비타민 나무(사극)의 페놀성 성분 분석 (Comparison of the Contents of Phenolic Compounds of Sea Buckthorn(Hippophae rhamnoides) Cultivated in Korea and Mongolia)

  • The purpose of this research is to provide basic informations to discriminate between sea buckthorn (Hippophae rhamnoides) cultivated in the Republic of Korea and Mongolia. The phenolic compounds of sea buckthorn, were measured by the HPLC analysis. Catechin, rutin, quercetin, isorhamnetin were found in methanol extracts of sea buckthorn. Total phenolic compound of Korean sea buckthorn leaves (1.852%) was about five times higher than those of Mongolian sea buckthorn berries (0.338%). As a result, the order of the total phenolic compound and Catechin content was 1) sea buckthorn leaves, 2) sea buckthorn stems, 3) sea buckthorn roots, and 4) sea buckthorn berries. Statistically no big differences in levels of phenolic compounds were consistently found in sea buckthorn cultivated in the Republic of Korea and Mongolia investigated in this work.

우엉 뿌리 추출물의 항산화 및 Tyrosinase 저해 활성과 Phenolic Compound 분석 (Antioxidative Activity and Tyrosinase Inhibitory Activity of the Extract and Fractions from Arctium lappa Roots and Analysis of Phenolic Compounds)

  • In this study, we investigated on antioxidative activities and tyrosinase inhibitory activities of methanol extract and its fractions from roots of Arctium lappa. The total phenolic compound and flavonoid content of the ethylacetate fraction was found to be 818.29 mg/g and 360.59 mg/g as the highest content. In the measurement of DPPH radical scavenging ability and tyrosinase inhibitory activity, the ethylacetate fraction was higher than the other fractions and the extract. In addition, comparative analysis of phenolic compounds by liquid chromatography-mass spectrometry (MS)/MS system under the multiple-reaction monitoring (MRM) with negative-ion electrospray ionization mode. The main phenolic compounds in the extract and fractions of roots from Arctium lappa were cynarin and chlorogenic acid. The main phenolic compound of the ethylacetate fraction was cynarin. n-Butanol fraction had a significantly higher chlorogenic acid content than other samples. In conclusion, DPPH radical scavenging ability and tyrosinase inhibitory activity of the cynarin-riched ethylacetate fraction showed the highest activity.

감잎의 Phenolic Compounds가 에탄올을 투여한 흰쥐의 항산화계 및 기타 효소활성에 미치는 영향 (Effects of Phenolic Compounds of Persimmon Leaves on Antioxidative System and Miscellaneous Enzyme Activities Related to Liver Function in Ethanol-Induced Hepatotoxicity of Rats)

  • 감잎 phenolic compounds가 횐쥐에서 에탄올투여로 생성된 oxygen free radical대사와 각종 효소활성에 미치는 영향을 검토하기 위하여 실험동물을 정상군, 감잎 phenolic compounds 투여군(70 mg/kg), 에탄을 단독투여군, 에탄올과 감잎 phenolic compounds 소량 병합투여군(70 mg/kg), 에탄올과 감잎 phenolic compounds 대량 병합투여군(140 mg/kg)의 5군으로 나누어 6주간 사육하여 체중증가율, 식이효율 및 혈청중 ALT, ALP 및 LDH활성 측정과 간손상 억제효과를 검토하기 위하여 간조직중 유리기 생성계 효소인 XO활성, 유리기 해독계 효소인 SOD, calaase 및 GSH-Px활성과 지질과산화물인 TBARS 및 GSH함량을 측정한 결과는 다음과 같다. 각 분획의 항산화력은 EtOAC, n-BuOH, chloroform, n-hexane, water 분획순으로 EtOAC분획의 항산화력이 가장 우수하였다. 에탄올만을 투여한 군은 정상군에 비하여 체중증가율과 식이효율이 유의적으로 감소되었고 감잎 phenolic compounds를 병합투여한 군은 체중증가율과 식이효율이 정상군에 근접하게 상승되었다. 간 손상의 지표로 알려진 ALT, ALP 및 LDH활성은 에탄올투여로 정상군에 비하여 유의하게 증가되었으나 감잎 phenolic compounds를 투여하여 Elf에 비하여 많은 감소를 보였다. 간조직중 유리기 생성계 효소인 XO활성은 에탄올투여로 정상군에 비하여 큰 폭의 증가를 보였으나 본 실험기간내에서는 유의성 있는 변화는 아니었으며 감잎 phenolic compounds를 투여하여 에탄올투여로 증가된 효소활성이 ETB에 비하여 감소는 되었으나 유의한 변화는 아니었다. 유리기 해독계 효소인 SOD, catalase 및 GSH-Px 활성은 에탄올투여로 정상군에 비하여 크게 증가되었으나 감잎 phenolic compounds투여로 ETB에 비하여 뚜렷한 저하효과를 나타냈다. TBA반응성생산물량에서 에탄올투여로 과산화지질함량이 CON에 비하여 많이 증가되어 유의한 변화를 나타냈으나 감잎phenolic compounds는 에탄올투여로 증가된 함량을 ETH에 비하여 1/3∼$\frac{1}{2}$정도 용량의존적으로 감소시켰으며 PEI의 경우 그 함량이 CON보다 오히려 낮았다. 또한 에탄올투여로 감소된 간조직중 GSH함량은 감잎 phenolic compounds 투여로 정상군보다 증가되었다. 이상의 실험결과 에서 감잎 phenolic compounds가 에탄올투여로 증가된 유리기 해독계 효소인 GSH-Px활성을 큰 폭으로 감소시키고 에탄올투여로 감소된 비효소적 항산화작용을 나타내는 GSH함량을 다량 증가시킴으로서 지질과산화물에 대한 방어력이 증가되어 나타난 결과로 여겨지며, 또한 혈청중의 ALT, ALP 및 LDH활성을 유의성있게 감소시키므로서 감잎 phenolic compounds가 에탄올에 의한 간세포 손상에 대한 해독 및 보호작용이 있는 것으로 사료된다.

