목적:. 방사선 감수성이 다른 마우스 조직에서 apoptosis 유도 수준을 확인하고 방사선 감수성에 관여 된 인자를 Proteomics를 통해서 확인한다. 대상 및 방법: C3H/HeJ 마우스에 10 Gy 방사선을 조사하고 8시간 후 비장과 간을 채취하여 apoptosis 유도 수준을 비교 분석하였다. 조직에서 단백질을 추출하여 2-dimension electrophoresis (2-DE)를 실시하였다. 2-DE에서 방사선에 의해 발현의 변화를 보이는 gel의 spot를 trypsin 처리하여 MALDI-TOF 측정한 후 Swiss-prot database를 통하여 단백질 을 동정하였다. 결과: Apoptosls index는 방사선 조사 후 비장 조직에서 $35.3{\pm}1.7{\%}$, 간조직은 $0.6{\pm}0.2{\%}$로 비장에 비해 간 조직이 낮게 나타났다. Proteomoics 결과에서 방사선 내성 조직인 간은 ROS대사에 관여되는 단백질인 glutathione Stransferase Pi, carbonic anhydrase, NADH dehydrogenase, peroxiredoxin VI, riken cDNA 등이 방사선 조사 후 증가되었고 apoptosis 관련된 단백질인 cytochrome c는 간과 비장 조직에서 확인되었다. 그러나 방사선 민감 조직인 비장에서는 방사선 조사 후 산화적 Stress에 관련된 단백질, apoptosis 관련 단백질, 신호 전달에 관련된 단백질, 면역반응, cell cycle, Ca 신호 전달, 대사 cycle에 관련된 단백질 등이 방사선에 관련하여 발현의 변화를 보여 주었다. 결론 : Apoptosis유도 수준이 다른 조직에서 apoptosis에 관련된 단백질과 redox에 관련된 단백질은 방사성 감수성 조절에 관련된 것으로 보인다.
Reactive oxygen species (ROS), which include superoxide anions and peroxides, induce oxidative stress, contributing to the initiation and progression of cardiovascular diseases involving atherosclerosis. The endogenous and exogenous factors hypercholesterolemia, hyperglycemia, hypertension, and shear stress induce various enzyme systems such as nicotinamide adenine dinucleotide (phosphate) oxidase, xanthine oxidase, and lipoxygenase in vascular and immune cells, which generate ROS. Besides inducing oxidative stress, ROS mediate signaling pathways involved in monocyte adhesion and infiltration, platelet activation, and smooth muscle cell migration. A number of antioxidant enzymes (e.g., superoxide dismutases, catalase, glutathione peroxidases, and peroxiredoxins) regulate ROS in vascular and immune cells. Atherosclerosis results from a local imbalance between ROS production and these antioxidant enzymes. In this review, we will discuss 1) oxidative stress and atherosclerosis, 2) ROS-dependent atherogenic signaling in endothelial cells, macrophages, and vascular smooth muscle cells, 3) roles of peroxidases in atherosclerosis, and 4) antioxidant drugs and therapeutic perspectives.
The peroxiredoxins (Prxs) constitute a very large and highly conserved family of thiol-based peroxidases that has been discovered only very recently. We consider here these enzymes through the angle of their discovery, and of some features of their molecular and physiological functions, focusing on complex phenotypes of the gene mutations of the 2-Cys Prxs subtype in yeast. As scavengers of the low levels of $H_2O_2$ and as $H_2O_2$ receptors and transducers, 2-Cys Prxs have been highly instrumental to understand the biological impact of $H_2O_2$, and in particular its signaling function. 2-Cys Prxs can also become potent chaperone holdases, and unveiling the in vivo relevance of this function, which is still not established, should further increase our knowledge of the biological impact and toxicity of $H_2O_2$. The diverse molecular functions of 2-Cys Prx explain the often-hard task of relating them to peroxiredoxin genes phenotypes, which underscores the pleiotropic physiological role of these enzymes and complex biologic impact of $H_2O_2$.
