Protective Effect of water extract Phellinus linteus-discard Schisandra chinensis solid fermented extracts on improvement of sarcopenia by Atorvastatin-induced muscle atrophy cell model (Atorvastatin으로 유도된 근위축 세포모델에서 상황-오미자박 고상발효물 열수추출물의 보호효과)

  • Objectives : This study is to effect of improving muscle atrophy through water extract on the solid-phase fermentation extraction with Phellinus linteus of discarded Schisandra chinensis in an atorvastatin-induced atrophy C2C12 cell. Methods : C2C12 myoblast were differentiated into myotube by 2% horse serum medium for 6 days, and then treated solid-phase fermentation(S-P) extract at different concentrations for 24h. To investigate the effect of S-P extract on the induction of muscle atrophy and expression of atrophy-related genes and apoptosis in differentiated C2C12 myotubes using a GSH, ROS, real-time PCR, western blots analysis. Results : As a result of treatment with atorvastatin at concentrations of 5, 10, and 20 uM on the 6th day of differentiation in C2C12 myotube cells, it was confirmed that the cell morphology was damaged in a concentration-dependent manner, and the length and thickness of the myotube also decreased in a concentration-dependent manner. Treatment with S-P extract (50, 100 and 200 ㎍/㎖) increased of GSH and inhibited ROS in the atorvastatin-induced muscle atrophy cell model at a concentration that did not induce toxicity. In addition, it was confirmed that it has an effect on muscle reduction by inhibiting apoptosis of muscle cells as well as being involved in protein production and degradation of muscle cells. Conclusions : Atorvastatin-induced atrophy C2C12 cell, S-P extract activates related to differentiation/generation and proteolysis, and inhibits cell death of atrophy in C2C12 cell. Based on this, it is necessary to prove its effectiveness through animal models and human application test, but it is considered to be discarded Schisandra chinensis can present the potential for development as a recycling industrial material.

Antibacterial, antioxidant and antitumor activities of mushroom mycelium mixed culture extracts (버섯 균사체 혼합 배양 추출물의 항균, 항산화 및 항암 활성)

  • In this study, we have investigated the antibacterial, antioxidant, and antitumor activities of mycelium cultural extract from mushroom. Mushroom mycelium was grown in a synthetic liquid media such as PD broth, YM broth or citrus extracts. In antibacterial activity test, the best result was achieved when mycelium cultural extracts from Phellinus linteus and Coriolus versicolor were incubated together on YM broth. On the other hand, mushroom mycelium cultured on citrus extracts showed better activity than that on PD broth. We have also tested the antioxidant activity at concentration up to 10 mg of mycelium cultural extract/mL. The more it is in higher concentration, the more the activity increases. The higher antioxidant activity was observed both on PD broth containing the Phellinus linteus and Coriolus versicolor mycelium and citrus extract containing the same. The complex culture extracts obtained from the synthetic medium and citrus extract medium showed 10-89% of the 1,1-diphenyl-2-picrylhydrazyl radical scavenger activity. The antitumor activity of mycelium cultural extract was examined by using MTT assay on A549 cells. Mushroom mycelium cultured on citrus extracts showed interestingly higher antitumor activity than that on synthetic liquid media.

Induction of Apoptotic Cell Death by Healthful Decoction Utilizing Phellinus Linteus in Human Lung Carcinoma Cells (상황을 이용한 보건기능 개선제의 인체폐암세포 apoptosis 유발에 관한 연구)

  • In the present study, we investigated the effects of aqueous extract of the healthful decoction utilizing Phellinus linteus (HDPL) on the cell growth of human lung carcinoma tumor cell line A549. Exposure of A549 cells to HDPL resulted in growth inhibition and induction of apoptosis in a dose-dependent manner as measured by hemocytometer counts, fluorescence microscopy and flow cytometric analysis. This increase in apoptosis was associated with inhibition and/or degradation of apoptotic target proteins such as poly(ADP-ribose) polymerase (PARP), b-catenin and phospholipase C- 1 (PLC- 1) protein. HDPL treatment induced the down-regulation of anti-apoptotic Bcl-2 expression, an anti-apoptotic gene, however, the level of Bax. a pro-apoptotic gene, was increased by HDPL treatment. In addition, HDPL-induced apoptotis of A549 cells was connected with activation of caspase-3 and caspase-9 protease in a dose-dependent manner, however, the levels of inhibitor of apoptosis proteins family were remained unchanged. Taken together, these results indicated that the anti-proliferative effects of HDPL were associated with the induction of apoptotic cell death through regulation of several major growth regulatory gene products such as Bcl-2 family expression and caspase protease activity, and HDPL may have therapeutic potential in human lung cancer.

