Saesongi (Pleurotus eryngii) was cultivated in both potato dextrose agar (PDA) and sawdust media supplemented with Ca salts. The addition of Ca phosphate and Ca carbonate to sawdust media did not affect the growth, whereas Ca sulfate addition suppressed the mycelial growth appreciably. The efficiencies of Ca accumulation in the fruiting were studied based on mycelial growth experiments on Ca-supplemented sawdust media. Supplementation with 0.1 to 5% Ca phosphate increased the Ca content in the fruiting body by 4.5-6.5 fold, to a level of $314.6{\pm}22.7$ to $449.7{\pm}29.3$.

The reaction proceeds through the accumulation of an intermediate (I). 2-Acetylpyridineketoxime (Ox) is released in the breakdown step of I. Phenol is not formed as a part of the products, and, therefore, diphenyl phosphate and Ox are produced by the breakdown of I. Based on the dependence of the rates for the formation and the breakdown of I on [Zn(Ⅱ)] and pH, and considering the UV spectral properties of I, the structure of I is tentatively assigned as the penta-covalent intermediate formed by the addition of a water molecule to S.

Owing to the growing interest in the production of fuels and chemicals from biomass the well-know butanol-acetone fermentation as carried out by Clostridium acetobutylicum has been intensely studied again in recent years. Several solvent-yielding fermentation processes were established which are operated by using batch cultures or continuous cultures. 1 could be shown that under conditions of phosphate limitation an asporogenous mutant of C. acetobutylicum establishes itself in a chemostat which produces the solvents continuously. Attempts have been made to change the butanol/acetone ratio in favor of butanol production. A corresponding shift of the product spectrum can be achieved by carbon monoxide addition to the head space of the fermentation (B.H. Kim et al., App. Envioron. Microbiol. 48, 764-770 1984) or by iron limitation. Progress has been made in understanding the mechanism underlying the shift from acid to solvent prodcction. Experimental results are in agreement with the view that intracellular accumulation of acetic and butyric acid results in a shortage of phosphate and coenzyme A. This shortage may serve then as signal for the synthesis of the enzymes involved in the formation of acetone and butanol.

The liver is an important organ in the regulation of glucose and lipid metabolism. It is responsible for systemic energy homeostasis. When energy need exceeds the storage capacity in the liver, fatty acids are shunted into nonoxidative sphingolipid biosynthesis, which increases the level of cellular ceramides. Accumulation of ceramides alters substrate utilization from glucose to lipids, activates triglyceride storage, and results in the development of both insulin resistance and hepatosteatosis, increasing the likelihood of major metabolic diseases. Another sphingolipid metabolite, sphingosine 1-phosphate (S1P) is a bioactive signaling molecule that acts via S1P-specific G protein coupled receptors. It regulates many cellular and physiological events. Since an increase in plasma S1P is associated with obesity, it seems reasonable that recent studies have provided evidence that S1P is linked to lipid pathophysiology, including hepatosteatosis and fibrosis. Herein, we review recent findings on ceramides and S1P in obesity-mediated liver diseases and the therapeutic potential of these sphingolipid metabolites.
The freshwater green alga Chlorococcum sp. grew on NH_4^{+},{\;}NO_3^{-}$, urea, yeast extract, and peptone as the nitrogen source showing similar pattens of growth and secondary carotenoid (SC) production. However, the most suitable nitrogen source for the induction fo SC was urea. The dffects of nutrient levels (urea, phosphate, sulfate, ferrous iron, and salt) on growth and SC production were stydied by varying the concentration of each nutrient in batch cultures. High biomass production was achieved in cultures containing 20-28 mM urea, 4.8-10 mM phosphate, 1.6 mM sulfate, 70 mM phosphate, 1.6 mM sulfate, 170 mM NACl, and $50{\;}\mu\textrm{M}$ iron. The optimum concentrations of nutrients for biomass and for the SC accumulation in biomass were evaluated and the two media for achieving high biomass production and SC production were thus developed. The extent to which each parameter to stimulate the formation of SC in the alga were varied and the potentially improned SC prodution by manipulating the nutrient levels in the modified media were descussed.
In order to investigate central metabolic changes in Beijerinckia indica, cells were grown on different carbon sources and intracellular flux distributions were studied under varying concentrations of nitrogen. Metabolic fluxes were estimated by combining material balances with extracellular substrate uptake rate, biomass formation rate, and exopolysaccharide (EPS) accumulation rate. Thirty-one metabolic reactions and 30 intracellular metabolites were considered for the flux analysis. The results revealed that most of the carbon source was directed into the Entner-Doudoroff pathway, followed by the recycling of triose-3-phosphate back to Hexose6-phosphate. The pentose phosphate pathway was operated at a minimal level to supply the precursors for biomass formation. The different metabolic behaviors under varying nitrogen concentrations were observed with flux analysis.