It is increasingly apparent that nature evolved peroxiredoxins not only as $H_2O_2$ scavengers but also as highly sensitive $H_2O_2$ sensors and signal transducers. Here we ask whether the $H_2O_2$ sensing role of Prx can be exploited to develop probes that allow to monitor intracellular $H_2O_2$ levels with unprecedented sensitivity. Indeed, simple gel shift assays visualizing the oxidation of endogenous 2-Cys peroxiredoxins have already been used to detect subtle changes in intracellular $H_2O_2$ concentration. The challenge however is to create a genetically encoded probe that offers real-time measurements of $H_2O_2$ levels in intact cells via the Prx oxidation state. We discuss potential design strategies for Prx-based probes based on either the redoxsensitive fluorophore roGFP or the conformation-sensitive fluorophore cpYFP. Furthermore, we outline the structural and chemical complexities which need to be addressed when using Prx as a sensing moiety for $H_2O_2$ probes. We suggest experimental strategies to investigate the influence of these complexities on probe behavior. In doing so, we hope to stimulate the development of Prx-based probes which may spearhead the further study of cellular $H_2O_2$ homeostasis and Prx signaling.
퍼옥시레독신은 티오레독신, 티오레독신 환원효소, NADPH로 이루어진 티오레독신 시스템의 환원력을 이용하여 과산화물을 제거하는 티오레독신 과산화효소 활성과 다른 단백질의 열변성에 의한 응집을 막아주는 샤페론 활성을 갖는 효소이다. 정형 2-Cys Prx군에 속하는 퍼옥시레독신 참고서열 1,024개 중 부분적인 서열 등을 제외한 967개 서열을 정렬하였을 때 75번과 103번 아스파트산 잔기는 99% 보존되었고, 73번 세린 잔기는 97% 보존되었음에도 불구하고 잘 보존된 아스파트산 잔기와 세린 잔기에 대해 알려지지 않았다. 이 잔기가 TSA1의 두가지 효소 활성에 미치는 영향을 알아보기 위해 재조합 단백질을 이용하여 활성도를 알아보았다. in vitro 실험을 통하여 잘 보존된 잔기인 103번 아스파트산은 75번 아스파트산보다 티오레독신 퍼옥시레독신 활성 및 분자 샤페론 활성에 더 영향을 미치고, 103번의 음전하는 분자 샤페론 활성에 중요한 역할을 하며 과산화효소활성에는 75번과 103번의 음전하가 관여함을 알 수 있었다. 또한 73의 세린 잔기 역시 과산화효소에 영향을 미치는 잔기임을 알 수 있었다. 최근 출아 효모 퍼옥시레독신인 TSA2의 79번과 109번의 세린 잔기를 시스테인 잔기로 변이시킨 경우 두 변이 단백질 모두 과산화효소 활성과 샤페론 활성이 증가되었는데 이는 ${\beta}$-sheet 구조의 증가와 관련되는 것으로 보고하였다[28]. 이들 두 세린 잔기는 TSA1 구조에 의하면 모두 ${\alpha}$-나선 구조에 위치하였다. 반면에 73번의 세린 잔기는 ${\beta}$-sheet의 C-말단에 위치하는 잔기로 과산화효소 활성에 대한 영향이 다르게 나타나는 것으로 추정된다. 추후 생체 내 실험을 통하여 아스파트산 잔기의 변이가 과산화물 저항성이 미치는 영향 및 열 저항성(thermal stress)에 미치는 역할을 살펴볼 필요가 있다. 또한 아스파트산 잔기와 과산화물과의 반응 및 분자 샤페론과의 반응에 장애가 되는 요인이 무엇인지에 대한 추가 연구가 필요할 것이다.
Alkyl hydroperoxide reductase subunit C from Pseudomonas aeruginosa PAO1 (PaAhpC) is a member of the 2-Cys peroxiredoxin family. Here, we examined the peroxidase and molecular chaperone functions of PaAhpC using a site-directed mutagenesis approach by substitution of Ser and Thr residues with Cys at positions 78 and 105 located between two catalytic cysteines. Substitution of Ser with Cys at position 78 enhanced the chaperone activity of the mutant (S78C-PaAhpC) by approximately 9-fold compared with that of the wild-type protein (WT-PaAhpC). This increased activity may have been associated with the proportionate increase in the high-molecular-weight (HMW) fraction and enhanced hydrophobicity of S78C-PaAhpC. Homology modeling revealed that mutation of $Ser^{78}$ to $Cys^{78}$ resulted in a more compact decameric structure than that observed in WT-PaAhpC and decreased the atomic distance between the two neighboring sulfur atoms of $Cys^{78}$ in the dimer-dimer interface of S78C-PaAhpC, which could be responsible for the enhanced hydrophobic interaction at the dimer-dimer interface. Furthermore, complementation assays showed that S78C-PaAhpC exhibited greatly improved the heat tolerance, resulting in enhanced1 survival under thermal stress. Thus, addition of Cys at position 78 in PaAhpC modulated the functional shifting of this protein from a peroxidase to a chaperone.
