Synthesis of an Amphiphilic $\pi$-Conjugated Triblock Copolymer of Poly(9,9-didodecylfluorene-2,7-diyl) and Poly(hydroxyl ethyl methacrylate)

  • A novel amphiphilic, symmetric rod-coil, triblock copolymer (denoted as PHEMA-b-PF-b-PHEMA) of poly(9,9-didodecylfluorene-2,7-diyl) (PF) and poly(hydroxyl ethyl methacrylate) (PHEMA) was synthesized. A $\pi$-conjugated poly(9,9-didodecylfluorene-2,7-diyl) (PF) was used as a rodlike midblock segment and connected with hydrophilic end blocks of poly(hydroxyl ethyl methacrylate) (PHEMA) by using an ATRP technique. The chemical structure of PHEMA-b-PF-b-PHEMA was confirmed by $^{1}H$-NMR and GPC, and its PL properties were investigated in selected solvents. Due to the dissimilarities in molecular conformation and solubility between PHEMA and PF blocks, both block segments were segregated to display a phase-separated morphology on a Si wafer.

Cloning and Characterization of a PI-like MADS-Box Gene in Phalaenopsis Orchid

  • The highly evolved flowers of orchids have colorful sepals and fused columns that offer an opportunity to discover new genes involved in floral development in monocotyledon species. In this investigation, we cloned and characterized the homologous PISTALLATA-like (PI-like) gene PhPI15 ($\underline{Ph}alaenopsis$ $\underline{PI}$ STILLATA # $\underline{15}$), from the Phalaenopsis hybrid cultivar. The protein sequence encoded by PhPI15 contains a typical PI-motif. Its sequence also formed a subclade with other monocot PI-type genes in phylogenetic analysis. Southern analysis showed that PhPI15 was present in the Phalaenopsis orchid genome as a single copy. Furthermore, it was expressed in all the whorls of the Phalaenopsis flower, while no expression was detected in vegetative organs. The flowers of transgenic tobacco plants ectopically expressing PhPI15 showed male-sterile phenotypes. Thus, as a Class-B MADS-box gene, PhPI15 specifies floral organ identity in orchids.

Synthesis, Crystal Structure, Spectra Characterization and DFT Studies on a Di-Cycle Pyrazoline Derivative

  • A dicycle pyrazoline derivative, 1-phenyl-5-(p-fluorophenyl)-3,4-($\alpha$-p-fluoro-tolylenecyclohexano) pyrazoline, was synthesized and characterized by elemental analysis, IR, UV-vis, fluorescence spectra and X-ray single crystal diffraction. Density function theory (DFT) calculations were performed by using B3LYP method with 6-$311G^{**}$ basis set. The optimized geometry can well simulate the molecular structure. Vibrational frequencies were predicted, assigned and compared with the experimental values, which suggest that B3LYP/6-$311G^{**}$ method can well predict the IR spectra. Both the experimental electronic absorption spectra and the predicted ones by B3LYP/6-$311G^{**}$ method reveal three electron-transition bands, with the theoretical ones having some red shifts compared with the experimental data. Natural bond orbital analyses indicate that the absorption bands are mainly derived from the contribution of n $\rightarrow\pi^*$ and $\pi\rightarrow\pi^*$ transitions. Fluorescence spectra determination shows that the title compound can emit blue-light at about 478 nm. On the basis of vibrational analysis, the thermodynamic properties of title compound at different temperature have been calculated, revealing the correlations between $C^0_{p,m}$, $S^0_m$, $H^0_m$ and temperature.

Suppressive Effect of Arazyme on Neutrophil Apoptosis in Normal and Allergic Subjects

