro growth conditions. The majority of the differentially expressed genes identified under in planta growth conditions were related to the nutrient transport, protease activity, stress tolerance, and pathogenicity. Among them, over 1,300 differentially expressed genes were determined to be secretory, including 184 putative type III effectors that may be involved in Xoo pathogenicity. Expression pattern of some of these identified genes were further validated by semi-quantitative RT-PCR. Taken together, these results provide a transcriptome overview of Xoo under in planta and in vitro growth conditions with a focus on its pathogenic processes, deepening our understanding of the behavior and pathogenicity of Xoo.

Physiological and Genetic Mechanisms for Nitrogen-Use Efficiency in Maize

  • Due to the strong influence of nitrogen(N) on plant productivity, a vast amount of N fertilizers is used to maximize crop yield. Over-use of N fertilizers leads to severe pollution of the environment, especially the aquatic ecosystem, as well as reducing farmer's income. Growing of N-efficient cultivars is an important prerequisite for integrated nutrient management strategies in both low- and high-input agriculture. Taking maize as a sample crop, this paper reviews the response of plants to low N stress, the physiological processes which may control N-use efficiency in low-N input conditions, and the genetic and molecular biological aspects of N-use efficiency. Since the harvest index(HI) of modern cultivars is quite high, further improvement of these cultivars to adapt to low N soils should aim to increase their capacity to accumulate N at low N levels. To achieve this goal, establishment and maintenance of a large root system during the growth period may be essential. To reduce the cost of N and carbon for root growth, a strong response of lateral root growth to nitrate-rich patches may be desired. Furthermore, a large proportion of N accumulated in roots at early growth stages should be remobilized for grain growth in the late filling stage to increase N-utilization efficiency. Some QTLs and genes related to maize yield as well as root traits have been identified. However, their significance in improving maize NUE at low N inputs in the field need to be elucidated.

옥상녹화에서 혼합식재에 따른 블루페스큐와 지피초화류의 생육 반응 (Effect of Companion Planting on Growth of Festuca glauca 'Elijah Blue' and Flowering Ground-cover Plants on Green Roofs)

  • This study was carried out to suggest an appropriate plant combination by evaluating the growth of flowering ground-cover plants planted with Festuca glauca 'Eljiah Blue' on the roof-top environment. As for the plant materials, Allium senescens and Chrysanthemum coreanum which are shorter than Festuca glauca 'Eljiah Blue' and Sedum takesimense and Agastache rugosa which are taller than Festuca glauca 'Eljiah Blue' were selected. Festuca glauca 'Eljiah Blue' was planted on Conrol, and Festuca glauca 'Eljiah Blue' with Allium senescens (T1), Festuca glauca 'Eljiah Blue' with Sedum takesimense (T2), Festuca glauca 'Eljiah Blue' with Agastache rugosa(T3), and Festuca glauca 'Eljiah Blue' with Chrysanthemum coreanum (T4) were planted in each experimental plot. Plant height and covering rate were measured to evaluate the growth of Festuca glauca 'Eljiah Blue'. Also, relative growth rate (RGR) of plant height, RGR of plant width, and mortality rate of the flowering ground-cover plants were estimated. Plant height and cover rate of Festuca glauca 'Eljiah Blue' was greatest in T3. RGR of plant height was greater in the order of Agastache rugosa, Allium senescens, Chrysanthemum coreanum, and Sedum takesimense. In particular, RGR of plant width was also greatest for Agastache rugosa. Mortality rates of Agastache rugosa and Allium senescens were lowest at 11%. Therefore, based on good growth of Festuca glauca 'Eljiah Blue' planted with Agastache rugosa, these results were suggested as a desirable combination of plant species for rooftop gardening.

Induction of Systemic Resistance against Bacterial Leaf Streak Disease and Growth Promotion in Rice Plant by Streptomyces shenzhenesis TKSC3 and Streptomyces sp. SS8

  • The genus Streptomyces demonstrates enormous promise in promoting plant growth and protecting plants against various pathogens. Single and consortium treatments of two selected Streptomyces strains (Streptomyces shenzhenensis TKSC3 and Streptomyces sp. SS8) were evaluated for their growth-promoting potential on rice, and biocontrol efficiency through induced systemic resistance (ISR) mediation against Xanthomonas oryzae pv. oryzicola (Xoc), the causal agent of rice bacterial leaf streak (BLS) disease. Seed bacterization by Streptomyces strains improved seed germination and vigor, relative to the untreated seed. Under greenhouse conditions, seed bacterization with consortium treatment TKSC3 + SS8 increased seed germination, root length, and dry weight by 20%, 23%, and 33%, respectively. Single and consortium Streptomyces treatments also successfully suppressed Xoc infection. The result was consistent with defense-related enzyme quantification wherein single and consortium Streptomyces treatments increased peroxidase (POX), polyphenol oxidase, phenylalanine ammonia-lyase, and β,1-3 glucanase (GLU) accumulation compared to untreated plant. Within all Streptomyces treatments, consortium treatment TKSC3 + SS8 showed the highest disease suppression efficiency (81.02%) and the lowest area under the disease progress curve value (95.79), making it the best to control BLS disease. Consortium treatment TKSC3 + SS8 induced the highest POX and GLU enzyme activities at 114.32 µmol/min/mg protein and 260.32 abs/min/mg protein, respectively, with both enzymes responsible for plant cell wall reinforcement and resistant interaction. Our results revealed that in addition to promoting plant growth, these Streptomyces strains also mediated ISR in rice plants, thereby, ensuring protection from BLS disease.
