Anatomical Characteristics of Three Korean Bamboo Species (국내산 대나무 3종의 해부학적 특성)

  • Bamboo is one of the major biomass resources in the world. To obtain valuable information for effective use of bamboo resources in Korea, the anatomical characteristics of the commercial Korean bamboo species (Phyllostachys pubescens, Phyllostachys nigra, and Phyllostachys bambusoides) were analyzed. The structures in bamboo culm were observed by optical and scanning electron microscopy. Also the crystalline properties as relative crystallinity and crystallite width were measured by an X-ray diffraction method. The three Korean bamboo species had the vascular bundle type I with tylosoid in intercellular space. In the outer part of culm, vascular bundles showed denser spacing than inner part. The fiber length in outer part samples of the three bamboo species showed longer than inner part samples. Furthermore, the fiber length showed a significant difference between inner part and outer part in three bamboo species, showing the longest fiber length in Phyllostachys bambusoides. Phyllostachys pubescens showed the greatest diameter in vessel and parenchyma on cross section. Parenchyma cells in Phyllostachys pubescens and Phyllostachys bambusoides showed similar length and width in both radial and tangential sections. The relative crystallinity and crystallite width in outer part samples of the three bamboo species showed higher values than those in inner part samples, with the greatest values from Phyllostachys bambusoides.

  • This study was performed to determine the effect of Phyllostachys nigra var. henenis Strapf leaf extract on jejunal crypt survival, endogenous spleen colony formation and apoptosis in jejunal crypt cells of mice irradiated with high and low dose of gamma-radiation. Phyllostachys nigra var. henenis Strapf administration before irradiation (I.P.: 125 mg/kg of body weight, at 24 hours before irradiation) resulted in an increase of the formation of endogenous spleen colony (p<0.01). The frequency of radiation-induced apoptosis was also reduced by pretreatment of Phyllostachys nigra var. henenis Strapf (I.P.: 280 mg/kg or 28 mg/kg of body weight, at 24 hours before irradiation, p<0.01). These results indicated that Phyllostachys nigra var. henenis Strapf might be a useful radioprotector, especially since it is a relatively nontoxic natural product. Further studies are needed to characterize better the promotion nature of Phyllostachys nigra var. henenis Strapf and its components.

Developement of Bamboo Strand Board Made from 3 Months Old Domesitic Bamboo Species (3개월생 국산 대나무를 이용한 대나무 스트랜드보드 개발)

  • This study was carried out to determine the suitability of 3 months old bamboo species of Phyllostachys bambusoides S. et Z., Phyllostachys Pubescens Mazel and Phyllostachys nigra var henonis Stapf as raw materials for the manufacture of strandboard. Total of 108 strandboards were made using urea-formaldehyde resin content level of 12% and one percent of liquid wax emulsion. The strandboard consisted of three layers the top and the bottom layer of which were oriented to the same direction and weighted 25% of the strandboard each. The middle core layer weighed 50% of the board and was perpendicular to the outer top and bottom layers. Analysis was performed to determine the effect of strand lengths and Uowing years of 3 months, 2 years and 3 years on strandboard properties. The physical and mechanical properties of bamboo species and boards were measured and compared to the standard requirements of strandboards. The results are as follows; 1. The more the growing years the higher the density of bamboo. Top part of bamboo indicated higher density value than that of bottom part. 2. Bamboo showed higher static bending strength compared to the main wood species. Longer growing years of bamboo generally inclosed the static tending strength out there were no statistical significancies for Phyllostachys bmbusoides S. et Z. and Phyllostachys pubescens Mazel. 3. Strand length indicated no difference on density and moisture content of strandboard. 5 cm of strand length gave the best static bending strength and internal bonding strength. Bamboo strandboard exhibited lesser extents of thickness swelling than that of CSA standard. 4. 3 months old bamboo gave higher static bending strength of strandboard than those of 2 years and 3 years old bamboo. In case of Phyllostachys nigra var henonis Stapf, 3 months old bamboo indicated higher internal bonding strength than those of 2 years and 3years old bamboo. but in Phyllostachys bambusoides S. et Z., Phyllostachys pubescens Mazel, there were no difference among growing years. Growing years showed no different physical properties of bamboo strandboard.

Phylogenic Study of Genus Phyllostachys (Phyllostachys) in Korea by Internal Transcribed Spacer Sequence (ITS) (ITS 서열에 의한 한국 왕대속 식물종의 계통분류학적 연구)

  • Phyllostachys consists of high and fast growing trees and is a genus in the family Gramineae. The genus has many species in Asia, with main distribution being in India and China. One of the most popular sequences for phylogenetic inference at the generic and infrageneric levels in plants is the internal transcribed spacer (ITS) region of the 18S-5.8S-26S nuclear ribosomal cistron. We evaluated four taxa with the ITS region to estimate phenotypic relationships within the genus Phyllostachys in Korea. Alignment of the DNA sequences required the addition of numerous gaps. Sequence variation within the Phyllostachys was mostly due to natural selection, although several indels and inserts were found. Within the genus Phyllostachys, P. nigra and P. nigra var. henonis were the relatives in the three phylogenetic analyses (MP, ML, and NJ). However, some external nodes were poorly supported. Morphological traits and simple repeats (ISSR) represented the result of a relationship similar to the that of ITS sequences in the genus Phyllostachys. This suggests that ITS sequences are very informative for identification of these taxa.

