We isolated a novel methylotrophic bacterium from soil collected in Dongducheon Environment Affairs Agency. The isolate was identified as Methylobacterium sp. WJ4 based on phylogenetic analysis. Plackett-Burman design was employed for screening eight parameters of nitrate mineral salts (NMS) medium for cell growth of a newly isolated Methylobacterium sp. WJ4 with experimental validation. Trace element solution and vitamin stock were found to affect cell growth, which can be further optimized for increased cell growth. This is the first report of screening parameters of NMS medium which affect cell growth of strain belonging to the genus Methylobacterium using Plackett-Burman design.
The Plackett-Burman experimental design was employed to evaluate the relative importance of medium constituents of each medium for enhanced griseofulvin production by Penicillium griseofulvum MTCC 1898 and Penicillium griseofulvum MTCC 2004. It was found that the medium constituents, sucrose, $K_2HPO_4,\;NaNO_3,\;and\;FeSO_4{\cdot}7H_2O$, significantly influenced the griseofulvin production by Penicillium griseofulvum MTCC 1898. In the case of Penicillium griseofulvum MTCC 2004, lactose, glucose, and $MnSO_4{\cdot}H_2O$ significantly influenced the griseofulvin production.
This paper proposes statistically designed experiments which provide a proactive means to implement safety and environmental applications. Minimal experimental designs such as fractional factorial design, Plackett-Burman design, Box-Behnken design are economical and can be achieved tremendous savings with relatively few experiments. These experimental designs and analysis methods are illustrated with cases.
The Plackett-Burman design and the response surface method (RSM) with a central composite design (CCD) were employed to develop a culture medium for ansamitocin P-3 production using Actinosynnema pretiosum ATCC 31565. Among the 11 nutrients tested using the Plackett-Burman design, two carbon sources, sucrose and dextrin, and two nitrogen sources, polypeptone and yeast extract, were selected. Optimization of the concentrations of the selected nutrients was then performed using RSM with CCD. After two rounds of RSM, the optimum concentrations ($\%w/v$) of sucrose, dextrin, polypeptone, and yeast extract were identified as 4.5, 4.5, 0.16, and 0.89, respectively. The maximum ansamitocin P-3 titer was 45.2 mg/l with the optimized medium, which was about 6 times higher than that (7.315 mg/l) obtained with an $R_{2}YE$ medium before optimization.
Plackett-Burman (PB) and Box-Behnken (BB) experimental designs were used to optimize fermentation variables for the biotransformation of glycyrrhizin (GL) to monoglucuronyl-glycyrrhetinic acid (MGGA). The PB design was first used to screen the important factors among the medium variables. The steepest ascent method was used to approach the optimum range for each of these factors. The BB design was finally used to analyze the response surfaces of the screened factors for further optimization. The optimized conditions for this system were 0.7 g/L $MgSO_4{\cdot}7H_2O$, 1.19 g/L GL, and cultivation for six days. The biotransformation of GL to MGGA could reach up to 35.72%, which is a good result for this kind of transformation.

Non-regular designs such as the Plackett-Burman(PB) design have traditionally been used for screening only main effects because of complex aliasing. But it was found that these designs could be used to estimate the 2-factor interactions as well as main effects through the hidden projection property. The goal of this paper is to propose the estimable model when projecting the 12-run PB design using the algebraic geometric method. The core of this method considers the design as a affine variety and the Grbner basis of the design ideal for this affine variety gives the estimable polynomial models. As the results of applying the 12-run PB design it is actually found that this design has the models not only with 2-factor interactions but with 3-factor. This design is the maximal fan in 4-factor projection.
The Cr(VI) removal capability of Acinetobacter junii VITSUKMW2 isolated from the Sukinda chromite mine site was evaluated and enhanced using statistical design techniques. The removal capacity was evaluated at different pH values (5-11) and temperatures ($30-40^{\circ}C$) and with various carbon and nitrogen sources. Plackett-Burman design was used to select the operational parameters for bioremediation of Cr(VI). Three parameters (molasses, yeast extract, and Cr(VI) concentration) were chosen for further optimization using central composite design. The optimal combination of parameters was found to be 14.85 g/l molasses, 4.72 g/l yeast extract, and 54 mg/l initial Cr(VI), with 99.95% removal of Cr(VI) in 12 h. A. junii VITSUKMW2 was shown to have significant potential for removal of Cr(VI).
Optimization for the production of Pest an was followed by the Plackett-Burman Design, using modified Czapek-dox medium as the starting point. At the flask level, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$, and aeration variables positively affected the Pestan production, DCW (dry cell weight), apparent viscosity, and flocculating activity response. KCI and $FeSO_4{\cdot}7H_2O$ negatively affected the Pestan production, DCW, apparent viscosity, and flocculating activity response. Aeration variable was shown to have a positive effect on only the flocculating activity response among Pestan production, DCW, and apparent viscosity responses. In comparison of the positive and negative variables media conditions, Pestan production and flocculating activity differed by about 9 and 125 times, respectively. In particular, at the jar fermentor level, the aeration variable was the most important factor of the all responses (pestan production, DCW, apparent viscosity, flocculating activity, and anionic charge density). The flocculating activity and apparent viscosity of Pestan were closely related to the molecular chain length and charge density.
As large-scale computational applications in various scientific domains have been utilized over many integrated sets of grid computing resources, the difficulty of their execution management and control has been increased. It is beneficial to refer job history generated from many application executions, in order to identify application‘s characteristics and to decide selection policies of grid resource meaningfully. In this paper, we apply a statistical technique, Plackett-Burman design with fold-over (PBDF), for analyzing grid environments and execution history of applications. PBDF design identifies main factors in grid environments and applications, ranks based on how much they affect to their execution time. The effective factors are used for selecting reference job profiles and then preferable resource based on the reference profiles is chosen. An application is performed on the selected resource and its execution result is added to job history. Factor's credit is adjusted according to the actual execution time. For a proof-of-concept, we analyzed job history from an aerospace research grid system to get characteristics of grid resource and applications. We built JARS algorithm and simulated the algorithm with the analyzed job history. The simulation result shows good reliability and considerable performance in grid environment with frequently crashed resources.

Leuconostoc citreum is one of the representative strains of Leuconostoc spp. that show fast growth rates in fermented vegetables. Sequential experimental designs including the Plackett-Burman design, fractional factorial design, steepest ascent analysis, central composite design and response surface methodology were introduced tooptimize and improve the medium for Leuconostoc citreum. Fifteen medium ingredients were examined and glucose (20 g/l), yeast extract (12.5 g/l), sodium acetate trihydrate (6.12 g/l), potassium phosphate (42.55 g/l) and dibasic ammonium citrate (4.12 g/l)were chosen as the best components to give a critical and positive effect for cell-growth. The biomass was increased to 2.79 g/l (169%), compared to the 1.65 g/l in MRS medium.
