광염색체이상시험의 광발암성 예측능력에 대한 평가 (Assessment of Sensitivity of Photo-Chromosomal Assay in the Prediction of Photo-carcinogenicity)

  • Photo-mutagenic compounds have been known to alter skin cancer rates by acting as initiators or by affecting subsequent steps in carcinogenesis. The objectives of this study are to investigate the utility of photo-chromosomal aberration (photo-CA) assay for detecting photo-clastogens, and to evaluate its ability to predict rodent photocarcinogenicity. Photo-CA assay was performed with five test substances that demonstrated positive results in photo-carcinogenicity tests: 8-Methoxypsoralen (photoactive substance that forms DNA adducts in the presence of ultraviolet A irradiation), chlorpromazine (an aliphatic phenothiazine an alpha-adrenergic blocking agent), lomefloxacin (an antibiotic in a class of drugs called fluoroquinolones), anthracene (a tricyclic aromatic hydrocarbon a basic substance for production of anthraquinone, dyes, pigments, insecticides, wood preservatives and coating materials) and Retinoic acid (a retinoid compound closely related to vitamin A). For the best discrimination between the test substance-mediated genotoxicity and the undesirable genotoxicity caused by direct DNA absorption, a UV dose-response of the cells in the absence of the test substances was firstly analyzed. All 5 test substances showed a positive outcome in photo-CA assay, indicating that the photo-CA test is very sensitive to the photo-genotoxic effect of UV irradiation. With this limited data-set, an investigation into the predictive value of this photo-CA test for determining the photo-carcinogenicity showed that photo-CA assay has the high ability of a test to predict carcinogenicity. Therefore, the photo-CA test using mammalian cells seems to be a sensitive method to evaluate the photo-carcinogenic potential of new compounds.

황기의 자외선에 의한 세포 손상을 막는 보호 효과 (The Protective Effects of Astragali Radix Against UV Induced Cellular Damage in Human Keratinocytes)

  • The root of Astragalus membranaceus Bunge (Leguminosae) has been used in the Korean oriental medicine for strengthening the vital energy. UV irradiation has been suggested as a major cause of photo aging in skin. In order to investigate protective effects against UV induced cellular damage, Astragali Radix was extracted with 70% ethanol and dissolved in DMSO. The protective effect was detected by MTT assay, LDH assay, and Comet assay in immortalized human keratinocyte cell line, HaCaT cell system after UV irradiation. Astragli Radix 70% EtOH extract reduced UV induced cellular damage in cell survival, membrane integrity and DNA damage.

Photo-protective and Anti-melanogenic Effect from Phenolic Compound of Olive Leaf (Olea europaea L. var. Kalamata) Extracts on the Immortalized Human Keratinocytes and B16F1 Melanoma Cells

  • Ethylacetate and butanol fractions of leaf extracts (OLE) showed the higher contents of total phenolic compounds than hexane and water fractions. Oleuropein contents were $4.21{\pm}0.57,\;3.92{\pm}0.43,\;0.32{\pm}0.03,\;5.76{\pm}0.32$, and $32.47{\pm}0.25mg$/100g for ethanol extract, and hexane, chloroform, ethyl acetate, and butanol fraction, respectively. Treatment of ultraviolet-B (UVB) irradiated cells with 3 OLEs prepared by using ethylacetate and butanol at concentrations 0.001, 0.005, and 0.01% respectively showed significant recovery of cell viabilities. Treatment of dexametason 1 mM reduced tumor necrotic factor (TNF)-${\alpha}$ secretion by about 40%. UVB irradiated immortalized human keratinocytes (HaCaT) cells were treated with 3 different OLEs at the same concentrations. Ethylacetate fraction showed the strongest inhibition activity with respect of reduction of the elevated (TNF)-${\alpha}$. Cytotoxicity of OLEs on the B16-F1 cells was evaluated through thiazolyl blue tetrazolium bromide (MTT) assay. Ethylacetate fraction has no cytotoxicity in the range of 0.005-0.01%. A slight cytotoxicity was observed at the concentration of 0.1% butanol fraction of OLE that caused 10% decrease in cell viability.

