Antioxidative Role of Geraniin in Lipid Peroxidation of Human LDL (사람 LDL의 지질과산화에 의한 geraniin의 항산화 효과)

  • Crowing evidence indicates that oxidized low density lipoprotein (LDL) nay promote atherogenesis. Therefore, inhibition of LDL oxidation may impede this process. The effect of geraniin on the susceptibility of human low density lipoprotein (LDL) to macrophages-induced oxidation was investigated by monitoring a thiobarbiruric acid reactive substrance (TBARS). The antioxidative activity of geraniin was higher than that of $\alpha$-tocopherol on low density lipoprotein (LDL) oxidation by thiobarbituric acid reactive substance (TBARS). Geraniin inhibited the C $u^{2+}$ mediated oxidation of human LDL in a dose dependent manner at concentration of 50 and 100 $\mu\textrm{g}$/mL. Geraniin, almost completely inhibited the macrophages mediated LDL oxidation in electrophoretic mobility and conjugate diene of LDL oxidation. Also, geraniin almost completely inhibited 0$_2$$^{[-10]}$ at concentration of 100 $\mu\textrm{g}$/mL. The physiological relevance of the antioxidative activity was validated at the cellular level where geraniin inhibited endothelial cell mediated LDL oxidation, When compound with several other antioxidants geraniin showed a high activity equal to natural antioxidants, $\alpha$-tocopherol and ascorbic acid, and the synthetic antioxidant, protocol. These results indicate that geraniin might play a protective antioxidant effects on LDL, probably affecting both the structural properties of macrophage and endothelial cell for the LDL oxidation..

Improvement of Preservation Quality of Chilled Bull Semen Using ${\alpha}$-tocopherol as an Antioxidant

  • Alpha-tocopherol as an antioxidant acts in preservation of chilled semen by preserving cell membrane damage from lipid peroxidation. Optimum concentrations of ${\alpha}$-tocopherol in egg yolk-citrate (EYC) extender need to be studied in crossbred bull's semen. Different concentrations of ${\alpha}$-tocopherol viz. 0, 1, 2, 4 and 6mg per ml of extender were used. Semen was collected once a week from four bulls used to regular collection, aged 4 to 7 years, weighing 320 to 450 kg, and with body condition score 4 to 4.5 and scrotal circumference 23 to 32 cm. Semen was evaluated routinely and sperm morphology was viewed under light microscope at ${\times}1,000$ magnification after fixing with buffered formal saline. Over 90% had normal head, acrosome, mid-piece and tail. Semen was diluted with egg-yolk-citrate extender to produce $15{\times}10^6$ spermatozoa/ml and 0, 1, 2, 4 and 6 mg/ml ${\alpha}$-tocopherol were added. The semens amples were kept at $8^{\circ}C$. Sperm motility and viability were examined daily up to 5 days under light microscopy at ${\times}200$ magnification. Sperm viability was acceptable (${\geq}40%$) up to the $4^{th}$ day with all concentrations of ${\alpha}$-tocopherol and up to the $5^{th}$ day with 2 mg/ml ${\alpha}$-tocopherol. Sperm motility was acceptable (${\geq}40%$) up to the $3^{rd}$ day irrespective of ${\alpha}$-tocopherol concentration, and up to the $4^{th}$ day with 2 mg/ml ${\alpha}$-tocopherol. It is suggested that the lifespan of chilled semen may be extended up to 4 days by adding 2mg/ml ${\alpha}$-tocopherol.

Effects of Green Tea, Buckwheat and Grape Leaves Extracts on Lipid Metabolism, Antioxidative Capacity, and Antithrombotic Activity in Rats Fed High Cholesterol Diets (녹차, 메밀, 포도잎 추출물이 고콜레스테롤 급여 흰쥐의 지질대사, TBARS 및 혈액응고에 미치는 영향)

  • Effects of green tea, buckwheat, and grape leaf extracts on factors related to blood circulation were studied using rats fed high-cholesterol diet for 4 weeks. Rats were randomly divided into five groups, and plant extracts were orally administered. Green tea extract increased bleeding time in rat tails, suggesting it could prevent platelet aggregation. Administration of green tea, buckwheat, and grape leaf extracts decreased total cholesterol level in liver. Grape leaf extracts decreased thiobarbituric acid-reactive substances in plasma, whereas buckwheat and grape leaf extracts decreased the substances in liver. These results showed extracts of green tea, buckwheat, and grape leaf were effective for improving lipid composition in blood and liver and inhibiting lipid peroxidation in animal tissue, suggesting they may have potential to prevent cardiovascular diseases.

