Antioxidant and Anti-inflammatory Effects of Phellinus linteus HN1009K (목질진흙버섯 Phellinus linteus HN1009K의 항산화 활성과 항염증효과)

  • Novel Phellinus linteus HN1009K (HK1009) has been characterized on speedy mycelial growth and yearly artificial cultivation. In this study, antioxidant and anti-inflammatory activities of ethanol extract from mycelium and fruiting body of P. linteus HN1009K (HK1009) were compared to those of P. linteus (PL, Korea Sangwhang) and P. baumii (PB, Jangsu Sanghwang) as controls. HK1009 extract showed higher DPPH free radical scavenging activity in both mycelial and fruiting body samples than those of PL and PB. However, in xanthine oxidase assay, antioxidant effect was highly measured on mycelial samples of PL and HK1009. Mycelial extract of HK1009 induced high inhibitory effect on LPS-induced nitric oxide (NO) production in RAW264.7 cells comparing to PL and PB without toxicity on the cells. The results suggest that P. linteus HN1009K possesses superior antioxidant activity and anti-inflammatory effects.

Inhibitory Effects of PLM-WE1 Formulated from Extract of Phellinus linteus Mycelium against Plant Viruses Infection and Identification of Active Compound (목질진흙버섯(Phellinus linteus) 균사체 추출물 제제 PLM-WE1의 식물 바이러스에 대한 감염억제 효과 및 활성성분의 동정)

  • Pepper mild mosaic virus(PMMoV) and Cucumber mosaic virus (CMV) are important pathogens in various vegetable crops worldwide. We have found that hot water extract of Phellinus linteus mycelium strongly inhibit PMMoV and CMV infection. Based on these results, the inhibitor named as 'PLM-WE1' formulated from extract of Phellinus linteus mycelium was tested for its inhibitory effects on PMMoV and CMV infection to each local lesion host plant (Nicotiana glutinosa: PMMoV, Chenopodium amaranticolor: CMV). Pretreatment effect of PLM-WE1 against infections of each virus (PMMoV and CMV) to local host plant was measured to be 99.2% to PMMoV and 80.3% to CMV, and its permeability effect was measured to be 45.0% to PMMoV and 41.9% to CMV. Duration of inhibitory activity of PLM-WE1 against PMMoV infection on N. glutinosa was maintained for 3 days at 75% inhibition level and CMV infection on C. amaranticolor maintained for 3 days at 62% inhibition level. Inhibitory effects on systemic host plants of PLM-WE1 were measured to be 75~85% to PMMoV and 75% to CMV. Under electron microscope, PMMoV particles were not denatured or aggregated by mixing PLM-WE1. It is suggested that the mode of action of PLM-WE1 differ from that of inactivation due to the aggregation of viruses. The methanol extract of P. linteus mycelium was sequentially partitioned with haxane, ethyl acetate, BuOH and $H_2O$. The $H_2O$ fraction was showed high activity than the other fractions. The active compound was isolated with a partial acid hydrolysis, fractional precipitation with ethanol. The inhibitory effect of the precipitate isolated from 70% ethanol fraction was 99.1% to PMMoV and 88.0% to CMV. The structure of isolated compound was determined by $^1H$-NMR and $^{13}C$-NMR. This compound was identified as a polysaccharide consisting alpha or beta-glucan.

Anti-Inflammatory Activity of Liquid Fermentation by Phellinus linteus Mycelium (상황버섯(Phellinus linteus) 균사체 액체발효물의 항염증 활성)

  • To investigate the industrial availability of liquid fermentation (PL-ferment) by Phellinus linteus mycelium as a postbiotics for the inhibition of inflammation, PL-ferment was fractionated into culture supernatant (CS), hot-water extract (HW) from PL-ferment, EtOH-precipitate (CP) fractionated from HW, and the dialysate (DCP) of CP. Compared to the other fractions, DCP which is expected to contain exopolysaccharide (EPS) as the major component, significantly decreased the production of NO, IL-6, and MCP-1 in LPS-induced RAW 264.7 cells, and IL-6 and IL-8 in TNF-α and IFN-γ-induced HaCaT cells. The general component analysis results showed that no significant difference in components was observed between the fractions, whereas sugar composition analysis revealed that DCP had decreased glucose and increased mannose contents compared to the other fractions. This suggests that mannose played an important role in the anti-inflammatory activity of the active fraction, DCP. Molecular weight distribution analysis revealed that DCP was mainly composed of low-molecular-weight material-removed high-molecular-weight polysaccharides of 18-638 kDa, suggesting that EPS originated from P. linteus EPS. In conclusion, our results suggest that the DCP of P. linteus mycelium fermentation using the anti-inflammatory activity could be used industrially as postbiotic material.