Skin protective effect of Indian gooseberry and barley sprout complex on skin dryness, wrinkles, and melanogenesis by cell models

  • BACKGROUND/OBJECTIVES: UV radiation is a major factor contributing to DNA damage in skin cells, including stem cells and mesenchymal stem cells, leading to the depletion of these crucial cells. This study examined whether a mixture of Indian gooseberry and barley sprout (IB) could inhibit UVB irradiation and 3-isobutyl-1-methylxanthine (IBMX)-induced photoaging and oxidative stress in the skin using HaCaT, Hs27, and B16F10 cells. MATERIALS/METHODS: The moisturizing-related factors, the collagen synthesis-related c-Jun N-terminal kinase (JNK)/c-Fos/c-Jun/matrix metalloproteinases (MMPs) pathway, and the melanogenesis-related cyclic adenosine monophosphate (cAMP)/protein kinase A (PKA)/cAMP-responsive binding protein (CREB)/melanocyte inducing transcription factor (MITF)/tyrosinase-related protein (TRP)/tyrosinase activation pathways were analyzed in vitro by an enzyme-linked immunosorbent assay, real-time polymerase chain reaction, and Western blot analysis. RESULTS: The IB complex increased the hyaluronic acid and sphingomyelin levels and the collagenase inhibitory activity, enhanced hydration-related factors, including collagen, hyaluronic acid synthase (HAS), elastin, long chain base subunit 1 (LCB1) (serine palmitoyltransferase; SPT), and delta 4-desaturase sphingolipid 1 (DEGS1), modulated the inflammatory cytokines levels, antioxidant enzyme activities and the NF-κB/MMPs/cyclooxygenase-2 (COX-2) pathway in UVB-irradiated HaCaT cells, and inhibited wrinkle formation by down-regulation of the JNK/c-Fos/c-Jun/MMP pathway and up-regulation of the transforming growth factor-𝛽 receptor I (TGF𝛽R1)/small mothers against decapentaplegic homolog (Smad3)/procollagen type I pathway in UVB-irradiated Hs27 cells. Moreover, the IB complex prevented melanin production by down-regulating the PKA/CREB/MITF/TRP-1/TRP-2 pathway in IBMX-induced B16F10 cells. CONCLUSION: These findings suggest that the IB complex has the potential to serve as a safeguard, shielding the skin from UVB radiation-induced photo-damage.

약용식물추출물의 광보호 효과와 항염증 효과 연구 (Photoprotection and Anti-inflammatory Effects of Chinese Medical Plants)

  • 자외선이 피부에 지속적으로 조사되면 썬번, 염증, 광노화와 같은 다양한 부작용이 생기게 된다. 따라서 광손상에 대한 보호작용은 점점 중요성이 인식되고 있으며, 천연 식물추출물에서 자외선에 대한 광보호효과가 있는 안전하고 효과적인 물질개발에 대한 연구가 진행되고 있다. 본 연구는 자외선에 의해 유도된 피부손상에 대한 보호효과가 우수한 천연식물추출물에 대한 것이다. 향나무, 능소화, 비자추출물에서 자외선에 의해 생성되는 라디칼에 대한 항산화효과, MMP 발현 및 활성 저해, 염증관련 사이토카인인 인터루킨 1알파, 6, 프로스타글란딘 E$_2$ 생성저해효과 등으로 피부세포보호 효과를 연구하였다. 실험결과 천연추출물 중 향나무추출물과 능소화추출물이 프리라디칼 및 슈퍼옥사이드 라디칼 소거효과가 우수하게 나타났으며 피부 콜라겐과 같은 메트릭스를 분해하는 효소인 MMP-1의 활성 및 발현 저해효과는 섬유아세포에서 UVA 조사에 의한 실험에서 우수하게 나타났으며, 피부세포 배양액에 대해 zymography를 실시하여 활성이 감소됨을 확인하였다. 피부 각질형성세포에서 자외선에 의해 유도된 염증관련 사이토카인인 인터루킨 6의 발현량 실험에서도 무처리군에 비해 향나무추출물이 인터루킨 6을 30% 정도 저해효과가 나타났으며, 염증반응 중 cyclooxygenase(COX)에 의한 경로에서 생성되는 프로스타글란딘 E$_2$의 생성량도 감소시켰다. 사람피부에서 SLS (0.5%) 첩포로 유발된 자극성 피부염의 항염증 효과 평가 시 SLS에 의해 유발되는 자극정도가 피검자의 피부상태에 따라 다양하게 나타났으며, 향나무 추출물 함유 에멀젼 제품 도포 실험에서는 피부 홍반 완화 효과와 피부장벽 회복효과가 우수하게 나타났다. 이상의 결과로 향나무추출물은 자외선 조사 및 SLS에 의한 피부손상에 대한 피부세포보호작용이 우수하여 광노화에 대응하는 자극완화 소재로서의 화장품 응용 가능성을 확인하였다.