마삭줄(Trachelospermum asiaticum var. intermedium nakai)로부터 추출한 pheonolic compounds의 생리활성 (Biological activities of isolated phenolic compounds from Trachelospermum asiaticum var. intermedium nakai)

  • 본 연구에서 마삭줄(Trachelospermum asiaticum var. intermedium Nakai)을 용매별과 ethanol 농도별로 추출한 결과, 용매별 추출에서 ethanol 추출물이 20.8 mg/g으로 가장 높은 phenolic compounds 용출량을 보였고, 농도별 추출에서 70% ethanol 추출물이 38.1 mg/g으로 가장 높았다. 물 추출물은 16.8 mg/g의 phenolic compounds 용출량을 보였다. 마삭줄을 물과 ethanol을 추출용매로 사용하여 항산화 효과와 항통풍 및 항당뇨 효과를 확인하여 건강 기능성 식품으로의 가능성을 증명 하고자 하였다. DPPH radical 소거능 측정 결과, $100{\mu}g/mL$ phenolic compounds의 농도에서 물 추출물은 80.9%, 70% ethanol 추출물은 83.1%의 활성을 나타내었다. ABTS radical 소거능 측정 결과 $200{\mu}g/mL$ phenolic compounds의 농도에서 물 추출물은 95.0%, 70% ethanol 추출물은 95.8%의 소거능력을 나타내었다. PF 측정 결과, $100{\mu}g/mL$ phenolic compounds의 농도에서 물 추출물은 2.43 PF, 70% ethanol 추출물에서는 2.45 PF를 나타내었다. TBARs 측정 결과, $200{\mu}g/mL$ phenolic compounds의 농도에서 물과 70% ethanol 추출물에서 89.9%와 89.3%의 활성을 나타내었다. 통풍 억제 효과 측정 결과, $200{\mu}g/mL$ phenolic compounds의 농도에서 물 추출물은 33.3%, 70% ethanol 추출물은 50.5%의 저해 효과를 나타내었다. ${\alpha}-glucosidase$ 저해 효과 측정 결과, 물 추출물에는 저해 효과가 나타나지 않았고, 70% ethanol 추출물에서는 $200{\mu}g/mL$ phenolic compounds의 농도에서 92.6%의 저해 효과를 나타내었다. 이러한 결과들로 미루어 보았을 때 마삭줄 추출물은 항산화 작용과 건강 기능성 식품으로 활용 가능성을 확인할 수 있었다.

번행초(Tetragonia tetragonioides)로부터 추출한 Phenolic Compounds의 미용 식품 활성 (Beauty Food Activities of Isolated Phenolic Compounds from Tetragonia tetragonioides)