  • In order to increase the shelf-life of Phyllostachys pubescens sap heating conditions were investigated. To evaluate the quality changes of Phyllostachys pubescens sap before and after heating at $70^{\circ}C$ for 30 min, pH, total microbial number and sensory characteristics were measured during storage periods at $2^{\circ}C$. During 10 days, the quality and sensory characteristics of Phyllostachys pubescens sap were maintained.

  • The leaf of Phyllostachys bambusoides S. et Z. (Gramineae) has been used in traditional herbal medicine as a antipyretics, antitussives and antidiuretcs, etc, in Korea, China and Japan. In order to investigate the effects on antilipoperoxidation and liver protective effect, the leaf of Phyllostachys bambusoides S. et Z. was extracted by water and then fractionated with butanol. The water extract and BuOH soluble fraction strongly exhibited antilipoperoxidatant effect in rat liver homogenate intoxicated with $CCl_4$. The BuOH fraction significantly suppressed the increases of s-GOT, s-GPT and s-LDH activities in injuried rats induced by $CCl_4$. And the BuOH fraction inhibited significantly the decrease of the body weight and showed the antilipoperoxidatant effect in liver and kidney in $CCl_4$ intoxicated rats.

Ultrasonic Properties of Phyllostachys bambusoides Sieb. et Zucc depending on Three directions (왕대의 3방향에 따른 초음파적 특성)

  • This research was carried out to examine the ultrasonic properties of $Phyllostachys$ $bambusoids$ Sieb et Zucc depending on three directions for providing the fundamental properties of the Korea traditional flute, Daekeum. The ultrasonic properties of $Phyllostachys$ $bambusoids$ Sieb. et Zucc were found to be different from those of wood, because of the distinctive anatomical structure of bamboo tissue. The ultrasonic velocity of peripheral zone showed higher values of all three directions. The ratio of longitudinal velocities to perpendicular velocities showed 1.16 at peripheral zone, 1.70 at central zone, 1.38 at inner zone respectively. The ratio of radial velocities to tangential velocities were measured as 1.286 at peripheral zone, 1.325 at central zone, 0.829 at inner zone respectively.

Genetic Diversity and Phylogenetic Relationship of Genus Phyllostachys by ISSR Markers (ISSR 분자 마커를 이용한 왕대속 대나무의 유전적 다양성 및 계통 관계)

  • Four taxa of the genus Phyllostachys were analysed with PCR-based molecular inter-simple sequence repeat (ISSR) determine markers to determine their phylogenetic relationships. Many species of this genus are regarded as economical and ecologically important in the world. With the nine primers screened, ISSR assay generated a total of 64 reproducible bands. Analysis of ISSR from individual plants of genus Phyllostachys resulted in 43 polymorphic bands with 70.49%. When species were grouped by four taxa, within group diversity was 0.092 (Hs), while among group diversify was 0.499 (Gst) on a per locus basis. The estimated gene flow (Nm) between the pairs of species was 0.559. Hence, we can expect weak or low gene flow among species. Total mean genetic diversity ($H_T$) for four taxa was 0.291. The ISSR data allowed us to resolve well-supported clades in the genus Phyllostachys. The four taxa of the genus Phyllostachys analyzed were distinctly related to a monophyletic.

Comparisons of Nutrient Concentration of Leaves, Roots, and Soils in Three Bamboo Stands

  • In bamboo, the nutrient status of tissues and associated soil is an important indicator of nutrient uptake by various bamboo species. In this study, the nutrient concentrations of leaves, roots, and mineral soil at 0-10 cm depths were examined in three bamboo stands [Phyllostachys bambusoides S ieb. et Zucc, Phyllostachys nigra var. henonis Stapf ex. Rendle, and Phyllostachys pubescens (Mazel) Ohwi] at a broad regional scale in southern Korea. In the three bamboo species, species-specific differences were observed in the carbon (C) and calcium (Ca) concentrations of leaves and in the nitrogen (N) and magnesium (Mg) concentrations of roots. Ca concentrations in leaves were significantly higher in P. bambusoides (11.94 g Ca kg-1) than in P. pubescens (7.83 g Ca kg-1), whereas potassium (K) concentrations were lowest in P. bambusoides among the three bamboo species. N concentrations in the roots were significantly lower in P. pubescens (5.23 g N kg-1) than in P. nigra var. henonis (7.72 g N kg-1). In contrast to bamboo tissues, soil nutrients, such as total N, organic C, exchangeable Ca2+, and Mg2+, did not differ significantly among the bamboo species. These results suggest that species-specific practices will be required for nutrient management of bamboo stands because nutrient concentrations vary considerably in the tissues of the three studied species.

Study on the Machinability of Korean Domestic Bamboo Species (국산 죽재의 기계 가공성에 관한 연구)

  • Machinability of three Korean domestic bamboo species, Phyllostachys pubescens Mazel, Phyllostachys nigra var. henonis Stapf. and Phyllostachys bambusoides S. et Z., was studied to investigate the possibility of utilization of Korean domestic bamboo species as raw material for industry. Splitting force reached a maximum within the first inch of knife travel and diminished thereafter. Parallel-sided wedge-type knife with sharpness angle of 15 degree required the lowest maximum splitting force. Roughnesses of surfaces developed during planing and sawing of bamboo were lower than those of Pinus radiata D. Don and quality of ripped surface was better than that of cross-cut surface on bamboo. 62.5 hours were required to dry 12mm-thick Phyllostachys pubescens Mazel from moisture content of 60% to moisture content of 10% at $70^{\circ}C$.
