Rapid Measurement of $NH_3$ and Weak Acid Permeation Through Liposomes and Renal Proximal Tubule Membranes

  • Using the methods of stopped-flow and epifluorescence microscopy with entrapped fluorophore, membrane permeability of $NH_3$ and weak acids in liposomes, renal brush border (BBMV) and basolateral membrane vesicles (BLMV), and primary culture cells from renal proximal tubule was measured. Permeability coefficient (cm/sec) of $NH_3$ was $(2.9{\times}10^{-2}$ in phosphatidylcholine liposome $25^{\circ}C)$, $5.9{\times}10^{-2}$ in renal proximal tubule cell $(37^{\circ}C)$, $4.0{\times}10^{-2}\;and\;2.4{\times}10^{-2}$ in BBMV and BLMV $(25^{\circ}C)$, respectively. Formic acid has the highest permeability coefficient among the weak acids tested, which was $4.9{\times}10^{-3}$ in liposome, $5.0{\times}10^{-3}$ in renal proximal tubule cell, $9.1{\times}10^{-3}$ in BBMV and $3.8{\times}10^{-3}$ in BLMV. There was a linear relationship between external concentration of nonionized formic acid and initial rate of flux of formic acid in liposome, and the slope coincided with the value of permeability coefficient of formic acid measured in pH 7.0. These results show that techniques of stopped-flow and epifluorescence microscopy with entrapped fluorophore provide the precise method of measurement of very rapid transport of nonelectrolytes through membranes with the advantages of instantaneous mixing effect, good resolution time and easy manipulation.

Study on the Effect of Sopungbosim-tang on Hypertension, Thrombosis and Brain damage (소풍보심탕이 고혈압, 혈전 및 뇌진탕에 미치는 영향)

  • This studt was investigated to prove the effect of SPBST on the hypertension, the thrombosis and the brain damage. The results were as follows; 1. SPBST affected the htpertension as adepressant, but insignificant. 2. SPBST decreased significantly dopamine, aldosterone but ineffective on the epinephrine, norepinephrine and renin activity. 3. SPBST increased the NO product but insignificant. 4. SPBST had a death suppression effect by 50% in pulmonary thrombosis inducement experiment and activated slightly on the fibrinolytic activity. 5. SPBST suppressed significantly platelet diminution and prolonged insignificantly PT and APTT. 6. On the measure of the blood flow rate induced by the thrombus, in vivo SPBST accelerated the blood flow rate, in vitro insignificant. 7. SPBST had no toxicity on the PC12 cell and B103 cell induced by amyloid β protein (-35) and a protective effect, in proportion to the density. 8. SPBST decreased significantly coma duration time in a Infatal dose of KCN and showed 50% of survival rate in a fatal dose. 9. SPBST decreased significantly ischemic area and edema incited by the MCA blood flow block. These results indicate that SPBST can be used in hypertension, the thrombosis, the brain damage, the ischemic cerebral infarction and the acute stage of the brain damage. Further study will be needed about the functional mechanism and etc.

Effects of Angelicae Gigantis and Cynanchum wilfordii Hemsley Extract on the Changes of Cerebral Flow (당귀와 백수오 추출물이 뇌혈류 변화에 미치는 영향)

  • This study was designed to investigate the effects of Angelicae Gigantis and Cynanchum wilfordii Hemsley extract (AAC) on the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and in the rats with cerebral ischemia induced by middle cerebral artery occlusion, and further to determine the mechanisms. The results were as follows. AAC significantly increased rCBF but significantly decreased MABP in a dose-dependent manner in normal rats. The increase of AAC-induced rCBF was significantly inhibited by pretreatment with methylene blue (0.01 mg/kg, i.p.), an inhibitor of guanylate cyclase, and AAC-induced MABP was decreased by pretreatment with methylene blue. In cerebral ischemics, rCBF was stably improved by AAC (10 mg/kg, i.p.) during the period of cerebral reperfusion, which was contrasted with the findings of rapid and marked increase in the control group. These results suggest that AAC can increase rCBF in the normal state, as well as improve the stability of rCBF in cerebral ischemic state.

Experimental Effects of Sibjeondaebo-tang and Gamy-Sibjeondaebo-tang on Cerebral Hemodynamics in Cerebral Ischemia Rats (십전대보탕(十全大補湯)과 가미십전대보탕(加味十全大補湯)이 뇌허혈 흰쥐의 뇌혈류역학에 미치는 실험적 영향)

