Hyperlipidemic Inhibitory Effects of Phellinus pini in Rats Fed with a High Fat and Cholesterol Diet

  • This study evaluated the in vitro and in vivo hypolipidemic effects of the medicinal mushroom Phellinus pini. The methanol extract (ME) of the fruiting body of Ph. pini was active against pancreatic lipase and cholesterol esterase with 99.14% and 67.23% inhibited activity at 1.0 mg/mL, respectively. It also inhibited 81.81% and 55.33% of ${\alpha}$-glucosidase and ${\alpha}$-amylase activities, respectively, at 2.0 mg/mL. Hyperlipidemia as induced by feeding rats with a high fat and cholesterol diet (HFC). HFC supplemented with a 5% fruiting body powder of Ph. pini (HFC + PhP) significantly reduced plasma total cholesterol, low-density lipoprotein cholesterol, and triglycerides in rats compared with HFC. The reduced levels were comparable to rats fed the normal control diet (NC). The atherogenic index of HFC + PhP rats was significantly lower than that of the HFC rats. The excretion of fecal total lipid and cholesterol in the HFC + PhP rats was significantly higher than those in the NC and HFC rats. Histopathological examinations demonstrated scant deposition of lipids in the liver of rats fed HFC + PhP. The dietary supplementation with the fruiting body powder provided natural plasma lipid and glucose lowering effects in experimental rats without adverse effects on the plasma biochemical parameters and liver function related enzyme activities. Therefore, the hypolipidemic effects of Ph. pini may be due to the inhibitory effects on pancreatic lipase, cholesterol esterase, ${\alpha}$-glucosidase, and ${\alpha}$-amylase, and excretion of excess lipids and cholesterol in the feces.

Immunomodulating Activity of the Exopolymer from Submerged Mycelial Culture of Phellinus pini

  • The immunomodulating activities and chemical characteristics of a water-soluble exopolymer from submerged mycelial culture of Phellinus pini were studied. Anticomplementary activity of this polymer was found to be $73.2\%$, and its activation system occurred through both classical and alternative pathways, where the classical pathway was detected to be the major one by crossed immunoelectrophoresis. Nitric oxide (NO) release ability and acid phosphatase activity of macrophage were increased by 1.6-fold ($100{\mu}g/ml$) and 3.4-fold ($500{\mu}g/ml$), respectively, and splenocyte proliferation in mixed lymphocyte reaction (MLR) was also increased by 2.6-fold ($200{\mu}g/ml$), compared to the control. The molecular weight of this polymer, determined by HPLC, was under 5 kDa. Total sugar and protein contents were 89.7 and 10.3%, respectively. Both sugar and amino acid compositions of the exopolymer were also analyzed.

  • 국내에 유통 중인 약용버섯의 분류체계 확립과 차가버섯 근연속간의 유연관계를 밝히기 위해 이들 버섯의 ITS 부위 염기서열을 비교 분석하였다. 조사결과, 약용버섯으로 주로 시판되고 있는 국내유통균주는 총 6개의 속(Phellinus, Inonotus, Sparassis, Fomes, Ganoderma, Hericium)으로 나누어짐을 알 수 있었고 그 중에서 기존에 잘 알려진 상황버섯(Phellinus linteus)과 최근 들어 수입양이 급증하고 있는 차가버섯이 대부분을 차지하고 있는 것으로 확인되었다 일명 차가버섯(Inonotus obliquus)으로 유통 중인 15개 제품 중 중국에서 수입되어진 한 균주(59번)가 P. pini로 확인되었으며 일본에서 수입되어진 균주(51, 52번)가 P. baumii와 P. linteus와 유사종 혹은 동일종으로 확인되었다.

  • The extract contents of various Phellinus species (Hymenochaetaceae) including P. linteus, P. baumii, P. pini and P. igniarius of domestic and foreign products, were examined to standardize the quality of these products. The differences of various extract contents were identified and compared between different species of Phellinus products. The thin layer chromatographic patterns were also analyzed and six unidentified samples were thought to originate from P. baumii and P. igniarius. The chemical standardization for the quality control of various origins could be suggested by this study.

