Transduced HSP27 protein protects neuronal cell death by enhancing FALS-associated SOD1 mutant activity

  • Familial Amyotrophic lateral sclerosis (FALS) is a progressive neurodegenetative disorder induced by mutations of the SOD1 gene. Heat shock protein 27 (HSP27) is well-defined as a stress-inducible protein, however the its role in ALS protection has not yet been established. To investigate the role HSP27 may have in SOD1 mutant-mediated apoptosis, human SOD1 or HSP27 genes were fused with a PEP-1 peptide in a bacterial expression vector to produce a genetic in-frame fusion protein, which was then transduced into cells. We found the purified PEP-1-HSP27 fusion proteins can be transduced efficiently into neuronal cells and protect against cell death by enhancing mutant SOD1 activity. Moreover, transduced PEP-1-HSP27 efficiently prevents protein aggregation produced by oxidative stress. These results suggest that transduced HSP27 fusion protein may be explored as a potential therapeutic agent for FALS patients.

Transdermal Penetration of Synthetic Peptides and Their Penetration Enhancement Caused by Some Terpene Compounds

  • The work presented in this paper represents a study of the rate and extent of transdermal penetration of three synthetic hexapeptides consisting only of glycine (Gly) and phenylalanine (Phe) as the constituent amino acids and they include Phe-Gly-Gly-Gly-Gly-Gly (Pep-1), Phe-Phe-Gly-Gly-Gly-Gly (Pep-2), and Phe-Phe-Phe- Gly-Gly-Gly (Pep-3). The present study demonstrated the extent to which the peptides having a high metabolic stability were transdermally transported from the various vehicles. The results of this study appear to indicate that minor differences in the lipophilicity of the synthetic hexapeptides have a slight influence on the rate and extent of transport. In the presence of terpene permeation enhancers, together with ethanol (i.e., menthone/ EtOH, carveol/EtOH or cineole/EtOH), the peptides were more rapidly penetrated through the skin and among the terpenes tested, cineole was the most effective for all three peptides. The maximum enhancement ratio of approximately 2 was achieved by cineole in 50% ethanol solution.

The Occurrence of Kranz Type Species Among the Noxious Weeds on Cultivated Land of Taiwan and their Biochemical Subdivision

  • One hundred and one noxious weeds on cultivated land of Taiwan were investigated for the occurrence of "Kranz" leaf anatomy and activities of PEP carboxylase and $C_4$ acid decarboxylating enzymes : NADP-malic enzyme, NAD-malic enzyme, PEP carboxykinase. Based on the leaf anatomy and a/b chlorophyll ratio, twenty-seven species exhibit "Kranz" type leaf anatomy, and seventy-four species were found without it. Among the species investigated, Digitaria radicosa (Presl) Miq., Leptochloa chinensis (L.) Nees, and Sporobolus fertilis (Steud.) W. D. Clayton in the Gramineae were first recorded as $C_4$ plants. Twenty-sven species of "Kranz" type leaf anatomy, include those of monocotyledon ; sixteen species in Gramineae, six species in Cyperaceae. Those of dicotyledon ; two species each in Euphorbiaceae and Amaranthaceae and one species in Portulacaceae. The subtype of fourteen previously uninvestigated species among twenty-seven species were further determined. The properties of the three decarboxylating enzyme from representative species were also characterized.

  • 본 논문은 전력증폭기의 효율을 개선하기 위한 도허티증폭기의 특성 분석에 관한 것이다. 도허티증폭기의 특성을 분석하기 위해 2-tone신호와 WCDMA신호를 사용하였고 180W PEP LDMOSFET을 사용하였다. 제작된 도허티증폭기는 출력 전력 20W에서 약 10%정도의 효율개선을 나타냈지만 선형성이 저하되는 특성을 나타내었다. 저하된 선형성을 개선시키기 위해 carrier 증폭기(Amp. A)와 peaking 증폭기(Amp. B)를 각각 변화하였으며, carrier 증폭기는 높은 PEP(Peak Envelope Power)와 적은 Id(drain current)가 되도록 하였고, peaking 증폭기는 높은 이득과, 낮은 gate bias에서 IMD(Intermodulation Distortion)가 상쇄되도록 하였다. 최종 측정 결과 출력전력 20W에서 선형성은 class AB증폭기와 비교하여 유사한 결과를 얻었으며, 효율은 약 10% 이상의 개선을 얻을 수 있었다.

