Measurement of the Anti-oxidative Properties of Extract from Medicinal Plants Using an On-line HPLC-DPPH Assay (HPLC와 DPPH radical 소거능 측정 방법의 결합에 의한 약용 식물 추출물의 항산화 활성 비교)

  • Natural anti-oxidative compounds have important disease prevention and food preservation properties, in addition to anti-bacterial, anti-inflammation, anti-cancer, and skin whitening effects. High-performance liquid chromatography (HPLC), with an ultra vilolet (UV) detector coupled to a reverse phase C18 column and an online measurement system for 1, 1-diphenyl-2-picryl hydrazyl (DPPH) radicals, was used to search for potent antioxidative compounds in crude extracts. The online HPLC-DPPH assay was then applied to confirm antioxidative compounds in water extracts from Radix of Pueraria lobata, Rhizoma of Zingiber officinale, Fructus of Chaenomeles sinensis, Cortex of Ulmus pumila, and Radix of Astragalus membranaceus. To determine the yields of the extracts, the Brix% of each extract solution was measured using a refractometer. When the relative DPPH radical scavenging ability values of the water extracts were compared with those of a positive control (ascorbic acid), the water extracts of P. lobata, C. sinensis, and U. pumila were 7.77%, 4.71%, and 4.19%, respectively. The results suggest that this method provides a useful assay for rapid measurement of DPPH radical scavenging abilities and conformation of antioxidative compounds in natural products. Moreover, it can reduce the time spent on the separation of active compounds from natural materials, such as medicinal plants, in addition to the use of reagents for separation.

A Study on the Torque Characteristics Depending on the Elastic Body Materials of a Hexadecagon Shaped Ultrasonic Motor (탄성체 재질 변화에 따른 16각형 초음파모터의 토크 특성 연구)

  • In this study, novel ultrasonic rotary motor of hexadecagon shape stator was proposed. Stator of the hexadecagon ultrasonic motor was composed of an elastic ring and ceramics. The elastic ring had sixteen sides and sixteen angular points. Eight ceramics were attached on the outer surface of the eight sides of the ring. When rotor of cylindrical shaft was inserted inside of the ring stator, central lines of the sixteen sides of the stator hold the shaft by the slight pressures(frictions). This slight pressure was a preload of the motor and it could be controlled by radius and thickness of the ring. When two sinusoidal voltages which have 90 degree phase difference were applied to each four ceramics, elliptical displacements of inner surface of the ring were obtained. These elliptical displacements of the inner surface rotated the shaft rotor through the frictions. The proposed hexadecagon ultrasonic motor was designed and analyzed by using the finite element method (FEM), depending on materials of the elastic ring. Based on the FEM results, one model of motor which showed maximum displacement at contact points was chosen and fabricated. And characteristics of the motor were compared with simulated results. When the motor was fabricated with these results, EL20ET0.5CT0.5CW2 model showed 115[rpm] speed about input voltage of 60[Vrms] at 65.6[kHz]. And the maximum torque of 6[gfcm] was obtained. From these results, the hexadecagon shaped ultrasonic motor can be used to actuator for optical device which needs detailed position control. Also it can be used to medical and portable device by reducing size and weight.

Bioequivalence Study of Toriem® Tablet to Motilium-M® Tablet (Domperidone Maleate 12.72 mg) Evaluated by Liquid Chromatography/Tandem Mass Spectrometry

  • The aim of the present study was to evaluate the bioequivalence of two domperidone maleate tablets, Motilium-$M^{(R)}$ Tablet (Janssen Korea Ltd., reference product) and $Toriem^{(R)}$ Tablet (Daewon Pharm. Co., Ltd., test product). Domperidone was extracted by liquid-liquid extraction using tert-butyl methyl ether and separated in less than 3 min on $C_{18}$ reverse-phase column using an isocratic elution. A tandem mass spectrometer, as detector, was used for quantitative analysis in positive mode by a multiple reaction monitoring mode to monitor the m/z $426.1{\rightarrow}119.1$ and the m/z $837.4{\rightarrow}158.2$ transitions for domperidone and the internal standard (roxithromycin), respectively. Calibration curves, from $0.05{\sim}50$ ng/mL of domperidone, showed correlation coefficients (r) higher than 0.9941. Intra day and inter day precision (C.V. %) for quality control were ranged from 10.04 to 16.09% and from 10.87 to 18.69%, respectively. The lower limit of quantification (LLOQ) of domperidone was 0.05 ng/mL. The method described is precise and sensitive and has been successfully applied to the study of bioequivalence of domperidone in 24 healthy Korean volunteers. Twenty-four healthy male Korean volunteers received a single dose of each medicine ($2{\times}12.72\;mg$ domperidone maleate) in a $2{\times}2$ crossover study. There was a one-week washout period between the doses. Plasma concentrations of domperidone were monitored for over a period of 24 hr after the administration. $AUC_{0-t}$ (the area under the plasma concentration-time curve) was calculated by the linear trapezoidal rule. $C_{max}$ (maximum plasma drug concentration) and $T_{max}$ (time to reach $C_{max}$) were compiled from the plasma concentration-time data. The 90% confidence intervals for the log transformed data were within acceptable range of log 0.8 to log 1.25 (e.g., $log\;0.92{\sim}log\;1.05$ for $AUC_{0-t}$, $log\;0.81{\sim}log\;1.05$ for $C_{max}$). The major parameters, $AUC_{0-t}$ and $C_{max}$ met the criteria of KFDA for bioequivalence indicating that $Toriem^{(R)}$ tablet is bioequivalent to Motilium-$M^{(R)}$ tablet.

