Quality standard of Agaricus bisporus in a market and changes of mushroom quality during storage (유통 양송이버섯의 등급별 품질규격 및 저장기간별 품질 변화)

  • This study was carried out to improve standardization of agricultural products and investigate quality changes during preservation at various treatments. The standardization does much to improve merchantable quality, distribution efficiency and fair dealings by shipping of the standard agricultural products. Mushrooms notified as the standard are five; Pleurotus ostreatus, Pleurotus eryngii, Flammulina velutipes, Agaricus bisporus and Ganoderma lucidum. But many farmers are suffering from strict standards. Therefore, modification of these standards is required to fit farmhouse situations. Fruit-body weight of A, B and C grades of the marketed Agaricus bisporus was 19.3g, 12.2g and 10.4, respectively. Average pileus diameter of A grades was 43.3mm but B and C grades did not show significant difference. Average stipe length of A, B and C grades was 9.8mm, 13.1mm and 11.6mm, and stipes thick was the highest in A grades. The weight loss rate of stipe cutting of Agaricus bisporus was 12.6% in harvested mushrooms and the rate of stipe cutting length was 60.8%. The diameter of Agaricus bisporus stored for 5 days was 3.5~4.5cm but decreased rapidly in 15 days. The weight loss rate of the high temperature samples lowered rapidly than that of the low temperature samples. Conversely, the L value of the low temperature samples was higher than that of the high temperature samples.

Artificial Cultivation of Tricholoma giganteum Collected in Korea (I) - Morphological Charateristics of Fruitbody and Environmental Condition in Habitat of T. giganteum - (한국산 왕송이버섯의 인공재배 (I) - 자실체 형태적 특징과 발생지 환경조사 -)

  • The result of study on the morphological feature of fruitbody, soil characteristics and enviromental condition of habitat of Tricholoma giganteum in Korea was as follows: The Pileus was $5.5{\sim}28.0\;cm$ in diameter and $1.5{\sim}3.7\;cm$ in thickness, indicating of yellowish white, beige, or ivory color. The lamellae were $18{\sim}20\;mm$ in diameter; the stipe was $9.0{\sim}35.7\;cm$ long and $1.0{\sim}3.0\;cm$ wide. The spores were $3.5{\sim}4.8{\times}5.7{\sim}7.4\;{\mu}m$ in size, ellipsoid, hyaline and the spore deposit white. The basidia were $6.1{\sim}7.0{\times}32.2{\sim}39.2\;{\mu}m$ in size. Cheiloystidia were $3.5{\sim}4.4{\times}30.5{\sim}33.1\;{\mu}m$ in size. Pileipellis were $3.3{\sim}4.4{\times}33.0{\sim}55.0\;{\mu}m$ wide and had clamp connection. Stipitipellis were $2.2{\sim}3.3{\times}88.0{\sim}93.1\;{\mu}m$ in size. Texture of soil in habitate of T. giganteum was silty loam, and organic materials content and available phosphate content in this soil were high as comparision to general soil in Korea. In the time of the mushrooms sprout, the room temperature indicated $25{\sim}27^{\circ}C$, their humidity showed $80{\sim}83%$, and the brightnesses were 328 Lux.

Estimated Gas Concentrations of MA(Modified Atmosphere) and Changes of Quality Characteristics during the MA Storage on the Oyster Mushrooms (느타리버섯의 환경기체조성 농도 예측 및 MA 저장 중 품질특성 변화)

  • This study was conducted to find out effective MA (Modified Atmosphere) gas compositions on the oyster mushroom through statistical analysis of the respiration rate and MA storage for the various packaging materials. Under the various gas compositions, the oxygen consumption rate of oyster mushroom was from 28.9 to 161.4mgO$_2$/kg$.$hr and the carbon dioxide evolution rate was from 53.4 to 166.9 mgCO$_2$/kg$.$hr at 20$^{\circ}C$. The estimated MA condition of oyster mushroom were 2.5∼4.5%O$_2$and 11.5∼l3%CO$_2$by the RSREG(Response Surface Regression). The gas compositions of MA packaging are following that 0.03mm LDPE were 1.6∼3.0%O$_2$and 3.9∼5.3%CO$_2$,0.05mm LDPE were 1.2∼1.3%O$_2$and 9.0∼11.1%CO$_2$and Nylon+PE were 0.9∼1.2%O$_2$and 33.5∼39.6%CO$_2$. The weight loss increased at 0.03mm LDPE but has the lowest value at Nylon+PE. The hardness of pileus and stipe was decreased with storage periods. The $\Delta$E-value increased with storage period and seriously changed in early storage period at 12 and 20$^{\circ}C$. In the 0.05mm LDPE, the gas compositions of packaging were similar to estimated gas compositions from the RSREG and the storage quality was superior to the other packaging materials in weight loss, hardness, and color difference at 4, 12 and 20 $^{\circ}C$.

