In this study, we aimed to observe the diversity of aquatic fungi by collecting deposits of soil, plants, and plant litter from streams and rivers. Three Penicillium strains were isolated from soil in streams. Based on the morphological characteristics and phylogenetic analysis using DNA sequences of the internal transcribed spacer, β-tubulin and calmodulin genes, the isolates were identified as Penicillium guanacastense, P. saturniforme, and P. scabrosum. These three fungi have not yet been reported in Korea.
Six interesting fungal strains were isolated during a survey of fungal diversity associated with freshwater; these strains were designated as CNUFC YJW2-22, CNUFC MSW11-6-2, CNUFC HRS5-3, CNUFC MSW242-6, CNUFC DMW2-2, and CNUFC CPWS-1. Based on a polyphasic approach including phylogenetic analyses of internal transcribed space (ITS), large subunit (LSU), beta-tubulin (BenA), and calmodulin (CaM) gene sequences, morphological analyses, the six strains were found to be identical to Acremonium guillematii, Cadophora novi-eboraci, Lectera nordwiniana, Mycoarthris corallina, Talaromyces siamensis, and Tetracladium globosum, respectively. To our knowledge, these are the first records of the rare Lectera, Mycoarthris, and Tetracladium genera in Korea, and the first reports of A. guillematii, C. novi-eboraci, L. nordwiniana, M. corallina, T. siamensis, and Te. globosum in a freshwater environment.
The study characterized Trueperella pyogenes (T. pyogenes) from caseous lymphadenitis (CLA) in Korean native goats (KNG). CLA samples (n=191) were obtained from abattoir-slaughtered KNG. Pus were obtained and Corynebacterium pseudotuberculosis was detected by PCR using PLD gene. Three PLD-negative samples (1.57%) from supramammary lymph node were subjected to bacterial culture. Isolates were identified using 16S rRNA and biochemically characterized by APIZYM. Phylogenetic and biochemical analyses identified the isolates as T. pyogenes, and have shown separate clusters and varied enzyme reaction, suggesting genotypic and phenotypic diversity. This first report of T. pyogenes-induced CLA in KNG provided information for further investigations.
During a survey of indigenous prokaryotic species diversity of the upstream Nakdong River, South Korea, 12 bacterial strains were isolated for further analysis. These bacterial strains were identified showing at least 98.7% 16S rRNA gene sequence similarity with known bacterial species that were previously unreported in South Korea. The 12 bacterial strains were phylogenetically diverse and assigned to four classes, eight orders, nine families, and ten different genera. The isolates were identified as Leucobacter holotrichiae (99.1%), Leucobacter tardus (99.9%), Rhodococcus rhodochrous (99.9%), Tessaracoccus oleiagri (100%), and Paeniglutamicibacter cryotolerans (99.3%), of the class Actinobacteria; Bacillus coagulans (99.7%) and Bacillus wudalianchiensis (99.1%) of the class Bacilli; Ochrobactrum pseudogrignonense (99.2%) and Paracoccus thiocyanatus (100%) of the class Alphaproteobacteria; and Ideonella azotifigens (99.0%), Polaromonas glacialis(99.3%), and Herbaspirillum seropedicae (99.5%) of the class Betaproteobacteria. The cellular and colonial morphology, biochemical properties, and phylogenetic position of these isolates were examined, and species descriptions are provided.