The production of astaxanthin in the microalga Haematococcus pluvialis has been investigated using a sequential methodology based on the application of two types of statistical designs. The employed preliminary experiment was a fractional factorial design $2^6$ in which the factors studied were: excessive irradiance and nitrate starvation, phosphate deficiency, acetate supplementation, salt stress, and elevated temperature. The experimental results indicate that the amount of astaxanthin accumulation in the cells can be enhanced by excessive irradiance and nitrate starvation whereas the other factors tested did not yield any enhancement. In the subsequent experiment, a central composite design was applied with four variables, light intensity, nitrate, phosphate, and acetate, at five levels each. The optimal conditions for the highest astaxanthin production were found to be $1040{\mu}E/(m^2{\cdot}s)$ light intensity, 0.04 g/L nitrate, 0.31 g/L phosphate, 0.05 g/L acetate concentration.

Purple acid phosphatase is important for phosphorus remobilization in plants, but its role in plant adaptation to low phosphorus availability is not known. The cDNA encoding O. sativa purple acid phosphatase (OsPAP1) has 1008 bp with an open reading frame of 335 amino acid residues. The amino acid sequence of OsPAP1 cDNA shows of 50-51% identity with other plant purple acid phosphatases. OsPAP1 was expressed in rice plants and in cell cultures in the absence of phosphate ($P_i$). The expression was organ-specific with the strongest expression in $P_i$-deprived roots. Functional expression of the OsPAP1 gene in the transgenic Arabidopsis line was confirmed by northern and western blot analysis. OsPAP1 overexpression lines had higher phosphatase activity than wild-type. Overexpression of OsPAP1 in Arabidopsis plants resulted in increased Pi accumulation under Pi sufficient condition. These results show that the OsPAP1 gene represents more efficient $P_i$ uptake and can be used to develop new transgenic dicotyledonous plants.
Proteomic analyses of differentially expressed proteins in rat retinal ganglion cells (RGC-5) following S-nitrosoglutathione (GSNO), an NO donor, treatment were conducted. Of the approximately 314 protein spots that were detected, 19 were differentially expressed in response to treatment with GSNO. Of these, 14 proteins were up-regulated and 5 were down- regulated. Notably, an increase in GAPDH expression following GSNO treatment was detected in RGC-5 cells through Western blotting as well as proteomics. The increased GAPDH expression in response to GSNO treatment was accompanied by an increase in Herc6 protein, an E3 ubiquitin ligase. Moreover, GSNO treatment resulted in the translocation of GADPH from the cytosol to the nucleus and its subsequent accumulation. These results suggest that NO stress-induced apoptosis may be associated with the nuclear translocation and accumulation of GAPDH in RGC-5 cells.
Herein, a new electrochemical method was described for the determination of ciprofloxacin based on the enhancement effect of an anionic surfactant: sodium dodecyl benzene sulfonate (SDBS). In pH 4.0 phosphate buffer and in the presence of 1.0 × 10-4 mol/L SDBS, ciprofloxacin yields a well-defined and sensitive oxidation peak at the carbon paste electrode (CPE). Compared with that in the absence of SDBS, the oxidation peak current of ciprofloxacin remarkably increases in the presence of SDBS. The experimental parameters, such as supporting electrolyte, concentration of SDBS, and accumulation time, were optimized for ciprofloxacin determination. The oxidation peak current is proportional to the concentration of ciprofloxacin over the range from 8.0 × 10-8 to 5.0 × 10-6 mol L-1. The detection limit is 2.0 × 10-8 mol L-1 after 2 min of accumulation. This new voltammetric method was successfully used to detect ciprofloxacin in drugs.
