The Influence of the Substituents for the Insecticidal Activity of N' -phenyl-N-methylformamidine Analogues against Two Spotted Spider Mite (Tetranychus urticae) (두 점박이 응애(Tetranychus urticae) 에 대한 N'-phenyl-N-methylformamidine 유도체의 살충활성에 미치는 치환기들의 영향)

  • To understand the influences of the substituents ($R_1{\sim}R_4$) on insecticidal activity of N'-phenyl-N-methylformamidine analogues (1~22) against two spotted spider mite (Tetranychus urticae), comparative molecular field analysis (CoMFA) model and comparative molecular similarity indices analysis (CoMSIA) model as three dimensional quantitative structure-activity relationships (3D-QSARs) model were derived and discussed quantitatively. From the results, the correlativity and predictability ($r^2{_{cv.}}=0.575$ and $r^2{_{ncv.}}=0.945$) of the CoMFA 1 model were higher than those of the rest models. The the CoMFA 1 and CoMSIA 1 model with the sensitivity of the perturbation and the prediction produced ($d_q{^{2'}}/dr^2{_{yy}}=1.071{\sim}1.146$ & $q^2=0.545{\sim}0.626$) by a progressive scrambling analysis were not dependent on chance correlation. The insecticidal activities from the optimized CoMFA 1 model were depend upon the steric field (62.5%), electrostatic field (28.9%), and hydrophobic field (8.6%) of N'-phenyl-N-methylformamidine analogues. Therefore, the inhibitory activities with optimized CoMFA 1 model were dependent upon steric factor. From the contour maps of the optimized models, it is predicted that the structural distinctions that contribute to the insecticidal activity will be able to applied new potent insecticides design.

Development of a Residue Analysis Method for Metamifop in Paddy Water, Soil, and Rice with HPLC (HPLC를 이용한 농업용수, 논토양, 및 현미 중 metamifop의 잔류분석법 개발)

  • An analytical method for detecting metamifop residue in paddy water, soil, and rice with high performance liquid chromatography (HPLC) was developed. Water was extracted with ethyl acetate before analyzing by HPLC. Soil residues were extracted with acetone under acidic condition and after purifying with $Extrelut^{(R)}$ NT, and silica SPE, the residue was analyzed by HPLC. For residue analysis in rice, the procedure involved extraction with acetone, purification with $Extrelut^{(R)}$ NT, partitioning between acetonitrile/hexane, purification with silica SPE cartridge, and analysis by HPLC. The limit of detection (LOD) was 1.0 ng, limit of quantitation (LOQ) was 3.0 ng, and method limit of quantitation (MLOQ) were 0.001 mg/L for paddy water, 0.01 mg/kg for rice and soil, respectively. Standard calibration curve shows linearity from 0.05 mg/kg to 5.0 mg/kg ($R^2=0.9999$). The recoveries in fortified paddy water were $91.3{\pm}3.5%$ (0.01 mg/L level) and $93.2{\pm}6.3%$ (0.05 mg/L level). The recoveries in fortified paddy soils were $92.5{\pm}4.0%$ (0.1 mg/kg level) and $92.7{\pm}4.0%$ (0.5 mg/kg level) in soil A, while, $102.3{\pm}4.4%$ (0.1 mg/kg level) and $98.9{\pm}7.9%$ (0.5 mg/kg level) in soil B, respectively. The recoveries in fortified rice were $93.0{\pm}6.9%$ (0.1 mg/kg level) and $85.0{\pm}3.5%$ (0.5 mg/kg level). This method was proved to be effective and can be used to determine the metamifop residue in paddy water, paddy soil, and rice.