  • This Study was designed to investigate the effects of Sibjeondaebo-tang (SDT) and Gamy-Sibjeondaebo-tang (GST, Sibjeondaebo-tang adding Cervi Pantotrichum Cornu) on the improvement in regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and in the rats with cerebral ischemia induced by middle cerebral artery occlusion, and further to determine the mechanisms. And, It was to investigate the effects of the SDT and GST with the change of histologic examination through the BDNF in the hippocampus CA1. In changes of cerebral hemodynamics, SDT and GST significantly increased rCBF in a dose-dependent manner but decreased MABP in normal rats. In mechanism of cerebral hemodynamics, Increase of GST-induced rCBF was significantly inhibited by pretreatment with methylene blue (0.01 mg/kg, i.p.), an inhibitor of guanylate cyclase, and Decrease of GST-induced MABP was significantly increased by pretreatment with methylene. These results suggested that the action of GST was mediated by guantlate cyclase pathway. In cerebral ischemics, the rCBF was stably improved by SDT (10 mg/kg, i.p.) significantly and stably increased by GST (10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrast with the findings of rapid and marked increase in Control group. These results suggested that GST had anti-ischemic action in cerebral ischemic state. In histological examination through TTC stain, Sample A group and Sample B group decreased discoloration in the cortical part at $28^{th}$ day after MCAO induction. In immunohistochemistric response of BDNF, Sample A group and Sample B group increased respondent effect at $28^{th}$ day after MCAO induction. These results suggest that GST can Increase rCBF in normal state, as well as improve the stability of rCBF in cerebral ischemic state. Furthermore, methylene blue inhibitor study suggested the mechanism of blood flow enhancement by GST may be mediated by guanylate cyclase pathway.

Influences of Wearing Far-infrared Indoor Clothing on Skin Blood Flow, Perceptual and Thermal Responses (원적외선 방사 기능 실내복 착용이 인체 피부혈류량, 온열 심리 및 온열 생리 기능에 미치는 영향)

  • The purpose of this study was to evaluate far-infrared clothing (FIR condition) with non-far-infrared clothing (Control condition) to assess the effects of FIR on thermo-physiological responses. Eight young healthy males (23.0±2.3 yr, 176.5±3.7 cm, and 69.0±4.3 kg) participated in this experimental trial, which consisted of a 20 min rest followed by a 40 min walk (4.0 km·hr-1) and a 20 min recovery at 20℃ with 50%RH. The results showed that finger skin blood flow and mean skin temperature were significantly higher for the FIR condition than the control during exercise and recovery (p<.001). Clothing microclimate temperature of the FIR condition was 0.5℃ higher on the back (p=.001) and 0.4℃ higher on the thigh (p=.015) during recovery. Clothing microclimate relative humidity of the FIR condition was 13% higher on the chest (p=.006) and 19% higher on the back (p<.001) during exercise than control. Subjects felt warmer and more comfortable in the FIR condition than in the control (p<.05). Perceived skin wettedness (%BSA) was less in the FIR condition than in the control (p=.001). These results indicate that ceramic-embedded clothing had significant effects on thermoregulatory responses for light activity in an indoor environment.

Study on the Effects of Fructus Schizandrae on the Cerebral Blood Flow and Blood Pressure (뇌혈유와 혈압에 미치는 오미자의 효능에 대한 연구)

  • The purpose of this present study is to measure the changes of regional cerebral blood flow(rCBF) and blood pressure(BP) in rats. following the intravenous injection of drug. The measurement was continually monitored by laser-doppler flowmeter (Transonic Instrument, USA) and pressure tranducer(Grass, USA) in anesthetized adult Sprague-Dawley rats about for 2h to 2h and a half through the data acquisition system composed of MacLab and Macintosh computer. The result of this experiment was as following. 1. Fructus Schizandrae increased the changes of rCBF in rats, significantly. 2. The rCBF of Fructus Schizandrae did not change by pretreated propranolol. 3. The rCBF of Fructus Schizandrae did not change by pretreated atropine. 4. The rCBF of Fructus Schizandrae did not change by pretreated I-NNA. 5. The rCBF of Fructus Schizandrae was decreased by pretreated methylene blue. 6. The rCBF of Fructus Schizandrae did not change by pretreated indomethacin. 7. Fructus Schizandrae decreased the changes of blood pressure significantly. 8. The BP of Fructus Schizandrae did not change by pretreated propranolol. 9. The BP of Fructus Schizandrae did not change by pretreated atropine. 10. The BP of Fructus Schizandrae did nol change by pretreated I-NNA. 11. The BP of Fructus Schizandrae was decreased by pretreated methylene blue. 12. The BP of Fructus Schizandrae did not change by pretreated indomethacin. These results indicate that Fructus Schizandrae can increase the rCBF and decrease the BP, that related to guanylyl cyclase activity.

  • Since arterial disease in the upper extremity is less common than that in the lower extremity, experimental and numerical investigations related to upper extremity have been rarely performed. We created a three-dimensional model of the arteries, larger than approximately 1 mm, in a Korean adult's left hand (from brachial to digital arteries), from 3T magnetic resonance imaging (MRI) data. For the first time, a three-dimensional computational fluid dynamic method was employed to investigate blood flow velocity, blood pressure variation, and wall shear stress (WSS) on this complicated artery system. Investigations were done on physiological blood flows near the branches of radial and deep palmar arch arteries, and ulnar and superficial palmar arch arteries. The flow is assumed to be laminar and the fluid is assumed to be Newtonian, with density and viscosity properties of plasma.

