Effect of the Cultivated Fruit body of Phellinus linteus on Low Density Lipoprotein Oxidation (배양상황버섯의 저밀도지질단백질 (LDL)산화에 미치는 영향)

  • Phellinus linteus (Hymenocaetaceae) has been used for the treatment of gastric cancer, noninsulin dependant diabetes, diarrhea, and menstrual irregularity. The antioxidative effect of cultivated fruit body of Phellinus linteus on low density lipoprotein oxidation was investigated. The MeOH and water extracts were examined by TEARS assay on human plasma LDL in vitro system. The MeOH ex. showed antioxidative activity, and then was fractionated into Precipitates, Water Insoluble fr., Water fro and ether fro The results showed that the lipophilic fractions, Water Insoluble fr. and Ether fr. of cu 1-tivated fruit body of Phellinus linteus, inhibited the oxidative modification of LDL.

  • Reactive oxygen species(ROS) or free radical-mediated oxidative stress plays an important role in the pathophysiologic process of disease state. This study investigated antioxidative effects of Phellinus linteus extract on the generation of 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, superoxide anion radical(${O_2}^-$), hydroxyl radical(${\cdot}OH$), nitric oxide(NO), and peroxynitrite($ONOO^-$) radical and free radical-mediated protein oxidation under in vitro assay systems. This results showed that Phellinus linteus extract effectively inhibited the generation of free radicals in the all assay system with dose-dependent manner and also significantly reduced the protein oxidative level. Thus, the present study indicates that Phellinus linteus extract possesses a potent antioxidant activity and plays a beneficial role against free radical-induced oxidative injury.

Quality Characteristics of Yanggaeng Supplemented with Sanghwang Mushroom (Phellinus linteus) Mycelia (상황버섯 균사체를 이용한 양갱 제조 및 품질 특성)

  • The principal objective of this study was to examine the quality characteristics of yanggaeng supplemented with powder derived from Sanghwang mushroom (Phellinus linteus) mycelia. We analyzed the potential of utilizing Phellinus linteus mycelia as a functional food material by estimating total polyphenol and flavonoid contents, electron-donating abilities, as well as antioxidative activities of the water and ethanol extracts of Sanghwang mushroom mycelia. The total phenol and flavonoid contents of ethanol extracts from Phellinus linteus mycelia were 0.69 mg/ml and 0.16 mg/ml, respectively, while the contents from the water extract of Phellinus linteus mycelia were 0.66 mg/ml and 0.22 mg/ml, respectively. The electron-donating abilities of ethanol and water extracts from Phellinus linteus mycelia were 88.64 and 90.29%, respectively. The ABTS radical scavenging activities of ethanol and water extracts from Phellinus linteus mycelia were 89.74 and 71.35%, respectively. In terms of color values, as the level of powder increased, the value of L (lightness) decreased, whereas those of a (redness) and b (yellowness) increased. In regard to the mechanical properties of the samples, we noted significant differences in hardness, springiness, chewiness, and gumminess (P<0.05). The results of the sensory evaluation showed that the score from SHM (Sanghwang mushroom mycelia) with 4% powder was significantly higher than other samples in terms of sweetness, color, taste, texture and overall quality (P<0.05). Taken together, the recommended level of Phellinus linteus mycelia powder in yanggaeng is 4% for optimal sensory characteristics.

Fermented Saccharina japonica with Phellinus linteus Mycelia Enhances its Radical Scavenging Activities (상황버섯 균사체로 발효한 다시마 추출물의 항산화 활성)

  • To develop new functional materials, Saccharina japonica (S. japonica) was cultivated with Phellinus linteus (P. linteus) mycelia by solid culture. S. japonica was fermented with P. linteus mycelia. The various radical scavenging activities of extracts of fermented S. japonica with P. linteus mycelia (FSPM) were investigated and compared to the antioxidant capabilities of unfermented S. japonica and P. linteus mycelia. The antioxidant activities of FSPM extracts were evaluated using ferric reducing antioxidant power (FRAP) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. FSPM showed stronger free radical scavenging abilities than P. linteus mycelia or S. japonica alone. These results indicate that fermentation of S. japonica changes its chemical nature and could provide beneficial antioxidant effects.

  • To examine the chemical properties of the polysaccharide from Phellinus linteus, we obtained the polysaccharide treated by the alkali and hot water. The carbohydrate and protein contents of each polysaccharide 81.1%, 37.4% and 6.2%, 21.8%, respectively. The carbohydrate consisted of six monosaccharides and the protein contained 16 amino acids.

  • Artificial cultivation of Phellinus linteus was studied. The best yield (B.E. $9.4{\pm}1.6$) of basidiocarps including basidiospore was obtained using Quercus acutissima among the logs of Morus alba, Quercus acutissima, Alnus japonica, Castanea crenata. Optimum temperature and required periods for the basidiocarps formation were $25^{\circ}C$ and about 10 months, respectively.