The Residue Property of Fungicide Dimethomorph and Pyraclostrobin in Green Onion under Greenhouse Condition (시설재배 쪽파에서 살균제 Dimethomorph와 Pyraclostrobin의 잔류특성)

  • In order to use in the classification of minor crop for the mutual application of safe use guideline, it was investigated the residue property of fungicide dimethomorph and pyraclostrobin in green onion, a stem-crop. After pesticides were applied 2 times with 1 week interval in that day of harvest, 3 days, 7 days, 10 days and 14 days before harvest, a green onion was harvested. The residue of dimethomorph in a green onion was 26.31 and 39.08 mg/kg in that day of harvest, however, in according to elapse time, it was reduced to 6.86 and 9.34 mg/kg in 14 days before harvest. In case of pyraclostrobin, it was also reduced from 13.46 and 39.08 mg/kg to 3.57 and 5.21 mg/kg. Based on the residue in that day of harvest, the deposit of spray solution in a green onion was calculated. The deposit of spray solution of dimethomorph was 274.35~345.84 mL/kg, in case of pyraclostrobin, it was calculated 213.65~343.33 mL/kg. When the amount of the deposit of both pesticides was compared in a green onion, it was so similar. On the other hand, it was estimated the predicted dissipation curve of pesticides in the green onion during cultivation. The half-life of dimethomorph was 6.95~7.45 days, in case of pyraclostrobin, 7.15~7.45 days. When both pesticides were compared with the residue property, the deposit of spray solution and half-life of dissipation were so similar.

Comparision of the Residue Property of Insecticide Bifenthrin and Chlorfenapyr in Green Onion and Scallion under Greenhouse Condition (시설재배 쪽파와 부추에서 살충제 Bifenthrin과 Chlorfenapyr의 잔류특성 비교)

  • In order to use in the classification of minor crop for the mutual application of safe use guideline, it was compared a green onion with a scallion on the residue property of insecticide bifenthrin and chlorfenapyr. After pesticides were applied 2 times with 1 week interval in that day of harvest, 3 days, 7 days, 10 days and 14 days before harvest, vegetables were harvested, and the residue of pesticides was investigated. Base on the residue in that day of harvest, the deposit of spray solution in vegetables was calculated. The deposit of spray solution of bifenthrin was 123.0 mL/kg in a green onion, and 74 mL/kg in a scallion. In case of chlorfenapyr, it was calculated 126.5 mL/kg in a green onion, and 70.0 mL/kg in a scallion. When the amount of the deposit of both pesticides was compared a green onion with a scallion, it was higher in a green onion. On the other hand, it was estimated the predicted dissipation curve of pesticides in a green onion and a scallion during cultivation. The dissipation curve of bifenthrin was y = 1.0334 $e^{-0.0602x}$ ($R^2$= 0.8606) in a green onion, and y = 0.7693 $e^{-0.1823x}$ ($R^2$= 0.9756) in a scallion. In case of chlorfenapyr, it was y = 2.2603 $e^{-0.0519x}$ ($R^2$= 0.9043) in a green onion, and y = 1.2940 $e^{-0.1051x}$ ($R^2$ = 0.9782) in a scallion. The half-life of bifenthrin was 11.51 days in a green onion, and 3.80 days in a scallion, respectively. Also, it was estimated half-life in chlorfenapyr, it was 13.35 days in a green onion, and 6.59 days in a scallion, respectively. The half-life of both pesticides in a green onion was longer than in a scallion. When both vegetables were compared with the residue property, the deposit of spray solution and half-life of dissipation in a green onion were more than those in a scallion.

Study on the Residue Analysis of Organophosphorus and Organochlorine Pesticides with Ion Trap and Quadrupole Mass Spectrometer (Ion Trap Mass Spectrometer와 Quadrupole Mass Spectrometer를 이용한 유기인제 및 유기염소제 농약 분석에 관한 연구)

  • Detection limit as well as calibration curves on organophosphorus pesticide(dimethoate, diazinon, parathion-methyl, fenitrothion, malathion) and BHCs were measured for evaluation of utility on qualitative or quantitative analysis of pesticides with ion Trap mass spectrometer and quadrupole mass spectrometer. As ionization source, EI and CI were adopted for qualitative analysis of pesticides by comparison of each fragmentation pattern. At the same time, the utility as trace analysis techniques through scan or selected ion monitoring(SIM) mode was evaluated. With ion trap for all pesticides, detection limit(DL, 1 ${\mu}L$ injection) on scan mode was ranged 0.008∼0.225 ng at signal to noise ratio 3. With quadrupole DL on scan mode was ranged 0.23∼3.1 ng over 0.032∼0.68 ng on SIM mode. The calibration curve with ion trap generated good linearity over 0.99 as correlation coefficient. As clean up procedure, Bio Beads S-X3 was used for the separation of oils from five organophosphorous pesticides in flour extractant showing more than 80% as recovery at most cases. In case of BHCs in jinseng with Florisil column, the recovery of pesticides has been 60% to 90%.

