Assessment of Organic Compound and Bioassay in Soil Using Pharmaceutical Byproduct and Cosmetic Industry Wastewater Sludge as Raw Materials of Compost (제약업종 부산물 및 화장품 제조업 폐수처리오니 처리토양에 대한 유기화합물 및 Bioassay 분석 평가)

  • This study was conducted to assessment organic compound and bioassay (density of inhabited animal, fluctuation of predominant fungi, and survival ratio of earthworm) for finding damage on red pepper by heavily amount application of sludges in soil, which was treated with 3 pharmaceutical byproducts and a cosmetic industry wastewater sludge as raw materials of compost, and for establishing estimation method. HEM contents in the soil treated with pharmaceutical byproducts sludge2 (PS2) and cosmetic sludge (CS) were 0.51, 1.10 mg/kg respectively. PAHs content of PS2 treatment in the soil was 3406.8 ug/kg on July 8. In abundance of soil faunas, the pharmaceutical byproducts sludge2 treatment was the most highest. The next was decreased in the order of pig manure (PM) and the cosmetic sludge treatment. However the other pharmaceutical sludge treatments were remarkably reduced populations of soil inhabited animals. In upland soil treated with organic sludges, the numbers of bacteria and fungi of the pharmaceutical sludge treatment were 736, 909 cfu/g and those of the cosmetic sludge treatment were 440, 236 cfu/g, respectively. The pharmaceutical sludge treatments and the cosmetic sludge treatment in identification of predominant bacteria were not any tendency to compare with non fertilizer and pig manure treatments, but they had diverse bacteria than NPK treatment. In microcosm tests, the survival of the tiger earthworm in five soil samples was hardly affected against the soil of PSI (20%) after three months treated in the upland But after six months, survival of PS1 was 80%. At present, raw material of compost was authorized by contents of organic matter, heavy metal (8 elements), and product processing according to 'The specified gist on possible materials of using after analysis and investigation among raw materials of compost', however, for preparing to change regulation of raw material of compost and for considering to possibility of application, this study was conducted to investigate toxic organic compound and bioassay methods using inhabited animal, fungi, and earthworm without current regulation.

Effect of Temperature Condition on Nitrogen Mineralization of Organic Matter and Soil Microbial Community Structure in non-Volcanic Ash Soil (온도가 유기물의 질소무기화와 미생물 군집구조에 미치는 영향)

  • This study was carried out to evaluate effect of temperature condition on nitrogen mineralization of organic matter, distribution of microbial group by PLFA profiles, and soil microbial community in non-volcanic ash soil. Dried soil 30 g mixed well each 2 g of pellet (OFPE) organic fertilizers, pig manure compost (PMC), and food waste compost (FWC). And then had incubated at $10^{\circ}C$, $20^{\circ}C$, and $30^{\circ}C$, respectively. Nitrogen mineralization rate increased with increasing temperature and that was in the order of FWC>OFPE>PMC. Distribution ratio of microbial group by PLFA profiles showed that was different significantly according to incubation temperature and the type of organic matter. As incubating time passed, density of microbial group decreased gradually. The Gram-bacteria PLFA/Gram+ bacteria PLFA, Fungi PLFA/Bacteria PLFA, and Unsaturated PLFA/saturated PLFA ratios were decreased according to the increasing temperature gradually. Principal component analysis using PLFA profiles showed that microbial community structures were composed differently by temperature factor at both 75 days ($10^{\circ}C$) and 270 days ($30^{\circ}C$). In conclusion, Soil microbial community structure showed relative sensitivity and seasonal changes as affected by temperature and organic matter type.

  • This research was carried out to investigate the moisture evaporation and property change depenging on the composting temperature for the escalator reversing composting system. The system was supported by the Y livestock cooperative composting factory. According to the test, the composting remperature eas varied in the range of $65\~80^{\circ}C$. The water content was de-creased to $75\%$ and became about $60\%$ at the end of the phase. Total nitrogen became $0.58\%$, total phosphoric acid $1.47\%$ and potashium $0.49\%$ at the end of the phase.

