Risk Assessment of Persicaria nepalensis Extract by Skin Irritation, Ocular Irritation, and Maximization Tests for Delayed Hypersensitivity (산여뀌 추출물의 피부자극, 안점막 자극 및 피부감작성에 대한 위해성 평가)

  • In this study, we evaluated the potential of 70% ethanol extract from Persicaria nepalensis (PNE) as a cosmetic ingredient by primary skin irritation, ocular irritation, and maximization tests for delayed hypersensitivity in New Zealand white rabbits and Hartley guinea pig. Skin safety study was performed to evaluate the potential toxicity of PNE using the primary irritation test. In the primary irritation test, 50% PNE was applied to the skin, and no adverse reactions such as erythema and edema were observed at the intact skin sites. Therefore, PNE was classified as a practically non-irritating material based on a primary irritation index of "0.0.". In the ocular irritation test, the 50% PNE applied did not show any adverse reactions in the different parts of rabbit eyes, including the cornea, iris, and conjunctiva. Thus, PNE was classified as a practically non-irritating material based on an acute ocular irritation index of "0.0.". Skin sensitization was tested by the Guinea Pig Maximization Test (GPMT) and Freund's Complete Adjuvant (FCA) using an intradermal injection of 10% PNE. Edema and erythema were not observed 24 and 48 h after the topical application of PNE in skin sensitization test, which exhibited a sensitization score of "0.0.". Therefore, it can be suggested that P. nepalensis could be used as potential candidates for cosmoceutical ingredients, without any major side effects.

Persicaria senticosa Ameliorates Imiquimod-induced Psoriasis-like Skin Lesions in Mice via Suppression of IL-6/STAT3 Expression and Proliferation of Keratinocytes

  • Psoriasis is one of the most common inflammatory skin disorders, with a global prevalence of 2% - 3%. It is an autoimmune skin disorder characterized by excessive generation of plaques on the skin with typical long-lasting red, itchy, and scaly lesions. In this study, we aimed to elucidate the anti-psoriatic effect of the methanolic extract of Persicaria senticosa (PS), a bioactive edible plant extract used in traditional medicine, using a mouse model of imiquimod (IMQ)-induced psoriasis. The daily topical application of IMQ could induce human psoriasis-like lesion. The extract ameliorated IMQ-induced psoriasis. Furthermore, hematoxylin and eosin staining and the Psoriasis Area and Severity Index (PASI) scores indicated that topical application of PS led to an improvement in erythema, scaling, and thickness scores of the mouse dorsal skin and a considerable decrease in the epidermal thickness of the ear and dorsal skin in the IMQ-induced psoriatic mouse model. We also studied the effect of PS on the proliferation of keratinocytes using HaCaT cells. The extract inhibited cell proliferation and IL-6 and pSTAT3 expression induced by M5 cocktail (comprising interleukin [IL]-1α, IL-17A, IL-22, oncostatin M, and tumor necrosis factor-α) in HaCaT cells. Thus, PS might serve as a potential therapeutic agent for the treatment of psoriasis.

Preparation Method of Indigo Standard Solution and Variation of Indigo Contents in Blue Dye Extract from Breeding Lines of Persicaria tinctoria H. Gross (인디고 분석 표준액의 제조법 및 쪽 육성계통의 인디고 함량변이)

  • This study was carried out to find out the optimum method of preparation of indigo standard solution and its stability, and to investigate the indigo contents in Niram, blue dye extract, from a total of 7 indigo plants and 34 breeding lines of Persicaria tinctoria H. Gross. Proper solvent for indigo standard was dimethyl sulfoxide (DMSO), and appropriate concentration was 1 mg of indigo in 10 mL of DMSO. Absorbance value of UV/Vis Spectrophotometer at 620 nm of standard solution was changed decreasingly 12 hours after the preparation of standard solution irrespective of the storage conditions such as temperature and light. Average value of absorbance of 8-fold diluted standard solutions prepared daily during 16 days was $0.210{\pm}0.005$, indicating the powder of indigo compound was stable chemically. Calibration curve was made for quantitative analysis of indigo of 7 Niram samples, and indigo contents ranged from 0.69% to 18.76% showing relatively larger variation. Across all 34 breeding lines, the range of indigo content was from 7.9 mg to 56.4 mg per 100 g of fresh leaves, averaging 25.2 mg of indigo content and showing a 47.7% coefficient of variation.