Preterm infants are vulnerable to the oxidative stress due to the production of large amounts of free radicals, antioxidant system insufficiency, and immature oligodendroglial cells. Reactive oxygen species (ROS) play a pivotal role in the development of periventricular leukomalacia. The three most common ROS are superoxide ($O2^{\cdot-}$), hydroxyl radical ($OH^{\cdot}$), and hydrogen peroxide ($H_2O_2$). Under normal physiological conditions, a balance is maintained between the production of ROS and the capacity of the antioxidant enzyme system. However, if this balance breaks down, ROS can exert toxic effects. Superoxide dismutase, glutathione peroxidase, and catalase are considered the classical antioxidant enzymes. A recently discovered antioxidant enzyme family, peroxiredoxin (Prdx), is also an important scavenger of free radicals. Prdx1 expression is induced at birth, whereas Prdx2 is constitutively expressed, and Prdx6 expression is consistent with the classical antioxidant enzymes. Several antioxidant substances have been studied as potential therapeutic agents; however, further preclinical and clinical studies are required before allowing clinical application.
Peroxiredoxin 6 (Prx 6)는 티올-특이적 항산화 단백질에 속하는 효소로서 산화적 스트레스로부터 세포를 보호할 뿐만 아니라 과산화물의 환원작용을 촉매한다. 본 연구에서는 인간 Prx 6의 promoter를 이용하여 항산화반응을 유발하는 추출물을 효과적으로 스크리닝하는 새로운 형질전환마우스를 개발하는 최종목적을 달성하기 위한 중간단계로서, hPrx 6/Luc 벡터를 개발하고, 이들 벡터의 안정적 발현과 성공적 반응성을 세포주를 이용하여 확인하고자 하였다. 이를 위해, 인간 Prx 6 promoter를 증폭하여 luciferase cDNA와 결합한 hPrx 6/Luc 벡터를 제조하였으며, 제조된 벡터를 제한효소 절단과 염기서열분석을 통해 확인하였다. hPrx 6/Luc 벡터는 NCI-H460 세포에 transfection한 후 인삼(KWG), 홍삼(KRG), 맥문동(LP), 홍문동(RLP)의 4가지 추출물을 처리하여 luciferase activity를 측정하였다. 그 결과, luciferase activity는 4가지 추출물에 의해 효과적으로 증가하였고, 특히 KRG과 LP를 처리한 그룹이 KWG과 RLP를 처리한 그룹보다 높았다. 또한, luciferase activity는 RLP 농도에 의존적으로 증가하였다. hPrx 6/Luc 벡터와 hPrx 6 mRNA반응의 차이를 비교하기 위해, 4가지 추출물을 처리한 후 hPrx 6 mRNA의 양을 RT-PCR로 분석하였다. 그러나, hPrx 6 mRNA의 양은 비록 고농도의 RLP 추출물에서는 약간의 증가가 관찰되었지만, 대조군에 비하여 4가지 추출물에서 유의적인 차이가 없었다. 한편, 4가지 추출물에 의한 superoxide dismutase (SOD) 활성의 변화는 비록 일부 차이는 있었지만 hPrx 6/Luc 벡터와 유사한 반응을 나타내었다. 따라서, 이러한 결과는 hPrx 6/Luc 벡터는 성공적으로 제조되었고, 세포내에서 안정적으로 발현하면서 항상화물질에 민감하고 정량적으로 반응할 수 있음을 제시하고 있다. 더불어, 이러한 세포주에서 확인결과를 바탕으로 형질전환마우스가 개발된다면, 항산화물질을 정량적으로 스크리닝하는 시스템으로 적용가능성이 매우 높음을 보여주고 있다.