Occurrence of Papaya ringspot virus Infecting Cucurbit Crops in Korea (박과작물에 발생하는 파파야원형반점바이러스의 발생 보고)

  • A flexuous rod-shaped virus was isolated from Cucurbita pepo leaves showing as green mosaic and puckering symptoms at Anseong, Korea. Based on the biological analysis, electron microscopy, and reverse transcription-polymerase chain reaction (RT-PCR), the virus isolate was identified as Papaya ringspot virus type watermelon (PRSV-W). From biological analysis, the host range of PRSV-W was limited to the families Cucurbitaceae and Chenopodiaceae. Most susceptible cucurbit species, such as Cucumis lanatus, Cucumis sativus, Cucurbita pepo, and Citrullus lanatus, showed symptoms of green mosaic, malformation, puckering, and narrow laminae by infection with PRSV-W. The local lesion were showed on the inoculated leaves of both Chenopodium amaranticolor and C. quinoa. Field survey of PRSV, Watermelon mosaic virus (WMV) and Zucchini yellow mosaic virus (ZYMV), three major viruses infecting cucurbit, was done during 2001 to 2003 on 173 commercial cucurbit cultivating fields distributed over the three regions of Gyeonggi, Gyeongbuk and Jeonnam Provinces where cucurbits are grown in different environmental conditions and cropping patterns. Typical viral symptoms were observed from 107 cultivating fields, and all three kinds of potyviruses were detected from 206 samples out of the 235 samples using RT-PCR. Watermelon mosaic virus (WMV) and Zucchini yellow mosaic virus (ZYMV) are the most widely distributed viruses in outdoor and retarding-culture fields, at an infection rating of 48 and 33 percents, respectively. PRSV was detected from 12 percent of 235 samples. The nucleotide and amino acid sequences of coat proteins (CP) of eight PRSV isolates, collected from several areas including Anseong, were determined and sequenced heterogeneity among the isolates was performed. The CP gene of PRSV showed 88.6~97.3 percent homology in nucleotide sequences and 95.1~99.3 percent homology in amino acid sequences with other PRSV isolates worldwide. The phylogenetic analysis indicated that the Korean PRSV isolates belong to the southern-east Asian cluster.

The residue property of fungicide boscalid and fluidioxonil at the same time harvest leafy-vegetables (일시수확 엽채류에서 살균제 Boscalid와 Fludioxonil의 잔류특성)

  • In order to use in the classification of minor crop for the mutual application of safe use guideline, it was investigated the residue property of fungicide boscalid and fludioxonil at the same time harvest leafy-vegetables, such as spinach, ulgaribaechu, vitaminchae and cheongkyungchae. After pesticides were applied 2 times with 1 week interval in that day of harvest, 2 days, 5 days and 7 days before harvest, vegetables were harvested, and the residue of pesticides was investigated. Base on the residue in that day of harvest, the deposit of spray solution in vegetables was calculated. The deposit of spray solution of boscalid was 253.9 mL/kg in spinach, 83.0 mL/kg in ulgaribaechu, 97.8 mL/kg in vitaminchae, and 88.3 mL/kg in cheongkyungchae, respectively. In case of fludioxonil, it was calculated 157.6 mL/kg in spinach, 67.6 mL/kg in ulgaribaechu, 64.8 mL/kg in vitaminchae, and 66.6 mL/kg in cheongkyungchae, respectively. When the amount of the deposit of both pesticides was compared in leafy-vegetables, it was the highest in the spinach. On the other hand, it was estimated the predicted dissipation curve of pesticides in leafy-vegetables during cultivation. The half-life of boscalid was 5.9 days in spinach, 7.4 days in ulgaribaechu, 4.6 days in vitaminchae, and 4.3 days in cheongkyungchae, respectively. Also, it was estimated half-life in fludioxonil, it was 3.0 days in spinach, 4.0 days in ulgaribaechu, 3.2 days in vitaminchae, and 3.5 days in cheongkyungchae, respectively. The half-life was the longest in the ulgaribaechu. When both pesticides were compared with the residue property, the deposit of spray solution and half-life of dissipation of boscalid were more than those of fludioxonil.