Analysis on the Flora and Vegetation for Ecological Restoration of Local Streams - In case of local streams in Namwon - (지방하천의 생태적 복원을 위한 식물상 및 식생분석 - 남원시 지방하천을 대상으로 -)

  • This study was carried out to offer ecological restoration plan of local streams by analyzing the vegetation and flora around four local streams (Pungchoncheon, Namchangcheon, Ganggicheon, Woncheoncheon) in Namwon. Flora and vegetation investigation was done at April 2007 to August 2007, and June 2009. The 194 of vascular plants were identified which contain 53 families, 143 genera, 194 species. The Family frequencies arranged by the order of Family Compositae (14.4%), Family Gramineae (13.4%) Family Leguminosae (6.7%) and Family Polygonaceae (5.2%). The naturalized plants were listed as 33 species, and their naturalization ratio and urban index were found to be 17.0%, and 12.2%, respectively. In this study a total of 21 relev$\acute{e}$s collected and analyzed. 6 plant communities are differentiated : Riverine softwood forests (Phragmites japonica-Salix gracilistyla community), Perennial herb vegetations on lotic water zone (Phragmites japonica community, Phalaris arundinacea community), Perennial herb vegetations on lentic water zone (Persicaria thunbergii-Typha orientalis community, Phragmites communis community), Annual or biennial vegetations on waterside (Persicaria thunbergii community). Species showing the higer value in the plant communities were Persicaria thunbergii, Phragmites japonica, Artemisia princeps, Humulus scandens, Bidens frondosa, Oenanthe javanica, Agropyron tsukusinense var. transiens, Stellaria aquatica, and Lactuca indica var. laciniata.

The Vegetation Characteristics of Small Palustrine Wetland in Rural Area (농촌지역 소규모 소택형습지의 식생특성)

  • A palustrine wetland is a type of a wetland that prevails in Korea as well as an ecosystem that provides various ecological functions and has affluent biodiversity. This study was conducted to understand the characteristics of vegetation in a palustrine wetland by analyzing a life form and naturalized plants, in order to present the maintenance and utilization plan of a small wetland. A total of 249 taxa including 76 families, 188 generics, 209 species, 38 varieties and 2 forma were found at six investigated wetlands. As a result of the appearance ratio analysis, Therophytes (37.8%) with 94 taxa and Hemicryptophytes (31.3%) with 78 taxa were in order of life form, and Gramineae (14.1 %) with 35 taxa and Compositae (11.2 %) with 28 taxa were in order of family level. These were deemed representative vegetation in a palustrine wetland. Equosetum arvense, Phragmites communis, Commelina communis, Persicaria thunbergii, Arenaria serpyllifolia, Stellaria media, Erigeron annuus, Erigeron canadensis and Artemisia princeps var, orientalis were found continuously for a period of investigation in all sites. Equosetum arvense, Phragmites communis and Persicaria thunbergii were species with high appearance frequency. Especially, Persicaria thunbergii was construed high in a cover ratio in May as it develop first among Hydrophytes and form a canopy sooner than anything else. We found that a degree of naturalization and an urbanization index appear comparatively high in Yeongok-ri, Cheonan, Gangjang-ri, Asan and Yodang-ri, Hwasung, which is construed due to artificial interference, such as fanning, fishing, swimming and green-tour program. These results would be used to utilize in restoring a deserted and damaged wetland and to provide fundamental data in creating a wetland as follows.

Inhibitory Effect of Persicaria perfoliata (L.) H. Gross on IgE Mediated Allergic Responses in RBL-2H3 Cells

  • Purpose : This study aimed to investigate the anti-allergic effect of Persicaria perfoliata water extract (PPWE) on IgE stimulated rat basophilic leukemia (RBL-2H3) cell line. Methods : P. perfoliata (L.) H. Gross has been used in traditional medicine as an anti-allergic agent, antipyretic, and diuretic and for respiratory disorders. To analyze the anti-allergic activity of PPWE, release of β-hexosaminidase in RBL-2H3 cells was estimated by enzyme linked immunosorbant assay (ELISA). Also, the cytotoxic effect of PPWE was identified by WST assay, and nuclear factor (NF)-κB and its upstream signaling molecules were assessed by western blot analysis. Results : PPWE treatment significantly attenuated β-hexosaminidase release in a dose dependent manner without any cytotoxicity. PPWE inhibited β-hexosaminidase activity by 38.4±1.2, 36.6±0.6, 32.5±2.2 and 26.5±1.2 at 500, 250, 100, and 50 ㎍/㎖ of PPWE, respectively, compared with the control group. In addition, an analysis of the expression level of NF-κB, an inflammation transcription factor, in RBL-2H3 cells upon IgE stimulation provided reults consistent with the results of β-hexosaminidase release. The phosphorylated status of upstream signaling molecules for transcription factor, mitogen activated protein kinases (MAPKs), was also analyzed. The results showed that PPWE treatment dose-dependently inhibited phosphorylation of extracellular regulatory kinase (ERK) and c-Jun N-terminal kinase (JNK). These results show that PPWE had a strong IgE-mediated degranulation inhibitory effect on RBL-2H3 cells. Conclusion : P. perfoliata ameliorated IgE-mediated allergic reaction via the modulation of MAPK and NF-κB signaling pathway in RBL-2H3 cells. These results indicate that P. perfoliata could be a potential candidate for a treatment strategy against various allergic disorders.

