A Study on the Laws and Regulations in Respect of Preparation and Processing of Herbal Medicines at Hospitals of Korean Medicine (한방의료기관 조제·포제 관련 법규에 대한 고찰)

  • Objectives : The purpose of this study is to analyze and identify the problems of current laws and regulations regarding preparation and processing of herbal medicines at hospitals of Korean Medicine. Possible solutions are proposed in the end Methods : Based on the status of hospitals of Korean Medicine and characteristics of Korean Medicine and Korean Medicine industry, I analyze the laws and regulations in regards to preparation and processing of herbal medicines and propose possible solutions. Results : Regulations for the agents, places, and cautions in respect of preparation and processing of herbal medicines are inadequate. Meanwhile, the definition of drug preparation in the Pharmaceutical Affairs Act does not apply to processing and preparation of herbal Medicines at hospitals of Korean Medicine, since herbal medicines accompanies chemical and physical change. Discussions & Conclusions : New regulations for on-site preparation at hospitals of Korean Medicine are necessary. Also, the definition for herbal medicines preparation, which states possible chemical and physical changes of herbal medicines, should be specified in Pharmaceutical Affairs Act.

In vitro and In vivo Activity of DW286, an Experimental Fluoronaphthyridione, against Gram-Positive Bacteria (새로운 Fluoronaphthyridinone계 항균제 DW286의 그람 양성세균에 대한 항균 활성)

  • In vitro antibacterial activity of DW286 was tested against recently collected clinical isolates of Gram-positive strains by the two fold agar dilution method. In vivo activity of DW286 was also determined against systemic infections in mice caused by Staphylococcus aureus. DW286 showed 16~64-fold more potent in vitro activities than ciprofloxacin against Gram-positive bacteria. Against systemic infection model caused by two S. aureus strains, one being methicillin-susceptible and the other methicillin-resistant, DW286 ($ED_{50}s$, 0.16 mg/kg and 4.36 mg/kg, respectively) was more potent than gemifloxacin (1.37 mg/kg, 26.58 mg/kg, respectively).

  • Pharmaceutical companies over the world are exposed to risks of lessened sales and negative impact on corporate image due to circulation of counterfeit drugs, which eventually threatens safety of patients. thus, in order to eradicate distribution of counterfeit drugs, many countries are taking initiatives to legalize and set serialization through e-Pedigree, which is based on RFID technology or barcode system to track and trace distribution of drugs from point of manufacturing to usage. therefore, this study contemplates domestic and international management system of medical drugs and suggests application methods of e-Pedigree based on RFID technology that promotes safety of patients and distribution of drugs.

Development of New Dihydropyran Linker for Solid-Phase Reaction

  • The linker which plays a role in connecting a polymer with a scaffold has become an important part n solid-phase reaction. To develop a new linker for alcohols and carbohydrates, dihydropyran moiety was selected in this study. New linker, 1-($4^{l},5^{l}$-dihydro-5H-pyranyl)-7-hydroxyheptan-3-one (5) was synthesized via four steps from $\delta$-valerolactone. This can be called as DDHP-linked Wang resin due to double dihy-dropyran rings. To the one pyran ring of new linker 5 was added Wang resin and other alcohols and carbohydrates as scaffolds were then added successfully to the another pyran ring. Carbohydrate and hydroxyl resins were connected via new linker in a 70% loading yield. The detachment of glucose moiety in the presence of PPTS (2 equiv.) in 1:1 n-buteanol/1,2-dichloroethane at $60^{\circ}C$ for 12 h was carried out quantitatively. When certain combinatorial chemical works are carried out using this dihydropyran linker, Wang resin itself can be recovered. Its fact is particularly very important in industry, because recovered resins can be recycled.

