The identification of optimum condition for direct regeneration in black raspberry

  • Adventitious buds appeared within 2 weeks on the base of the petiole explants and increased for two months. A maximum of regeneration (15.6%) was obtained on the medium containing $1.5\;{\mu}M$ TDZ in combination with $1\;{\mu}M$ IBA. To know which explants are the best for the induction of regeneration, three explants such as leaf, petiole and leaf-petiole were used. Among the explant types, the leaf-petiole explant was significantly more effective than leaf and petiole for promoting adventitious shoots, with leaf-petiole inducing at the highest regeneration frequency (33.7%). The regeneration frequency of adventitious shoots in the leaf-petiole explants was significantly affected by leaf size and the position of explants. The leaf-petiole smaller than 5 mm leaf in width was induced at the highest regeneration frequency (68.9%). The smaller leaf sizes, the greater regeneration frequency. Also when the leaves are nearer to the shoot tip, the regeneration frequency is higher. When the rooted micro-shoots were transferred to the soil after growing for 6 weeks in the media, the survival rate was 90%.

Determination of Daily Amount of N and K Required in Various Growth Stages and Establishment of Diagnostic Criteria Using Petiole Sap Analysis in the Semi-Forcing Culture of Cucumber (반촉성 관비재배 오이의 생육단계별 시비관리를 위한 일일시비량 및 엽병즙액의 농도 기준 설정)

  • This study was conducted to determine the daily application rate and amount of N and K with fertigation during different growth stages in semi-forcing culture of cucumber plants (Cucumis sativus L. cv. Eunseongbaekdadagi). The diagnostic criteria for N and K also investigated based on petiole sap analysis. The dry weight increased slowly until 30 days after transplanting. The highest dry weights were observed at 60 days after planting, then it decreased. As the plant grew, the contents of N and K in the petiole sap and fruit of cucumber decreased. The daily uptake of N and K were highly correlated with the growing days. The $NO_3$ concentrations in petiole sap were in the range from 3,500 to 4,500 mgㆍ$L^{-1}$ in the early growth stage, but those were in the range from 2,000 to 3,000 mgㆍ$L^{-1}$ after then. However, K concentration in petiole sap were in the rang from 5,000 to 7,000 mgㆍ$L^{-1}$ The fluctuation in petiole sap concentration of K was severe in the monthly fertigation and moderate in the daily fertigation. The fertigation by petiole sap diagnosis forced EC of soil to be low and yield to increase compared to the control.

Effects of root nodules on the plant type in soybean-Especially internode length and petiole length on the main stem

  • The plant type is generally one of the most important factor for crop production and be influenced by nitrogen absorption. Soybean plants have nodules in their roots, supplying nitrogen at the vegetative and reproductive stages. Root nodules seem to effect plant type of soybean plants, but there are few reports on the relation nodules and plant type. We tried to clarify the effects of root nodules on the plant type, especially internode length and petiole length, comparing non-nodule soybean with normal soybean. The pot experiment and field experiment were carried out at Mie University and Utsunomiya University in 2015 and 2016. Enrei, a popular cultivar in central Japan, and En1282, non-nodulating isogenic line of Enrei, were used. The petiole length on main stem was measured after defoliation and internode length and yield components were measured after harvest. In the field experiment, the patterns of the final length in internode and petiole on main stem were consistent in both cultivars, and a positive correlation was found between the Nth petiole length and the N-1th internode length, belong to the same phytomere. Therefore, the petiole and internode on the main stem make similar response for environmental factors. In pot experiment, Enrei grew with the same pattern as field experiments, but in En1282, the elongation of petiole and internode in the upper part was suppressed, especially the petiole was suppressed greatly. The main stem becomes the basis of the plant type. These results were considered that the nitrogen is distributed preferentially to the internode than the petiole. It seems that the pot cultivation restricted the rhizosphere and caused nitrogen deficiency in En1282. These results suggested that the slight nitrogen deficiency provided from the root nodules was compensated by the increase of the amount of inorganic nitrogen absorption due to the expansion of the rhizosphere, and the severe nitrogen deficiency suppressed elongation of petiole and internode. It is clear that root nodules effect the plant type by supplying nitrogen to internodes and petioles.

  • 1. Comparative studies, the number, form and pattern of ramification, on the petiole stele types of 3 orders, 11 families, 41 genera and 104 species of ferns found in Korea were carried out. 2. The number, form and pattern of the ramified steles were found to be different according to the taxa studied. 3. The stele types of petiole may be classified as unibranch, bibranch, tribranch, and polybranch. The species belonging to each stele type were found to have similar embyrological characteristics among them. Therefore, it might be reasonable to assume that the stele type can be used as a basis for classifing family lines. 4. The number of ramified steles in the petiole were found to be in general agreement with that of the leaf traces, though a few exceptional cases were found. 5. It is well known that there is a large degree of disgreement among the taxanomists on the classification of ferns. The classification of ferns by means of petiole stele types may ease this difficulty in certain extent.

The petiole anatomy of the genus Spiraea L. (Rosaceae) in Korea (한국산 조팝나무속(Spiraea L. 장미과)의 엽병 해부)

  • The petiole anatomy of three sections (sects. Calospira, Chamaedryon, and Spiraria) in the genus Spiraea (Rosaceae), which are distributed/cultivated in Korea, was studied and described in details. The vascular bundle system of the distal, medial, and proximal parts of the petiole were studied using LM with the aim of documenting any differences in vascular anatomy. Significant variation of anatomical characters was found in petiole shape (dichotomy, lens or subcircular in outline), presence of two distinct lateral wings, the thickness of epidermis, number of vascular bundles, length of ventral axis, length of dorsiventral axis, width of median vascular bundle, and length of median vascular bundle. All features were compared and we concluded that petiole anatomical characters can be useful for providing diagnostic features to distinguish the studied taxa. A key for identifying the Spiraea taxa in Korea based on their petiole anatomical characters is also provided.