Antibacterial Activities of Mushroom Liquid Culture Extracts Against Livestock Disease-Causing Bacteria and Antibiotic Resistant Bacteria (가축질병 세균 및 항생제 내성 세균에 대한 버섯 균사체 배양 추출물의 항균 활성)

  • The ethyl acetate extracts from the liquid cultures of Coriolus versicolor, Phellinus linteus, and Hericium erinaceus showed significant antibacterial activities against Escherichia coli K88, E. coli K99, E. coli 987P, and Salmonella typhimurium 14058 causing bacterial diarrhea in Korean house pigs and chicken. Of these extracts, Coriolus versicolor extract showed the highest antibacterial activity. In addition, these extracts also showed significant growth inhibition against Staphylococcus aureus CARM3230 and E. coli CARM1381 which are known as kanamycin and ampicillin-resistant strains. These results showed that the mushroom extracts could be developed as a livestock feed additives that can replace commercial antibiotics, and also could be good resources for the development of a new antibacterial agent.

Down-regulation of COX-2 and hTERT Expression by Healthful Decoction Utilizing Phellinus Linteus in Human Lung Carcinoma Cells (상황을 이용한 한의학적 보건기능 개선제에 의한 인체폐암세포의 증식억제에 관한 연구)

  • The objective of the present study was to investigate the effects of aqueous extract from the healthful decoction utilizing Phellinus linteus (HDPL) on the growth of human lung carcinoma A549 cells. HDPL treatment declined the cell viability of A549 cells in a concentration-dependent manner and the anti-proliferative effects by HDPL treatment were associated with morphological changes such as membrane shrinking and cell rounding up. HDPL treatment did not affect the distribution of the cell cycle. Western blot analysis and RT-PCT data revealed that the levels of tumor suppressor p53 and cyclin-dependent kinase inhibitor p21WAF1/CIP1 in HDPL-treated A549 cells were remained unchanged. However, HDPL treatment inhibited the expression of cyclooxygenase-2 (COX-2) mRNA and protein in a concentration-dependent fashion. Additionally, the expression of human telomerase reverse transcriptase (hTERT), a main determinant of the telomerase enzymatic activity, was progressively down-regulated by HDPL treatment. Taken together, these findings suggest that HDPL-induced inhibition of human lung cancer cell proliferation is associated with the inhibition of several major growth regulatory gene products, such as COX-2 and hTERT, and HDPL may have therapeutic potential in human lung cancer.

Single- and Repeated-dose Toxicities of Acanthopanax senticosus Fermentation Products in Rats (흰쥐에서 가시오가피 발효물의 단회 및 반복투여 독성평가)

  • In this study, the products of Acanthopanax senticosus fermentation were derived from the mycelia of 2 mushrooms, Ganoderma lucidum and Phellinus linteus, to determine their safety in Sprague-Dawley rats. Rats were orally administered the water extracts of A. senticosus fermentation products with G. lucidum (FM-5111) or P. linteus (FM-5131) at dose levels of 0.5, 1.0, or 2.0 g/kg for the single-dose toxicity test and 0.25, 0.5, or 1.0 g/kg for the repeated-dose toxicity test. There were no significant differences in body weight gain, feed intake, or water consumption between control and FM-5111- or FM-5131-treated rats. Hematological and blood biochemistry analysis revealed that none of the investigated parameters were affected by the A. senticosus fermentation products, and no remarkable lesions were observed upon histopathological analysis. We conclude that the A. senticosus fermentation products obtained from mushroom mycelia are safe for long-term administration and could be considered as multi-functional nutrients for the improvement of liver function and immunity.

Anti-inflammation Activity of Water Extracts from Hericium Erinacium among Medicinal Mushrooms (약용 버섯 중 노루궁뎅이 버섯 열수 추출물의 항염증 효과)

  • Macrophage-derived nitric oxide (NO) plays an important role in immune responses. Overproduction of NO by inducible nitric synthase(iNOS) is known to be closely correlated with the pathology of a variety of diseases and inflammations. This study investigated the inhibitory effect of extract from various mushrooms(Phellinus linteus(PL), Inonotus obliquus(IO), Sarcodon aspratus(SA), Hericium erinacium(HE), and Russula cutefracta(RC)) on NO production and its molecular mechanism in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Among the mushrooms, HE extracts exhibited the strongest NO-inhibition activity. It was also found that HE extract inhibited LPS-induced iNOS and COX-2 expression in RAW264.7 cells. The present results give the understanding of biological activities of mushrooms and encourage their application for supplements and cook.

