Anti-oxidant and Anti-inflammatory Potentials of Sasa quelpaertensis Leaf Residue Extracts (제주조릿대 잎 잔사 추출물의 항산화 및 항염 활성)

  • Sasa quelpaertensis Nakai is a native Korean plant that grows only on Mt. Halla of Jeju Island. Its leaf is used for a popular bamboo tea due to its various health-promoting properties, and it has been increasingly used as food and cosmetic ingredients. To utilize the S. quelpaertensis leaf efficiently, the preparation method for phytochemical-rich extract (PRE) using the leaf's residue was previously reported, which was produced after hot water extraction. This study was undertaken to evaluate the anti-oxidant and anti-inflammatory potential of PRE and its solvent fractions. The ethyl acetate fraction of PRE (EPRE) showed higher DPPH, ABTS, and superoxide radical scavenging activities, and it effectively inhibited intracellular reactive oxygen species (ROS) and nitric oxide (NO) production in lipopolysaccharides (LPS)-stimulated RAW 264.7 cells. EPRE also induced the expression of heme oxygenase-1 (HO-1) by increasing the level of nuclear factor E2-related factor 2 (Nrf2) in a nuclear fraction. The inhibiting effect of EPRE on LPS-induced NO production was partially reversed by the HO-1 inhibitor (zinc protoporphyrin, ZPP), suggesting that HO-1 is involved in suppressing NO production. Taken together, the results suggest that EPRE has potential as a promising anti- oxidant and anti-inflammatory agent.

Genetic Organization of the hrp Genes Cluster in Erwinia pyrifoliae and Characterization of HR Active Domains in HrpNEp Protein by Mutational Analysis

  • The disease-specific (dsp) region and the hypersensitive response and pathogenicity (hrp) genes, including the hrpW, $hrpN_{Ep}$, and hrpC operons have previously been sequenced in Erwinia pyrifoliae WT3 [Shrestha et al. (2005a)]. In this study, the remaining hrp genes, including the hrpC, hrpA, hrpS, hrpXY, hrpL and hrpJ operons, were determined. The hrp genes cluster (ca. 38 kb) was comprised of eight transcriptional units and contained nine hrc (hrp conserved) genes. The genetic organization of the hrp/hrc genes and their orientation for the transcriptions were also similar to and collinear with those of E. amylovora, showing ${\geq}80%$ homologies. However, ORFU1 and ORFU2 of unknown functions, present between the hrpA and hrpS operons of E. amylovora, were absent in E. pyrifoliae. To determine the HR active domains, several proteins were prepared from truncated fragments of the N-terminal and the C-terminal regions of $HrpN_{Ep}$ protein of E. pyrifoliae. The proteins prepared from the N-terminal region elicited HR, but not from those of the C-terminal region indicating that HR active domains are located in only N-terminal region of the $HrpN_{Ep}$ protein. Two synthetic oligopeptides produced HR on tobacco confirming presence of two HR active domains in the $HrpN_{Ep}$. The HR positive N-terminal fragment ($HN{\Delta}C187$) was further narrowed down by deleting C-terminal amino acids and internal amino acids to investigate whether amino acid insertion region have role in faster and stronger HR activity in $HrpN_{Ep}$ than $HrpN_{Ea}$. The $HrpN_{Ep}$ mutant proteins $HN{\Delta}C187$ (D1AIR), $HN{\Delta}C187$ (D2AIR) and $HN{\Delta}C187$ (DM41) retained similar HR activation to that of wild-type $HrpN_{Ep}$. However, the $HrpN_{Ep}$ mutant protein $HN{\Delta}C187$ (D3AIR) lacking third amino acid insertion region (102 to 113 aa) reduced HR when compared to that of wild-type $HrpN_{Ep}$. Reduction in HR elicitation could not be observed when single amino acids at different positions were substituted at third amino acids insertion region. But, substitution of amino acids at L103R, L106K and L110R showed reduction in HR activity on tobacco suggesting their importance in activation of HR faster in the $HrpN_{Ep}$ although it requires further detailed analysis.