Canopy-Related Characteristics of Korean Soybean Cultivars (한국 콩 품종의 초형관련 형질의 특성)

  • This study was conducted to obtain basic information on mainstem, branch and leaf characteristics related to canopy for development of high yielding cultivar using 70 Korean soybean cultivars developed from 1913 to 2000. Variations of canopy width, branch length, and canopy width/length ratio were higher compared to other characteristics among 12 mainstem and branch characters. Variations of petiole angle, leaflet width/length ratio and compound leaf dry weight were higher than other characteristics among eight leaf characters related to plant canopy. Three classifications of soybean cultivars were used based on usage: I)soy sauce and tofu, II)bean sprout, and III)cooking with rice. Canopy width/length ratio was higher in group III, cooking with rice than group I, soy sauce and tofu, and group II, bean sprout, and there was no difference between the two, group I and group II. The total branch length/main stem height ratio was higher in group II, bean sprout and group III, cooking with rice than group I, soy sauce and tofu. Mainstem and branch characteristics related to plant canopy were classified into four groups by ratio of canopy width/length and total branch length/main stem length, respectively. Soybean cultivars with narrow canopy and high dependence of mainstem were Danweonkong, Keumkangkong, Shelby, and Shinpaldalkong. Soybean cultivars with broad canopy and high dependence of mainstem were Kanglim, Keumkangdaelip, and Jinyulkong, and a cultivar with broad canopy and high dependence of branch were Geomjeongkong 2. Leaflet length/width ratio was lowest in cooking with rice and there was no difference between soy sauce and tofu and bean sprout. Compound leaf area was largest in cooking with rice and smallest in bean sprout. Leaf petiole length was short in bean sprout and there was no difference between soy sauce and tofu and bean sprout. Leaf petiole angle was highest in cooking with rice and lowest in bean sprout. Leaf type was classified into four groups based on leaflet width/length ratio and compound leaf area, respectivly. Buseok and Taekwangkong had an oval leaflet and largest area of compound leaf. Eunhakong and Sohokong had extreme narrow leaflet and smallest area of compound leaf. Leaf petiole type was classified into three and four groups based on leaf petiole length and angle, respectively. A soybean cultivar with the shortest petiole length and smallest petiole angle was Eunhakong and cultivars with short petiole length and large petiole angle were Alchankong, Muhankong, and Pureunkong. A soybean cultivar with long petiole length and small petiole angle was Sinpaldalkong 2. Among a total of 70 Korean soybean cultivars, Eunhakong had an extreme narrow type in leaf, smallest compound leaf area, shortest petiole length, and smallest petiole angle of compound leaf.

Leaf Anatomy of Parkia clappertoniana Keay(Mimosaceae)

  • The bipinnate-compound leaf of Parkia clappertoniana has prominent, long petiole with a pulvinal base. The pulvinus has wood, periderm, lenticels and extensive cortical layer. Its vascular bundles are elongated with alternating rays. The vascular bundles of the petiole, rachis and veins are oval, with conspicuous bundle caps. Stomatal complex is predominantly paracytic with occasional occurrence of anomocytic stomata near the midrib. The oval stomata have conspicuous stomatal ledges. Trichomes are unicellular, acicular and restricted to the rachis, petiole and pinnule mid-ribs and margin. Ecological and taxonomic significance of features is discussed.

  • This study was investigated to find out the optimum planting densities of Aptum graveolns L. about growth characters, yield components and yi-eld at 9 planting densities by transplanting cultureThe results obtained are summarized as follows ;1 . Effects of Auxins end Cytokinins on Callus Induction from Leaf Blade, Petiole, and Stem Segments of in Vitro-grown 'Sheridan' Grape Shoots

  • To establish an the mass production system of grape anthocyanin pigments through callus and cell suspension culture, the effects of various combinations of auxins and cytokinins on friable callus production were studied. for friable callus production, 2,4-D was superior to other regulators. IAA at 2 mg/L induced callus from stem and petiole while NAA resulted in rooting. Callus induction rate increased with the 2,4-D level, and stem segments were superior to leaf blade or petiole, showing nearly 100% with 1 and 2 mg/L 2,4-D from petiole and stem. Combined treatments of 2,4-D + kinetin and NAA + BA also yielded friable callus from stem segments. In treatments with 1 mg/L 2,4-D + 1 mg/L kinetin and 1 mg/L NAA + 1 mg/L BA, callus induction rate was nearly 100%. The combination effect of 2,4-D and BA on anthocyanin production was not significant.

Plant Regeneration from Turnip (Brassica rapa ssp. rapifera) Organs

  • Shoot induction system was developed in the recalcitrant plant species, Brassica rapa ssp. rapifera by using optimum selection of profit organ, phytohormone combination, seedling age and kind of culture container. Out of in vitro cultured leaf segment, petiole, hypocotyl, and cotyledon with petiole, only cotyledon with petiole derived from 4 day-old seedlings induced multiple shoot. The optimum combination of auxin and cytokinin for the multiple shoot induction was MS medium containing 5mg/L BA and 0.5mg/L NAA. The major factors for multiple shoot propagation were part of plant organ, age of seedling, and ratio of auxin and cytokinin. In addition, shoot regeneration was promoted in the 100ml Erlenmeyer flask compared with the $90mm{\times}20mm$ Petri-dish. The induced shoots formed roots easy on MS medium containing 0.1mg/L IBA and the whole plants were successfully cultivated in soil.