Characterization of a new commercial strain 'Mongdol' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 느타리 신품종 '몽돌'의 육성 및 그 특성)

  • A new commercial strain "Mongdol" of oyster mushroom was developed by hyphal anastomosis. It was improved with hybridization between monokaryotic strain derived from Pleurotus ostreatus ASI 0627 and dikaryotic strain derived from P. ostreatus ASI 2929. The optimum temperature of mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $12{\sim}18^{\circ}C$, respectively. When two different media including PDA (potato dextrose agar medium) and MCM (mushroom complete medium) were compared, the mycelial growth of this mushroom was faster in MCM than in PDA. Similar result was observed with the control strain P. ostreatus ASI 2504. Analysis of the genetic characteristics of the new cultivar "Mongdol" showed a different DNA profile as that of the control strain ASI 2504, when RAPD (Random Amplified Polymorphic DNA) primer URP3 and URP6 were used. Fruiting body production per bottle was about 106 g using demonstration farms. The color of pileus was blackish gray and the stipe was long. Therefore, we expect that this new strain "Mongdol" will satisfy the consumer's demand for high quality mushrooms.

Quality Changes in Oyster Mushrooms during Modified Atmosphere Storage as Affected by Temperatures and Packaging Materials (저장 온도와 포장재에 따른 느타리버섯의 MA 저장 중 품질변화)

  • Modified atmosphere packaging was applied to oyster mushrooms (Pleurotus ostreatus) to study the effect of storage temperatures and packaging materialso. Whole mushrooms (200g) were package with polyethylene film $(PE,\;60{\mu}m\;thickness)$, ethylene vinyl acetate (EVA), or ceramic film (containing 5% zeolite) and stored at 0, 5, 10 and $20^{\circ}C$. Weight loss, color, firmness, gas composition $(O_2,\;CO_2)$ inside the film package and ethanol content in the tissue of MA packaged mushrooms were examined. Mushroom that were packed unwrapped in a conventional hardboard box (2 kg) lost marketability at a very early stage of storage due to weight loss, shrinkage, browning, and spore formation. During storage, film packaging prevented or retarded the deterioration of the mushrooms in the aspects of appearance, texture, and discoloration. Firmness slightly decreased with storage time. Total color difference was much higher in the control than in the film-packaged mushroom and rapidly increased at the early of storage. Correlation analysis showed a high correlation between total color difference and b values. These results were characterized by the reduced respiration rate resulting from elevated carbon dioxide and reduced oxygen levels in the package. At all storage temperatures, ethanol content in the tissue increased slightly at the early part of storage and rose considerably towards the end of the storage period. Ethanol content in the oyster mushrooms was higher in the stipe than in pileus tissues. The shelf life of the oyster mushrooms was about $8{\sim}11$ days at $0^{\circ}C$, about $4{\sim}6$ day at $5^{\circ}C$, about $2{\sim}3$ days at $10^{\circ}C$, and about $1{\sim}2$ days at $20^{\circ}C$.

Analysis of growth environment of Flammulina velutipes using the smart farm cultivation technology (병재배 팽이버섯의 스마트팜 재배를 통한 생육환경 분석)

