The effects of Gamisipjeontang-gamibang on injured tissue after burn elicitation (가미십전탕(加味十全湯) 가미방(加味方)이 화상(火傷)의 조직 손상에 미치는 영향)

  • Objective : The purpose of this study is to examine the effects of Gamisipjeontang-gamibang(GT) on an experimental bum elicitation. Method : The absorbance of the photo cell mixed with GT at the Abs 560 nm was measured after irradiation for 1 min. In order to know the antioxidant effect on skin cell of mice after burn elicitation, superoxide dismutase(SOD) activity was measured. Also, in order to know effects on the skin regeneration in the burned mice, we counted the eosinophil in blood from animals via cardiac puncture and observed the histological structure in the epidermal basal layer and the dermal section on the 3rd, 7th and 14th day after burn elicitation. We also studied changes in angiogenesis in the capillary surrounding the basal layer and dermal papilla. The changes of HSP70 distribution and changes of p53 positive reaction were observed to investigate the changes of the stress in the skin as well. Result : The results indicated that GT has a significant impact on the antioxidant effect on skin cells of mice after bum elicitation by increasing SOD activity in vitro test. GT seems to decrease MIP-2 which induces neutrophil infiltration and promotes the angiogenesis dose-dependently. Furthermore, GT decreased HSP70, the expression of which was induced by elevated temperatures, and p53 which induce a apoptosis after stress. Conclusion : GT can be applied to burned skin through its antioxidant effect and skin regeneration property.

Analysis on Werner Bischof's Korean War Documentary Photos (베르너 비숍의 한국전쟁 다큐멘터리 사진 분석)

  • This research analyzed military journalist activities and key photos of Werner Bischof, the military journalist during Korean war. Werner Bischof entered Korea twice in 1951 and 1952 for military journals. Through Korean war pictures, essay, multiple letters and photobooks, I shed a light on the background of his military journals and activities, and analyzed topics and characteristics based on key photos that he took during his first and second military journalist activities. Bischof created a work centered on 'Human'. Especially, he has this consciousness of 'What happens to civilians in war-ridden area?'. Bischof took civilians in Sanyangri in his camera to announce pain of civilians and tragedy of war coming from war during his first military journalist activity. During his second military journalist activity, he critiqued ideological brainwashing, life of prisoners of war in a humanist point of view. The latter showed characteristics of clear contrast of black and white, image arrangement based on characters, and outstanding photo views. His military journal activity is in search of civilian suffering, and humanism in Korean war. This consciousness crosses through generation over generation that cannot be compared to other military journalists.

Protective Effects of Complex Diet of Korean red ginseng and fish oil on UV-induced Photoaging (홍삼과 어유(魚油) 복합식이에 의한 UV-유도 광노화 억제효과)

  • Objective : UV irradiatiion causes skin-aging involving coarse wrinkles, thickning, dyspigmentation, and rough skin surface. This study was aimed to elucidate the anti-winkle activity of complex diet of Korean red ginseng (RG) and fish oil (FO) on UV-induced Photoaging. Methods : To investigated photo-protective effects of Korean red ginseng and fish oil on UV-induced damaged skins, SKH hairless female mice were randomly divided into six groups [control, UV, UV/RG, UV/FO, UV/RG/FO(low), UV/RG/FO(high)]and orally administered three times a week respectively. UV radiation was applied to the backs of the mice three times a week for 8 weeks. Expressions of matrix metalloproteinase (MMP)-3, MMP-13 and tissue inhibitor matrix metalloproteinase (TIMP)-1 in skin were measured by immunohistochemical staining. Results : In this study, UVB-induced epidermal hypertropy was diminished by RG group or FO group or complex group of RG and FO. Expression levels of MMP-3 and MMP-13 were reduced and expression level of TIMP-1 was increased by RG group or FO group or complex group of RG and FO. Especially MMP-3 and MMP-13 were markedly reduced by diet of FO and complex diet of RG and FO compared with untreated group. Conclusions : This results suggest that complex diet of RG and FO have a anti-wrinkle activity on UV-induced photo-aging and intrinsic aging.

