An Experimental study on the effects of Insambakhab-tang on the Anti-allergic effect and Pulmonary Function of $O_3$ intoxicated Mice (인삼백합탕이 알레르기와 폐손상에 미치는 영향)

  • Experimental studies were done to research the Clinical effects of Insambakhab-tang on the Anti-allergic effect and pulmonary function of O₃ intoxicated Mice. Anti-allergic effect experiment consisted of vascular permeability responses to intradermal histamine and serotonin, 48hrs homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against egg white albumin, and delayed type hypersensitivity responses to Picryl Chloride and SRBC. Pulmonary function of O₃ intoxicated Mice experimental consisted of pulmonary thromboembolism (Sodium Arachidonate-induced and ADP-induced), lung TBA value, and serum Na/sup +/, K/sup +/, Cl/sup +/ level. The results obtained as follows; 1. In the effects of Insambakhab-tang on the pulmonary thromboembolism by Sodium Arachidonic acid and ADP, Insambakhab-tang group revealed significant effect. 2. In the effects of Insambakhab-tang on the vascular permeability responses to intradermal histamine, Insambakhab-tang group revealed significant effect. 3. In the effects of Insambakhab-tang on the vascular permeability responses to intradermal serotonine, Insambakhab-tang group revealed significant effect. 4. In the 48hrs homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against egg white albumin, Insambakhab-tang group revealed significant effect. 5. In the delayed type hypersensitivity responses to Picryl Chloride, Insambakhab-tang group revealed significant effect. 6. Insambakhab-tang group revealed significant effect on decrease of the lung TBA value of lung. 7. In the effects of Insambakhab-tang on Serum Na/sup +/, K/sup +/ Level in O₃-intoxicated Mice. Insambakhab-tang group revealed none significant effect, but In the effects of Insambakhab-tang on Serum Cl/sup +/ Level in O₃-intoxicated Mice, Insambakhab-tang group revealed significant effect.

  • Allergic rhinitis and allergic dermatitis are common allergic diseases. Galgeun-tang has been used for treating various diseases, which include allergic rhinitis and allergic dermatitis. Experimental sutdies have been done to research the anti-allergic effects of Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang. We have observed: the vascular permeability response induced by serotonin and histamine, the contact dermatitis response induced by picryl chloride, the delayed type hypersensitivity response to SRC, the mice paw edema induced by carrageenin, and the rectal temperature in febrile rats induced by endotoxin, as well as the writhing syndrome induced by $0.7{\%}$ acetic acid. The results were as follows: 1. In the vascular permeability response to intradermal serotonin and histamine, Galgeun-tang and Gamigalgeun-tang showed a significant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect. 2. In the contact dermatitis response induced by picryl chloride, Galgeun-tang and Gamigalgeun-tang showed a signigicant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect. 3. In the delayed type hypersensitivity response to SRC, Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang showed a significant inhibitory effect. 4. In the mice paw edema induced by carrageenin, Galgeun-tang and Gamigalgeun-tang showed a significant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect. 5. In the rectal temperature in febrile rats induced by endotoxin, Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang showed a significant inhibitory effect. 6. In the writhing syndrome induced by a $0.7{\%}$ acetic acid solution. Galgeun-tang and Gamigalgeun-tang showed a significant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect.

Effects of polyphenol and anthocyanin contents of Punica granatum fruit parts on their antioxidant and anticancer activities (석류 과실 부위별 폴리페놀 및 안토시아닌 함량이 항산화 및 항암 활성에 미치는 영향)

  • The purpose of this study was to determine the content of phenolics and various anthocyanin compounds, and antioxidant and anticancer activities of the extracts from pomergranates, which were produced in two different regions including Goheung, Korea and California, USA. These pomergrantes were divided into juice, pericarp, and seed parts and each part was extracted with 95% methanol. Content of total phenolics [mg chlorogenic acid equivalents/kg DW] was highest in pericarp, followed by juice and seeds from pomergrantes in both regions. The anthocyanins identified in pomegranate fruit were cyanidin-3,5-diglucoside chloride (Cy3,5G), cyanidin-3-O-glucoside chloride (Cy3G), delphini din-3,5-di-O-glucoside chloride (Dp3,5G), delphinidin-3-O-glucoside chloride (Dp3G), pelargonidin-3,5-di-glucoside chloride (Pg3,5G), and pelargonidin-3-glucoside chloride (Pg3G). Among these, cyanidin-3,5-diglucoside chloride (Cy3,5G) was the major anthocyanin in California pomegranate fruit juice and Goheung pomegranate fruit pericarp. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was dose-dependently increased, and was higher in pericarp part than juice or seed parts from pomergrantes in both regions. By MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay, pericarp extracts from pomergrantes in both regions showed the highest anticancer activity, which was higher on Calu-6 for human pulmonary carcinoma than SNU-601 for human gastric carcinoma. Correlation between polyphenols and anticancer activity on Calu-6 was determined to be in the range of $r^2=0.8904$ to 0.9706.

In the delayed type hypersensitivity resposes to SRBC, Hoichunyanggyuksan revealed significant effect. 5. In Anti-pyretic action by yeast method, Hoichunyanggyuksan showed significant effect. 6. In Anti-inflammatory action by carrageenin method, Hoichunyanggyuksan showed significant effect. 7. In analgesic action by acetic acid method, Hoichunyanggyuksan was recognized significantly.