Fungi are cosmopolitan and they occupy diverse niches as consumers, producers, and decomposers. They play critical roles in the environment by enabling nutrient cycling and generating a plethora of secondary metabolites. This study aimed to identify and characterize fungal strains isolated from diverse sources on Muui Island, Republic of Korea. In 2023, a total of 86 fungal strains were collected and examined. Investigation of the morphological features and phylogenetic analyses of multiple barcode loci identified one putative novel species and two species previously unrecorded in the Republic of Korea: Colletotrichum sp., Colletotrichum guizhouense, and Fusarium brachygibbosum. This study provides a comprehensive description of their molecular phylogenies and morphological characteristics. These findings will contribute to the existing knowledge about fungal species in the Republic of Korea and future research on the fungal diversity on Muui Island.
The genus Parornix Spuler, 1910 is a small group within the subfamily Parornichinae belonging to the family Gracillariidae in Korea. The subfamily Parornichinae was recently established as a taxonomic category by Kawahara & Ohshima, 2017, based on the largest dataset in a phylogenetic study. In this study, we reviewed the Korean species of the genus Parornix. In total, 5 species were recognized from Korea. Among them, one species, Parornix loganella (Stainton, 1848) is recorded for the first time from Korea. Descriptions and illustrations of the adults and genitalia of them are provided herein.
This study introduces one specimen each of the genera Gymnopilus, Marasmius, and Mycena as an unrecorded species found in the Hwasun Gotjawal and Jeju Gotjawal Provincial Park of the Hangyeong-Andeok terrain, South Korea. Morphological analyses of macroscopic and microscopic features, and molecular analysis of the nuclear ribosomal internal transcribed spacer region were conducted to identify the specimens. Our results indicated that the newly identified specimens were Gymnopilus dilepis, Marasmius tenuissimus, and Mycena seminau. Here, we describe these newly identified mushroom species and provide baseline data on their diversity in Gotjawal, South Korea.
The diverse microbial communities that colonize distinct segments of the gastrointestinal tract are intimately related to aspects of physiology and the pathology of human health. However, most recent studies have focused on the rectal or fecal microbiota, and the microbial signature of the duodenum is poorly studied. In this study, we compared the microbiota in duodenal and rectal samples to illustrate the characteristic microbial signatures of the duodenum in healthy adults. Nine healthy volunteers donated biopsies and luminal contents from the duodenum and rectum. To determine the composition and diversity of the microbiota, 454-pyrosequencing of bacterial 16S rRNA was performed and multiple bioinformatics analyses were applied. The α-diversity and phylogenetic diversity of the microbiota in the duodenal samples were higher than those of the rectal samples. There was higher biodiversity among the microbiota isolated from rectal biopsies than feces. Proteobacteria were more highly represented in the duodenum than in the rectum, both in the biopsies and in the luminal contents from the healthy volunteers (38.7% versus 12.5%, 33.2% versus 5.0%, respectively). Acinetobacter and Prevotella were dominant in the duodenum, whereas Bacteroides and Prevotella were dominant in the rectum. Additionally, the percentage of OTUs shared in biopsy groups was far higher than in the luminal group (43.0% versus 26.8%) and a greater number of genera was shared among the biopsies than the luminal contents. Duodenal samples demonstrated greater biological diversity and possessed a unique microbial signature compared with the rectum. The mucosa-associated microbiota was more relatively conserved than luminal samples.

We have studied the eukaryotic plankton species diversity to compare the community structure of fresh and brackish waters in the lower reaches of the Nakdong River using metagenomic methods. We constructed 18S rDNA clone libraries of total DNAs extracted from environmental water samples collected at Mulgeum (MG100929, fresh) and Eulsukdo bridge (ES, brackish). Through the steps of colony PCR, PCR-RFLP, sequencing and similarity analysis, we discovered the diverse species composition of eukaryotic plankton. Total 338 clones (170 at MG100929 and 168 at ES) were analyzed, and then we found 74 phylotypes (49 for MG100929 and 25 for ES). From the phylogenetic analysis, we confirmed various eukaryotic plankton of broad range of taxonomic groups, including Stramenopiles, Cryptophyta, Viridiplantae, Alveolata, Rhizaria, Metazoa, and Fungi. We also found several unreported species in Korea and candidates of new taxonomic entities at levels higher than genus. Especially, the cryptic species diversity including unreported phylotypes of Pirsonia (Stramenopiles) and Perkinsea (Alveolata) suggests that the molecular monitoring method can produce new informative biological data in monitoring the changes in the Nakdong River Mouth ecosystem.

Objective: As a charismatic species, cashmere goats have rich genetic resources. In the Inner Mongolia Autonomous Region, there are three cashmere goat varieties named and approved by the state. These goats are renowned for their high cashmere production and superior cashmere quality. Therefore, it is vitally important to protect their genetic resources as they will serve as breeding material for developing new varieties in the future. Methods: Three breeds including Inner Mongolia cashmere goats (IMCG), Hanshan White cashmere goats (HS), and Ujimqin white cashmere goats (WZMQ) were studied. IMCG were of three types: Aerbas (AEBS), Erlangshan (ELS), and Alashan (ALS). Nine DNA samples were collected for each population, and they were genomically re-sequenced to obtain high-depth data. The genetic diversity parameters of each population were estimated to determine selection intensity. Principal component analysis, phylogenetic tree construction and genetic differentiation parameter estimation were performed to determine genetic relationships among populations. Results: Samples from the 45 individuals from the five goat populations were sequenced, and 30,601,671 raw single nucleotide polymorphisms (SNPs) obtained. Then, variant calling was conducted using the reference genome, and 17,214,526 SNPs were retained after quality control. Individual sequencing depth of individuals ranged from 21.13× to 46.18×, with an average of 28.5×. In the AEBS, locus polymorphism (79.28) and expected heterozygosity (0.2554) proportions were the lowest, and the homologous consistency ratio (0.1021) and average inbreeding coefficient (0.1348) were the highest, indicating that this population had strong selection intensity. Conversely, ALS and WZMQ selection intensity was relatively low. Genetic distance between HS and the other four populations was relatively high, and genetic exchange existed among the other four populations. Conclusion: The Inner Mongolia cashmere goat (AEBS type) population has a relatively high selection intensity and a low genetic diversity. The IMCG (ALS type) and WZMQ populations had relatively low selection intensity and high genetic diversity. The genetic distance between HS and the other four populations was relatively high, with a moderate degree of differentiation. Overall, these genetic variations provide a solid foundation for resource identification of Inner Mongolia Autonomous Region cashmere goats in the future.