Unit Loading Factor of Nitrogen and Phosphorus for Controlling Eutrophication of Youngsan Lake (영산호의 부영양화방지를 위한 질소, 인의 배출원단위에 관한 조사연구)

  • In order to estimate unit loading factors of N, and P for controlling eutrophication of Youngsan lake. This study was performed in 4 kinds of pollutant sources from domestic sewage, industrial waste water, livestock stall waste water and drainage of agricultural area during the period from april to october 1988. These results were as follows: 1. The sewage flow for domestic waste water was 191.2 l/capita, day and that of the gray and toilet waste water among the domestic waste water were shown 152.9 l/capita, day(80%) and 38.3 l/capita, day(20%), respectively. 2. The unit loading factor total nitrogen(T-N) for domestic waste water was 7.582g/capita, day, and that of the gray and toilet waste water among the domestic waste water were 1.826g/ capita, day(24.1%) and 5.756g/capita, day(75.9%), respectively. The other hand, the unit loading factors of total phosphorus(T-P) for domestic waste water was 0.925g/capita, day, and that of gray and toilet waste water among the domestic waste water were 0.470g/capita, day(50.8%) and 0.455g/capita, day(49.2%), respectively. 3. In offering Price per million won, the T-P loading factor for drinking manufacture, confectionery manufacture, beer-manufacture and fibre manufacture in the industrial pollutant sources estimate to be 0.350g/day, 0.099g/day, 32.351g/day and 1.536g/day, while T-N loading factor about them in the industrial pollutant sources estimate to be 4.117g/day, 2.414g/day, 106.726g/day and 60.504g/day, respectively. 4. The T-P loading factor according to wash-water of milch cow and pig were 6.735g/day and 18.526g/day, in case of T-N they were 42.397g/day and 27.226g/day, respectively. 5. The T-P loading factor for pollutants drainage in the Paddy fields, fields and forests area were 0.082g/are, day, 0.014g/are, day and 0.002g/are, day, and the T-N loading factor were 0.309g/are, day, 0.158g/are, day and 0.064g/are, day, respectively. The diffrent of the loading factor for pollutants discharges in the agricultural area were resulted from the rainful intensity, the rainful, the amount of fertilization manure, and etc.

  • The application of sludge wastes into agricultural fields has been increasing annually in Korea. In particular, sewage sludge application has been widely accepted in decades. Sewage sludge application aid in the recycling of essential nutrients and act as a source of organic matter improving the structure and water-holding properties of the soil. The efficient use of sludge wastes, however, requires an individual assessment of waste products. This study assessed the biological characteristics of organic waste-treated lysimeter soils and develop its indicator to assess the soil health of organic waste-treated lysimeter soils. Several analytical techniques more recently developed such as restriction fragment length polymorphism (RFLP), phospholipid fatty acid (PLFA), and community level substrate utilization (CLSU) fingerprints allow for detailed analyses of soil microbial communities. PLFA and RFLP was, therefore, used in the study to characterize the microbial communities in soil without the need to isolate individual fungi and bacteria. PLFA, RFLP and CLSU have been utilized to assess microbial characteristics of the lysimeter soils with four different sludge wastes for eight consecutive years. Each of these methods was analyzed for a different aspect of soil microbial characteristics. The study would disclose those methods yielded highly reproductive results for each soil and allow distinguishing the soils based on the structures of specific geneand PLFA-pools more than CLSU fingerprints. PLFA methods, especially, revealed the same relative similarities of the treated soils based on cluster analysis of the biological characteristics. Pig manure compost-treated soil, however, was only the same relative resemblance among the three methods. These results indicated that PLFA easily assessed the biological soil characterization.

Optimal Mixing Ratio of Wastewater from Food Waste and Cattle Manure and Hygienic Aspect in Batch Type Anaerobic Digestion (음식물폐수와 축산분뇨의 혼합소화에서 적정 혼합비 및 소화슬러지의 위생성 연구)

  • This research was performed to figure out the optimal mixing ratio of food- to livestock wastewater for the best degradation of organic matter in the anaerobic digestion. The presence of various microorganisms, such as Escherichia coli and Staphylococcus aureus, was also investigated in both wastewater in this process. Enteric bacteria were only found in livestock wastewater, whereas pathogenic bacteria like S. aureus were detected in both wastewater. The optimal mixing ratio of food- to livestock wastewater for the best mineralization was found to fifty to fifty, with reduction ratios of $BOD_5$, CODcr SS as 23.2%, 24.7%, 19.7%, respectively. Hygiene of the digested sludge was also analyzed by counting the number of total colonies and various pathogens. Enterobacteriaceae including E. coli were barely detected in 10 days after reaction. Meanwhile, S. aureus was gradually reduced during reaction, even showing 1,000~5,000 CFU/mL in final days.