Assessment of Fertilizer Efficiency of Pharmaceutical Byproduct and Cosmetic Industry Wastewater Sludge as Raw Materials of Compost (제약업종 부산물 및 화장품 제조업 폐수처리오니의 비효검정)

  • Pharmaceutical byproduct sludge and cosmetic industry wastewater sludge can be used as a raw material of compost. Effects of three types of pharmaceutical byproduct sludge and one type of cosmetic industry wastewater sludge on soil properties and red pepper growth were investigated in a field based concrete pot ($2{\times}2m$). These sludges and pig manure ($5Mg\;ha^{-1}$, dry basis) were incorporated into the upper of loam soil 30 days prior to transplanting red pepper. Changes in soil properties and contents of heavy metals and toxic organic compounds in soil and plant were measured. And also plant growth measurement and bioassay of soil phytotoxicity were included. Contents of heavy metals were increased in the soils treated with the sludges. Plant growth in the sludge treatments were mostly inferior to that of NPK treatment, especially in early stage. Content of N in plant was lower in all sludge treatments at early and middle growth stages, and it was especially caused by characteristics and concentration of nitrogen and organic matter of sludges. Total yield of red pepper was highest in the NPK treatment and followed by pharmaceutical sludge 3, pig manure, pharmaceutical sludge 1, and pharmaceutical sludge 2, and the yield of cosmetic sludge treatment was considerably lower than others. HEM and PAHs contents in soil of cosmetic sludge treatment were $4.80mg\;kg^{-1}$ and $2,263.2{\mu}g\;kg^{-1}$, respectively. Root elongation of lettuce exposed to the water extract of soil treated with cosmetic sludge was about 20% of that found in the test with soil extract of non fertilization treatment. At present, raw materials of compost were authorized according to the contents of organic matter, heavy metals and product processing. Toxic organic compounds analysis and bioassay would be helpful for authorization and assessment of suitability of raw materials of compost.

APPLICATION STUDY OF CHEMOINFOMETRICAL NEAR-INFRARED SPECTROSCOPY IN PHARMACEUTICAL INDUSTRY

  • A chemoinfometrical method for evaluating the quantitative determination of crystallinity one polymorphs based on fourie-transformed near-infrared (FT-NIR) spectroscopy was established. A direct comparison of the data with the ones collected from using the and compared with the conventional powder X-ray diffraction method was performed. [Method] The pPure a and g forms of indomethacin (IMC) were prepared by reportedusing published methods. Six kinds of standard samples obtained by physically mixing of a and g forms. After the powder X-ray diffraction profiles of samples have been measured, the intensity values were normalized to against the intensity of silicon powder as the as an external standard. The calibration curves for quantification of crystal content were based upon the total relative intensity of four diffraction peaks from of the form g crystal. FT-NIR spectra of six calibration sample sets were recorded 5 times with the NIR spectrometer (BRAN+LUEBBE). Chemoinfometric analysis was performed on the NIR spectral data sets by applying the principal component regression (PCR). [Results] The relation between the actual and predicted polymorphic contents of form g IMC measured using by the X-ray diffraction method shows a good straight linen linear relation., and it has slope of 0.023, an intercept of 0.131 and a correlation coefficient of 0.986. PCR analyses wereis was performed based on normalized NIR spectra sets offer standard samples of known content of IMC g form. IMC. A calibration equation was determined to minimize the root mean square error of the predictionthe prediction. Figure 1 shows a plot of the calibration data obtained by NIR method between the actual and predicted contents of form g IMC. The predicted values were reproducible and had a smaller standard deviation. Figure 2 shows that the plot for the predicted transformation rate (%) of form a IMC to form g as measured by X-ray diffractomeoy against to those as measured by NIR method. The plot has a slope of 1.296, an intercept of 1,109, and a correlation coefficient of 0.992. The line represents a satisfactory correlation between the two predicted values of form g IMC content. Thus NIR spectroscopy is an effective method for the evaluation to the pharmaceutical products of quantitative of polymorph.