  • This study was peformed to determine the antioxidant, antimicrobial activity and antifungal of the cambodian mushroom, Phellinus linteus, in various concentrations from the mushroom extracts. It's extracts were found to cause significant free radical scavenging effects on DPPH (1,1-diphenyl-2-picrylhydrazyl). This extracts exhibited in vitro broad-spectrum antimicrobial activity of Gram-negative, Gram-positive bacteria and without antifungal activity. On the other hand, the extracts had not significant hemolytic activity against human red blood cells. These results indicate cambodian mushroom, Phellinus inteus having antioxidant and antimicrobial activity without hemolytic activity maybe useful as therapeutic agents.

Purification of Anti-obesity Lipase Inhibitor from the Fruiting Body of Phellinus linteus (상황버섯 자실체로부터 항비만성 라이페이스 저해물질의 정제)

  • For development of anti-obesity nutraceuticals from mushrooms, new anti-obesity lipase inhibitor from Phellinus linteus was purified by systematic solvent extraction, TLC and HPLC and characterized. Methanol extract from P. linteus most effectively inhibited(72.5%) porcine pancreatic lipase and ethyl acetate fraction showed the highest inhibitory activity in the systematic solvent extraction. A lipase inhibitor from the ethyl acetate fraction was purified through following steps including 3 times silica gel chromatography and preparative HPLC. The purified lipase inhibitor was a yellowish geen powder and its $IC_{50}$ value was 175 ng. Its molecular weight by MALDI-TOF-MS was 523.06 Da and showed maximal absorption spectrum at 225.1 nm.

Antimutagenic and Cytotoxicity Effects of Phellinus linteus Extracts (상황버섯(Phellinus linteus) 추출물의 항돌연변이원성 및 세포독성 효과)

  • This study was performed to determine the antimutagenic and cytotoxic effect of the Phellinus linteus methanol extract on Salmonella typhimurium TA98, TA100 and human cancer cell lines. In the Ames test, methanol extract of P. linteus alone did not exhibit any mutagenicity but showed substantial inhibitory effects against mutation induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), 4-nitroquinoline-nitroquinoline-1-oxide(4NQO), 3-amino-1,4-dimethyl-5H-pyrdo[4,3-blindol(Trp-P-1) and benzo(α)pyrene(B(α)P). The methanol extracts of P. linteus(200㎍/plate) showed approximately 78.3%, 78.7% and 88.1% inhibitory effect on the mutagenesis induced by 4NQO, Trp-P-1 and B(α)P. The anticancer effects of P. linteus extract against human breast adenocarcinoma(MCF7), human lung carcinoma (A549), human fibrosarcoma (HT1080), human hepatocelular carcinoma (Hep3B) and human epitheloid carcinoma (HeLa) were investigated. The treatment of 1mg/mL P. linteus extracts had the highest cytotoxicity against MCF7 (92.0%), followed by Hep3B (84.9%), A549 (84.2%) and HT1080 (82.9%). In contrast 1mg/mL treatment of P. linteus extracts had only 10∼40% cytotoxicity on normal human liver cell (WRL68).

Antioxidant and Anticancer Effects of Fermentation Vinegars with Phellinus linteus, Inonotus obliquus, and Pleurotus ostreatus (상황버섯, 차가버섯, 느타리버섯 발효식초의 항산화와 항암활성 효과)

  • The antioxidant and anticancer activities of Phellinus linteus, Inonotus obliquus, and Pleurotus ostreatus according to extraction methods were measured. The contents of polyphenolic compounds were higher in fermented extracts than those in water extracts. The contents were 192.4mg/100 $m{\ell}$ for Inonotus obliquus vinegar, 186.9 mg/100 $m{\ell}$ for Phellinus linteus vinegar, and 156.5 mg/100 $m{\ell}$ for Pleurotus ostreatus vinegar, respectively. EDA(electron donating ability) in Phellinus linteus that was highest among mushrooms was 83.9% in the water extract and 96.8% in the fermented extract. The growth of stomach cancer cells, SNU-719 was inhibited 62.4%, 65.5%, and 53.0% by Phellinus linteus vinegar, Inonotus obliquus vinegar, and Pleurotus ostreatus vinegar, respectively and also the growth of liver cancer cells, Hep3B was inhibited 67.1%, 68.3%, and 57.6% by Phellinus linteus vinegar, Inonotus obliquus vinegar, and Pleurotus ostreatus vinegar, while the growth of normal cell, DC2.4 was not affected. Even though Phellinus linteus and Inonotus obliquus showed higher inhibition in cancer cell growth, Pleurotus ostreatus can be efficiently used for antioxidant and anticancer activities due to their cheap price in the market.