Quality Characteristics of Porridge Containing Rice incubated with Phellinus linteus Mycelium (상황버섯균사체 쌀을 이용한 죽의 품질특성)

  • In this study, we investigated the quality characteristics and optimum conditions of porridge containing rice incubated with Phellinus linteus mycelium (PLMR). Composition measurements of PLMR were moisture 13.05%, ash 0.88%, carbohydrate 78.65%, crude protein 6.29%, and crude lipid 1.13%, also glutinous rice composition was moisture 13.99%, ash 0.32%, carbohydrate 77.18%, crude protein 7.85%, and crude lipid 0.66%. The water absorption rate of PLMR increased with increasing soaking time during 6 hours. However, the water absorption rate of glutinous rice increased until 30 minutes of soaking time. The pH of porridge was decreased as PLMR contents increased. The viscosity of porridge was increased as PLMR contents increased, and it was the lowest for the control at 914.77 cP. However, spreadability of porridge was decreased as PLMR contents increased. In terms of color, lightness was the highest for the 50% level, redness was the highest for the 75% level, and yellowness was the highest for the 25% level. In terms of textural characteristics, hardness and fracturability were increased as PLMR contents increased, whereas springiness, cohesiveness and resilience showed reverse results. In terms of consumer acceptance, flavor, appearance, viscosity, delicate taste, and overall acceptability were not significantly different between samples. Hot taste was decreased as PLMR contents increased, whereas grittiness showed reverse results. In conclusion, the results indicate that adding 25~50% PLMR to porridge is optimal for a recipe.

Immune-Enhancing Effects of Polysaccharides Isolated from Phellinus linteus Mycelium on Mori ramulus (상지에 배양한 상황버섯 균사체로부터 분리된 다당류의 면역증강 효과)

  • The objective of this study was to examine the immune-enhancing effects of polysaccharides isolated from Phellinus linteus mycelium on Mori ramulus. Crude polysaccharides were isolated by pressurized extraction ($121^{\circ}C$, $1.2kgf/cm^2$, 3 h), ethanol precipitation, and lyophilization. In addition, crude polysaccharides were further fractionated into unabsorbed fractions (PF-1, fraction No. 3~15) and absorbed fractions (PF-2, fraction No. 24~33) by DEAE-sepharose CL-6B column chromatography in order to isolate immune-regulating polysaccharides. The major constituents in PF-1 and PF-2 were total sugar (75.51% and 52.38%), total protein (1.63% and 8.41%), uronic acid (17.53% and 15.04%), and ${\beta}-glucan$ (28.33% and 25.04%), respectively. PF-1 increased production of nitric oxide (NO) and cytokines, such as tumor necrosis factor-alpha ($TNF-{\alpha}$) and interleukin-6 (IL-6) in a dose-dependent manner. The mRNA expression levels of inducible NO synthetase, cyclooxygenase-2, $TNF-{\alpha}$, and IL-6 markedly increased as determined by polymerase chain reaction analysis. The above data led us to conclude that macrophage activation of purified polysaccharides was higher than that of crude polysaccharides. The polysaccharides isolated from P. linteus mycelium on M. ramulus investigated herein are useful as natural immune-enhancing agents.

Immunostimulating Activity and Characterization of Polysaccharides from Mycelium of Phellinus linteus

  • Hot-water extract, Fr. 1, of Phellinus linteus mycelium was fractionated into Fr. 2, 3, 4, and 5 by the difference of solubility in ethanol. The polysaccharide fractions were studied for their immunostimulating activity on in vitro T-independent polyc1onal antibody response to trinitrophenyl-haptened SRBC (sheep red blood cell). The Fr. 4 with the highest immunostimulating activity was subjected to DEAE-cellulose ion exchange chromatography and gave five fractions, 4-I, II, III, IV, and V. The in vitro immunostimulating assay of the five fractions showed that 4-I and 4-III had a similar activity to that of LPS but the other fractions had low activity. By analyses of chemical composition and HPLC, all fractions obtained were found to be heteropolysaccharide-protein complex. The molecular weights ranged from 9, 000 to 15, 000. Sugar analyses showed that glucose, galactose, mannose, arabinose, and xylose were main component. Uronic acid and amino sugar were also detected in the fractions. It should be noted that the molecular weight (15, 000) of 4-III was very small and the structure of 4-III may be different from the known immunostimulating branched $\beta$-(1longrightarrow3)-glucan.