Fabrication of TiO2 Impregnated Stainless Steel Fiber Photocatalyts and Evaluation of Photocatalytic Activity (TiO2 담지 스테인리스 강 섬유 광촉매 제조 및 광촉매 활성 평가)

  • $TiO_2$ impregnated stainless steel fiber photocatalysts ($TiO_2/SSF$) were fabricated to overcome inherent problems of powdery $TiO_2$ photocatalysts in water treatment. Adhesion strength of the impregnated $TiO_2$ was examined using an ultrasonic-cleaner. Photocatalytic activity was evaluated through decomposition experiment of methylene blue and formic acid. Bactericidal efficiency was evaluated through sterilization experiment of E. Coli and Vibrio Vulnificus. Adhesion strength of the impregnated $TiO_2$ was so high that more than 95% was left over even after the treatment in an ultrasonic-cleaner for 30 min. Methylene blue and formic acid were decomposed as much as 60% and 38% of the initial concentration and more than 99.9% of E. Coli and Vibrio Vulnificus were killed after 1 hour exposure to the prepared photocatalyst under UV irradiation. In the case of decomposition of formic acid, decomposition ratio increased if oxidants were added. Especially the decomposition ratio increased as high as 80% when hydrogen peroxide was added as an oxidant.

New Synthesis of the Ternary Type Bi2WO6-GO-TiO2 Nanocomposites by the Hydrothermal Method for the Improvement of the Photo-catalytic Effect (개선된 광촉매 효과를 위한 수열법에 의한 삼원계 Bi2WO6-GO-TiO2 나노복합체의 쉬운 합성 방법)

  • A novel material, $Bi_2WO_6-GO-TiO_2$ composite, was successfully synthesized using a facile hydrothermal method. During the hydrothermal reaction, the loading of $Bi_2WO_6$ and $TiO_2$ nanoparticles onto graphene sheets was achieved. The obtained $Bi_2WO_{6-GO-TiO2}$ composite photo-catalysts were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive X-ray (EDX) analysis, transmission electron microscopy (TEM), Raman spectroscopy, ultraviolet-visible diffuse reflectance spectroscopy (UV-vis-DRS), and X-ray photoelectron spectroscopy (XPS). The $Bi_2WO_6$ nanoparticle showed an irregular dark-square block nanoplate shape, while $TiO_2$ nanoparticles covered the surface of the graphene sheets with a quantum dot size. The degradation of rhodamine B (RhB), methylene blue trihydrate (MB), and reactive black B (RBB) dyes in an aqueous solution with different initial amount of catalysts was observed by UV spectrophotometry after measuring the decrease in the concentration. As a result, the $Bi_2WO_6-GO-TiO_2$ composite showed good decolorization activity with MB solution under visible light. The $Bi_2WO_6-GO-TiO_2$ composite is expected to become a new potential material for decolorization activity. Photocatalytic reactions with different photocatalysts were explained by the Langmuir-Hinshelwood model and a band theory.

Inhibitory Effects of Lespedeza cuneata Ethanol Extract on Ultraviolet-Induced Photo Aging (야관문 에탄올 추출물의 자외선 조사에 의한 피부 광노화 억제 효과)

  • To investigate the inhibitory effects of Lespedeza cuneataextract (LC) on photo aging, skin aging was induced by UVB irradiation of hairless mice for 5 weeks. The skin erythema indices for the positive control (PC) and Lespedeza cuneata extract (LC-1%, 3%, and 5%) groups were lower than that of the control group. However, both lipid and water capacities of the PC and LC-3% groups were significantly higher than those of the control group. Skin wrinkles in the PC and LC-3% groups formed in a pattern of shallow furrows and thin crests. Skin TBARS contents and XO activity in the LC group were lower than those of the control group. The activities of GSH, SOD, and CAT in the LC-3% group were significant higher than those of the control group. Therefore, Lespedeza cuneata extract could be an effective natural herbal material for the inhibition of skin aging in hairless mice skin.

The Effect of Salicylic Acid (SA) on the Photosynthetic Activity in Commelina communis L. (Salicylic acid가 닭의장풀의 광합성에 미치는 영향)

  • The effect of Salicylic acid (SA) on the leaf growth, chlorophyll content, photosynthesis, stomatal conductance in Commelina communis L. was investigated. C. communis which was grown in Hoagland solution containing 1 mM SA during 3 weeks resulted in a significant reduction of leaf length, width and area. 1 mM SA reduced about 10% of the leaf area at 1 week and 2 weeks, but inhibited 22% of the leaf area at 3 weeks. SA also showed great effect on the reduction of chlorophyll content. SA reduced 47% and 53% of chlorophyll content each at 2 weeks and 3 weeks when it was compared with the control. Chlorophyll a/b ratio in SA treated sample was increased at 3 weeks representing that SA was sensitive to chlorophyll b. The treatment of SA did not inhibit the activity of PSII+PSI, PSII and PSI. This result indicates that SA did not show direct effect oil the photosynthetic electron transport activity. However, when C. communis was grown in Hoagland solution containing I mM SA fer long period, SA showed clear inhibition of photosyhthesis in intact leaves. The treatment of SA for 3 weeks showed about 23~65% inhibition of photosynthetic activity at various light intensity (100~1000$\mu$mo1 m­$^2$$^1$). Similar effect was found on stomatal conductance. The treatment of SA caused an almost closure of stomata. Similar effects of stomatal conductance at various light intensity indicate a loss of normal control on stomatal mechanism. These results indicate that the effect of SA on photo-synthetic activity was not by SA itself but by indirect metabolic pathway.