  • In this study, smart farm technology was used by farmers cultivating 'CHIKUMASSHU T-011' in order to develop an optimal growth model for the precision cultivation of bottle-grown winter mushroom and the results of the same are mentioned herein. Farmers participating in the experiment used 60 ㎡ of bed area with 4 rows and 13 columns of shelf shape, 20 horsepower refrigerator, 100T of sandwich panel for insulation, 6 ultrasonic humidifiers, 12 kW of heating, and 20,000 bottles of Flammulina velutipes mushroom spores. The temperature, humidity, and carbon dioxide concentrations, which directly affect the growth of the mushroom, were collected and analyzed from the environmental sensors installed at the winter mushroom cultivation area. The initial temperature was found to be 14.5℃, which was maintained at 14℃ to 15℃ until the 10th day. In the restriction phase, the initial temperature was 4℃ and was maintained between 2℃ and 3℃ until the 15th day, while during the growth phase, it was maintained between 7.5℃ to 9.5℃. Analysis of the humidity data revealed initial humidity to be 100%, which varied between 88% to 98% during primordia formation period. The humidity remained between 77% to 96% until the 15th day, in the restriction phase and between 75% to 83% during the growth phase. The initial carbon dioxide concentration was 3,500 ppm and varied between 3,500 ppm to 6,000 ppm during primordia formation period and was maintained at 6,000 ppm until the 15th day. During the growth phase, the carbon dioxide concentration was found to be over 6,000 ppm. Fruiting body characteristics of 'CHIKUMASSHU T-011' cultivated in the farmhouse were as follows: Pileus diameter of 7.5 mm and thickness of 4.1 mm, stipe thickness of 3.3 mm, and length of 154.2 mm. The number of valid fruiting bodies was 1,048 unit per 1,400 mL bottle, and the individual weight was 0.71 g per unit. The yield of fruiting bodies was 402.8 g per 1,400 mL bottle.

Cultivation of Oyster Mushroom[Pleurotus ostreatus (Jacq. ex Fr.) Kummer] at Medium with Ca(OH)2 Treatment (수산화칼슘[Ca(OH)2] 처리 배지를 이용한 느타리버섯의 재배)

  • This research is aimed to investigate the cultivation method of oyster mushroom[Pleurotus ostreatus (Jacq. ex Fr.) Kummer] at the non-sterilized medium with $Ca(OH)_2$ treatment. Therefore, experiments were carried out to develop non-sterilization method of medium by addition of $Ca(OH)_2$ for omission of heat sterilization progress of medium. General components, minerals and amino acid in Jiri wild type No. 1 (Pleurotus ostreatus) and production cost were analyzed. For the purpose of omission of heat sterilization progress, treatment ratio of $Ca(OH)_2$ (purity 95%) was 5%(w/w) of dry medium. Initial pH of this medium was 11 and then the pH was changed by 9 after the uniform mixing of the medium for half an hour. The various germs occurred 50% and 100% at pH 8 and pH 7 of the non-sterilized medium, respectively. Production of oyster mushroom increased by $2,030\;ton\;ha^{-1}$ when the main raw material used corn pith instead of waste cotton. The time required of mycelium culture was 30 days when hypha was cultured at the non-sterilized medium, and pinhead occurred when 2 or 3 days was passed after the time required of mycelium culture. Occurrence of pinhead was most rapid at the condition of $22{\sim}26^{\circ}C$, 65% humidity and pH $6.5{\sim}7.0$ and required of $22{\sim}28$ days at $70{\sim}80\;mm$ thickness of non-sterilized medium. Ca content in 1st harvest oyster mushroom was higher than that in 2nd harvest one, and its difference was $30.3\;mg\;kg^{-1}$. Amino acid content by stipe thickness of oyster mushroom was ranged from 411.2 to $343.9\;mg\;kg^{-1}$ both in a pileus and a stipe of 1st harvest mushroom, and from 402.4 to $498.2\;mg\;kg^{-1}$ and from 442.6 to $470.4\;mg\;kg^{-1}$ in those of 2nd harvest one, respectively. The results of the present study suggest that the non-sterilization medium by addition of $Ca(OH)_2$ is usable with the cultivation of oyster mushroom.