Changes in Myrosinase Activity and Total Glucosinolate Levels in Korean Chinese Cabbages by Salting Conditions (배추 절임조건에 따른 Myrosinase 활성 및 Total Glucosinolates 함량 변화)

  • Korean Chinese cabbage (Brassica campestris L. ssp. pekinensis) is one of the major cruciferous vegetables. Cruciferous vegetables contain a series of relatively unique secondary metabolites of amino acids called glucosinolates. Although glucosinolates do not appear to be bioactive, they are hydrolyzed by plant myrosinase when the cells in plants are damaged, and release biologically active compounds such as isothiocyanates, nitriles, and thiocyanates. The objective of this study was to determine the myrosinase activity and total glucosinolate levels of Korean Chinese cabbages by different salting times (0, 12, 18, and 24 h) and salt concentrations (6, 10, 14%). The total water content, salt content, and pH of brined cabbages decreased with increasing salting time. The myrosinase activity as determined by a glucose kit, decreased with increasing salting time and salt content. The total glucosinolates were purified using an anion exchange column and measured by UV-visible spectrophotometer. The fresh Korean Chinese cabbages contained $25.38{\pm}1.45\;{\mu}mol/g$ dry weight of glucosinolates. However, the total glucosinolates of brined cabbages decreased with increasing salting time and salt concentration. After 24 h of salting time, the total glucosinolates of brined cabbages rapidly decreased by $16.12{\pm}11.09$, $11.25{\pm}10.91$, $9.29{\pm}10.73\;{\mu}mol/g$ in 6%, 10%, and 14% salt solution, respectively. Overall, the total glucosinolate levels of Korean Chinese cabbages were found to vary inversely with salting time and salt concentration.

Isolation and Characterization of a Basic Leucine Zipper Gene in Poplar (Populus alba × P. glandulosa) (현사시나무에서 Basic Leucine Zipper 유전자의 분리와 특성 구명)

  • Basic leucine zipper (bZIP) protein is a regulatory transcription factor that plays crucial roles in growth, development and stress response of plant. In this study, we isolated a PagbZIP1 gene that belonged to Group SE3 of bZIP from Populus alba ${\times}$ P. glandulosa, and investigated its expressional characteristics. The PagbZIP1 is 844 base pairs long and encodes a putative 144-amino-acid protein with an expected molecular mass of 16.6 kDa. The PagbZIP1 has two conserved domains including the basic and leucine zipper portions. Southern blot analysis revealed that two copies of the gene are presented in the poplar genome. PagbZIP1 was specifically expressed in the root and suspension cells. Moreover, the expression of PagbZIP1 was induced by drought, salt, cold and ABA. Therefore, our results indicated that PagbZIP1 might be expressed in response to abiotic stress through the ABA-mediated signaling pathway in poplar.

The Comparison between Hot-Water Extracts and Microwave Extracts of Scutellaria radix for Antioxidant and Neuroprotective Effects (가열 추출법과 마이크로웨이브 추출법을 이용한 황금 물 추출물의 뇌세포 보호 및 항산화 효과 비교 연구)