  • RTT(Round Trip Time)가 큰 위성링크에서는 TCP의 정상적인 동작이 어렵다. PEP(Performance Enhanced Proxy)은 위성과 지상 통신간의 성능 향상을 위해 개발되었으나. PEP의 위성링크 구간에 적용되는 TCP Cubic, TCP Hybla등 기존의 Enhanced TCP는 충분한 윈도우사이즈를 확보하는데 시간이 걸리는 문제점이 있다. 본 논문에서는 Cubic의 초기 윈도우 값을 높게 설정해 위성 대역폭을 보다 빨리 점유할 수 있는 수정된 혼잡제어 알고리즘을 제안한다. 제안한 방법은 기존 혼잡제어 알고리즘보다 전송률이 약 26% 향상되었다.

A Case Study of Job-Shadowing Internship Program for Information Specialists (전문가양성을 위한 직무관찰식 인턴쉽 시행연구 - 문헌정보학 전공사례 -)

  • This study focuses on two analysis: direct cognition of effectiveness of internship practicum, difference of career attitude maturity. The purpose of this experimental case study is to find out the practical problems of Internship education as a field-study of LIS by applying Job-Shadowing Internship methods. The results reveal that 71% of interns responded the positive effects of Job-Shadowing, but the effects to the career attitude maturity was not statistically significant. The implications of the case study are as follows; (1) systematic pre-instruction and orientation from the department are the critical factor of internship, (2) practicum guidelines(PEP), which is provided by the person in charge of tasks in internship institutions is the another important factor, and finally, (3) tools for measuring the effectiveness of internship is required for LIS practicum major course development.

A STUDY ON COMORBID DISORDERS AND ASSOCIATED SYMPTOMS OF PERVASIVE DEVELOPMENTAL DISORDER CHILDREN (전반적 발달장애 아동들의 공존질환 및 동반증상에 대한 연구)

  • Objective:The purpose of this study was to investigate the prevalence and characteristics of comorbid disorders and associated symptoms in pervasive developmental disorder(PDD) and to examine the correlation between associated symptoms and developmental characteristics in PDD children. Method:The sample consisted of 209 cases of PDD and 143 cases of developmental language disorder(DLD)(control group) who were treated at the Seoul National Mental Hospital from Jan. 1996 to Mar. 1999. The diagnostic work based on DSM-IV criteria was performed by one or two child psychiatrists, while the clinical feature was evaluated by doctors’s notes, occupational/speech therapy reports, and results of social maturity scale(SMS), childhood autism rating scale(CARS), and psycho-educational profile(PEP). Two groups were compared on a wide range of measures including comorbid disorders, associated symptoms, treatment drugs, and PEP. The relation between associated symptom & PEP was investigated in total(106 cases) and in each dignostic group. Sixty-four cases of PDD were divided into three groups by CARS and then compared on associated symptoms. Result:The prevalence of comorbid disorder was 19.6% in PDD, 41.2% in DLD. The rate of manifestation of 13 associated symptoms was 31.47% in PDD, 22.13% in DLD on the average. Associated symptoms significantly high in PDD were preoccupation, obsession, self-mutilation, stereotypy, sleep problems, and odd response. In total patient group, associated symptoms that significantly influenced PEP were preoccupation, self-stimulation, stereotypy, inappropriate affect, sleep problems, and odd response. But, in each diagnostic group, no associated symptom influenced PEP. Associated symptoms significantly different between the 3 groups of CARS were stereotypy, anxiety, and sleep problems. Conclusion:These preliminary results suggest that developmental characteristics may influence associated symptoms in PDD children and a realistic approach considering minute diagnosis by associated symptoms and comorbid disorders is required.

