Effect of Beta-glucans Extracted from Phellinus baumii on the Growth of Caenorhabditis elegans (예쁜꼬마선충의 생육에 관한 장수상황버섯의 베타글루칸 함유 추출물의 영향)

  • This study investigates the effect of ${\beta}$-glucans on the growth of Caenorhabditis elegans. Comparison was made among lipopolysaccharide (LPS) and ${\beta}$-glucans extracted from Phellinus baumii, in the presence of peptidoglycans which is available as the major carbon source from OP50, a non-pathogenic strain of Escherichia coli. When the three sources of carbohydrate were added singularly or in mixture to the culture media, a significant level of variation was observed with respect to fecundity. Addition of ${\beta}$-glucans appeared to increase the fecundity. When ${\beta}$-glucans was reinforced in the culture media, the fecundity increased at least 20 percent compared to the OP50-only media which exclusively contains peptidoglycans. In terms of life span, C. elegans showed a modest reduction when treated especially with ${\beta}$-glucans. C. elegans accumulated less fat in the ${\beta}$-glucans containing media different from the OP50 media. Based on the Sudan black staining, fat deposition significantly decreased corresponding to the ${\beta}$-glucans content in the media. On LPS-supplemented media, no difference was observed in fat deposition compared to the normal OP50 media. At the level of motility, ${\beta}$-glucans-treated worms moved more distance as well as LPS-treated worm. They also showed a comparable degree of motility under similar treatment with each source of carbohydrate. In conclusion, LPS and ${\beta}$-glucans, extracted from P. baumii, may not entirely replace the food required for C. elegans; however, it might be utilized as valuable alternative food source which C. elegans use as forms of carbohydrates in stead of peptidoglycan of OP50.

For a better and healthy aging, it is even more important for active seniors to manage their health. The development of functional ingredients targeted for active seniors, which may improve cardiovascular and menopausal women's health, and their certification as individually recognized functional materials could serve as a new growth engine for the functional foods industry as well as contribute to the improvement of public health.

  • 상황버섯은 오랜 역사와 전통이 있는 소재로서 폴리페놀류가 함유되어 있어 항암, 혈중콜레스테롤 저하, 혈압강하 등의 약리효과가 있는 것으로 알려져 있다. 본 연구에서는 in vivo 실험을 통해 CPA (Cyclophosphamide)로 유도된 면역저하 마우스 모델에서 상황버섯 투여 농도에 따른 cytokine 활성 효과를 확인하여 상황버섯의 면역 증진 효과를 평가하고, 유효한 농도를 선정하여 시제품을 제작하고자 하였다. 연구결과, 림프기관 장기 무게(흉선, 비장, 악하림프절)와 혈청과 비장의 cytokine이 CPA군(CPA control)에 비해 상황버섯 추출물 투여군에서 유의적으로 증가하였다. 또한 실험을 퉁해 선정한 유효한 농도인 50 mg/kg의 농도로 시제품을 제작하였으며, 상황버섯 특유의 떫은 맛을 감쇄하기 위해 아스코르빈산과 NaHCO3을 첨가하여 최적 배합 비율을 선정하여 음료 200 mL당 상황버섯 추출물 55.6 mg (0.0278%), 아스코르빈산 200 mg (0.1%), NaHCO3 60 mg (0.03%)이 함유 되도록 제작하였다. 본 연구 결과 상황버섯추출물은 CPA로 인해 감소된 사이토카인 분비를 상향 조절함으로써 면역저하 상태를 개선시켜 면역 증진 효과가 있는 것으로 판단된다.

Screening of Anti-gout Xanthine Oxidase Inhibitor from Mushrooms (버섯류로부터 항통풍성 Xanthine oxidase 저해물질의 탐색)

  • Anti-gout xanthine oxidase inhibitory activity of water extracts from various mushrooms were determined. The highest xanthine oxidase inhibitory activity was 72.9% in the water extract from fruiting body of Agaricus brazillensis and also were high in the extract from fruiting bodies of Pleurotus salmoneostramineus(60.1%), Phellinus baumii(57.7%), Agaricus bisporus(56.7%) and Hericium erinaceum(53.4%). The xanthine oxidase inhibitor was maximally extracted when Agaricus brazillensis fruiting body was treated with water at $30^{\circ}C$ for 24 h.