Moisturizing and Dryness Reduction Effect of Face Cream Containing Persicaria Perfoliata (L.) Extract (며느리배꼽추출물을 함유하는 페이스 크림의 보습 및 건조함 감소 효과)

  • Purpose : This study was aimed to analyze the effects of cosmetics containing Persicaria perfoliata water extract (PPWE) on the skin moisturizing and improvement of skin condition in clinical trials. Methods : Clinical trial was conducted for five weeks after IRB approval at Dong-Eui University. Out of a total of 64 people, 15 people each were assigned to four groups as follows; control group A, B, C and the experimental group A that using cosmetic containing PPWE. Skin condition was measured two times, before and after clinical trial, by a professional skin analyzer, SDM (skin diagnosis system). Moisture and oil value of participants was analyzed twice, each morning and evening, using a portable device on their cheeks. In addition, the survey was investigated subjective satisfaction on change in skin condition and the satisfaction on the use of cosmetics. Result : The experimental group exhibited subjectively significant changes before and after clinical trials on skin its dryness (p=.039), blush (p=.017), and redness (p<.001). In addition, subjective evaluation was also the highest satisfaction in aspects of number of application (p=.003), amount of application (p=.002), moisture maintenance, and skin scratching frequency. The satisfaction on the use of cosmetics was the highest in the intention to repurchase (p=.045), recommendation willingness to others (p=.020), and intention to use various products (p=.001). Skin moisture of the clinical trial participants using the SDM, moisture level and elasticity of the experimental group increased by 12.94 and 10.28. Moisture level, which was measured by a portable device, was the most potently increased in the experimental group. Conclusion : Consequently, PPWE containg cosmetics exhibited the effects of moisturization and attenuated skin dryness in clinical trials, which might be utilized as a fundamental data to develop numerous lines of cosmetics.

STUDIES ON THE DIMORPHISM OF THE PERSICARIA SENTICOSA NAKAI (Persicaria senticosa Nakai의 Dimorphism에 관한 연구)

Bot. 3(1) 16-25 During his researches regarding the morphological and physiological properties of Polygonecenae, the author has found that the species, Persicaria senticosa, aiso, besides the heterostylous plants of Polygonum family, Fagopyrum esculentum, and Persicaria japonica which was recently verified by the author as dimorphic, shows the typical floral structure of heterostylism, the description of which is not found in taxonomical works. Further research on this species have revealed that this plant, despite pressessing characteristic structural dimorphism, does not exhibit even the slight signs of heterostlylous properties physiologically. This is a deviation from the usual behavior of normal heterostylous plants. What is interesting is the fact that the physiological behavior of this species is quite contrary to that of P. japonica which is considered to be the most highly specialized dimorphic plant. Thus it is assurred that if some species of this family had taken a heterostylic form in its course of development from autogamy to allogamy, P. seoticosa would be the least differentiated type of dimorphic forms among the three heterostylous plants, including buckwheat, of this family. The results obtained in this experiment are summarized as follows: 1) P. senticosa has two forms of flower, one, long style-short stamened; the other, short style-long stamened. Not only conspicuous is this primary difference, but the secondary difference, such as pollen grain size, is noticeable between long and short styled individuals, thus expressing structurally the definite trait of a dimorphic plant. 2) Structural alteration of floral parts towards dimorphism has preceeded far less in comparison with those of the P. japonica and F. esculentum. 3) Elaborate studies on fertility reveal that this species does not show the slightest sign of the physiological characteristics of dimorphic plants. In other words, regardless of the modes of combinations, legitimate and illegitimate, fertilization and fruit setting flourish unimpaired. 4) Growth of pollen tubes apparently parallels the results in the fertility, tubes reaching ovary approximately 30 minutes after pollination both in legitimate and illegitimate combinations. Pollen tube penetration appears to be comparatively rapid. 5) A slight difference in the growth of pollen tube seems to exist between legitimate and illegitimate combinations, legitimate union giving slightly faster tube penetration. 6) In the present experiment it was clarified that P. senticosa, known to possess one form of flower in taxonomy, is in reality dimorphic plant having two of flowers. Although this species is definitely heterostylous in floral structure, physiological evidence and pollen tube behavior show that the differentiation of this plant toward the dimorphism has apparently proceeded slightly except for some parts of floral organ. In ordinary heterostylous plants it is a matter of common occurrence that when illegitimately cmbined, there is poor or no fertility, Contrary to the universal property of heterostylous plants, no difference is observed in the fertility and pollen tube growth between the legitimate and illegitimate combinations in the case of P. senticosa. Compared to the P. japonica and F. esculentum, which are supposed to have undergone high degree of dimorphic differentiation, it is an unavoidable conclusion that P. senticosa has not yet developed as a heterostyle plant except for some of its floral parts. If P. japonica is assumed to be the most differentiated type of hetenostylous plant, thenthe P. senticosa would be regarded as the primitive, retaining still the self-fertile nature so common to the Polygonum genus. In nature, however, this plant has a better chance to be pollinated legitimately owing to the two forms of flowers than to be pollinated illegitimately. A Study of Shelf-Life and Antimicrobial Activity on Putrefactive Microorganisms related to Soybean Curd of Persicaria hydropiper L. extracts (두부 부패 미생물에 대한 여뀌 추출물의 항균활성과 저장성 연구)