  • Microwaves are non-ionizing electromagnetic waves of frequency between 300MHz to 300GHz and positioned between the X-ray and infrared rays in the electromagnetic spectrum. In recent years, the use of microwave for extraction of ingredient from plant material has shown remarkable research interest and potential. Scutellaria radix has been used as a traditional medicine for a variety of diseases. It has been reported to exert beneficial health effects, such as anti-bacterial, antiviral, anti-inflammatory, and free-radical scavenging. Oxidative stress or the accumulation of reactive oxygen species (ROS) leads neuronal cellular death and dysfunction, and it contributes to neuronal degenerative disease such as Alzheimer's disease, Parkinson's disease and stroke. In this study, we aimed to compare the neuroprotective and antioxidant effect of Scutellaria radix extracted by different methods using hot-water extraction (SBE-DW) or microwave extraction (SBE-DW-MW). As a result, we first examined HPLC analysis of hot-water and microwave extracts of Scutellaria radix. The hot-water and microwave extracts of Scutellaria radix showed the discernible difference patterns of HPLC analysis. Microwave-water extracts of Scutellaria radix increased DPPH radical scavenging activity more than hot-water extraction. Microwave-water extracts of Scutellaria radix also showed neuroprotective effects and ROS inhibition against glutamate-induced oxidative stress in mouse hippocampal HT22 cells, but hot-water extraction not showed. In addition, the phosphorylation of MAPKs induced by glutamate insult was prevented by microwave-water extracts of Scutellaria radix. Thus, these results suggested that microwave extraction can be utilized for improving the extraction efficiency and biological activity of Scutellaria radix.

Performance of Northern Exposure Index in Reducing Estimation Error for Daily Maximum Temperature over a Rugged Terrain (북향개방지수가 복잡지형의 일 최고기온 추정오차 저감에 미치는 영향)

  • The normalized difference in incident solar energy between a target surface and a level surface (overheating index, OHI) is useful in eliminating estimation error of site-specific maximum temperature in complex terrain. Due to the complexity in its calculation, however, an empirical proxy variable called northern exposure index (NEI) which combines slope and aspect has been used to estimate OHI based on empirical relationships between the two. An experiment with real-world landscape and temperature data was carried out to evaluate performance of the NEI - derived OHI (N-OHI) in reduction of spatial interpolation error for daily maximum temperature compared with that by the original OHI. We collected daily maximum temperature data from 7 sites in a mountainous watershed with a $149 km^2$ area and a 795m elevation range ($651{\sim}1,445m$) in Pyongchang, Kangwon province. Northern exposure index was calculated for the entire 166,050 grid cells constituting the watershed based on a 30-m digital elevation model. Daily OHI was calculated for the same watershed ana regressed to the variation of NEI. The regression equations were used to estimate N-OHI for 15th of each month. Deviations in daily maximum temperature at 7 sites from those measured at the nearby synoptic station were calculated from June 2006 to February 2007 and regressed to the N-OHI. The same procedure was repeated with the original OHI values. The ratio sum of square errors contributable by the N-OHI were 0.46 (winter), 0.24 (fall), and 0.01 (summer), while those by the original OHI were 0.52, 0.37 and 0.15, respectively.

  • In SS branch of Korea District Heating Corporation, Combined Heat & Power power plant with 99MW capacity and 98Gcal / h capacity is operated as a district electricity business. In this region, it is difficult to operate the generator due to the problem of surplus heat treatment between June and September due to the economic recession and the decrease in demand, so it is urgent to develop an economical energy new business model. In this study, we will develop an optimized operation model by introducing a renewable energy hybrid system based on actual operation data of this site. In particular, among renewable energy sources, fuel cell (Fuel Cell) power generation which can generate heat and electricity at the same time with limited location constraints, photovoltaic power generation which is representative renewable energy, ESS (Energy Storage System). HOMER (Hybrid Optimization of Multiple Energy Resources) program was used to select the optimal model. As a result of the economic analysis, 99MW CHP combined cycle power generation is the most economical in terms of net present cost (NPC), but 99MW CHP in terms of carbon emission trading and renewable energy certificate And 5MW fuel cells, and 521kW of solar power to supply electricity and heat than the supply of electricity and heat by 99MW CHP cogeneration power, it was shown that it is economically up to 247.5 billion won. we confirmed the results of the improvement of the zone electricity business condition by introducing the fuel cell and the renewable energy hybrid system as the optimization process model.

