Glycosylation Enhances the Physicochemical Properties of Caffeic Acid Phenethyl Ester

  • In this study, we synthesized a glycosylated derivative of caffeic acid phenethyl ester (CAPE) using the amylosucrase from Deinococcus geothermalis with sucrose as a substrate and examined its solubility, chemical stability, and anti-inflammatory activity. Nuclear magnetic resonance spectroscopy showed that the resulting glycosylated CAPE (G-CAPE) was the new compound caffeic acid phenethyl ester-4-O-${\alpha}-{\small{D}}$-glucopyranoside. G-CAPE was 770 times more soluble than CAPE and highly stable in Dulbecco's modified Eagle's medium and buffered solutions, as estimated by its half-life. The glycosylation of CAPE did not significantly affect its anti-inflammatory activity, which was assessed by examining lipopolysaccharide-induced nitric oxide production and using a nuclear factor erythroid 2-related factor 2 reporter assay. Furthermore, a cellular uptake experiment using high-performance liquid chromatography analysis of the cell-free extracts of RAW 264.7 cells demonstrated that G-CAPE was gradually converted to CAPE within the cells. These results demonstrate that the glycosylation of CAPE increases its bioavailability by helping to protect this vital molecule from chemical or enzymatic oxidation, indicating that G-CAPE is a promising candidate for prodrug therapy.

일부 농약의 생물농축계수의 측정 (Determination of Bioconcentration Factor in Some Pesticides)

  • The present study was performed to investigate the bioconcentration of BPMC, chlorothalonil, dichlorvos and methidathion. The BCFs(bioconcentration factors) and depuration rate constants for four pesticides in zebrafish(brachydanio rerio) were measured under semi-static conditions(OECD guideline 305-B) in a concentration of one-hundredth of the 96 hours LC50 of each pesticide at the equilibrium condition. The results obtained are summarized as follows : The BCFs of BPMC, chlorothalonil, dichlorvos and methidathion were 1.44$\pm$0.09, 2.223$\pm$0.063, 0.81$\pm$0.08 and 5.53$\pm$0.13, respectively. Depuration rate constants of BPMC, chlorothalonil, dichlorvos and methidathion were 0.028, 0.015, 0.220 and 0.152, respectively. The concentrations of BPMC, dichlorovs and methidathion in zebrafish reached an equilibrium in 3 days, and the equilibrium of chlorothalonil was reached after 14 days. Depuration rate of dichlorvos was the fastest followed by methidathion, BPMC and chlorothalonil. The lower BCF of BPMC was due to its relatively high KOW, slow KDEP, and low SW and VP, compared to chlorothalonil and methidathion. The BCF of chlorothalonil was much lower than that excepted on the basis of high KOW, slow KDEP, SW and VP. The reason is that the experimental concentration for chlorothalonil is 1/100~1/1000 lower than that of BPMC, dichlorvos and methidathion. The BCF of dichlorvos was lower than that of other pesticides due to its very rapid KDEP, very high VP and SW, and very low KOW. The BCF of methidathion was higher than that of other pesticides due to its very low VP and SW. Therefore, these data suggest that physicochemical properties of pesticides may be important in the bioconcentration.

아마씨 가루를 첨가한 쿠키의 품질특성 (Quality Characteristics of Cookies Made with Flaxseed Powder)

  • 본 연구에서는 아마씨 가루를 밀가루 중량기준 0%, 6%, 12%, 18%의 비율로 첨가한 쿠키를 제조한 다음, 반죽의 밀도, pH, 수분 함량, 쿠키의 퍼짐성, 손실률, 팽창률, 경도, 색도, 관능평가, 과산화물가, 산가 등을 측정하였다. 반죽의 밀도는 대조군과 실험군 간에 유의적인 차이가 없었고 pH는 첨가군이 대조군보다 높게 나타났으며 수분 함량은 낮게 나타났다. 퍼짐성은 아마씨 가루 첨가량이 증가할수록 증가하였고(p<0.05) 손실률은 대조군과 6% 첨가군 간에 유의적인 차이가 없었고 12% 첨가군이 가장 높았으며 18% 첨가군이 가장 낮았다. 팽창률은 대조군과 6%, 12% 첨가군 간에 차이가 없는 반면, 18% 첨가군은 대조군보다 감소하였다. 경도는 아마씨 가루 첨가군이 대조군보다 유의적으로 낮았고(p<0.05) 명도와 황색도는 아마씨 가루 첨가량이 증가할수록 감소하였다. 관능검사결과 아마씨 가루 첨가량이 증가할수록 쿠키의 표면색, 고소한 맛이 강하게 평가되었고 전반적인 기호도는 12% 첨가군이 대조군보다 높은 점수로 평가되었다(p<0.05). 저장 기간에 따른 산화 안정성을 측정한 결과 과산화물가는 모든 시료에서 저장 30일까지 증가하다가 감소하였고 산가는 저장기간 동안 지속적으로 증가하였으며 각 시료 간에 유의적인 차이는 없었다.

