오죽잎차와 오죽죽순차의 성분 분석 및 항산화 효과 (Composition Analysis and Antioxidant Activity of Ojuk (Phyllostachys nigra Munro) Leaf Tea and Shoot Tea)

  • 본 논문에서는 오죽(Phyllostachys nigra Munro)의 잎과 죽순을 사용하여 차 가공공정을 통한 오죽잎차와 오죽죽순차가 제조되었고, 이들에 대한 영양성분 분석을 실시하였다. 뿐만 아니라, 일반적인 차를 음용하는 조건($80^{\circ}C$ 물, 10분)에서 이들 차로부터 얻을 수 있는 수용성 물질의 추출율과 총 폴리페놀 함량, 총 플라보노이드 함량이 가공되지 않은 오죽 원재료 및 마테차, 녹차와 비교되었다. 마지막으로 차 음료에 포함된 항산화능과 각 항산화 화합물을 탐색하고자 온라인, 오프라인 항산화 분석이 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and di(phenyl)-(2,4,6trinitrophenyl)iminoazanium (DPPH) 라디칼에 대해서 이루어졌다. 본 논문의 결과는 오죽으로부터 제조된 이들 차가 다양한 영양성분을 함유하고 있고, 건강에 이로운 전통차로 음용될 수 있음을 보여주었다.

SH-SY5Y 인간 신경모세포종 세포에서 MPTP 유발 세포 독성에 대한 거저리(Tenebrio molitor) 추출물의 보호효과 (Protective effects of mealworm (Tenebrio molitor) extract on N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced cellular toxicity in SH-SY5Y neuroblastoma cells)

  • Purpose: Edible insect extracts have been used as an alternative source for medicinal supplements due to their significant antioxidative and anti-inflammatory activity. Recent studies have reported that anti-microbial peptides from insects have neuroprotective effects on dopamine toxins. The purpose of this study was to investigate the protective functions of mealworm (Tenebrio molitor) extract (MWE) on N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced cellular toxicity in SH-SY5Y neuroblastoma cells. Methods: Cellular toxicity induced by the MPTP toxin and the impact of MWE on cell survival were analyzed using MTT assays. DAPI staining was performed to observe apoptotic phenomena caused by MPTP. Changes in caspase-3 activity and protein expression were observed using enzyme activity assays and western blot assays, respectively. Results: MWE exerted significant antioxidant activity, which was measured by both DPPH and ABTS radical assays, with a dose-dependent relationship. Furthermore, MWE resulted in cellular proliferation in SHSY5Y cells in a dose-dependent manner. Furthermore, MWE pretreatment significantly inhibited MPTP-induced cytotoxicity, with a dose-dependent relationship. The morphological characteristics of apoptosis and increased reactive oxygen species induced by MPTP were also significantly reduced by MWE pretreatment. Conclusion: MWE treatment significantly attenuated MPTP-induced changes in the levels of proteins associated with apoptosis, such as caspase-3 and PARP. These findings suggest that MWE exerts neuroprotective effects on human neuroblastoma SH-SY5Y cells subject to MPTP-induced dopaminergic neurodegeneration.

  • 제주도에서 생산된 로즈마리(Rosmarinus officinalis L.)와 라벤더(Lavandula angustifolia Mill.) 두 종의 허브 에센셜 오일과 하이드로졸 성분을 GC-MS로 분석하였다. 로즈마리 에센셜 오일에는 ${\alpha}$-pinene (40.96%)의 함량이 가장 높았으며, 라벤더 에센셜 오일에는 camphor (34.44%)가 가장 높았다. 로즈마리 하이드로졸에는 verbenone이 (45.31%) 가장 높게 나타났으며, 라벤더 하이드로졸에서는 camphor (67.04%)로 가장 높게 나타났다. 또한 로즈마리와 라벤더 에센셜 오일과 하이드로졸의 항산화 효능을 검증해 본 결과, 두 종류의 에센셜 오일 모두 DPPH, ABTS 라디칼 소거능과, $Fe^{2+}$ ion 킬레이팅 활성을 나타냈으나, alkyl 소거능은 거의 나타나지 않았다. DPPH 라디칼 소거능은 로즈마리 에센셜 오일이 라벤더 에센셜 오일보다 우수한 반면, $Fe^{2+}$ ion 킬레이팅 실험에서는 라벤더 에센셜 오일이 로즈마리 에센셜 오일보다 더 우수했다. 로즈마리, 라벤더 하이드로졸 모두 alkyl 라디칼 소거능을 나타냈으나, 라벤더 하이드로졸만이 $Fe^{2+}$ ion 소거능을 나타냈다. 특히 $H_2O_2$에 의해 유도된 Fibroblast와 HaCaT에서의 세포독성 실험에서 하이드로졸이 에센셜 오일보다 우수한 세포사멸 억제능을 나타냈다.