Detection of Differentially Expressed Genes in Glioblastoma by Suppression Subtractive Hybridization

  • Objective: A variety of genetic alterations in human glioblastoma comprises signal transduction and cell cycle arrest control of cellular processes. Subtractive hybridization is potentially a faster method for identifying differentially expressed genes associated with a particular disease state. Using the technique of subtraction, we isolated novel genes that are overexpressed in glioblastoma tissue as compared to normal brain tissue. Methods: We evaluated the differential expression of genes in each of hybridizing tester and driver cDNAs to digested 130 clones. After sequencing of 130 clones and homology search, this study performed to determine mRNA expression of the unknown gene, "clone 47", in brain tissue, glioblasoma, and several cancer cell lines by reverse transcription-polymerase chain reaction (RT-PCR). To test the time course for Go-phase arrest, serum stimulation and expression at various times for RT-PCR performed. Results: We identified 23 novel genes by BLAST of the digested 130 clones. The expressions of "clone 47" mRNA of glioblastoma and several cancer lines were significantly higher than normal brain tissues and several normal cell lines. We confirmed the mRNA expression of "clone 47" was up-regulation for $0.5{\sim}1hr$ of WI-38 cell differentiation. Conclusion: The novel gene, "Clone 47" is upregulated in glioblastoma tissue and several cancer cell lines. This gene is time dependent activation during time course of serum stimulation. This result suggests that "clone 47" playa role in brain tumorigenesis and the activation of this "clone 47" may be necessary for the development of cancer.

  • The market volume of urban regeneration projects has steadily increased, thereby mixed-use development also shows significant market growth. However, previous researches on urban regeneration mainly deduced suggestions and plans for institution improvement based on comparison between projects implemented the inside and outside of the country. Whereas, researches on project management closely related to project success were limited, particularly in performance management. Accordingly, this research aims at developing a program performance management framework for public clients so as to succeed mixed-use development projects. Through extensive literature review and expert interview, this research developed performance indices for diverse facilities, risk management framework and integrating method of program performance score. The proposed framework is able to consider total life cycle from basic planning to maintenance phase and to check real-time performance level. Moreover, risk management framework can periodically assess and control the level of inherent risks within performance indices. The results can contribute to improve existing performance management practices and be basis for a web-based system developed in future.

Assessment Method of Step-by-Step Cyber Security in the Software Development Life Cycle (소프트웨어 생명주기 단계별 사이버보안 평가 방법론 제안)

  • Instrumentation and control(I&C) system has been mainly designed and operated based on analog technologies in existing Nuclear Power Plants(NPPs). However, As the development of Information Technology(IT), digital technologies are gradually being adopted in newly built NPPs. I&C System based on digital technologies has many advantages but it is vulnerable to cyber threat. For this reason, cyber threat adversely affects on safety and reliability of I&C system as well as the entire NPPs. Therefore, the software equipped to NPPs should be developed with cyber security attributes from the initiation phase of software development life cycle. Moreover through cyber security assessment, the degree of confidence concerning cyber security should be measured and if managerial, technical and operational work measures are implemented as intended should be reviewed in order to protect the I&C systems and information. Currently the overall cyber security program, including cyber security assessment, is not established on I&C systems. In this paper, we propose cyber security assessment methods in the Software Development Life Cycle by drawing cyber security activities and assessment items based on regulatory guides and standard technologies concerned with NPPs.