THE TREATMENT RESULTS OF A DAY TREATMENT PROGRAM FOR CHILDREN WITH DEVELOPMENTAL DISORDERS (발달 장애 아동에게 시행된 주간 치료실 프로그램의 치료 결과에 대한 고찰)

  • Objective:This was conduced to evaluate the effect of early intervention to promote the development of children with developmental disorders. Methods:49 children(31 in PDD group and 18 in DLD group) participated in a one year day treatment program conduced from 1996 to 1999. They were performed PEP, CARS, and SMS. They were grouped by diagnosis, comorbidity, chronological age and CARS score at the beginning of the program and the treatment effect was compared. Results:The children who participated in the day treatment program showed significant increase in their PEP, SMS score, and decrease trend in their CARS score. When children were grouped by diagnosis, comorbidity, chronological age, and severity in CARS score we did not find out significant difference between groups. Conclusion:Our data suggest that the day treatment program which emphasis on development is effective in treating children with developmental disorders.

Post-exposure Prophylaxis against Varicella Zoster Virus in Hospitalized Children after Inadvertent Exposure (수두-대상포진 바이러스에 노출된 소아 환자의 예방 조치)

  • Purpose: This study described the post-exposure prophylaxis (PEP) and secondary varicella infection in children inadvertently exposed to varicella zoster virus (VZV) in the hospital. Methods: We retrospectively analyzed data from patients with VZV infection who were initially not properly isolated, as well as children exposed to VZV at the Seoul National University Children's Hospital between January 2010 and December 2015. The PEP measures were determined by the presence of immunity to VZV and immunocompromising conditions. Patient clinical information was reviewed via medical records. Results: Among 147 children hospitalized between 2010 and 2015, 13 inadvertent exposures were notified due to VZV infection. Five index children had a history of VZV vaccination. Eighty-six children were exposed in multi-occupancy rooms and 62.8% (54/86) were immune to VZV. The PEP measures administered to 27 exposed patients included varicella zoster immunoglobulin and VZV vaccination. Four children developed secondary varicella, which was linked to a single index patient, including one child who did not receive PEP and three of the 27 children who received PEP. The rates of secondary varicella and prophylaxis failure were 4.7% (4/85) and 11.1% (3/27), respectively. The secondary varicella rates were 1.9% (1/54) and 9.7% (3/31) among immunocompetent and immunocompromised children, respectively. Conclusions: Delayed diagnosis of VZV infection can lead to unexpected exposure and place susceptible children and immunocompromised patients at risk for developing varicella. The appropriateness of the current PEP strategy based on VZV immunity may require re-evaluation.

Specific Primer Sets for RT-PCR Detection of Major RNA Viruses of Tomato Plants in Korea (국내의 토마토 주요 바이러스 진단을 위한 역전사중합반응법용 프라이머 세트)

  • Major tomato viruses in Korea are Tomato chlorosis virus (ToCV), Tomato spotted wilt virus (TSWV), Cucumber mosaic virus (CMV), Pepper mottle virus (PepMoV), and Tomato mosaic virus (ToMV). RT-PCR conditions for the viruses were examined, especially in primer set and RT-PCR mixture. Total 46 primer sets from the unique sequence of the viruses were tested for nonspecific background products in a RT-PCR mixture without template. Among them 16 primer sets were applied to healthy tomato RNA, resulting the compatibility between RT-PCR mixture and primer set influenced RT-PCR to reduce nonspecific background products. Based on the combinations among cDNA synthesis parameters and RT-PCR mixtures, two reaction mixtures were finally selected for ToCV detection. The condition allowed to determine more specific primer sets; C029 (ToCV), C072 (TSWV), C070 (CMV), C048 (PepMoV), and C065 (ToMV). These primer sets are expected to be of use to specific detection of the major viruses in tomato plants.