  • 본 연구는 물과 ethanol을 추출용매로 사용한 번행초(Tetragonia tetragonioides) 추출물이 미용 식품 소재로 사용 가능한 기능성을 증명하고자 하였다. Phenolic compounds는 물 추출물에서 3.29 mg/g, 50% ethanol 추출물에서 4.14 mg/g의 결과를 각각 나타내었다. 전자공여능 측정 결과 $200{\mu}g/mL$ phenolic compounds의 농도에서 물 추출물은 98.45%, 50% ethanol 추출물은 91.20%의 활성을 나타내었다. ABTS radical 활성 측정 결과 $100{\mu}g/mL$ phenolic compounds의 농도에서 물 추출물은 97.28%, 50% ethanol 추출물은 97.03%의 활성을 나타내었다. Antioxidant protection factor(PF) 측정 결과 $200{\mu}g/mL$ phenolic compounds의 농도에서 물과 ethanol 추출물 모두 1.77 PF의 같은 결과를 나타내었다. Thiobarbituric acid reactive substance(TBARS) 측정 결과 $100{\mu}g/mL$ phenolic compounds의 농도에서 물과 ethanol 추출물은 94.77%와 95.64%의 높은 활성을 나타내었다. 미백과 관련 있는 tyrosinase 저해 활성을 측정한 결과 $200{\mu}g/mL$ phenolic compounds의 농도에서 ethanol 추출물은 34.96%의 억제력을 나타내었다. 주름 개선 효과를 측정하는 elastase 저해 활성과 collagenase 저해 활성을 측정한 결과 $200{\mu}g/mL$ phenolic compounds의 농도에서 ethanol 추출물만 78.90%와 61.3%의 높은 저해 활성을 나타내었다. 수렴 효과를 나타내는 astringent effect에서 $200{\mu}g/mL$ phenolic compounds 농도의 ethanol 추출물은 7.82%의 효과를 나타내었다. Hyaluronidase 저해 효과 측정 결과 물 추출물 $200{\mu}g/mL$ phenolic compounds의 농도에서 81.04%의 염증 억제 효과를 나타내었다. 이러한 결과들로 미루어 보았을 때 번행초 추출물은 항산화 작용과 미용 식품으로서의 기능성을 기대할 수 있다.

  • Currently in Korea, standard operating procedure for the analyses of phenolic compounds in water is the spectrometric comparison of colors developed by 4-amino antipyrin with phenolic compounds. It is however that this method cannot identify individual compound and that some phenolic compounds do not react with 4-amino antipyrin. Spectrometric determinations of phenolic compounds were compared with chromatographic analyses of gas chromatography (GC) and high pressure liquid chromatography (HPLC) of various phenolic compounds. Individual phenolic compounds could be determined by both chromatographic methods but HPLC methods were more precise with lower detection levels in general.

Encapsulation of Anthocyanin from Purple Potato by the Application of Food Polymers

  • Anthocyanins has a strong antioxidant capacity but exhibit poor stability in water. Therefore, stability of anthocyanin from purple potato (Solanum tuberosum L.) was encapsulated by the application of food polymers. Solid formulation of purple potato was prepared using whey protein, tapioca and lecithin by capillary rheometer at $80^{\circ}C$. The ratio of the polymer and potato powder was 2:8. Total phenolic compound, total flavonoid, total anthocyanin and antioxidant activity was investigated by the spectrophotometer. Result revealed that total phenolic compound (TP) ($5321{\mu}g/100g$), total flavonoid (TF) ($1352{\mu}g/100g$) total anthocyanin (TA) ($764{\mu}g/100g$) and free radical antioxidant activity (DPPH) (86%) was higher in 0.01 M acetic acid mediated lecithin based formulation compared to control (Potato powder) (TP: $1357{\mu}g/100g$; TF) ($634{\mu}g/100g$, TA) ($264{\mu}g/100g\;DPPH$) (64%). Lecithin is a strong emulsifier having capacity to extract bioactive compound and encapsulate extracted compound by nonpolar tail and negatively charged head. Therefore, it would be concluded that lecithin might be used as an encapsulating agent for the bioactive compound from purple potato.

  • A phenolic compound isolated from the ethylacetate extract of Duchesnea indica (Andr.) Focke (Rosaceae) showed antimicrobial activities against Staphylococcus aureus, Shigella dysentriae and Pseudomonas aeruginosa. This compound was identified as ellagic acid by spectral analysis.

  • Light fermentation has been conducted under different light conditions such as normal dark light, white light, and light emitting diodes (LEDs) various color (blue, green, red, white on blueberry powder with fermenting bacteria Bacillus subtilis (B2). The bacteria B2 was isolated and identified by 16S rRNA sequencing method. RYRP biologically converted to secondary metabolites through light fermentation in the presence of Bacillus subtilis, the bacteria actively involved in bioconversion process. LEDs fermentation to enhance the production of phenolic content while comparing to normal dark and white light. Among the different color LEDs, blue LEDs mediated fermentation showed higher amount of total phenolic and flavonoid content. Then blue LEDs mediated fermented compound were characterized by FTIR and GC-MS, subsequently the compound was analyzed antioxidant activity tests and the antioxidant activity exhibited higher. This is the first study to demonstrate that B. subtilis-LEDs mediated fermentation is useful for facilitating phenolic compound production and enhancing antioxidant activity, which may have greater application fermentation fields.