  • The AlGaN layer has direct wide bandgaps ranging from 3.4 to 6.2 eV. Nowadays, it is becoming more important to fabricate optical devices in an UV region for the many applications. The high quality AlGaN layer is necessary to establish the UV optical devices. However, the growth of AlGaN layer on GaN layer is difficult due to the lattice mismatch and difference thermal expansion coefficient between GaN layer and AlGaN layer. In this paper, we attempted to grow the LED structure on GaN template by mixed-source HVPE method with multi-sliding boat system. We tried to find the optical and lattice transition of active layer by control the Al content in mixed-source. For the growth of epi layer, the HCl and $NH_3$ gas were flowed over the mixed-source and the carrier gas was $N_2$. The temperature of source zone and growth zone was stabled at 900 and $1090^{\circ}C$, respectively. After the growth, we performed the x-ray diffraction (XRD) and electro luminescence (EL) measurement.

  • We propose an advanced transaction scheduling protocol for concurrency control of multilevel secure databases in wireless mobile network environment. Wireless communication is characterized by frequent spurious disconnections. So short-lived transaction must quickly access database without any delay by long-lived one. We adapted two-phase locking protocol, namely traditional syntax-oriented serializability notions, to multilevel secure databases in wireless mobile network environment. Altruistic locking, as an advanced protocol, has attempted to reduce delay effect associated with lock release moment by use of the idea of donation. An improved form of a1truism has also been deployed for extended a1truistic locking. This is in a way that scope of data to he early released is enlarged to include even data initially not intended to be donated. Our protocol is based on extended altruistic locking, but a new method, namely bi-directional donation locking for multilevel secure databases (MLBiDL), is additionally used in order to satisfy security requirements and concurrency. We showed the Simulation experiments that MLBiDL outperforms the other locking protocols in terms of the degree of throughput and average waiting time.

Use of Bacteriocin Produced by Lactococcus sp. CU216 with pH Sensitive Liposome Entrapment (Lactococcus sp. CU216이 생산하는 박테리오신을 함유한 pH Sensitive Liposome의 응용)

  • The objective of this study was to control Kimchi fermentation using pH sensitive bacteriocin entrapping liposome(bacteriocin-liposome). The liposomes were prepared by the reverse-phase evaporation method from a mixture of DPPC(dipalmitoyl phosphatidylcholine, DPPE(dipalmitoyl phosphatidylethanolamine), DOPC(dioleoyl phosphatidylcholine) and cholesterol in a molar ration of 4:2:1:4. The bacteriocin-liposome was disruptured at pH 4 of buffer and was stable at alkaline pHs(6 and 7). Irrespective of the addition of the bacteriocin-liposomes, the pH of every Kimchi sample decreased to 5 during 5 days storage at 5$^{\circ}C$. Kimchi samples treated with bacteriocin-liposomes maintained pH 4 or higher, while Kimchi samples not treated with bacteriocin-liposomes exhibited pH 3.58 or lower. In general, the pH of Kimchi samples stored at 20$^{\circ}C$ decreased faster, compared to that of Kimchi samples stored at 5$^{\circ}C$. The pH of Kimchi samples treated with the bacteriocin-liposomes was 3.9 during 90 days storage, while that of the samples not treated with the bacteriocin-liposomes was 3.68 and 3.32 during 30 days and 90 days storages, respectively. Lactic acid bacteria in Kimchi treated with the bacteriocin-liposome grew relatively slow at 5$^{\circ}C$. The viable cell number of lactic acid bacteria increased up to 4${\times}$10$\^$7/ cells/ml and then decreased to 8${\times}$10$\^$6/ cells/ml during 90 days storage at 5$^{\circ}C$.

  • An analytical diffusion model for flood routing with backwater effects and lateral flows is developed. The basic diffusion equation is linearized about an average depth of (H + h), and is solved using the boundary conditons which take into account the effects of backwater and lateral flows. Scouring phenomenon around pier which affects on the support function of pier and the stabilization if river bed is a complex problem depending on flow properties and river bed state as well as pier geometry. therefore, there is no uniting theory at present which would enable the designer to estimate, with confidence, the depth of scour at bridge piers. The various methods used in erosion control are collectively called upstream engineering, HEC-RAS Model, underwater blasting. They consist of reforestation, check-dam construction, planting of burned-over areas, contour plowing and regulation of crop and grazing practices. Also included are measures for proper treatment of high embankments and cuts and stabilization of streambanks by planting or by revetment construction. One phase of reforestation that may be applied near a reservoir is planting of vegetation screens. Such screens, planted on the flats adjacent to the normal stream channel at the head of a reservoir, reduce the velocity of silt-laden storm inflows that inundate these areas. This stilling action causes extensive deposition to occur before the silt reaches the main cavity of the reservoir.