Antioxidant Activities of Red Hamcho(Salicornia herbacea L.) against Lipid Peroxidation and the Formation of Radicals (빨간 함초의 지질 과산화 및 라디칼 형성에 대한 항산화 활성)

  • Hamcho(Salicorinia herbacea, glasswort), a halophyte, is an annual succulent shrub that grows on coastal wetlands and has been regarded as a functional food for good health. Natural dried red and green Hamcho were extracted with 25% ethanol and water at 70$^{\circ}C$. The antioxidant activities of these four extracts were examined by six different assays, including the measurement of total phenolics, radical scavenging effects on 1,1-diphenyl-2-picrylhydrazyl(DPPH), nitric oxide(NO) and nitrite(NO$_2$) scavenging effects, reducing power, and the inhibitory effect on tyrosinase activity. The total phenolic contents of the four extracts were high at 237 ${\sim}$ 255 mg of gallic acid equivalents per 1 g of dried sample tested. The green Hamcho extracts contained more phenolic compounds than the red Hamcho extracts. When they were compared to tocopherols, the antioxidant activities of the green and red Hamcho powders were significantly higher at the same concentration levels(5 mg and 3 mg, respectively) in a POV test. The inhibition effects of the four extracts at a level of 75 ${\mu}l$ were higher than 98% in the POV test. The red Hamcho 25% ethanol extract showed a high significant effect on DPPH radical scavenging(SC$_{50}$, 90.1 ${\mu}l$). The green Hamcho 25% ethanol extract, however, showed a high significant effect on NO radical scavenging(SC$_{50}$, 6.1 ${\mu}l$). The NO$_2$ radical scavenging effect was assayed at pH 1.2, 4.2 and 6.0, and all the Hamcho extracts scavenged the NO$_2$ radical much more effectively at pH 1.2. The NO$_2$ scavenging effect of the red Hamcho 25% ethanol extract(64%) was as high as that of 5 mM vitamin C at pH 1.2(p<0.05). In the reducing power test, the red Hamcho 25% ethanol extract revealed the highest ferric ion reducing activity among the Hamcho extracts, and its activity was as high as that of 0.33 mg/ml of vitamin C. The four Hamcho extracts showed high tyrosinase inhibition effects of more than 80%, and their activities were higher than 50${\mu}$g of kojic acid. The green and red Hamcho 25% ethanol extracts totally inhibited tyrosinase activity(100%). Therefore, the results suggest that red Hamcho extracts may serve as useful natural antioxidants along with green Hamcho extracts.

Free Radical Scavenging Activities and Inhibitory Effect on Xanthine Oxidase by Acetone Extract from Buckwheat (수원 5호 메밀 채소 추출물의 유리라디칼 소거 및 Xanthine Oxidase 활성 저해)

  • To examine the characteristics of the antioxidative property of Buckwheat components, acetone extracts from a buckwheat, Suwon 5, was fractionated using five solvents. Hexane, ethylacetate, ether, butanol and water fractions were obtained. Butanol fraction showed the greatest electron donating ability and inhibitory effect on lipid peroxidation. It also showed the most excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. Spectrophotogram of butanol fraction was similar to that of rutin. Superoxide radical scavenging activity was related to the contents of rutin. Inhibitory effect of each fraction on xanthine oxidase was also measured. Butanol fraction had the strongest inhibitory effect on xanthine oxidase and $IC_{50}\;was\;3.1\;{\mu}g$. The inhibition type of butanol fraction on xanthine oxidase turned out to be a mixture of the uncompetitive and non-competitive modes.

Effect of Microwave on the Antioxidant Activity of Barley Bran (맥강의 항산화능에 대한 Microwave의 영향)

  • Effect of microwave treatment to barley bran was tested on antioxidative ability. Treatments of microwave at 100 W and 200 W, changed color of barley bran to brown, but carbonation was happening at 440 W. The electron donating abilities of barley bran was not reduced when treated with 100 W and 200 W microwave treatment for 10 minute. According to TBARS, the barley bran extract treated with 100 W and 200 W intensity of microwave for 10 min maintained the inhibition of lipid peroxidation, while the extract from 440 W microwave treatment for 5 min did not. Total phenolic contents were reduced with increased intensity of microwave treatment. The antioxidant activity of barley bran was not affected by microwave treatment at 100 W and 200 W, while the 440w microwave treatment reduced the activity due to carbonation of barley bran and decreased phenolic compounds.

