Antimicrobial Effect of Methanol Extracts from Some Medicinal Herbs and the Content of Phenolic Compounds (약용식물 추출물에 대한 항미생물 활성 검색과 폴리페놀 함량)

  • Methanol extracts were prepared from 32 medicinal herbs of the extracts were tested their microbial inhibition activities against food borne pathogens and/or food poisoning microorganisms, food-related bacteria and yeast. Methanol extracts of Cinnamomum cassia, Paeonia suffruticosa, Alnus japonica, Eugenio caryophyllata and Illicium verum exhibited antimicrobial activity for the microorganisms tested, except lactic acid bacteria and yeast. Minimum inhibitory concentrations(MIC) were about 5 mg/mL for Bacillus subtilis, Staphylococcus epidermidis and Pseudomonas aeruginosa. Cell growth of lactic acid bacteria was inhibitied, but greatly on Leuconostoc mesenteroides. The phenolic compound contents were 10.98 mg/g, 10.31 mg/g, 8.55 mg/g and 6.69 mg/g in Thea sinensis, Eugenia caryophyllata, Alnus japonica and Artenisia capillaris, respectively. Antimicrobial activity appeared to be related to phenol compound content in medicinal herbs. The methanol extracts of medicinal herbs could be suitable for the development of a food preservative.

Analysis of Nutritional Composition and Phenolic Compound in Propolis Collected from Falseacacia and Chestnut Tree in Korea (국내산 아까시나무와 밤나무 유래 propolis의 영양성분 및 페놀성 화합물 분석)

  • Nutritional composition and phenolic compounds of raw propolis collected from falseacacia (Robinia pseudoacacia L.) and chestnut tree (Castanea crenata), and their 70% ethanol extracts of propolis (EEP) were analyzed. Propolis had high crude lipid content, but no significant differences in general compositions in terms of collection area and plant origins. Mineral contents varied greatly depending on the plant origins, with falseacacia propolis showing the highest mineral content. Sixteen amino acids were analyzed, among which aspartic acid content was the highest at $328.4{\sim}410.6\;mg%$ and methionine the lowest at $0{\sim}21.1\;mg%$. Extraction yield for EEP was relatively high at $64.2{\sim}81.9%$, and total polyphenol and flavonoid contents were $13.9{\sim}23.7$ and $8.6{\sim}10.8%$, respectively. HPTLC and HPLC analysis on the phenolic compounds revealed the overall chromatographic patterns were almost equal, showing similar polyphenol compositions between the propolis. About 16 peaks were identified by HPLC analysis, among which 6 peaks of p-hydroxy benzoic acid, caffeic acid, ferulic acid, benzoic acid, cinnamic acid, and chrysin were identified.

Determination of Total Content of Phenolic Compounds in Chinese Matrimony Vine's Accessions (국내외 수집종 구기자 잎과 줄기의 페놀화합물 함량)

  • This study was conducted to determine the variation of phenolic compounds in the leaf and stem of 131 accessions of Lycium chinesis Miller. The levels of total phenolic compounds in the leaf of L. chinesis ranged between 8.8 to $14.9mg\;g^{-1}$ and among them 60% of the accessions belong between 11.6 and $13.5mg\;g^{-1}$ for the content of phenolic compounds in the leaf. The accession CB03286-89 contained the highest total phenolic compounds among the accessions tested, which was 1.7-fold higher than that of the lowest content accession CBP03310-250. In the stem, the total phenolic compound of 131 accessions of L. chinesis ranged from 6.8 to $12.4mg\;g^{-1}$, showing slightly lower level than that in the leaf. The content of (+)catechin was highest in the leaf and stem of accession CB03286-89 and Japan No.1, respectively. Myricetin was detected in the leaf of seven accessions (i.e. Geumsan jaerae, Japan No.1, China collection No.1, CL32-13, CB04329-13, China collection No.12 and CB03286-89) and in the stem of five accessions (i.e., Japan No.1, China collection No.1, China collection No.12, CB03286-89 and 99797). Accessions had a great influence on the content of phenolic compounds. So, accessions-specific phenolic compound profiles might be helpful for commercial use or production of phenolic compounds in L. chinesis.