PCR-Based Sensitive Detection of Wood-Decaying Fungus Phellinus linteus by Specific Primer from rDNA ITS Regions

  • Based on the rDNA ITS sequences data, specific primer set for PCR detection of wood-decaying fungus Phellinus linteus was designed. The length of PCR products using designed primer set(SHF and SHR) was about 540 bp. Among 11 species, 17 isolates of Phellinus spp. including Phellinus linteus, P. pomaceus, P. spiculosus, P. baumi, P. pini, P. igniarius, P. gilvus, P. biscuspidatus, P. weirii, P. johnsonianus, P. robutus, and P. igniarius, seven isolates of Phellinus linteus showed about 540 bp-sized single band. This molecular technique could offer a useful tool for detecting and identifying Phellinus linteus.

낙엽진흙버섯 자실체 유래 열수 추출물과 메탄을 침전물의 항바이러스활성 (Antiviral Activity of Hot-Water Extract and Its Ethanol Precipitate of Phellinus pini Fruiting Body)

  • 알래스카산 낙엽진흙버섯[Porodaedalea pini (Brot.) Murrill] (syn. Phellinus pini)의 자실체 조분쇄물로부터 $100^{\circ}C$에서 4시간 추출하여 건량 수율 20.5%의 건조 분말 열수추출물을 제조하여, HeLa 세포에서 CVB3에 대한 항바이러스 활성을 조사한 결과 plaque 형성을 현저히 억제하였다. 또한, 다른 여러 가지 버섯 추출물에 비해 neuraminidase 활성 저해능이 가장 높았다. 열수 추출물로부터 75% 에탄올 침전으로 건량 수율 28.3%의 저분자 상등액 건조물(ES)과 수율 43.3%의 고분자 침전물 건조물(EP)을 얻었다. 상등액인 ES에서는 항바이러스 활성이 없었지만, 침전물인 EP는 HeLa 세포에서 CVB3바이러스에 대해 농도 의존적으로 plaque 형성을 현저히 억제하였고, $EC_{50}$은 0.45 mg/mL 이었으며, HeLa 세포에 대한 세포독성 $CC_{50}$은 2.25mg/mL 이었다. 또한, EP는 neureminidase 활성을 농도 의존적으로 저해하였으며, 1.7mg/mL에서 약 75%의 효소활성 억제효과를 보였다. 이러한 결과는 낙엽진흙버섯 자실체의 별수추출물로부터 얻어진 에탄을 침전물 EP가 CVB3 뿐만 아니라, influenza virus(Flu)등에 대해서도 광범위하게 항바이러스 활성을 나타낼 가능성을 보여 주었다. EP는 다당류로서 glucose의 함량이 79.8%로 가장 높았고 galactose, xylose, mannose와 fucose를 소량 포함한 수용성 heteroglycan의 일종으로, 소량(12.7%, w/w)의 단백질 또는 작은 펩타이드를 함유한 당단백의 일종일 것으로 사료된다.

Phylogenetic Analysis of Phellinus linteus and Related Species Comparing the Sequences of rDNA Internal Transcribed Spacers

  • The phylogenetic tree displayed the presence of five groups in the Phellinus genus, which were distinguished based on their morphology. Most of the p. linteus appeared a cluster which was highly significant with the exception of P. linteus KACC 500122 and KACC 500411. They formed the sister taxa of P 1inteus where P. baumii, Phellinus sp. MPNU 7003, MPNU 7007, and MPNU 7010 had similar morphological characteristics. Also, P. nigricans IMSNU 32024 and P. pini var, carniformans IMSNU 32031 were grouped in the same cluster with P. igniarius KCTC 6227, KCTC 6228, and P. chrysoloma KCTC 6225 extracted from the Gen-Bank database. P. torulosus IMSNU 32028 and Phellinus sp. MPNU 7011 formed a closed group, however, these species had a distant taxa when compared with the other Phellinus species. The nucleotide sequences of the internal transcribed spacer (ITS) regions of ribosomal DNA (rDNA) including the 5.85 rDNA were determined from 24 strains of the Phellinus genus in order to analyze their phylogenetic relationship. These fungi were divided into two basic groups based on their ITS length, however, this grouping was different from that based on their morphological characteristics. Although various ITS sequences were ambiguously aligned, conserved sites were also identified. Accordingly, a neighbor-joining tree was constructed using the nucleotide sequence data of the conserved sites of the ITS regions and the 5.8S rDNA.