Effects of Fermented Rice Wine Using Mycelium of Phellinus linteus on the Gastric Mucosa of Rat (상황버섯 균사체를 이용한 발효주가 흰쥐의 위점막에 미치는 영향)

  • It was examined the effect of fermented rice wine using mycelium of Phellinus linteus (FWPL) on the gastric mucosa of rat. The gastric mucosal lesions were not seen macroscopically in normal, but ethanol-administrated rats produced congestion and edema with a few local lesions. The administration of FWPL showed a similar pattern as like normal except trace histopathological changes. The results of Western blot analyses showed that the higher expression of inducible nitric oxygenase (iNOS), cyclooxygenase (COX)-1, COX-2, tumor necrosis factor-$\alpha$ and c-fos, especially COX-2, in the ethanol-administrated rat compared with normal rat. But FWPL-administrated rat showed a trace increase of these expression compared to normal rat. About immunohistochemical observations, weaker iNOS reactions were detected in mucous cells of epithelim of ethanol administrated rat compared with normal and FWPL-administrated rat. These results suggested that FWPL-administrated rat showed a trace changes on the mucus barrier-related protein expression compared with ethanol-administrated rat and thus FWPL may be use to develop a functional alcoholic beverage.

Effects of Traditional Wine by using Mycelium of Phellinus linteus on the Expression of Inflammation-Related Proteins in Rat Liver (상황버섯 균사체를 이용한 전통주의 장기투여가 흰쥐 간장에 미치는 영향)

  • It was examined that the effect of fermented traditional wine made by using mycelium of Phellinus linteus (TWPL) on the expression of inflammation-related proteins in rat liver. Levels of aspartate aminotransferase (AST) was significantly increased in the serum of ethanol-treated rats compared to normal. However, the level of AST showed no significant changes in the TWPL-treated rat compared normal. Slight histopathological changes of liver such as cloudy swelling, inflammatory cells infiltration, Kupffer cell reaction were demonstrated in the rats challenged with ethanol compared with normal. Fewer scores of these changes were observed in TWPL-treated rat with recovered glycogen in hepatocytes of whole hepatic lobule. The RT-PCR and Western analysis showed that the expression of inflammatory proteins such as cyclooxygenase-2, inducible nitric oxide synthase, tumor necrosis factor (TNF)-${\alpha}$ were decreased in the TWPL-treated rat compared with ethanol-treated ones. Immunohistochemical analysis showed that the expression of interleukin-lf and TNF-${\alpha}$ tended to decrease in TWPL-treated rat compared with ethanol-treated ones. These results suggest that TWPL may contains some protective agent for alcohol-induced liver injury through a regulating inflammation-related proteins.

Enzyme activity of Basidiomycetes products in each cereals (곡물별 담자균 배양물의 효소활성)

  • In this study, growth rates and enzyme activities of mushroom mycelium were investigated in each cereals. Cultivation on hulled barley resulted in slightly faster mycelial growth as compared to other substrates. Enzyme activities were measured in different periods. In result, ${\alpha}$-amylase activity was higher in Pleurotus ostreatus and Phellinus linteus cultured cereals, whereas ${\beta}$-amylase activity was higher in Pleurotus ostreatus and Fomitella fraxinea cultured. Protease level did not affect kinds of cereal except Phellinus linteus. The color values lightness decrease by Basidiomycetes cultivation and redness and yellowness increased.

Antioxidant activity of extracts with extraction methods from Phellinus linteus mycelium on Mori ramulus (추출 방법에 따른 상지에 배양한 상황버섯 균사체 추출물의 항산화 활성)

  • This study was conducted to examine the antioxidant activities of extracts derived with different extraction methods (HE, hot water extraction; UE, ultrasonic extraction; and PE, pressured extraction) from Phellinuslinteus mycelium in Mori ramulus. The extraction yield of PE (7.73%) was higher than that of UE (6.03%) and of HE (5.44%). The total phenolic and flavonoid contents of PE were 9.87 g/100 g and 1.90 g/100 g, respectively. The oxygen radical absorbance capacity (ORAC) of PE ($769.63{\mu}M/g$ FW) was higher than that of HE ($622.96{\mu}M/g$ FW) and of UE ($249.06{\mu}M/g$ FW). The DPPH and ABTS radical scavenging activities of PE at $1,000{\mu}g/mL$ were 35.85% and 51.42%, respectively. The superoxide radical scavenging activity of PE, 29.16-93.89%, was higher than that of other extracts. The ferric-reducing antioxidant power and the reducing power of PE were $180.60-607.93{\mu}M$ and 0.14-0.51, respectively. The tyrosinase inhibition activity of PE (9.66-24.05%) improved with an increase in the treatment concentration. The antioxidant and tyrosinase inhibition activities of PE were significantly higher than those of the other extracts. In conclusion, we provided experimental evidence that Phellinus linteus mycelium extracts from Mori ramulus have potential as functional materials.