Effects of Ethyl Acetate Fraction from Melothria Heterophylla on Antioxidant Activity and Matrix Metalloproteinase-1 Expression in Ultraviolet A-irradiated Human Dermal Fibroblasts (백렴 에틸 아세테이트 층의 항산화 활성과 Matrix Metalloproteinase-1 발현 저해효과)

  • UV irradiation leads to distinct changes in skin connective tissue, which is degradation of collagen. Many of these alterations in the extracellular matrix are mediated by matrix metalloproteinases. In this study, to develop a new anti-aging agent, we screened the antioxidant activity of solvent fractions from ethanolic extract of Melothria Heterophylla. Among the four solvent fractions tested, the EtOAc fraction exhibited the highest antioxidant activity. It was investigated the inhibitory effect of the EtOAc fraction on the expression and activity of MMP-1 in UVA-irradiated human dermal fibroblasts. The EtOAc fraction inhibited the activity of MMP-1 in a dose dependent manner with the $IC_{50}$ values of $9{\mu}g/mL$. Also, UVA-induced MMP-1 expression was reduced about $90\%$ by $100{\mu}g/mL$ of the EtOAc fraction but MMP-1 mRNA expression was not inhibited. Therefore, we conclude that the EtOAc fraction significantly inhibits MMP-1 expression at the protein level. From these results, we suggest that the EtOAc fraction from M. heterophylla could be used as a new anti-aging agent for the photo-damaged skin.

  • UV-B radiation gives harmful effects on plants, such as production of several types of DNA lesions, and growth inhibition. On the other hand, plants have some protective mechanisms, including filtering effect due to accumulation of phenolic compounds in epidermal cells and reactivation of DNA lesions, which are enhanced by UV-B irradiation. We have investigated the mechanism of UV-B effects on plants using cucumber seedlings as plant materials. Cucumber plants were cultivated in an artificially lit growth chamber. Supplemental UV-B irradiation, of which intensity was almost equal to the level of natural sunlight, retarded the growth of first leaves. The growth retardation must result trom the inhibition of cell division and/or cell growth. Microscopical observation of leaf epidermis suggested that the growth retardation might be mainly caused by cell growth inhibition. The retardation was, however, restored within 2 or 3 days after the termination of UV-B irradiation. It is known that UV-B irradiation lowers the activity of photo system II (PS II). In the present experimental conditions, however, UV-B irradiation has little effect on PS II activity as estimated by chlorophyll fluorescence. The stomatal conductance, a major factor determining photosynthetic rate, of first leaves increased during the growth. The increase of stomatal conductance was suppressed by UV-B irradiation and restored by termination of the irradiation. It has not been clear, however, what mechanisms are involved in the suppression of increase of stomatal conductance.

Anti-wrinkle Activity of Acanthopanax senticosus Extract in Ultraviolet B (UVB)-induced Photoaging (가시오가피 추출물의 광노화에 의한 주름형성 억제 효과)

  • This study was performed to investigate anti-wrinkle effects of Acanthopanax senticosus (AS) on ultraviolet B (UVB)-induced photoaging with wrinkle formation. AS extract showed higher DPPH radical scavenging activity (3 ${\mu}g/mL$ as $IC_{50}$) and collagenase inhibition (1.52 mg/mL as $IC_{50}$) than those of ascorbic acid (50 ${\mu}g/mL$ and 2.17 mg/mL, respectively). Cell proliferation and type I pN collagen synthesis were increased by 11.4% and 96.4%, respectively, compared with non treatment control. In vivo, SKH-1 hairless mice were administrated AS 400 mg/kg for 10 weeks with UVB irradiation three times a week. After 10 weeks, a visual assessment and replica assay were performed on each mouse. According to visual assessment of close-up photos and skin replica, oral administration of A. senticosus affected on inhibition of wrinkle formation caused by UVB irradiation on the skin of mice as compared to the vehicle treated control mice. These results indicated that A. senticosus could protect skin wrinkle formation caused by collagen synthesis of fibroblast cells and photo-irradiation of UVB in hairless mice.