Globalization and Independency of Populist Nations' Welfare Policies: Focusing on the Influences of Multinational Pharmaceutical Companies on the Korean Government's Policy on the Pharmaceutical Industry (세계화와 국민국가의 복지정책 자율성: 다국적 제약자본이 우리나라 제약정책 결정에 미친 영향을 중심으로)

  • Globalization has conflicting effects on pharmaceutical policies. A change into a 'populist competitive nation' due to globalization strengthens policies to reduce drug manufacturing costs while the WTO's TRIPS Agreement that is affected by multinational pharmaceutical companies increases drug manufacturing costs by bolstering the patent rights on new drugs. Currently, the independency of populist nations' policies to reduce drug manufacturing cost is being compromised because multinational organizations(such as the European Union) which represents the interests of the multinational pharmaceutical companies put restrictions on the pharmaceutical policies of populist nations for purposes of promoting the industrial goals of the multinational companies. Korea is no exception. Up until the late 1990s, the main feature of the pharmaceutical policies in Korea was keeping the drug price at the cost level based on a growth-driven ideology, and this was Korea's unique policy tools as a developing nation. However, the increase in the power of multinational pharmaceutical companies currently infringes on the independency of Korea's pharmaceutical policies. Expensive imported drugs were originally covered by the national health insurance plan, but starting from 1999 such drugs began to be covered by the plan. After separation of medical and pharmaceutical services, the use of expensive drugs was increased, and the Korean government planned to introduce the reference price policy in order to contain the cost of the national health insurance plan. However, due to pressures from the U.S. government as well as multinational pharmaceutical companies, implementation of the policy has been postponed. In addition, due to a pressure from the U.S. government, a working group was created which would affect the health care policy of the Korean government. Discussions so far on globalization was about whether the change into populist competitive nations due to globalization resulted in the reduction of welfare spending. However, this study shows not only the reduction of health care cost through policies to reduce drug manufacturing costs but increase in welfare spending by raising the strengths of multinational pharmaceutical companies that are for-profit providers of welfare service. While focusing on the contradictory effects of globalization on pharmaceutical policies of a nation, this study looked at how these conflicting effects end up promoting the interests of multinational pharmaceutical companies by examining the Korean case.

e-Business Strategy of Healthcare Industry (보건의료산업에서의 전자상거래 가치 및 활성화 전략)

  • This paper examines the current situation and major impacts of e-business on healthcare industry: hospital, pharmaceutical, medical device and health-related internet sectors. Of the 137 samples collected with mail survey, the utilization rate of B2C e-commerce is 31.4% and B2B is 13.1%. And 74.5% of respondents remains the first development stage which represents the simple advertisement such as the one-way information offering. The key obstacle of expansion of health care e-commerce turns out to be the illogical and outdated medical-related law and institution. Finally, policy recommendations are proposed based on the evaluation of the current policy implemented by government.

  • A new ceramide alomacrorrhiza A was isolated from the ethanolic extract of the plant Alocasia macrorrhiza (L.) Schott. Its chemical structure was elucidated as (2S,3S,4R)-2N-[(2'R)-2'- hydroxy-hexacosanoyl]-tetradecane-1,3,4-triol based on extensive 1D, 2D NMR, EI-MS, FABMS, HR-FAB-MS spectroscopic data and chemical degradation studies.

Isocitrate dehydrogenase mutations: new opportunities for translational research

  • Over the last decade, comprehensive genome-wide sequencing studies have enabled us to find out unexpected genetic alterations of metabolism in cancer. An example is the identification of arginine missense mutations of isocitrate dehydrogenases-1 and -2 (IDH1/2) in glioma, acute myeloid leukemia (AML), chondrosarcomas, and cholangiocarcinoma. These alterations are closely associated with the production of a new stereospecific metabolite, (R)-2-hydroxyglutarate (R-2HG). A large number of follow-up studies have been performed to address the molecular mechanisms of IDH1/2 mutations underlying how these events contribute to malignant transformation. In the meanwhile, the development of selective mutant IDH1/2 chemical inhibitors is being actively pursued in the scientific community and pharmaceutical industry. The present review article briefly discusses the important findings that highlight the molecular mechanisms of IDH1/2 mutations in cancer and provides a current status for development of selective mutant IDH1/2 chemical inhibitors.