  • Arazyme is a metalloprotease secreted by Aranicola proteolyticus that was previously shown to suppress cytokine expression of keratinocytes and endothelial cells and inhibit histopathological features in an atopic dermatitis-like animal model. However, the regulatory effects of arazyme in other allergic diseases have yet to be elucidated. In this study, we investigated whether arazyme is effective against neutrophil apoptosis in allergic diseases such as allergic rhinitis and asthma. Arazyme inhibited neutrophil apoptosis of normal subjects in a dose-dependent manner. However, the antiapoptotic effect of arazyme was reversed by LY294002, an inhibitor of PI3K, AKTi, an inhibitor of Akt, PD98059, an inhibitor of MEK, and BAY-11-7085, an inhibitor of NF-${\kappa}B$. Arazyme induced activation of NF-${\kappa}B$ via PI3K/Akt/ERK pathway. The anti-apoptotic effect of arazyme is associated with inhibition of cleavage of caspase 3 and caspase 9. Arazyme inhibited constitutive apoptosis of neutrophil in a dose-dependent manner in allergic subjects, and its mechanism was shown to be associated with PI3K/Akt/ERK/NF-${\kappa}B$. The results presented here improve our understanding of neutrophil apoptosis regulation and will facilitate development of drugs for treatment of allergic diseases.

HCl/ethanol로 유발한 급성 위염 마우스에서 PI3K/Akt/NF-κB 신호전달경로를 통한 진피 열수 추출물의 보호 효과 (The protective effect of Citrus unshiu Peel water extract through PI3K/Akt/NF-κB signaling pathway in mice with HCl/ethanol-induced acute gastritis)

  • 본 연구에서는 우선 진피 열수 추출물의 in vitro 항산화능을 평가하기 위해 total polyphenol, total flavonoid 함량, DPPH 및 ABTS radical 소거능을 분석한 후, 150 mM/60% ethanol로 유발한 급성 위염 동물 실험을 진행하여 급성 위염 완화 효과를 검증하였다. 약물투여군의 혈청 내 ROS와 MPO 수준, 조직 내 MDA 수준의 유의성 있는 감소를 확인하였으며, western blot을 통해 NOX2와 p22phox를 포함한 산화적 스트레스 관련 단백질을 억제하였고, PI3K/Akt/NF- κB 신호 전달 경로를 통한 염증성 단백질의 현저한 감소를 확인하였다. 따라서 이러한 결과는 진피 열수 추출물이 급성 위염에 대한 완화 효과를 나타냈으며, 위염 및 천연 치료제의 후보 소재로서 가능성이 있다고 판단된다. 또한, 향후 만성 위염, 위암과 같은 위장 질환에 관한 추가 연구에서 진피 열수 추출물의 활용 가능성을 시사한다.

수직자기기록매체용 Fe-Co-B/M 하지연자성층의 미세결정구조 및 자기특성 (Microstructure and Magnetic Properties in Fe-Co-B/M Films for Soft Magnetic Underlayer of Perpendicular Magnetic Recording Media)

  • It is necessary to develop soft magnetic layer with high saturation magnetization 4 $\pi{M}_s$ and in-plane magnetic anisotropy field Hk for soft magnetic underlayer of perpendicular magnetic recording media with high signal to noise ratio. Fe-Co-B layer with high 4 $\pi$Ms of about 23 kG deposited on Ni-Fe and Ni-Fe/Si seedlayer exhibited very high in-plane magnetic anisotropy filed Hk of about 280 and 380 Oe, respectively, In-plane XRD studies clarified that the lattice spacing of planes along the easy axis direction was longer than that along the hard axis direction in the Fe-Co-B layers with high Hk. These results indicate that high Hk of Fe-Co-B/Ni-Fe and Fe-Co-B/[Ni-Fe/si] layers were resulted from magnetoelastic anisotropy owing to a residual stress. Moreover, the high Hk in the Fe-Co-B/Ni-Fe layer was maintained until 30$0^{\circ}C$ annealing temperature.

In this paper, we obtain sharp results concerning coefficient estimates, distortion theorem and radius of convexity for the class $T^{\alpha}_{\lambda}$(p, A, B). It is further shown that the class $T^{\alpha}_{\lambda}$(p, A, B) is closed under "arithmetic mean" and "convex linear combinations". We also obtain class preserving integral operators of the form $F(z)=\frac{p+c}{z^c}{\int^z_0t^{c-1}}f(t)dt$, c>-p, for the class $T^{\alpha}_{\lambda}$(p, A, B). Conversely when $F(z){\in}T^{\alpha}_{\lambda}$(p, A, B), radius of p valence of f(z) has also determined.