당귀분말을 첨가한 쿠키의 품질특성 (Quality Characteristics of Cookies Prepared with Angelica gigas Nakai Powder)

  • 연구는 당귀분말을 첨가한 쿠키의 품질특성과 관능특성을 평가한 것으로써, 당귀분말 첨가 쿠키 반죽의 pH는 6.13~6.26의 범위였으며, 반죽의 밀도는 0.99~1.15의 범위였다. 당귀분말 첨가의 증가에 따라 쿠키의 퍼짐성은 작아졌으며, 명도와 황색도의 감소, 적색도의 증가로 인해 전반적으로 쿠키가 어두워지고 진해지는 경향을 나타내었다. 기계적 조직감의 특성은 강도와 경도가 당귀분말의 첨가비율에 따라 높아지는 경향을 나타내어 쿠키 제조에 있어 당귀분말의 첨가는 쿠키를 단단하게 하는 역할을 하였다. 관능 선호도에서는 색, 냄새, 조직감, 맛, 전반적인 선호도의 항목에서 당귀분말 무첨가 쿠키보다 일정 범위의 당귀분말을 첨가한 쿠키의 선호도가 높아졌으며, 그 중에서도 쿠키의 전체 재료 배합량 대비 1.5%의 당귀분말을 첨가하는 것이 가장 이상적인 것으로 평가되었다.

피부 재생능력 촉진을 위한 물리적 복합자극의 활용 연구 (Combinatorial Physical Stimulation and Synergistically-Enhanced Fibroblast Differentiation for Skin Regeneration)

  • For proper wound healing, dermal contraction and remodeling are critical; during the natural healing process, differentiated fibroblasts called "myofibroblasts" typically undertake these functions. For severe wounds, however, a critical mass of dermal matrix and fibroblasts are lost, making self-regeneration impossible. To overcome this impairment, synthetic wound patches with embedded functional cells can be used to promote healing. In this study, we developed a polydioxanone (PDO)-based cell-embedded sheet on which dermal fibroblasts were cultured and induced for differentiation into myofibroblasts, whereby the following combinatorial physicochemical stimuli were also applied: aligned topology, electric field (EF), and growth factor. The results show that both the aligned topology and EF synergistically enhanced the expression of alpha smooth-muscle actin (${\alpha}$-SMA), a key myofibroblast marker. Our proof-of-concept (POC) experiments demonstrated the potential applicability of a myofibroblast-embedded PDO sheet as a wound patch.

Superoxide Dismutase Mimetic Activity of Cu(II)-Salicylic Acid Analogs

  • The superoxide dismutase (SOD)-mimetic activities of copper complexes of a series of salicylic acid (SA) analogs were tested and compared to the activity of bovine erythrocyte SOD using ferricytochrome c reduction assay. Stability constants of copper complexes were measured potentiometrically using SCOGS2 program. In the presence of 10 g/l albumin, all the copper complexes lost their SOD mimetic activities. Multiple regression analysis was employed for the statistical comparisons between the SOD mimetic activity and their physicochemical properties. Correlation exists for the SOD mimetic activity and steric parameter $(E_s)$ and/or electronic parameter $({\Sigma}{\sigma})$ in xanthine/xanthine oxidase (XOD) system, demonstrating that E, plays a key role in SOD activity whereas ${\Sigma}{\sigma}$ influences it to a lesser extent. The protective effect of copper complexes against membrane damage was measured by counting D-glucose released frm $EG_s$. D-glucose and XOD were entrapped within $EG_s$ and acetaldehyde was used as a substrate for XOD. In this membrane model system using $EG_s$, hydrophobic parameter $({\Sigma}{\pi})$ is of most importance, producing parabolic equation while $E_s$, and ${\Sigma}{\sigma}$ appear to playa minor role in protection against D-glucose release. In summary, to design an efficient SOD mimetic, stability, steric factor, lipophilicity and redox potential should be considered.