Application of Multiparametric Flow Cytometry (FCM) to Enumerate the Diagnosis of Pseudomonas aeruginosa and Escherichia coli

  • In this study, multiparametric flow cytometry (FCM) was installed to enumerate the diagnosis of Pseudomonas aeruginosa ATCC 10145 and Escherichia coli K12 (IFO 3301). The nucleic acids (DNA/RNA) were double stained by a LIVE/DEAD bacLight viability kit, involving green SYTO 9 and red propidium iodide (PI), based on the permeability of two chemicals according to the integrity of plasma membrane. As the results showed, the gate for dead bacteria was defined as the range of $0.2{\times}10^0$ to $6.0{\times}10^1$ photo multiplier tube (PMT) 2 fluorescence (X-axis) and $2.0{\times}10^0$ to $2.0{\times}10^2$ PMT 4 fluorescence (Y-axis), and the gate for live bacteria was defined as the range of $6.0{\times}10^0$ to $6.0{\times}10^2$ PMT 2 fluorescence (X-axis) and $2.0{\times}10^0$ to $4.0{\times}10^2$ PMT 4 fluorescence (Y-axis). In the comparison of the number of the tested bacteria detected by FCM (viability assessment) and plate culture (cultivability assessment), the number of bacteria detected by FCM well represented the number of bacteria that was detected by the colony forming unit (CFU) counting method when bacteria were exposed to isopropyl alcohol and silver/copper cations. Consequently, it is concluded that the application of FCM to monitor the functional effect of disinfectants on the physiological status of target bacteria can offer more rapid and reliable data than the plate culture colony counting method.

Mechanism of Jaeumgenby-tang on the Regional Cerebral Blood Flow, Mean Arterial Blood Pressure and Cardiac Muscle Contractile Force in Rats (자음건비탕이 국소뇌혈류량, 평균혈압, 심근수축력에 미치는 작용기전)

  • Jaeumgenby-tang(JGT) have been used in oriental medicine for many centries as a a therapeutic agent of vertigo caused by deficiency of qi and blood. The effects of JGT on the regional cerebral blood flow(rCBF), mean arterial blood pressure(MABP) and cardiac muscle contractile force(CMF) is not known. The purpose of this Study was to investigate effects of JGT on the rCBF, MABP, CMF and mechanism of JGT induced changed rCBF, MABP, CMF. The changes of rCBF, MABP and CMF were determinated by Laser-Doppler Flowmetry(LDF). The results were as follows; JGT extract was increased rCBF, MABP and CMF in a dose-dependent, specially JGT extract was significantly increased rCBF and MABP. Pretreatment with propranolol was significantly inhibited JGT induced increase of rCBF but pretreatment with indomethacin and methylene blue were accelerated JGT induced increase of rCBF. Pretreatment with propranolol and indomethacin were inhibited JGT induced increase of MABP, but pretreatment with methylene blue was accelerated JGT induced increase of MABP. Pretreatment with propranolol was significantly inhibited JGT induced increase of CMF but pretreatment with indomethacin and methylene blue were accelerated JGT induced increase of CMF. This results suggest that JGT increased rCBF by increasing MABP and CMF and the action of JGT is mediated by adrenergic β-receptor.

Effects of Mixture of Bambusae Caulis in Liquamen and Bamboo Extract on the Regional Cerebral Blood Flow and Mean Blood Pressure in Rats (대나무 추출액과 죽력의 혼합물이 국소 뇌혈류량 및 평균 혈압에 미치는 영향)

  • This Study was designed to investigate the effects of Mixture of Bambusae Caulis in Liquamen and Bamboo Extract on the change of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal and cerebral ischemic rats. Experimental materials were as follows ; BE- 1 was Bamboo Extract (BE) extracted with 70% ethyl alcohol, BE-11 was BE extracted with distilled water at $121^{\circ}C$ for 30 min, BE-111 was BE extracted with distilled water at $121^{\circ}C$ for 3 hrs, MLC was mixture of Bambusae Caulis in Liquamen (BCL) and BE-111 mixed at the ratio of 1 to 100 (MLC100), 1 to 50 (MLC50), 1 to 20 (MLC20), 1 to 10 (MLC10), 1 to 5 (MLC5). The results were as follows , The Changes of BE- 1 on the rCBF and MABP in normal rats were not showed, BE- 11 significantly decreased rCBF in a dose-dependent manner Dut increased MABP in a dose-dependent manner. BE-111 increased rCBF in a dose-dependent manner, MLC significantly increased rCBF in a dose-dependent manner and increased MABP in a dose-dependent manner. rCBF was significantly and stably increased by MLC5 (1 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. As results above ; The present author thought that BE- 111 and MLC increased rCBF by dilating pial arterial diameter.