  • Application of sludge wastes into the field may help soil fertility with physical, chemical and biological properties. Efficient use of sludge waste, however, requires an individual assessment of the waste products. A lot of experiment into the organic waste-treated soils has been done for decade. However, studies have not been carried out on the assessment of agricultural soil by Basidiomycota. This study was assessed the influence of sludge application on soil healthy in agricultural upland soils. The organic wastes selected for long-term application experiment in this study were municipal sewage sludge (MSS), industrial sewage sludge (ISS), leather processing sludge (LS), alcohol fermentation processing sludge (FS), and pig manure compost (PMC). To develop the soil healthy assesment method, soil samples were diluted by 20X with distilled water. After shaking at 200rpm for 30 minutes, the shaked sample was mixed on PDA(Potato Dextrose Agar). And sterilized at $121^{\circ}C$ for 20 minutes. Coriolus hirsutus (MKACC 50560) was inoculated on petri-dish including PDA mixed sample. After the media was incubated at $25^{\circ}C$ for five days, the mycelial growth of C. hirsutus was measured. When the mycelial growth on sample media was compared with growth on media contained PDA only, well grown media contained sample soil was assesed as healthy soil. The results suggest that the simple method by Coriolus hirsutus is a handy way to assess the healthy of waste sludge-applied upland soils.

  • Agricultural recycling of livestock wastes by composting has lots of beneficial effects on crop production and soil fertility. Most of composts are made from pig manure and water content controller such as saw dust, bark, rice hulls etc. by aerobic processing. But the insufficient supply of saw dust, bark etc. cause the indiscreet use of industrial waste having heavy metals and toxic synthetic chemicals. This research investigated the present quality status of organic by-product fertilizers, and suggested the way of quality interpretation under the statistical approaches based on median, mean and weighted average value. Since the data of one hundred of samples for heavy metal contents (Cd, Pb, Cu, and Cr) and OM/N showed extremely left-skewed distribution, the median was more useful than the mean in representing the characteristics of distribiition for each items. The weighted average value will be a useful index for the quality based on the total amount of producing.

As incubating time passed, microbial community structure shifted gradually.

The Assessment of Toxicity on organic Sludge Using Acetylcholinesterase, Cytochrome P450, and Hsp70 Extracted from Earthworm (Eisenia fetida) (지렁이에서 추출한 Acetylcholinesterase, Cytochrome P450, and Heat Shock protein 70을 이용한 유기성슬러지 독성 평가)

  • The toxicitiy of organic sludge such as municipal sewage sludge (MSS), industrial sewage sludge (ISS), alcohol fermentation processing sludge (AFPS) and leather processing sludge (LPS) were evaluated with three environmental biomarkers as acetylcholinesterase, cytochrome P450, and heat shock protein 70 extracted from earthworm (Eisenia fetida). Their toxicities were compared with those of pig manure compost (PMC). MSS, ISS, LPS, and AFPS did not significantly affect the acetylcolinesterase activity, whereas only the elutriate of PMC slightly was increased the activity. MSS, AFPS, and PMC tended to slightly inhibit the cytochrome $P_{450}$ activity, but ISS and LPS showed significantly the inhibitory effect on cytochrome $P_{450}$. The hsp70 expression began to increase after treatments and showed high induction at 6 hour, followed by zero level at around 12 hour. The quantity of the hsp70 expressed by elutriate treatments of PMC, AFPS, MSS, ISS, and LPS was 1.9, 3.0, 3.3, 4.4, and 4.7 fold higher than that of distilled water. These results indicate that in toxicity tests of five organic waste materials, four kinds of sludge materials appeared more toxic than PMC. Results of AChE, P450, and hsp70 of earthworm might be useful for expecting or assessing an effect by exposure of organic wastes to earthworms in soil.