디클로페낙 프로드럭들의 흰쥐 피부 투과 (Rat Skin Permeation of Diclofenac and its Prodrugs)

  • Various alkyl ester prodrugs of diclofenac were synthesized in order to investigate the relationship between their skin permeation characteristics and physicochemical properties. Solubility in various vehicles was measured at room temperature. 1-Octanol/water partition coefficients (Log P) and capacity factors (k') were measured to determine the lipophilicity of the prodrugs. Stability of prodrugs in the skin extract and homogenate was also investigated before conducting the skin permeation studies. Increases in the Log P and capacity factor values were observed when alkyl esters of diclofenac were prepared. Since the aqueous solubility of the prodrugs was not high enough, they were saturated in propylene glycol (PG) for skin permeation studies. Prodrugs were rapidly metabolized to diclofenac, both in skin homogenate and in dermal extract of skin. The skin permeation rate of alkyl ester prodrugs was significantly higher than diclofenac with shorter lag time. Moreover, a parabolic relationship was observed between the permeation rate and the log P values of prodrugs, and the maximum flux was achieved at a log P value of around 4.0.

Influence of Purple Sweet Potato Powder Addition on the Quality Characteristics and Oxidative Stability of Cookies

  • Cookies were prepared by adding different levels (0, 5, 10, 15, and 20%) of purple sweet potato powder (PSPP) and their physicochemical and sensory characteristics were examined. With increase of the PSPP level, moisture content decreased and ash content increased (p<0.05). The spread factor of control cookies was higher than that of cookies containing PSPP. The incorporation of PSPP in cookies lowered the lightness and yellowness but increased redness (p<0.05). Texture measurement showed that the addition of PSPP did not affect the hardness of cookies. Results of acceptance test revealed that overall acceptability score was not significantly different between the control and the PSPP added cookies. Therefore, it is suggested that PSPP can be incorporated into cookies up to 20% without affecting the cookie quality. Total polyphenol content and antioxidant activity were assessed to examine the functionality of cookies. In addition, substitution of wheat flour with PSPP yielded cookies with higher polyphenol content and antioxidative activity (p<0.05). The peroxide value (POV) and acid value increased during storage at $60^{\circ}C$ for 80 days. The POV was significantly lower in cookies containing PSPP than the control cookies, suggesting that the addition of PSPP deterred the lipid oxidation (p<0.05).

흡수촉진제를 이용한 펌제의 웨이브 효율 증가 및 모발손상 억제 (Increase of Permanent Wave Efficacy and Decrease of Hair Damage by using Enhancer of Permanent Wave Lotion)

  • Human hair could be damaged by various physicochemical conditions and treatment. Permanent and decoloring treatment were the most serious factor on hair damage. The new permanent wave lotion containing Permeation enhancers such as Cremophor EL, Transcutol and propylene glycol based on cysteine permanent wave lotion were prepared. Efficiency of permanent wave and hair damage following pH of permanent wave lotion and addition of permeation enhancer were investigated. PH of solution, wave efficiency, loss of protein from hair, morphology of hair by SEM and solubility of alkaline solution were evaluated. The addition of Cremophor EL and Transcutol with ethanol increased permanent wave efficacy and decreased hair damage effectively. They diminished permanent wave lotion's pH and augmented permanent wave lotion's penetration compare to cysteine permanent wave lotion. new permanent wave lotion containing permeation enhancers such as Cremophor EL could be a good candidate for a new permanent wave lotion.

PLGA 미립구를 이용한 새로운 단회 접종 항원 전달 시스템의 개발 (Improved Antigen Delivery Systems with PLGA Microsphere for a Single-Step Immunization)

  • A promising approach to the development of a new single-step vaccine, which would eliminate the requirement for multiple injections, involves the encapsulation of antigens into microspheres. Biodegradable poly(lactide-co-glycolide) (PLGA) microspheres gave us a bright insight for controling antigen release in a pulsatile fashion, thereby mimicking two or tree boosting injections. However, in spite of the above merits, the level of immunization induced by a single-shot vaccination is often lower tan two doses of alum-adsorbed antigen. Therefore, optima modification of the microsphere is essential for the development of single-step vaccines. In the review, we discuss the stability of antigen in microsphere, safety and non-toxic in human and encapsulation technology. Also, we attempted to outline relevant physicochemical properties on the immunogenicity of microsphere vaccine and attainment of pulsatile release pater by combination of different microsphere, as well as to analyze immunological data associated with antigen delivery by microsphere. Although a lot of variables are related to the optimized microsphere formulation, we could conclude that judicious choice of proper polymer type, adjustment of particles size, and appropriate immunization protocol along with a suitable adjuvant might be a crucial factor for the generation of long-lasting immune response from a single-step vaccine formulation employing PLGA microsphere.