Characterization of a new commercial strain 'Goni' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 백색느타리 품종 '고니'의 육성 및 그 특성)

  • To develop new white variety of Oyster mushroom, all white varieties which have been collected and kept in the lab were revived and screened their cultural characteristics. 84 intra-specific Oyster mushroom hybrids between the white-colored mutants Suhan and Wonhyeong were developed using hyphal anastomosis technique in 2007. The Po2007-63 ($2842-7{\times}0205-7$) was shown the best cultural characteristics, selected to be a new variety and named as 'Goni'. The new commercial strain, 'Goni' has white pilei and grows well under spring and autumn conditions in Korea. The fruiting bodies of 'Goni' are of an excellent quality in that not only the stipe is thick and long but also the pileus is small and hard. The optimum temperatures for mycelial growth and fruiting body development were $25-30^{\circ}C$ and $10-16^{\circ}C$, respectively. Time period required for the initiation of the first fruiting body is about 3 to 5 days depending on the temperatures. The shape of fruiting body is thin funnel shape. Fruiting body production per bottle was about $91{\pm}13$ g which is almost 97% quantity compared to that of other variety 'Miso'. Relatively low temperature incubation ($11^{\circ}C$) resulted in the development of better quality of 'Goni' mushrooms. When two different media including potato dextrose medium and mushroom complete medium were compared, the growth of mushroom were much faster in mushroom complete medium at $20-25^{\circ}C$, but not at $25^{\circ}C$. Similar results were observed with other variety 'Miso'. Analysis of the genetic characteristics of the new commercial strain 'Goni' showed a major DNA profile as that of the parental Suhan when primer URP 1 was used, but different to 'Miso' that was used as a control. When screens were performed with primer URP 2, DNA patterns were similar both to that of the parents and 'Miso'. This new variety of the white Oyster mushroom has a clean and fresh image that corresponds well to "health food". We therefore expect that this new strain will satisfy the consumers demand for variety and excellent mushrooms.

Studies on the development of mushroom mediums of Pleurotus eryngii using ginko leaf pomace (은행잎박을 이용한 큰느타리버섯 배지 개발)

  • Ten to thirty percent of ginko leaf pomace(GLP) were added to pine sawdust to investigate mycelial growth. Mycelial growth on the medium with GLP was 86~101(mm/28 days) and was slower than that of the control without GLP as, 102(mm/28 days). Mycelial growth time on medium of sawdust with 10 to 20% GLP was similar to the control without GLP as 29 days but that on medium with 30% GLP was delayed for 2 days. The time of pinhead formations of sawdust with 10 to 20% GLP was 7 days, the growth time to harvest was 11 days. These results were similar to those of the control without GLP. The sizes of pileus of treatments with 10 to 30% GLP were tend to be smaller compared with the control. The length of stipe of 10% GLP treatment was 98mm which was longer than that of the control with 91mm. The thickness of stipes of 10 to 30% GLP treatments were 45 to 48mm which were tend to be thicker than that of the control with 45mm. The hardness of stipe were 35,062($g/cm^2$) to 4,1065($g/cm^2$)) which were harder than that of the control with 32,156($g/cm^2$)). GLP treatments seem to be favorable to store or distribute. Yields per volume of GLP added treatments were 88.7g to 95.6g, which was increased 3 to 12% compare to 85.8g of the control without GLP.

Characteristics and breeding of a new multi-generation oyster mushroom (Pleurotus ostreatus) variety 'Dagul' (다발성 신품종 느타리 '다굴'의 육성 및 자실체 특성)

  • To develop a new variety of oyster mushroom(Pleurotus ostreatus), parental strains was selected by the method of Mon-Mon crossing between monokaryotic strains derived from ASI 2596(Suhan No.3) and ASI 2782(Black pileus mutant). The SB-73(ASI 2596-11 x 2782-8) was shown the best cultural characteristics, selected to be a new variety and named as 'Dagul'. The 'Dagul' was formed incompatibility line distinctly in the confrontation growth of parental strains Suhan No.3 and ASI 2782. The optimum temperature for mycelial growth, fruiting body development and pH arrange were $25{\sim}30^{\circ}C$, $14{\sim}17^{\circ}C$ and pH5~8, respectively. Fruiting body production per bottle was about $68.0{\pm}24.1$ g which is almost 115% quantity compared to that of other variety Suhan No.3. And also the stipe is long and individual generation is multiple. Analysis of the genetic characteristics of the new variety 'Dagul' showed different DNA bands as that of the control strains, Suhan No.3 and ASI 2782, when RAPD(Random Amplified Polymorphic DNA) primers URP7 and Rcb1 were used. This new variety 'Dagul' of oyster mushroom is characterized by multiple of individual generation and the stipe is long. We therefore expect that this new strain will increase of the income by cultivation of field.
