Effects of Jiyutang with Fructus Immaturus Ponciri on the immune response in the Mouse (지유탕가(地楡湯加) 지실(枳實)이 생쥐의 면역반응(免疫反應)에 미치는 영향(影響))

  • This study was done to know the effect of Giyuetang on the inflammatory response, contact hypersensitivity (CH), and rosette forming capacity of spleen cells. The effects of Jiyutang with Fructus Immaturus Ponciri on the inflammatory response were evaluated by measuring the production of such reactive oxygen intermediate(ROI) as $O_2^-$ and $H_{2}O_2$ in the peritoneal neutrophils and macrophages. The effects of Jiyutang with Fructus Immaturus Ponciri on the CH were evaluated by checking the ear swelling response against dinitrofluorobenzene(DNFB). The administration of Jiyutang with Fructus Immaturus Ponciri on the mouse decreased the amount of ROI in both neutrophils and macrophages. Jiyutang with Fructus Immaturus Ponciri depressed CH without affecting the rosette forming capacity of spleen cells. The results of this study showed that Jiyutang with Fructus Immaturus Ponciri might have anti-inflammatory effects by decreasing the amounts of ROI in the phagocytes and suppressing the CH, without affecting the rosette-forming capacity of spleen cells.

  • Fever has been recognized as a cardinal feature of disease since antiquity, but only recently has the pathophysiology of fever come to be understood. It became clear that the ultimate cause of fever is not a bacterial product (a so-called exogenous pyrogen) but a product of host inflammatory cells (i.e., an endogenous pyrogen). Many studies have demonstrated that mononuclear phagocytes are the principal source of endogenous pyrogen and that a variety of mononuclear cell products can mediate the febrile response. Cytokines are also important as mediators of the acute-phase response to infection and inflammation.

  • Ketoconazole is an imidazole antifungal agent which inhibits the biosynthesis of fungal cellmembrane ergosterol and has immunosuppressive properties in vitro. Biphenyl dimethyl dicarboxylate (PMC) has been utilized for antioxidative action and for liver-protective purposes. Studies were undertaken to investigate effects of biphenyl dimethyl dicarboxylate (PMC) on the immunosuppression of ketoconazole in ICR mice. In the combination of PMC and ketoconazole, as compared with the treatment of ketoconazole alone, there were significant increases in activities of natural killer (NK) cells and phagocytes along with circulation leukocytes. The elevation of serum glutamic-pyruvic transaminase (S-GPT) and total protein levels caused by ketoconazole were reduced by the combination of PMC and ketoconazole. In addition, lower serum albumin and albumin/globulin (A/G) ratio were also increased to normal level.

  • In present work an attempt was made to synthesize various analogues of levofloxacin by introducing new functionality at carboxylic group position via nucleophilic substitution reaction. For this purpose the carboxylic group at C-6 was esterified and later subjected to nucleophilic attack at the carbonyl carbon by various aromatic amines. Structure of the analogues was confirmed by different techniques i.e. IR, $^1H$ NMR and mass spectrometry. The antibacterial activity of the derivatives was also assessed and compared with the parent against a series of Gram-positive and Gramnegative bacteria. A synergistic as well as antagonistic behavior was observed in these derivatives. Additionally unlike levofloxacin, the derivatives were also found to modulate oxidative burst response of phagocytes exhibiting moderate to significant inhibitory activity.

  • Dose-dependent responses indicated by the increase of leukocyte, peritoneal exudate cell and weights of immunorgans revealed the improvement of immunity. In the effect of macrophage on phagocytes, there were not substantial differences in the phagocytic and corrected phagocytic index. The administered group being compared with the controlled group, there were no significant changes in SGOT, S-GPT, alaline phosphatase, total protein, albumin, globulin, cholesterol, triglyceride, blood urea nitrogen and glucose. Effect of Ginseng Saponins on Phagocytosis of Feline Peripheral Blood Phagocytes (고양이 말초혈액 탐식세포의 탐식능에 있어서 인삼 사포닌의 효과)

  • 고양이 말초혈액 탐식세포(단핵구세포(MNC) 및 다형핵백혈구(PMNC))의 탐식능 에 있어서 인삼 saponin(ginseng total saponin(G75), ginseng PT saponin(GPT) 및 ginseng PD saponin(GPD))의 면역증강 효과를 flow cytometry를 이용하여 분석하였다. 인삼 ssponins을 직접 첨가하여 배양한 MNC 및 PMNC에서는 탐식중강 효과가 나타나지 않았다. 각각의 인삼 saponin을 첨가하여 배양한 PMNC 및 MNC 배양상충액의 존재하에 PMNC 및 MNC의 탐식능을 겅토한 결과, MNC의 탐식능은 Gff 첨가 PMNC 배양상충액과 GTS 및 GPT 첨가 MNC 배양상충액의 존재하에서 약간의 탐식증강 효과를 보였다. PMNC 탐식능의 경우에는 GPD 첨가 PMNC 배양상충액에서 미약한 탐식증강 효과가 나타났으나, 각각의 인 삼 saponin 첨가 MNC 배양상충액 존재하에서는 모두 현저한 탐식중강 효과를 나타내었다. 이상의 결과로부터 고양이 말초혈액 탐식세포의 탐식증강 효과는 인삼 saponin의 직접적인 작용보다는 인삼 saponin에 의해 활성화된 단핵구세포에서 분비되는 가용성물질에 의해 단 핵구세포보다는 다형핵백혈구에서 현저하게 탐식효과가 증강되는 것으로 판단되었다.

Effect of Biphenyl dimethyl Dicarboxylate on the Cellular and Nospecific Immunosuppressions by Ketoconazole in Mice

  • The effect of biphenyl dimethyl dicarboxylate (PMC) on the cellular and nonspecific immunosuppressions by ketoconazole (KCZ) was investigated in ICR mice. PMC at a dose of 6 mg/kg was administered orally to mice daily for 14 consecutive days. KCZ was suspended in RPMI 1640 medium and orally administered at 160 mg/kg/day 2 hrs after the administration of PMC. Immune responses of the delayed-type hypersensitively (DTH) reaction to sheep red blood cells (SRBC), phagocytic activity and natural killer (NK) cell activity were evaluated. DTH reaction to SRBC was enhanced to normal level by the combination of PMC and KCZ, as compared with treatment of KCZ alone. In the combination of PMC and KCZ, as compared with treatment of KCZ alone, there were also significant increases in activities of natural killer (NK) cells and phagocytes along with circulating leukocytes. These findings indicate that PMC shows a significant restoration from the immunotoixc status induced by KCZ.

Extracellular Mechanisms of Neutrophils in Immune Cell Crosstalk

  • Neutrophils are professional phagocytes that provide defense against invading pathogens through phagocytosis, degranulation, generation of ROS, and the formation of neutrophil extracellular traps (NETs). Although long been considered as short-lived effector cells with limited biosynthetic activity, recent studies have revealed that neutrophils actively communicate with other immune cells. Neutrophils employ various types of soluble mediators, including granules, cytokines, and chemokines, for crosstalk with immune cells. Additionally, ROS and NETs, major arsenals of neutrophils, are utilized for intercellular communication. Furthermore, extracellular vesicles play a crucial role as mediators of neutrophil crosstalk. In this review, we highlight the extracellular mechanisms of neutrophils and their roles in crosstalk with other cells.
