Mycoplasma hyopneumoniae Induces Grap, Gadd45β, and secreted phosphoprotein 1 Gene Expression as Part of the Inflammatory Response in RAW264.7 Cells

  • Genes related to Mycoplasma hyopneumoniae-induced inflammation were identified using the genefishing technology, an improved method for identifying differentially expressed genes (DEGs) using an annealing control primer (ACP) system in RAW264.7 cells. After treatment with M. hyopneumoniae, 16 DEGs were expressed in RAW264.7 cells using a pre-screening system. Among these 16 DEGs, 11 DEGs (DEGs 1, 4, 5-10, 12-15) were selected and sequenced directly, revealing that DEG12 (Grap), DEG14 (Gadd45), and DEG15 (secreted phosphoprotein 1) were related to inflammatory cytokines. This is the first report that intact M. hyopneumoniae induces the expression of Grap, Gadd 45${\beta}$, and secreted phosphoprotein 1 in RAW264.7 cells. Subsequently, these genes may be targets for screening novel inhibitors of the mycoplasmal inflammatory response.

The Role of SH2 Domain-containing Leukocyte Phosphoprotein of 76 kDa in the Regulation of Immune Cell Development and Function

  • Recent years have seen an explosion of new knowledge defining the molecular events that are critical for development and activation of immune cells. Much of this new information has come from a careful molecular dissection of key signal transduction pathways that are initiated when immune cell receptors are engaged. In addition to the receptors themselves and critical effector molecules, these signaling pathways depend on adapters, proteins that have no intrinsic effector function but serve instead as scaffolds to nucleate multimolecular complexes. This review summarizes some of what has been learned about one such adapter protein, SH2 domain-containing leukocyte phosphoprotein of 76 kDa (SLP-76), and how it regulates and integrates signals after engagement of immunoreceptors and integrins on various immune cell lineages.

  • 진균 속에 속하는 종인 Cordyceps는 중국의 전통약제로서, 그 유효성분인 cordycepin이 혈소판 응집에 관여한다는 보고가 있지만 phosphoprotein 조절에 관련된 연구는 미흡하다. 본 연구에서는, cordycepin이 fibrinogen binding에 관여한다고 알려진 PI3k/Akt와 $TXA_2$ 분비 및 과립방출에 관여한다고 알려진 p38와 같은 phosphoprotein의 인산화를 어떻게 조절하며 혈소판응집을 억제시키는지 규명하고자 하였다. 그 결과, cordycepin가 $261.1{\mu}M$$IC_{50}$으로 collagen이 유도한 혈소판 응집을 강력하게 억제하였고, PI3K와 Akt의 인산화를 감소시키며 ${\alpha}IIb/{\beta}_3$에 대한 fibrinogen 결합을 농도의존적으로 억제하였다. 또한, cordycepin은 collagen이 촉진시킨 p38의 인산화를 억제함으로써, 과립방출의 지표인 ATP 과 serotonin의 방출을 억제하였고 COX-1과 TXAS의 활성 및 $PLC-{\gamma}_2$ 인산화에 대한 영향없이 $TXA_2$ 생성량을 감소시켰다. 따라서, cordycepin은 PI3K/Akt, p38와 같은 phosphoprotein의 인산화를 억제함으로써 혈소판 응집억제를 나타내는 항혈전 치료 및 예방약물로서 유용한 가치가 있다고 여겨진다.

Identification of nucleolar and coiled-body phosphoprotein 1 (NOLC1) minimal promoter regulated by NF-κB and CREB

  • Nucleolar and coiled-body phosphoprotein 1 (NOLC1) is a phosphoprotein that transiently associates with the mature nucleolar H/ACA and C/D box small nucleolar ribonucleoproteins (snoRNPs). Several lines of evidence indicate that NOLC1 plays an important role in the synthesis of rRNA and the biosynthesis of ribosomes. In the present study, we examined the transcriptional regulation mechanisms that govern the expression of NOLC1. We first performed functional dissection of the NOLC1 promoter. We demonstrated that transcription factors NF-${\kappa}B$ and CREB could bind to the minimal NOLC1 promoter. This was demonstrated by electrophoretic mobility shift assays and chromatin immunoprecipitation. Mutagenesis and overexpression assays revealed that NF-${\kappa}B$ and CREB positively regulated the NOLC1 promoter. These findings may provide new insight into the mechanisms that regulate NOLC1 expression.

  • The overexpression of Phosphoprotein Enriched in Astrocytes (PEA15) gene is commonly found in human diabetic patients. The overexpression of this gene in skeletal muscle and fat tissues have been reported to cause insulin resistance, thereby impairing insulin stimulated glucose uptake. We introduced a gene of mouse PEA15 (mPEA15) and enhanced green fluorescent protein (EGFP) into fertilized one cell pig zygotes using microinjection, and produced a piglet that showed overexpression of mPEA15 in the muscle tissues and expression of EGFP in the ear tissues and hooves. RT-PCR RFLP, southern blot and FISH analysis showed that the tissues carried the transgene. Real-time RT-PCR and western blots demonstrated that PEA15 gene was overexpressed in the various tissues and muscle tissues, respectively. These fads suggest that expression vector system is normally expressed in the transgenic (TG) pigs. To use as animal diseases model for type 2 diabetes, further study is necessary to confirm whether diabetes occur in these TG pigs, especially insulin resistance.