A Novel Benzoyl Glucoside and Phenolic Compounds from the Leaves of Camellia japonica

  • A novel benzoyl glucoside (4) and 13 known phenolic compounds were isolated from the leaves of Camellia japonica by a guided 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay. The structure of 4 was determined to be 4-hydroxy-2-methoxyphenol 1-O-$\beta$-D-(6'-O-p-hydroxylbenzoyl)-glucopyranoside (camelliadiphenoside). The 13 known compounds were identified as (E)-coniferyl alcohol (1), (-)-epicatechin (2), 4-hydroxyphenol 1-O-$\beta$-D-(6-O-p-hydroxybenzoyl) glucopyranoside (3), naringenin 7-O-$\beta$-D-glucopyranoside (5), quercetin 3-O-$\beta$-L-rhamnopyranosyl(1$\rightarrow$6)-$\beta$-D-glucopyranoside (6), kaempferol 3-O-$\beta$-L-rhamnopyranosyl(1$\rightarrow$6)-$\beta$-D-glucopyranoside (7), (+)-catechin (8), 1,6-di-O-p-hydroxybenzoyl-$\beta$-D-glucopyranoside (9), phloretin 2'-O-$\beta$-D-glucopyranoside (10), quercetin 3-O-$\beta$-D-glucopyranoside (11), quercetin 3-O-$\beta$-D-galactopyranoside (12), kaempferol 3-O-$\beta$-D-galactopyranoside (13), and kaempferol 3-O-$\beta$-D-glucopyranoside (14). Their chemical structures were determined by the spectroscopic data of fast atom bondardment mass spectrometry (FABMS) and nuclear magnetic resonance (NMR). Flavonoids having the catechol moiety showed significantly higher DPPH radical scavenging activity than other isolated compounds having monohydroxy phenyl group.

  • 리튬이온 이차전지에는 폴리에틸렌 격리막이 주로 사용되어 오고 있다. 전지의 고온 안정성 확보를 위해서는 폴리에틸렌 격리막보다 높은 멜트다운(melt down) 온도를 갖는 격리막이 요구된다. 이를 위해 폴리에틸렌 격리막을 단량체인 2,2-bis[4-(2-hydroxy-3-methacryloyloxypropoxy)phenyl]propane(bis-GMA) 혹은 이의 유도체들로 코팅하고 이를 라디칼 중합반응시켰다. 점도가 높은 bis-GMA를 사용한 경우에는 라디칼 중합반응이 진행되지 않은 반면 점도가 낮은 bis-GMA 유도체를 사용한 경우에는 라디칼 중합반응이 진행되었다. 적절한 함량의 bis-GMA 유도체를 포함한 반응액으로 격리막을 코팅하고 이를 중합반응시켜 격리막을 제조한 결과, 격리막의 통기도 감소없이 멜트다운 온도를 $160^{\circ}C$까지 향상시킬 수 있었다.

Antioxidant activity of sesame cake extracts obtaining using various ethanol extraction conditions

  • The antioxidant activities of sesame cake extracts prepared using ethanol and hot water were evaluated. Seventy percent-ethanol extracts yielded the highest total phenolic (96.56 mg/g extract) and flavonoid (8.35 mg/g extract) contents. In the ${\beta}$-carotene bleaching and ferric thiocyanate test, 30%-, 50%-, and 70%-ethanol extracts exhibited higher antioxidant activity than that exhibited by 90%-ethanol extracts. However, 90%-ethanol extracts showed greater antioxidant activities in phenyl-2-picryl-hydrazyl free radical scavenging test and thiobarbituric acid test using corn oil. The antioxidant activities of sesame cake extracts did not correlate with the total phenolic and flavonoid contents; however, the results suggest that oxidative stability may be improved by sesame cake extracts.

A Study on the Copolymerization Kinetics of Phenylethyl Acrylate and Phenylethyl Methacrylate

  • Copolymers of phenyl alkyl acrylates/methacrylates are used clinically as soft materials for the foldable intraocular lens (IOL) to treat cataracts. In this study, copolymers of 2-phenylethyl acrylate (PEA) and 2-phenylethyl methacrylate (PEMA) of various compositions were prepared using free radical polymerization in solution. The composition of the copolymers was determined by $^1H$-NMR analysis. The reactivity ratios of the monomers were calculated using the conventional Fineman-Ross or Kelen-Tudos method. The reactivity ratio of PEA ($r_1$) and PEMA ($r_2$) were estimated to be 0.280 and 2.085 using the Kelen-Tudos method, respectively. These values suggest that PEMA is more reactive in copolymerization than PEA, and the copolymers will have a higher content of PEMA units. The glass transition temperature ($T_g$) of the copolymers increased with increasing PEMA content. The molecular weight and polydispersity indices ($M_w/M_n$) of the polymers were determined by GPC. Overall, these results are expected to be quite useful in applications to foldable soft IOL materials.