Molecular Detection of Phellinus linteus and P. baumii by PCR Specific Primer

  • Specific primer sets based on ribosomal DNA(rDNA) internal transcribed specer(ITS) sequences were designed for rapid detection of Phellinus linteus and P. baumii. Polymerase chain reaction(PCR) with these primers produced unique bands for each Phellinus species. The annealing temperature range is from $40^{\circ}C\;to\;55^{\circ}C$. The length of PCR products(P. linteus and P. baumii) using designed combinative primer sets of PL1F, PL2R, PB1F, PB2R, ITS5F and ITS4R, were from 520 by to 730 bp. Fifteen strains of Phellinus species including P. linteus, P. baumii, P. weirianus, P. johnsonianus, P. rhabarberinus, P. pini, P. gilvus, P. igniarius, P. nigricans and P. laevigatus were examined in this study. Five strains, including two isolated strains of P. linteus(MPNU 7001 and MPNU 7002), and two isolated strains of P. baumii(MPNU 7004 and MPNU 7005) were shown to have about 520 bp (PL1F-PL2R), 700 bp (TTS5F-PL2R) and 600 bp (PB1F-ITS4R) -sized PCR single bands respectively. This molecular genetic technique provided a useful method for rapid detection and identification of P. linteus and P. baumii.

The Determination of the Partial 28S Ribosomal DNA Sequences and Rapid Detection of Phellinus linteus and Related species

  • Species of Phellinus were known to harmful fungi causing white pocket rot and severe plant disease such as canker or heartrot in living trees in the West, but some species have been used to traditional medicines in the Orient for a long time. In this study the partial D1-D2 nucleotide sequences of 28S ribosomal DNA from 13 Phellinus strains were determined and compared with the sequences of 21 strains obtained from GenBank database. According to the neighbor-joining(NJ) method comparing the sequence data the phylogenetic tree was constructed. The phylogenetic tree displayed the presence of four groups. Group I includes P. ferreus, P. gilvus and P. johnsonianus, Group II contains P. laevigatus, P. conchatus and P. tremulae, Group III possesses P. linteus, P. weirianus, P. baumii, P. rhabarbarinus and P. igniarius, and Group IV comprises P. pini, P. chrysoloma. P. linteus and P. baumii, which were used mainly in traditional medicine, belong to the same group, but exactly speaking both were split into two different subgroups. To detect P. linteus only, we developed the PCR primer, D12HR. The primer showed the specific amplification of P linteus, which is permitted to medicinal mushroom in the East. The results make a potential to be incorporated in a PCR identification system that could be used for the rapid identification of this species from its related species, P. linteus especially.

Hypolipidemic Effect of Exo-Polymer Produced in Submerged Mycelial Culture of Five Different Mushrooms

  • The hypolipidemic effect of exe-polymer produced in submerged mycelial culture of Hericium erinaceus (HE), Auricularia auricula-judue (AA), Flammulina veluripes (FV), Phellinus pini (PP), and Grifola frondosa (GF) was investigated in dietary-induced hyperlipidemic rats. The animals were administered with exe-polymers at the level of 100 mg/kg body weight daily for four weeks. Hypolipidemic effect was achieved in all the experimental groups, however, HE exo-polymer proved to be the most potent one, which significantly reduced the plasma triglyceride ($28.9\%$), total cholesterol ($29.7\%$), low-density lipoprotein (LDL) cholesterol ($39.6\%$), phospholipid ($16.0\%$), and liver total cholesterol ($28.9\%$) level, when compared to the saline administered (control) group. The results of the present investigation strongly demonstrate the potential of HE exe-polymer in combating hyperlipidemia in the experimental animals.