Linarin enhances melanogenesis in B16F10 cells via MAPK and PI3K/AKT signaling pathways

  • In this study, we discovered for the first time that linarin, a flavonoid compound, enhances melanin biosynthesis in B16F10 cells, and subsequently elucidated the underlying mechanism of linarin-induced melanogenesis. Linarin showed no cytotoxicity at a concentration of 42 μM and significantly increased intracellular tyrosinase activity and melanin content in B16F10 cells. Mechanistic analysis showed that linarin increased the expression of tyrosinase, tyrosinase-related protein 1 (TRP-1), and microphthalmia-associated transcription factor (MITF) that are related to melanogenesis. Moreover, linarin decreased the phosphorylation of extracellular signal-regulated kinase (ERK) and protein kinase B (AKT). Finally, we evaluated the effect of the structure-activity relationship of linarin and its aglycone on melanogenesis. The results indicated that linarin enhances the expression of melanogenic proteins by activating MITF expression via the modulation of mitogen-activated protein kinase (MAPK), phosphatidylinositol 3-kinase (PI3K), and protein kinase B signaling pathways in B16F10 cells, thereby enhancing melanogenesis.

칸막이 구조를 이용한 단일 평면상의 38 GHz용 도파관 슬롯 배열 안테나 (A Single-Layer Waveguide Slot Array Antenna using Diaphragms for 38 GHz Frequency Band)

  • 본 논문은 내부 칸막이(diaphragm) 구조를 갖는 38 GHz용 도파관 슬롯 16$\times$16 배열 안테나의 설계와 측정결과를 보여준다. 우선, 도파관 윗면에 전파 진행방향의 슬롯이 갖는 복사특성을 연구하고, 도파관 내부의 양쪽벽면에 가로로 위치한 칸막이들을 이용하여 슬롯 배열 안테나의 임피던스를 조절함으로써 반사손실을 최소화 시켰다. 이 칸막이들의 위치와 크기를 최적화하여 20 dB Chebyshev 16배열 도파관 슬롯 안테나를 설계하였다. 다음에는, $\pi$-와 T-접합 전력 분배기로 구성된 직렬 급전구조를 이용하여 20 dB Chebyshev 16포트 전력 분배기를 설계하였다. 38 GHz 대역의 광대역 무선 멀티미디어 서비스용으로 설계된 16$\times$16 슬롯 배열 안테나를 컴퓨터 제어 밀링기술로 제작하고, 측정한 결과 31.2 dBi의 이득을 얻었고, 부엽준위는 -19 dB 이하이었으며, 740 MHz의 밴드 폭을 얻었다.

Genistein attenuates isoflurane-induced neurotoxicity and improves impaired spatial learning and memory by regulating cAMP/CREB and BDNF-TrkB-PI3K/Akt signaling

  • Anesthetics are used extensively in surgeries and related procedures to prevent pain. However, there is some concern regarding neuronal degeneration and cognitive deficits arising from regular anesthetic exposure. Recent studies have indicated that brain-derived neurotrophic factor (BDNF) and cyclic AMP response element-binding protein (CREB) are involved in learning and memory processes. Genistein, a plant-derived isoflavone, has been shown to exhibit neuroprotective effects. The present study was performed to examine the protective effect of genistein against isoflurane-induced neurotoxicity in rats. Neonatal rats were exposed to isoflurane (0.75%, 6 hours) on postnatal day 7 (P7). Separate groups of rat pups were orally administered genistein at doses of 20, 40, or 80 mg/kg body weight from P3 to P15 and then exposed to isoflurane anesthesia on P7. Neuronal apoptosis was detected by TUNEL assay and FluoroJade B staining following isoflurane exposure. Genistein significantly reduced apoptosis in the hippocampus, reduced the expression of proapoptotic factors (Bad, Bax, and cleaved caspase-3), and increased the expression of Bcl-2 and Bcl-xL. RT-PCR analysis revealed enhanced BDNF and TrkB mRNA levels. Genistein effectively upregulated cAMP levels and phosphorylation of CREB and TrkB, leading to activation of cAMP/CREB-BDNF-TrkB signaling. PI3K/Akt signaling was also significantly activated. Genistein administration improved general behavior and enhanced learning and memory in the rats. These observations suggest that genistein exerts neuroprotective effects by suppressing isoflurane-induced neuronal apoptosis and by activating cAMP/CREB-BDNF-TrkB-PI3/Akt signaling.