Correlation between physicochemical properties and biological half-life of triazole fungicides in perilla leaf (들깻잎에서 Triazole계 살균제의 생물학적 반감기와 물리화학적 특성과의 상관관계)

  • The biological half-life of pesticides applied on crops is the key indicator for ensuring the safety of agricultural products. The biological half-life is affected by the several factors like growing conditions of the crop, climate, application method, and physicochemical properties of pesticides. In this study, the biological half-life was calculated and the degradation rates of six triazole fungicides sprayed on perilla leaves were evaluated. Moreover, the statistical analysis confirmed the correlation between the biological half-life and physicochemical properties of six triazole pesticides. The recoveries of the six pesticides were between 84.8-104.9%, which satisfied the residual pesticide analysis criteria. The biological half-life of six pesticides sprayed on perilla leaves, calculated using the first-order kinetics model, ranged between 6.4-15.1 days. When the biological half-life and the physicochemical properties were correlated using the principal component analysis: pKa and Log P, the biological half-life was found to be affected by PC1. The correlation coefficient between biological half-life and physicochemical properties (pKa), calculated by Spearman rank-order correlation, was R2 = -0.928 (p <0.01). Biological half-life has been shown to correlate with pKa. In conclusion, it can be used as a database for the relationship between biological half-life and physicochemical properties and will contribute to ensure safe supply of agricultural products.

The Analysis of Food Safety Incidents from 1998 to 2008 in Korea (1998 - 2008 발생한 식품안전관련 사건.사고 분석)

  • This study was conducted the analysis of food safety incidents between January 1998 and October 2008 using media reports. Total number of food safety incident was 569 through the study period. The average of food safety incident per year and month was 51.7and 4.9, respectively. The top 10 food types involved in the lists of food safety incidents were as follows; marine products, meat and meat products, confectionaries, beverages, special nutritional food, teas, noodles, soy and bean paste sauces, and milk and milk products etc. The top 10 single foods also were as follows; ready-to eats, meat, confectionary, health support foods, steeping tea, infant formula, meat products, ginseng products, foods for body weight control etc. Of the total 569 incidents, 247 (43.4%) were related with chemical hazards involving pesticide, food additives etc, biological hazards were 126 (22.1 %), and physical hazards were 97 (17.0%) incidents. In analysis stage in the food chain at which breakdown in food safety occurred, primary production were the most common stage with 364 (64%) incidents, and incidents at the manufacture handling and distribution stages were with 151 (26.5%), and 44 (7.7%), respectively. The results of this study can be used as a better data for risk analysis or food safety strategies.

Analysis of 2002 year's Pesticides Poisoning Deaths' data of Korea National Statistical Office and Counterplan to Prevent Deaths Due to Herbicides Poisoning (2002년 농약중독 사망자 통계청 자료 분석 및 제초제 중독으로 인한 사망사고 예방 대책)

  • Backgrounds: Deaths from pesticides poisoning has increased annually, but no studies have been reported. Especially paraquat is suspected to be a major material in pesticide poisoning deaths. In Korea, simple epidemiology of pesticides poisoning is not well known. So, there is no effective method to prevent pesticides poisoning. We tried to investigate the present status of pesticides poisoning with data from Korea National Statistical Office and suggest an effective counterplan. Methods: We analysed death cause data derived from Korean Statistical association, deaths causes being pesticides poisoning in the year 2002. Also, we investigated data from internet, mainly from Korea National Statistical Office home page. Results: Total 2,875 persons died after pesticides poisoning in 2002 year. Average age is 54.2 year, standard deviation being 17.7 year. Many victims (about $41\%$) died during the herbicides' non-necessary period (JanuaryMarch, October-December) Conclusion: Herbicides sale should be restricted only during herbicides' necessary period, and so called a pesticides safety supervisor system shall effectively prevent herbicides poisoning.