Evaluation of antioxidant property of heat shock protein 90 from duck muscle

  • Objective: The objectives of this study were to investigate the direct antioxidative effect of 90 Kda heat shock protein (Hsp90) obtained from duck muscle. Methods: The interaction of Hsp90 with phospholipids and oxidized phospholipids was studied with surface plasmon resonance (SPR), and their further oxidation in the presence of Hsp90 was evaluated with thiobarbituric acid reactive substances (TBARS) assay. The scavenging effect on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis (3-ethylbenzthiazoline-6-sulfonic acid (ABTS) was measured, and the electron paramagnetic resonance (EPR) spectroscopy in combination with 5-tert-Butoxycarbonyl-5-methyl-1-pyrroline-N-oxide and 2-phenyl-4,4,5,5,-tetramethylimidazoline-1-oxyl-3-oxide (PTIO) was utilized to determine the abilities of Hsp90 in scavenging hydroxyl and PTIO radicals. Results: SPR showed Hsp90 could bind with both phospholipids and oxidized phospholipids, and prevent their further oxidation by the TBARS assay. The DPPH and ABTS scavenging activity increased with Hsp90 concentration, and could reach 27% and 20% respectively at the protein concentration of 50 μM. The EPR spectra demonstrated Hsp90 could directly scavenge ·OH and PTIO· radicals. Conclusion: This suggests that Hsp90, a natural antioxidant in meat, may play an important role in cellular defense against oxidative stress, and may have potential use in meat products.

효소 발효 자이언트 흑마늘 페이스트를 첨가한 양갱의 항산화 활성 및 품질 특성 (Antioxidative Capacity and Quality Characteristics of Yanggaeng with Fermented Aged Black Giant Garlic (Allium ampeloprasum L. var. ampeloprasum auct.) Paste)

  • 본 연구에서는 효소 발효 자이언트 흑마늘 페이스트의 비율(0%, 3%, 6%, 9%, 12%)을 다르게 첨가하여 양갱을 제조하고, 항산화 활성 및 품질 특성을 평가한 결과, pH는 대조군이 6.79로 가장 높았고, 효소 발효 자이언트 흑마늘 페이스트의 첨가비율이 증가함에 따라 유의적으로 감소하는 경향을 보였다. 환원당은 대조군이 5.88%로 가장 낮았고, 첨가군이 7.7~11.98%로 효소 발효 자이언트 흑마늘 페이스트의 첨가량이 증가할수록 환원당이 점점 유의적으로 높게 나타났다. 색도 측정 결과, L값과 b값은 효소 발효 자이언트 흑마늘 페이스트 첨가 비율이 증가할수록 유의적으로 감소하였다. 기계적 조직감은 효소 발효 자이언트 흑마늘 페이스트의 첨가비율이 증가할수록 낮은 조직감을 나타내었고, 총 폴리페놀 함량과 DPPH radical 소거 활성은 효소 발효 흑마늘 페이스트 첨가량이 증가할수록 높은 값을 보였다. 기호도 평가 결과, 효소 발효 자이언트 흑마늘 페이스트 첨가 비율이 3~6%일 때, 모든 평가항목에서 유의적으로 높은 점수를 받았으며, 9%를 초과함에 모든 기호도가 확연하게 낮게 나타났다. 이상의 결과를 종합하여 볼 때 효소 발효 자이언트 흑마늘 페이스트를 6%까지 첨가하여 양갱을 제조한다면 양갱의 관능적 품질을 최대한 유지하면서 기능성 식품으로 소비자의 요구를 충족시킬 수 있을 것이라 여겨진다.

Synthesis, Structure and Biological Properties of a Novel Copper (II) Supramolecular Compound Based on 1,2,4-Triazoles Derivatives

  • A novel mononuclear supramolecule of copper(II) has been synthesized with Ippyt ligand (Ippyt=3-(4'-imidazole phenyl)-5-(pyrid-2''-yl)-1,2,4-triazole) (1). Compound 1, namely [$Cu(Ippyt)_2(H_2O)_2$], has been characterized by single-crystal X-ray diffraction, IR spectrum, elemental analysis and thermogravimetric analysis. Structure determination reveals that the elongated-octahedral geometry is formed in the vicinity of the copper (II) atom being coordinated by four nitrogen atoms from two Ippyt ligands occupying the equatorial position and two oxygen atoms from two coordinated water molecules in the axial position, which together form the $N_4O_2$ donor set. Hydrogen bonding interactions between nitrogen and oxygen atoms result in the set up of a supramolecular network architecture. Biological properties including antibacterial activity and superoxide dismutase (SOD) mimetic activity of compound 1 have been investigated by agar diffusion method and the modified Marklund method, respectively. The results indicate that compound 1 exhibits a stronger antibacterial efficiency than the parent ligand and it also has a certain radical-scavenging activity.