Antioxidative Activities by Water-Soluble Extracts of Morus alba and Cudrania tricuspidata (뽕나무(Morus alba)와 꾸지뽕나무(Cudrania tricuspidata)의 수용성 추출물에 의한 항산화 활성)

  • The antioxidative activities of water-soluble extracts from leaves and stem bark of Morus alba and Cudrania tricuspidata were compared in vitro experimental models. Antioxidative activities were measured by inhibition activity against lipid peroxidation of mouse liver microsome, and they were showed in the following order; stem bark of C. tricuspidata(53%)>stem bark of M. alba(43%)>leaves of C. tricuspidata(38%)>leaves of M. alba(43%). In antioxidative activities determined by thiocyanate method and TBA method, the water-soluble extract of stem bark of C. tricuspidata showed the highest antioxidative activity. The water-soluble extracts of leaves were slightly stronger than other extracts in DPPH$({\alpha},{\alpha}'-diphenyl-{\beta}-picrylhydrazyl)$ method. The concentrations of total polyphenolic compound from water-soluble extracts of leaves and stem bark of M. alba and C. tricuspidata were 1.32%, 1.28%, 1.34% and 1.30% respectively. In these results, the water-soluble extract of stem bark from Cudrania tricuspidata showed the highest antioxidative activity.

  • The polyphenol fractions of Korean ginseng were purified using Sephadex LH-20, MCI gel, Bondapak $C_{18}$ TLC, and HPLC from the 60% acetone soluble fraction. Fraction I showed 48.16%, 79.71% and 43.55% inhibition at 150 ppm against lipid oxidation in the presence of copper ion, superoxide and hydrogen peroxidation. Electron donating abilities of fraction II showed 35.17% inhibition at 200 ppm. Fraction III showed 48.49% and 25% inhibition at 150 ppm against lipid oxidation in the presence of ferrous ion and hydroxy radical ion. The phospholipase $A_2$ inhibitory effect of fraction III was 48.9% at the concentration of $60\;{\mu}g/ml$. The cytotoxic effects of fraction II was the highest (73.29% at 0.25 mg/ml) among the tested polyphenol fractions.

Biochemical Aspect of Superoxide Toxicity to Plant Mitochondria (식물 미토콘드리아에 대한 Superoxide독성의 생화학적 측면)

  • Biochemical consequence of the accumulation in cells of superoxide $(O^{-}_{2})$ which was proposed to be probably a common chemical factor in the secondary process of the mechanism of chilling injury as well as in the visible light photodamage in cells of higher plants, has been investigated in the present work. Especially focused was the destructive effect of $O^{-}_{2}$ on the biochemical activity of mitochondria, as informations which support the suggestion that mitochondrial inner membrane is the major site of $O^{-}_{2}$ production have been collected. Mitochondria and submitochondrial particles (SMP) were prepared from soybean hypocotyls for this case study. When SMP were treated with the electrolytically produced $O^{-}_{2}$ they suffered not only inhibition of the membrane-bound enzymes as demonstrated by cytochrome c oxidase, but also lipid peroxidation of membrane as proved by malondialdehyde production. Malate dehydrogenase present in the protein extract from mitochondrial matrix was also inhibited by the $O^{-}_{2}$ treatment. These results exhibited the chaotic effect of the overproduction and accumulation of $O^{-}_{2}$ in cells under a certain abnormal circumstance such as environmental stress on the physiological function of mitochondrial; disruption of the cellular metabolic pathways and the structural integrity of membrane.

The Effect of Lipid Peroxidation of White Ginseng(WG) and Fermented Ginseng(FG) Extracts on the Liver of Mice by R-ray Irradiation (감마선을 조사한 생쥐 간에서 발효인삼이 지질과산화에 미치는 영향)

  • The effects of White Ginseng and Fermented Ginseng on liver damage induced by $^{60}Co$ r-ray irradiation were investigated. To one group of ICR male mice were given in White Ginseng(150 mg/kg/day for 7days, orally) before 5Gy(1.01Gy/min) dose of $^{60}Co$ r-ray irradiation. To another group were given in Fermented Ginseng (150 mg/kg/day for 7days, orally)before 5Gy(1.01Gy/min)dose of $^{60}Co$ r-ray irradiation was investigated. Radiation irradiation group were given with saline(0.1 ml) and 5Gy. Contrast group were given with saline(0.1 ml). The levels of $H_2O_2$, catalase and MDA in liver tissue were measured. In Fermented Ginseng (FG+Rad) group and white Ginseng(WG+Rad) group than irradiation group(Rad), the catalase level were significantly increased, and the catalase levels were appeaWhite at radiation protection. It was significantly decreased to MDA and $H_2O_2$ level to Fermented Ginseng (FG+Rad) group and white Ginseng(WG+Rad) group than irradiation group(Rad). Therefore, Fermented Ginseng and white Ginseng were very excellent protector on radiation of liver in mice.