Antioxidant Activity of Major Cultivars Prunus mume in Korea (국내 주요 재배 매실 품종의 항산화 활성)

  • This study examined the total phenolic and flavonoid compounds, and the antioxi dant activity of major cultivars of Prunus mume in Korea. The total phenolic content ranged from 64.13 to $93.43mg{\cdot}100g^{-1}$, with the content in 'Osuku' being higher than that in the other cultivars; a difference in the content among cultivars was observed. The total flavonoid content was $6.16{\sim}18.57mg{\cdot}100g^{-1}$, showing a significant difference, with the highest content being noted in 'Gojirou'. The DPPH radical scavenging activity was measured and was relatively high for Nanko, Kamakami, Natsuka, Dana, and Osuku cultivars. When ABTS radical scavenging activity was measured, all the cultivars showed a scavenging activity over 80%, with the scavenging activity of 'Osuku' being the highest. With respect to nitrite scavenging ability, all cultivars exhibited a high scavenging activity of over 50%, with the highest activity observed for 'Nanko' (76.03%), 'Gojirou' (70.56%), and 'Yourou' (70.32%). Based on the study results, it is considered that Prunus mume can be a useful resource because of various substances with functional physiological activities.

Nutritional Characteristics of Calystegia japonica (메꽃(Calystegia japonica)의 영양학적 특성)

  • In the present study, the proximate composition, sugar, minerals, total phenolic and flavonoid compounds, and amino acids in Calystegia japonica (C. japonica) were measured to determine if it can be used as a nutritional and functional material for the development of valuable foods. The mean crude protein, fat, and ash contents of the leaves were 5.75, 2.46, and 7.77%, respectively. The soluble-protein contents of the leaves and roots were 146.78 and 33.67 mg%, respectively. The reducing-sugar and free-sugar contents of the leaves were 682.70 and 166.00 mg%, respectively, and those of the roots were 2,934.89 and 37.70 mg%. The mineral content of the leaves was 3,122.13 mg%, and that of the roots was 1,540.85 mg%. The three elements Ca, K, and Mg were very rich in all their parts, with minerals accounting for 96-99% of their total mineral contents. The total phenolic compound of the leaves was 3,028.89 mg%, and the total flavonoid compound was 382.67 mg%. The phenolic and flavonoid compounds in the leaves were more than 7.6 times those in the roots. The free-amino acid levels in the leaves and roots were 2,467.15 and 1,334.81 mg%, respectively. The results of the comparison of the leaves and roots of C. japonica showed that the leaves had a rich proximate composition consisting of minerals, total phenolic and flavonoid compounds, and amino acid. This suggests that C. japonica leaves are potentially useful sources of functional and favorite foods and nutraceuticals.

Chemical identification and antioxidant activity of phenolic compounds extracted from the fruiting body of 'Hankyong Sanghwang', Phellinus linteus KACC 93057P (Phellinus linteus KACC 93057P, '한경상황버섯'인공재배 자실체 페놀추출물의 항산화 활성 및 화학적 동정)

  • The aim of this study was to identify chemical composition and antioxidant activity of phenolic extract of fruiting bodies of an artificially cultivated Hankyong Sanghwang mushroom, Phellinus linteus KACC 93057P (PLHS). The total phenolic content of 60% ethanolic extract of fruiting bodies of two-year-old PLHS grown on Oak wood logs was $19.05{\pm}0.32mg$ gallic acid equivalent (GAE)/g, which was 4-10 times high compared to the other species of mushrooms. 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbisthiazoline-6-sulfonic acid) (ABTS) radical scavenging activities and the ferric reducing antioxidant power (FRAP) value of PLHS were 2-10 and 5 times higher, respectively, than those of the other species of mushrooms. High performance liquid chromatography (HPLC) analysis of ethyl acetate fraction of 70% methanolic extract of the fruiting bodies of PLHS revealed the presence of styrylpyrone-class compounds, davallialactone, hispidin, hypholomine B, and caffeic acid, a compound of the phenylpropanoid pathway.