  • The Aphyllophorales is a large order containing about 2,000 known species. Many of these are the bracket and coral fungi. The vast majority of these fungi are saprophytic on the plant debris. Many species are significant in decomposing plant remains, as they are able to digest cellulose or lignin that occurs in plant cell walls. Many of these fungi have been involved in everyday human affairs. A few were used medicinally by the Greeks and Romans as a remedy for many complaints, including colic, fractured limbs and bruises. Other bracket fungi have been used as curry combs for horses, as snuff, as razor strops and as a source of dye for clothing. The texture of the basidiocarp may be similar to that of cork, wood, leather, paper, or cartilage. Unlike the basidiocarps of the Order Agaricales, the basidiocarps of the Aphyllophorales are not fleshly and moist. Division of the members of the Aphyllophorales into genera was originally made on the basis of gross morphology of the basidiocarp and hymenium and Donk(1964) recognizes 22 families in this order. The species and genus whose typical in Aphylloporales were listed in Table. with related information. The ITS region sequence of some genus were found by BLAST search. Sequences retrieved from GenBank were visually aligned by the program CLUSTAL G. As a result, the medicinal mushroom was separated in four groups. In this multiple alignment, the sequence analysis among Fomes group, Inonotus group and Phellinus group showed high genetic similarity except Hericium group and Sparassis group.

The physicochemical characteristics and antioxidant capacities of commercial tea products from Phellinus baumii, Ganoderma lucidum (상황버섯과 영지버섯 차류 제품의 이화학적 특성 및 항산화능)

  • This study was conducted to investigate the physicochemical characteristics, antioxidant capacities of Phellinus linteus and Ganoderma lucidum commercial tea products. The physicochemical characteristics included pH, Hunter's color values, soluble solid contents, evaporation residues, and ${\beta}$-glucan contents. The antioxidant capacities were measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities, ferric reducing antioxidant power (FRAP), and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities, total phenolic contents (TPC), and total flavonoid contents (TFC). The pH, soluble solid contents, evaporation residues, and ${\beta}$-glucan contents were in the range of 4.43-7.05, $0.40-0.73^{\circ}Brix$, 62.04-258.84 mg/100 g, and 15.51-62.32 mg%, respectively. Hunter's color values (L, a, and b) indicated 41.76-55.02, -0.49-5.06, and 17.41-28.32, respectively. The antioxidant capacities showed $32.63-367.81{\mu}M$ GAE (DPPH radical scavenging activities), $321.86-1,035.19{\mu}M$ TE (FRAP), $703.50-1,091.83{\mu}M$ (ABTS radical scavenging activities), $286.56-916.00{\mu}M$ (TPC), and $85.33-635.33{\mu}M$ (TFC). Overall, P. linteus liquid tea 2 (PL2) and G. lucidum liquid tea 1 (GL1) showed high antioxidant capacities (p<0.05). The TPC and TFC were highly correlated with DPPH radical scavenging activities, FRAP, and ABTS radical scavenging activities (r=0.7298-0.9743), but the ${\beta}$-glucan contents were not correlated well with antioxidant activities tested (r=0.3146-0.6663).

Quality Characteristics and Antioxidant Activity of Fermented Milk containing Mushroom Extracts (버섯 추출물을 첨가한 발효유의 품질특성 및 항산화 활성)

  • This study was carried out to investigate the quality characteristics and antioxidant activity of fermented milk containing mushroom (Phellinus baumii, Ganoderma lucidum, and Lentinus edodes) extracts. As the ratio of the mushroom extract increased, the pH of the fermented milk decreased proportionally and titratable acidities increased significantly. The number of lactic acid producing bacteria was the highest in the fermented milk sample containing 1.0% Lentinus edodes extract. The DPPH and ABTS radical scavenging activities of the fermented milk containing mushroom extracts were higher than that of the controls. The quality characteristics, such as pH, titratable acidity, and the number of lactic acid producing bacteria were not remarkably different between the milk samples subjected to treatments with and without the addition of mushroom extracts during the storage period. From the sensory evaluation of the fermented milk samples containing mushroom extracts, the color, flavor, taste, texture, and overall acceptability of the fermented milk sample containing 1.0% Lentinus edodes extract was found to be considerably better than those of the other groups. In conclusion, the present study indicated that the fermented milk containing mushroom extracts could be used as a functional antioxidant containing food.