In the effect of Hyunggaeyeungyotang on vascular permeability responses to intradermal serotonin in rats, Hyunggaeyeungyotang revealed significant effect. 3. In the 48hr homologous passive cutaneous anaphylaxis in rats provoked by the IgE-like antibody against egg albumin, Hyunggaeyeungyotang revealed significant effect. 4. In the effect of Hyunggaeyeungyotang on Delayed-type hypersensitivity responses to picryl chloride in mice, Hyunggaeyeungyotang revealed none of significant. 5. In the effect of Hyunggaeyeungyotang on Delayed-type hypersensitivity responses to SRBC in mice, Hyunggaeyeungyotang revealed none of significant. According to above-stated results, Hyunggaeyeungyotang is concluded to be effective as anti-allergic regimen and recommended to be used for treatment of allergic disease.

  • This experimental research has been done to study the effects of Bojungikgi-tang(BJIKT) on the anti-allergic reaction. We found the several important results from the research which has been performed by two experiments toward immediately type and delayed type in order to study the effects of BJIKT on hypersensitivity response to mice. The results obtained from our research are as following: 1. The survival rate of one group to which we injected only the compound 48/80 is almost 0% according to its density and timing test. In the other hand, the survival rates of the other group to which we injected both of the compound 48/80 and BJIKT are 20%, 10%, 30%, 10%, 40%, and 70% according to 0.025, 0.05, 0.1, 0.25 0.5 and 1(mg/g) of compound 48/80. Time dependency test also shows the 10% and 0% survival rates in 5 and 10 minutes. 2. BJIKT revealed the significantly inhibitory effect on Compound 48/80 induced Mast cell degranulation. 3. BJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to picryl chloride. 4. BJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to sheep red blood tell. Our research provides the important evidence that BJIKT is benificial to the prevention and treatment of allergy related diseases.

Effects of Baicalein on Picryl Chloride-induced Contact Dermatitis in BALB/c Mice (Baicalein의 BALB/c Mice에서의 접촉성 피부알레르기 예방효과)

  • Contact dermatitis is induced by allergy response from a lot of chemical substances brought on by environmental contamination. In this study, we investigated allergy prevention effect of baicalein which is Scutellaria radix extract. Scutellaria radix is the dried roots of Scutellaria baicalensis Georgi (Labiatae). It has been used in oriental traditional medicine for treatment of fever, diarrhea and inflammation. We have induced allergy by PCL after oral administration of 0 mg/kg, 50 mg/kg or 100 mg/kg baicalein dose for 8 times during 8 days in BALB/c mice and measured ear thickness, IgE level, histamine level, mast cell number and histopathological changes. The ear thickness of the 100 mg/kg baicalein group was thinner than that of the 0 mg/kg baicalein group. IgE level and histamine level of the 100 mg/kg baicalein group were lower than those of 0 mg/kg baicalein group. The number of mast cells of 0 mg/kg baicalein group more than that of 100 mg/kg baicalein group in the ear tissue. In addition, intradermal edema with prominent inflammatory cell infiltration was observed at 0 mg/kg baicalein group more than 100 mg/kg baicalein group in the ear tissue. These results suggest that baicalein has allergy prevention effect in BALB/c mice.

In the effects of Chihyosan and Chihyosangamibang on vascular permeability responses to intradermal histamine, both of chihyosan and Chihyosangamibang group revealed significant effect. 2. In the effects of Chihyosan and Chihyosangamibang on vascular permeability responses to intradermal serotonin, both of chihyosan and Chihyosangamibang group revealed significant effect. 3. In the 48hrs homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against egg white albumin, Chihyosan groups revealed significant effect, but Chihyosangamibang groups revealed none significant effect. 4. In the delayed type hypersensitivity responses to Picryl Chloride, Chihyosan and Chihyosangamibang groups revealed none significant effect. 5. In the delayed type hypersensitivity responses to. SRBC, Chihyosan revealed none significant effect, but Chihyosankamibang revealed significant effect. 6. Both of Chihyosan and Chihyosangamibang groups revealed significant effect on decrease of the lung TBA value of lung. 7. Both of Chihyosan and Chihyosangamibang groups revealed significant effect on decrease of the water contents of right and left lung. 8. Both of Chihyosan and Chihyosangamibang groups revealed significant effect on decrease of oxygen consumption time. 9. In the decrease effect of arterial blood $pCO_2$ level, both of Chihyosan and Chihyosangamibang groups revealed none significant effect. 10. In the increase effect of arterial blood $pO_2$ level, both of Chihyosan and Chihyosangamibang groups revealed none significant effect. 1. In the decrease effect of arterial blood $HCO_3^-$ level, both of Chihyosan and Chihyosangamibang groups revealed significant effect. 12. In the increase of arterial blood pH level, Chihyosangamibang groups revealed none significant effect, but Chihyosan groups revealed significant effect. According to above stated results, both of Chihyosan and Chihyosangamibang are very usefully for treatment of cough, asthma, chronic obstructive pulmonary diseases and allergic pulmonary diseases.