  • The objective of this study was to provide basic data that would help develop the natural preservatives that could replace them. This study examined antibacterial activity and preservative effects in soybean curd. When the concentration of Persicaria hydropiper L. was 1,000 ppm in the antibacterial activity by the paper disk law of three cultures(Bacillus subtilis KY-3, Bacillus sp. KY-7, Bacillus methylotrophicus KY-11) selected from 21 kinds of microorganism separated from tofu anaerobes, the clear zone (mm), which was similar to benzoic acid 0.1 M, the comparison group, was measured. The results were as follows. The result of impeding growth in liquid culture indicated considerable suppression of the growth of bacteria in the concentration of 800 ppm and 1,000 ppm respectively. In the measure of MIC, KY-3 and KY-7 were 0.06%, and KY-11 was 0.05%. For the changes in pH by concentration, three cultures and tofu anaerobes were not increased to $10^{5-6}/g$ in 1,000 ppm. In addition, in the search of total number was employed to find the preservative effects of tofu. Therefore, this study expect the Persicaria hydroper L. extract on preservative effects of tofu, which can be easily exposed to food poisoning bacteria in the food safety as well as improving the possibility of natural alternative preservatives.

Effect of Gibberellin on the Adventitious Root Formation from the Leaves-derived Calli in Persicaria perfoliata (며느리배꼽 잎 유래 캘러스의 부정근 형성에 미치는 지베렐린의 작용)

  • This study was carried out to investigate the action of phytohormones which influence the adventitious root formation of calli originating from the leaves of Persicaria perfoliata. The optimal medium condition for callus formation was ½-strength MS, 1% sucrose, and 4.5 μM 2,4-D. In order to determine which phytohormones had an effect on the adventitious root formation, the calluses were cultured in various media with different kinds of phytohormones. As a result, the medium with GA3 or IAA was shown to induce root formation. To deeply investigate the effects of GA3 and IAA, calli were cultured in 0.1, 1, and 10 mg/l levels of phytohormones. Numbers of roots formed per callus were 10.9, 14.2, 22.6 in GA3, 5.8, 3.9, 1.1 in IAA, respectively. Therefore, the higher GA3 or the lower IAA concentration, the more roots formed. To confirm this role of GA3 we tested with inhibitors PBZ and NPA. GA3 with PBZ resulted in reduction by 52.4~69.4% compared to GA3 alone. In contrast, GA3 with NPA resulted in an increase by -8~45.6% compared to GA3 alone in root formation. Also, results were determined on the effect of GA3 with other phytohormones on root formation. Kinetin, 2iP and ABA with GA3 had a negative effect, but IAA with GA3 showed a similar result to GA3 alone. From these results we infer GA plays a key role and auxin has subsidiary activity on adventitious root formation. This is the first report that indicates GA3 promotes adventitious root formation from calli in P. perfoliata.
