Role of the PLA2-Activated Neutrophilic Oxidative Stress in Oleic Acid-Induced Acute Lung Injury

  • Background: The underlying pathogenesis of fat embolism-induced acute lung injury (ALI) has not been elucidated. In the present study, the pathogenesis of fat embolism-induced ALI was probed in association with neutrophilic oxidative stress in oleic acid (OA)-induced ALI of S-D rats. Methods: OA was injected intravenously to provoke ALI in experimental rats. Five hours later, indices of ALI were measured to confirm the role of the neutrophilic respiratory burst. The effect of an inhibition of phospholipase A2 (PLA2) was also evaluated. Results: The accumulation of neutrophils in the lung due to OA caused increased neutrophilic oxidative stress in lung, which was ameliorated by mepacrine. What were the results from inhibition of PLA2. Conclusion: Excess neutrophilic oxidative stress contributes to OA-induced ALI, which is lessened by the inhibition of PLA2.

Characteristics of Cytosolic Calcium-Independent Phospholipase $A_2$ Isolated from Rat Liver

  • A calcium-independent phospholipase $A_2$ ($iPLA_2$) was identified from the cytosolic fraction of rat liver cells. On gel filtration chromatography, the $iPLA_2$ activity was eluted as broad peaks of 150 to 500 kDa. The enzyme was maximally active at pH 7.5, retained 75% of its original activity after heating at $50^{\circ}C$ for 5 h, and was inhibited by $Ca^{2+}$, $Mg^{2+}$, and $Zn^{2+}$ ions, but was not affected by $Na^+$ and $K^+$ ions. The enzymatic activity was increased up to 150% by 1 to 4 mM DTT and was inhibited up to 25% by 0.1 to 1 mM PMSF. The $iPLA_2$ activity had preference for the head group of phospholipids, where phosphatidylethanolamine was preferred to phosphatidylcholine. The results suggest that the $iPLA_2$ may be a novel enzyme distinct from the previously reported $iPLA_2s$.

Enhancement of a Liver Form of Cytosolic Phospholipase $A_2$ Activity by Methylmercury

  • Methylmercury (MeHg), which is widely distributed in the environment, is well known for both its acute and chronic poisoning effects on the human health; however, the precise biochemical mechanisms by which this compound elicits its toxicity in a cellular level are still poorly understood. To examine whether MeHg-induced liver injury involves activation of Phospholipase $A_2$ ($PLA_2$), the $PLA_2$ activity of control and MeHg-administrated livers was measured. MeHg stably enhanced a liver form of cytosolic $PLA_2$ activity, which exhibited several biochemical properties similar to those of the 100 kDa $cPLA_2$, except in its elution profile of a DEAE-5PW HPLC, and it migrated as a molecular weight of 80 kDa in Western blot analysis. This blotting analysis also indicated that the MeHg-induced enhancement of the activity could be due to the increase in the amount of the enzyme protein rather than a stable modification of the enzyme such as phosphorylation. Our data also showed the higher myeloperoxidase activity in MeHg-administrated liver than in the control, suggesting that this increase in the amounts of the 80 kDa $PLA_2$ and its activity may be resulted from infiltration of neutrophils into the liver during a hepatic injury process such as MeHg-induced inflammation. Taken together, these data suggest that MeHg-induced liver injury may be mediated by activation of the 80 kDa form of liver cytosolic $PLA_2$.

장의 허혈-재관류로 유도된 급성 폐손상에서 산화성 스트레스에 관여하는 group II phospholipase $A_2$의 역할 (Role of Group II Phospholipase $A_2$ in the Pulmonary Oxidative Stress of the Acute Lung Injury Induced by Gut Ischemia-Reperfusion)