Golgi Phosphoprotein 2 Down-regulates the Th1 Response in Human Gastric Cancer Cells by Suppressing IL-12A

  • Golgi phosphoprotein 2 (GOLPH2) is a very important biomarker in a variety of diseases. Its biological function is not clear, particularly in gastric cancer. To investigate the role of GOLPH2 in human gastric cancer, and determine its effect on the Th1 lymphocyte response, its expression and that of IL-12A were measured by real-time PCR and immunohistochemistry. The relationship between GOLPH2 and IL-12A was analysed statistically. The effect of GOLPH2 on the Th1 lymphocyte response was investigated with an in vitro co-culture system. The results showed that in human gastric cancer, the expression of GOLPH2 was significantly higher and the expression of IL-12A was lower than in normal gastric mucosal tissues, and the expression levels of GOLPH2 and IL-12A were negatively correlated. In addition, obvious down-regulation of the Th1 response was observed when lymphocytes were co-cultured with gastric cancer SGC7901 cells over-expressing GOLPH2. GOLPH2 down-regulated the expression of IL-12A, and inhibited the expression of TNF-${\alpha}$ and IFN-${\gamma}$. The results indicated that GOLPH2 down-regulates the Th1 response via suppression of IL-12A in human gastric cancer, and this might provide a target for the prevention and treatment.

  • Fe(III) IDA -PEG/ dextran 액 이상 분계 시스템에서 phosphoprotein의 분배 예측을 위한 모델식이 유도되었다. 본 모델식에는 단백칠 분배에 수소이온과 hydroxide 이온의 저해효과가 포함되어 었다. 표변에 각각 1,27~의 phosphoryl 그 룹을 갖고 있는 ovalbumin의 AI, A2, 성분을 정제한 후 단백질 분배 실험을 수행하였으며 실 험결과와 모텔식에의한 예측이 잘 얼치하는 것 을 확인하였다. Fe(III)IDA-PEG와 단백질 표면 p phosphoryl group과의 결합상수는 PEG 상과 dextran상에 셔 각각 $6.1{\times}, 10^3M^{-1} and 2.3{\times}10^4M^{-1}$이었으며,이것은 Cu(II)IDA-PEG와 단백질 표면 histidine과의 결합상수 보다 3-5배 높은 값이 였 다.

심근세포로의 분화에 관여하는 새로운 생리활성 단백질 SPP2의 발굴 (Identification and Characterization of Secreted Phosphoprotein 2 as a Novel Bioactive Protein for Myocardial Differentiation)

  • 심장 발생과정에 관여하는 주요 전사인자들의 기능에 대한 규명 등의 발전에도 불구하고 줄기 세포에서 매우 효율적인 심근 세포로의 분화를 촉진하는 새로운 생체 활성 분자를 찾는 것이 여전히 필요하다. 마우스배아줄기세포(mESC) 유래 심근세포의 Illumina 발현 마이크로어레이 데이터를 분석하였다. 미분화 mESCs와 비교하여 mESC 유래 심근세포에서 4배 이상 유전자 발현이 증가한 276개 유전자가 스크리닝되었다. Secreted phosphoprotein 2 (Spp2)는 후보물질 중 하나이며 bone morphogenetic protein 2 (BMP2)에 대한 슈도수용체로서 BMP2 신호 전달을 억제하는 것으로 알려져 있다. 그러나 심근 형성과의 연관성은 알려진 바 없다. 우리는 mESC 세포주인 TC-1/Kh2와 E14를 이용하여 기능성 심근세포로 분화하는 동안 Spp2 발현이 증가함을 검증하였다. 흥미롭게도, Spp2 분비는 배아체(embryoid body, EBs) 형성 후 3일차에 일시적으로 증가했는데, 이는 Spp2의 분비가 ESCs의 심근세포로의 분화에 관여함을 시사한다. Spp2의 기능을 분석하기 위해, 우리는 BMP2를 처리하면 분화 경로를 근모세포에서 골모세포로 전환되는 특성을 가진 C2C12 마우스 근모세포 세포주를 사용하여 실험을 수행하였다. mESCs의 분화와 유사하게, Spp2의 전사는 C2C12 근모세포가 근관으로 분화됨에 따라 증가하였다. 특히, 분화 초기 단계에서 Spp2의 세포외 분비가 극적으로 증가하였다. 또한, Spp2-Flag 재조합 단백질로 처리하면 C2C12 근모세포의 근관으로의 분화가 촉진되었다. 종합하면, ESCs를 심근 세포로 분화시키는 새로운 생체 활성 단백질로 Spp2를 제안한다. 이것은 심근형성의 분자 경로를 이해하고 허혈성 심장질환에 대한 줄기세포 요법의 실험적 또는 임상적 발전을 촉진하는 역할을 할 것으로 기대한다.