Ecological and Morphological Characteristics of Zostera caulescens Miki (Zosteraceae) in Korea (한국산 수거머리말(Zostera caulescens Miki.)의 형태 및 생태적 특성에 대한 연구)

  • Ecological characteristics on habitats and morphological features of the seagrass Zostera caulescens Miki in Korea were examined. The biogeographical distribution of Z. caulescens was confirmed in the south coast of the Korea. Zostera caulescens usually inhabits at the inner bay, where is sheltered from wave action and 3.0 to 6.5 m deep. The sediment in habitats is composed of very fine muddy sand or sandy mud. In morphology, Z. caulescens is easily distinguished from other species of the genus by the formation of vegetative canopy on the top of reproductive stems. The number of longitudinal ribs in testa was 24 revealed by scanning electron microscope (SEM) while the number of ribs in Z. marina has been often reported to be 16-20. Purplish anthocyanin spots were displayed on the surface when the testa was removed. We found that the size and shape of leaf epidermal cells in Z. caulescens were very different from those of Z. marina when the leaf cuticles were removed by maceration which could be useful characters for identifying Zostera species. The leaf of Z. caulescens displays two different casts of color when the fresh plant is closely observed. The margins of leaves appear brighter than the center of leaves due to the thickening by the development of paralleled venation in the middle of leaves. The comparison of two populations of Z. caulescens in Korea showed that they were considerably different in their shoot density and biomass. The shoot density and biomass at Gabae population was $367.3 m^{-2}\;and\;725.7g$ dry wt $m^P{-2}$ respectively, while those at Jukrim population were $112.5m^{-2}\;and\;392.0g\;dry\;wt\;m^{-2}$, respectively.

Ship Sewage Treatment Using Fixed Media Method (고정식 메디아법을 이용한 선박의 오폐수 처리)

  • The purpose of this study is to develop Sewage Treatment Plant that treat sewage which occurred in ship using fixed media method and to consider applicable to the Pilot Scale device of the STP regulations in MLTM(Ministry of Land, Transport and Maritime Affairs) and MEPC(Marine Environment Protection Committee). In test results, pH geometric mean was 7.68, $BOD_5$(Biochemical Oxygen Demand) geometric mean was 7.28 mg/l, $COD_{cr}$(Chemical Oxygen Demand) geometric mean was 48.39 mg/l, TSS(Total Suspended Solid) geometric mean was 18.00/l, Residual chlorine geometric mean was 0.19 mg/l, and E. coli geometric mean was 1CFU/100 ml. In addition, about 97.4% of $BOD_5$ was reduced, the $COD_{cr}$ reduction averaged 96.4%and the TSS reduction averaged 97.6%. STP have been determined by the MLTM and MEPC regulation of the marine pollution prevention equipment for performance testing product.

  • The methanol extract from the aerial parts of Aceriphyllum rossii was fractionated into ethyl acetate, n-BuOH and $H_2O$ layers through solvent fractionation. Repeated silica gel column chromatography of EtOAc and n-BuOH layers afforded five flavonol glycosides. They were identified as astragalin (1), kaempferol 3-O-${\alpha}$-L-rhamnopyranosyl (1${\rightarrow}$6)-${\beta}$-D-glucopyranoside (2), rutin (3), kaempferol 3-O-${\alpha}$-L-rhamnopyranosyl (1${\rightarrow}$4)-${\alpha}$-L-rhamnopyranosyl 1${\rightarrow}$6)-${\beta}$-D-glucopyranoside (4), and quercetin 3-O-${\alpha}$-L-rhamnopyranosyl (1${\rightarrow}$4)-${\alpha}$-L-rhamnopyranosyl (1${\rightarrow}$6)-${\beta}$-D-glucopyranoside (5) on the basis of spectroscopic data. All of them showed an inhibition in farnesyl protein tranferase (FPTase) activity, and rutin (3) inhibited the growth of rat H-ras cell and the cell migration of human umbilical vein endothelial cells (HUVECs).