Effects of Proteins on the Reactivity of Various Phenolic Compounds with the Folin-Ciocalteu Reagent (다양한 페놀성 물질과 Folin-Ciocalteu 시약과의 반응성에 미치는 단백질의 영향)

  • The Folin-Denis assay using the Folin-Ciocalteu (F-C) reagent has been commonly used for analyzing the total phenolic compound content in various food products. In the present study, the effects of proteins on the reactivity of the F-C reagent with different phenolic compounds were investigated. Bovine serum albumin (BSA) or skim milk proteins showed a concentration-dependent increase in color response in the Folin-Denis assay; these proteins decreased the color response of most phenolic compounds tested. The reactivity of phenolic compounds was significantly less pronounced in the presence of BSA and this interference was greater at higher concentrations of phenolic compounds. The reactivity of phenolic compounds with the F-C reagent was reduced significantly by their oxidation; the reaction of the oxidized products with the F-C reagent was more severely affected by BSA. The interfering effects in the Folin-Denis assay might be attributable to binding interactions of phenolic compounds with proteins.

Selection of Cell Lines for High Yields of Antioxidants from Callus of Ginseng Superior Lines (인삼 육성계통 캘러스로부터 항산화물질 고함유 세포주의 선발)

  • Cell growth and production of phenolic compounds by callus cultures of Panax ginseng C. A. Meyer were investigated under various phytohormones concentrations and inoculum size. The results indicated that the cell growt was improved by a MS medium supplemented with 2 mg/L of CPA. The maximum cell yield was obtained at inoculum size of 1 g/flasd. The production of phenolic compounds in the callus cultures was higher than those in the ginseng root. Especially, one cell line (20601) showed the highest content of phenolic compounds and antioxidant activity.

Effects of elevated CO2 on growth of Pinus densiflora seedling and enzyme activities in soil

  • Atmospheric $CO_2$ concentrations have increased exponentially over the last century and, if continued, are expected to have significant effects on plants and soil. In this study, we investigated the effects of elevated $CO_2$ on the growth of Pinus densiflora seedling and microbial activity in soil. Three-year-old pine seedlings were exposed to ambient as well as elevated levels of $CO_2$ (380 and 760 ppmv, respectively). Growth rates and C:N ratios of the pine seedlings were also determined. Dissolved organic carbon content, phenolic compound content, and microbial activity were measured in bulk soil and rhizosphere soil. The results show that elevated $CO_2$ significantly increased the root dry weight of pine seedling. In addition, overall N content decreased, which increased the C:N ratio in pine needles. Elevated $CO_2$ decreased soil moisture, nitrate concentration, and the concentration of soil phenolic compounds. In contrast, soil enzymatic activities were increased in rhizosphere soil, including ${\beta}$-glucosidase, N-acetylglucosaminidase and phosphatase enzyme activities. In conclusion, elevated $CO_2$ concentrations caused distinct changes in soil chemistry and microbiology.

Component Comparision of Artemisia selengensis and Artemisia sp. for Artemisia selengensis Utilization (물쑥의 유용화(有用化)를 위(爲)한 수집종(蒐集種)쑥과의 함분비교(咸分比較) 연구(硏究))

  • This experiment was carried out to get the basic information on utilizing artemisia selengensis, the chemical component of artemisia selengensis and those Artemisia sp. were analyzed and compared. The results are as followed; The contents of water, crude ash and crude fiber in Artemisia selengensis were higer than in Artemisia sp. the contents of thiamin, riboflavin and ascorbic acid in Artemisia selengensis were a little lower than those in Artemisia sp. while niacin in Artemisia selengensis was a little higher than those in Artemisia sp. The content of dietary fiber were 40.91% for Artemisia selengensis and 4166.7mg% for Artemisia sp. and the contents of Na, Fe and Mn in Artemisia selengensis were over twice as a high as those in Artemisia sp. While the content of Cu was Artemisia selengensis was lower than that in Artemisia sp. The content of phenolic compound in Artemisia selengensis was a little higher than those in Artemisia sp.