  • 배경: 급성 호흡곤란증후군은 다양한 병인에 의해 발병하지만 그 병인론이 아직까지 확립되어 있지 않다. 본 연구에서는 장의 허혈-재관류시에 발병하는 급성 호흡곤란증후군에서 group II phospholipase $A_2$ ($PLA_2$)의 역할을 알아보기 위하여 시행되었다. 특히 폐장내의 호중구의 침윤과 더불어 유발되는 산화성 스트레스에서 group II $PLA_2$의 역할을 규명하려 하였다. 대상 및 방법: 체중 300g 내외의 Sprague-Dawley 종 흰쥐에서 급성 폐손상을 유발하기 위하여 상장간막동맥을 60분간 차단한 후 120분간 재관류를 시행하였다. Group II $PLA_2$가 폐장의 손상, 특히 혈관 내피세포의 손상에 미치는 영향을 호중구의 작용과 연관하여 알아보기 위하여 폐누출지수, 폐장내 myeloperoxidase의 활성도, 폐포세척액내의 단백함량을 측정하였다. 또한 장의 허혈-재관류에 따른 폐장내 $PLA_2$ 활성도의 변화를 검사하였고, 호중구에서의 산소기 형성에 미치는 group II $PLA_2$의 역할은 분리된 호중구에 rutin, manoalide, scalaradial과 같은 group II $PLA_2$ 억제제를 이용하여 산소기 생성이 억제됨을 확인함으로써 알아보았다. 장의 허혈-재관류에 따른 폐장 조직의 산화성 스트레스를 확인하기 위해 광학현미경법 및 cerium chloride를 이용한 세포화학적인 전자현미경법을 이용하여 폐장내 산소기의 생성을 확인하였다. 결과: 장의 허혈-재관류 후 폐장내 호중구의 침윤과 함께 급성 폐손상이 유발되었고, 폐장내 myeloperoxidase 활성도, 폐누출지수 및 폐세척액내의 단백함량이 대조군에 비해 유의하게 증가하였다(p<0.001). 폐장 및 장에서의 group II $PLA_2$ 활성도는 허혈-재관류 후 폐장, 장 모두에서 유의하게 증가하였고, rutin에 의해서 현저히 감소하였다(p<0.001). 사람의 혈액에서 분리된 호중구에서의 산소기 생성을 cytocrhome-c reduction assay를 통해 알아본 결과 rutin, manoalide, scalaradial 같은 group II PLA, 억제제에 의해 호중구의 산소기 생성이 감소함을 알 수 있었다. 허혈-재관류 후 광학현미경적 소견은 폐장내 염증세포의 침윤 및 모세혈관 주위의 부종이 관찰되었으나 rutin에 의해 이러한 변화는 억제되었다. $CeCl_3$을 이용한 세포화학적 전자현미경 실험에서 허혈-재관류 후 과산화수소의 생성이 증가하고 rutin에 의해서는 억제됨을 확인하였다. 결론: Croup II $PLA_2$의 억제는 침윤된 호중구로부터 산화기 생성을 억제함으로써 급성 폐손상을 완화하는 것으로 보이며, 따라서 group II $PLA_2$는 장 허혈-재관류로 유도된 급성 폐손상의 산화성 스트레스에서 중요한 역할을 하는 것으로 보인다.

  • In our continuing effort to investigate compounds having anti-inflammatory activity from natural products, Ailanthus altissima was examined. Among six compounds isolated from Ailanthus altissima, Luteolin-7-O-glucoside (L7G) along with ethanol extract of Ailnathus altissima (EAa) were chosen to determine their inhibitory activity on secretory recombinant phospholipase $A_2s$ enzyme activity in vitro. As a results, EAa inhibited human recombinant $sPLA_2-V$ ($IC_{50}$ of about 100 ${\mu}g/ml$) and $cPLA_2$, ($IC_{50}$ of about 59 ${\mu}g/ml$), while L7G showed strong inhibitory effect on $sPLA_2-A$, V and $cPLA_2$ with an $IC_{50}$ value of approximately 40 ${\mu}M$, respectively.