Effects of Fermented Rice Wine by Using Mycelium of Phellinus linteus on the Expression of Inflammation-Related Proteins in Human Hepatoma Cells and Rat Liver (상황버섯 균사체를 이용한 발효주가 인체간암세포와 흰쥐 간의 염증관련 단백질 발현에 미치는 영향)

  • We have recently discovered that mycelium of Phellinus linteus, popular medical mushrooms in Korea, possess alcohol dehydrogenase and produce alcohol. In the present study, it was examined that the effect of fermented rice wine made by using mycelium of P. linteus (FLMP) on the expression of in-flammation-related proteins in both $HepG_2$ cells and rats. To examine the effect of FLMP on the morphology and expression of inflammatory proteins in $HepG_2$ cells, the cells were incubated with ethanol, and FLMP for 24 hours, and then analyzed by microscopic observation and Western blot and reverse transcription polymerase chain reaction (RT-PCR). While ethanol induced the morphological change accompanied with cell debris formation and scattering on $HepG_2$ cells, FLMP had no effect. The results of Western blot and RT-PCR analyses showed that the level of inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-1 and COX-2 was induced by ethanol, however, FLMP inhibited the expression of these proteins and its mRNAs. In the animal model, the value of flutamate oxaloacetate transaminase and glutamate pyruvate transaminase was significantly increased by administration with ethanol. But the group administrated with FLMP showed lower levels on the changes of these markers compared with ethanol-administrated group. Besides, the results of Western blot and RT-PCR analyses showed that the expression of inflammatory proteins such as iNOS, COX-1 and COX-2 was not affected by FLMP administration in rat liver. About histopathological and immunohistochemical observations, inflammatory loci were markedly decreased in the FLMP-administrated rat compared to ethanol-administrated rats and showed weaker COX-2 and iNOS jmmunoreactions. These results suggested that FLMP showed slight changes on the inflammatory proteins expression compared to ethanol and FLMP may be used as a functional alcoholic beverage.

Enhancement of Immunostimulation by Fractionation of Active Polysaccharide from Fermented Ginseng with Phellinus linteus Mycelium in Solid Culture (면역활성 증진을 위한 수삼의 상황버섯 균사체 고체배양으로 조제된 수삼발효물로부터 활성 다당류 분획)

  • Crude polysaccharide (WG-PL-CP) was fractionated from fermented ginseng with Phellinus linteus in solid culture to enhance the immunostimulation of ginseng. WG-PL-CP produce three active polysaccharide-rich fractions (WG-PL-CP-II, III, and IV) on DEAE-Sepharose CL-6B ($Cl^-$ form). WG-PL-CP-III displayed higher mitogenic activity (1.98-fold of the saline control at $100\;{\mu}g/mL$) than did WG-CP-III or PL-CP-III (1.60- or 1.65-fold, respectively), and potent intestinal immune system modulating activity through Peyer's patch was obtained by WG-PL-CP-IV only (1.56-fold). Meanwhile, WG-PL-CP-II and III significantly enhanced macrophage stimulating activity (2.01- and 1.94-fold) compared to WG-CP-II and III (1.73- and 1.66-fold) or PL-CP-II and III (1.79- and 1.72-fold). In addition, WG-PL-CP-III and IV mainly contained neutral sugar (73.5 and 67.3%) and uronic acid (23.2 and 24.6%). Component sugar analysis also showed that WG-PL-CP-III consisted mainly of uronic acid as well as the neutral sugars Glc, Ara, Gal, Rha and Xyl (molar ratio of 0.81:1.00:0.49:0.42:0.28:0.20), whereas WG-PL-CP-IV was mainly comprised of uronic acid, Ara, Rha, Gal, Xyl and Glc (1.00:0.75:0.69:0.63:0.42:0.34). Therefore, it is assumed that these active polysaccharides play an important role in enhancing the immunostimulation of fermented ginseng with P. linteus in solid culture.

Taxonomical Classification and Species-specific Detection of Genus Some Phellinus using Phylotype (Phylotype에 의한 수종의 Phellinus속의 분류체계 확립 및 종간구별을 위한 신속동정법 개발)

  • This study was carried out to identify the phylogenetic relationship of Phellinus species and to know its distribution by comparing the DNA sequences of internal transcribed spacer regions(ITS1 and IST2) and 5.8S ribosomal DNA (rDNA) repeat unit. The Phellinus species had their specific sequences in IST1 and 2 regions depending on suedes. The comparison of the ITS sequences of standard strains indicated that the sequences of ITS1 were more variable than those of ITS2. Nine strains of the commercial products of Phellinus species used in this study were identified as P. lintues, P. baumii, P. igniarius, and P. pini. Most of commercial species were P. pini and P. baumii, and P. gilvus was not found. Also, P. linteus was only found in form of mycelial culture rather than fruiting body. Moreover, the species-specific primers were designed based on ITS sequence data. Each species-specific primers were bound in P. lintues(ITSF-PL2R), P. baumii(PB1F-ITS4R), P. igniarius(IF1-IR3), P. pini(PF1-PR3), and P. gilvus(GF2-GR4), respectively. These primer sets would be useful fer the detection of specific-species among unidentified Phellinus species rapidly.