  • Objective : The purpose of this study was to investigate the effect of Bee Venom on the lipopolysaccharide, sodium nitroprusside and hydrogen peroxide induced expression phospholipase $A_2$ and calcium concentration in RAW 264.7 cells, a murine macrophage cell line. Method : The expression of phospholipase $A_2$ was determined by western blotting with corresponding antibodies, and the generation of intracellular calcium concentration was investigated by delta scan system in RAW 264.7 cells. Results : 1. Compared with control, expressions of lipopolysaccharide-induced phospholipase $A_2$ were decreased significantly by $1\;{\mu}g/{\mu}l$ of bee venom and decreased by 0.5, $5\;{\mu}g/{\mu}l$ of bee venom. 2. Compared with control, expressions of sodium nitroprusside-induced phospholipase $A_2$ were decreased significantly by $5\;{\mu}g/{\mu}l$ of bee venom but increased by 0.5, $5\;{\mu}g/{\mu}l$ of bee venom. 3. Compared with control, expressions of hydrogen peroxide-induced phospholipase $A_2$ were decreased significaltly by $1{\mu}g/{\mu}l$ of bee venom and decreased by $0.5\;{\mu}g/{\mu}l$ of bee venom but increased by $5\;{\mu}g/{\mu}l$ of bee venom. 4. Compared with control, lipopolysaccharide, sodium nitroprusside and hydrogen peroxide- induced intracellular calcium concentrations were decreased by 0.5, 1, $5\;{\mu}g/{\mu}l$ of bee venom and by indomethacin

  • 생체막의 주요 구성성분인 인지질의 2번 위치에 결합한 불포화지방산은 각종 전이금속이나 각종 활성산소들의 공격을 쉽게 받아 지질과산화반응이 일어나서 생체에 유독한 화합물을 생성하게 된다. 생체는 이러한 기구의 해독을 위하여 크게 2가지 방어기전을 갖고 있다. 즉 Vitamin- C, $\alpha$-tocopherol, flavonoid, SOD, catalase 등과 같이 생성된 활성산소를 제거시키는 기구와. 활성산소에 의해 생성된 과산화물을 제거시키는 기구로 glutathione peroxidase (GPX)가 알려졌으며 GPX에 의해 독성이 낮은 수산화물까지 환원시키는 기구가 보고되었다. 그러나 인지질의 과산화물 그대로는 GPX의 기질이 쥘수 없으므로, 산화된 지방산을 절단하는 효소에 대한 기구의 해석이 요구되고 있다. 최근 여러질병에 관련되어 있는 인지질 2번위치의 지방산을 분해하는 phospholipase $A_2$ (PLA$_2$)가 과산화지질의 분해에 관여한다는 주장이 제기되었다. 따라서 본 연구에서는 rat liver microsome에 $CCl_4$투여로 일어나는 과산화반응에 있어서 PLA$_2$의 역할을 규명하기 위하여 본 실험을 행하였다.

One-Step Purification of Melittin Derived from Apis mellifera Bee Venom

  • The concern over the use of melittin in honey bee venom due to its adverse reaction caused by allergens such as phospholipase A2 ($PLA_2$) and hyaluronidase (HYA) has been an obstacle towards its usage. We developed a novel single-step method for melittin purification and the removal of $PLA_2$ and HYA. This study explores the influence of pH, buffer compositions, salt concentration, and types of cation-exchange chromatography resins on the recovery of melittin and the removal of both HYA and $PLA_2$. Melittin was readily purified with a strong cation-exchange resin at pH 6.0 with sodium phosphate buffer. It resulted in a recovery yield of melittin up to 93% (5.87 mg from a total of 6.32 mg of initial melittin in crude bee venom), which is higher than any previously reported studies on melittin purification. $PLA_2$ (99%) and HYA (96%) were also successfully removed. Our study generates a single-step purification method for melittin with a high removal rate of $PLA_2$ and HYA, enabling melittin to be fully utilized for its therapeutic purposes.

  • Chronic inflammatory processes are associated with pathology of Alzheimer's disease(AD). The expression of both cyclooxygenase-2(COX-2) and phospholipase A2(PLA2) appears to be strongly activated during AD, indicating the importance of inflammatory gene pathways as a response to brain injury. Stimulation of heterotrimeric G protein-coupled receptors including muscarinic receptors activates cytosolic PLA2 and receptor-mediated activation of PLA2 generates free fatty acids (i.e., arachidonic acid).
