열처리에 의한 작약의 Paeoniflorin 함량 변화 (Changes of Paeoniflorin Content in Peony Roots by Heat-treatment)

  • Peony is a medicinal herb which have utilized widely as chineses medicine. The paeoniflorin is the predominant component In peony root, monoterpene glucoside containing pinane structure. The effective components were extracted with the cold water from the intact peony roots, and effectively extracted with 70% ethanol from the dry powder of peony roots. The changes of paeoniflorin contents were investigated during the drying process of peony roots and processing of peony extract by the heat-treatment. Air-drying was the best condition for the preservation of paeoniflorin content among the drying processes of peony roots. But convective drying at 6$0^{\circ}C$ was recommended for the drying process of peony roots in large scale. The paeoniflorin in peony extracts was not destroyed by the treatment at 6$0^{\circ}C$ and 8$0^{\circ}C$ for 5 hrs, but destroyed 30%, 28% and 40% of paeoniflorin by treatment at 10$0^{\circ}C$ for 5 hrs, 115$^{\circ}C$ and 121$^{\circ}C$ for 10 minutes, respectively. The paeoniflorin was continueously extracted for 4 hrs from the dry pieces of peony roots(0.5$\times$0.5$\times$0.5cm) in boiling water but destroyed gradually after 4 hrs at 10$0^{\circ}C$. Paeoniflorins in 70% ethanol extracts of peony root were not destroyed at all in the process of concentration to dry powder at 60"C on vacuum.cuum.

작약류(芍藥類)의 진통(鎭痛) 진경(鎭痙)에 대(對)한 연구(硏究) (A Study on the Analgesic and Anti-convulsional Effect of Paeoniae Radix)

  • Both wild peony and cultivated peony demonstrated a slight effect and tranquilizer of anti-convulsion on the strychinine and picrotoxine but the wild plant was more effective. The methanol extract of cultivated peony root demonstrated a weak effect of muscle loosening but its mixture with water extractable fraction did not have this effect in contrast to the case wild type whose methanol extract mixture with water extractable fraction still retained a strong anti-convulsional muscle loosening effect. Therefore, it is concluded that wild peony root has more analgesic and muscle loosening effect than the cultivated counterpart has.

Antioxidative Constituents from Paeonia lactiflora

  • The ethanol extract of the peony root (Paeonia Lactiflora Pall, Paeoniaceae) as well as its major active components including gallic acid and methyl gallate were evaluated for their protective effects against free radical generation and lipid peroxidation. In addition, the protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were examined. The ethanol extracts of the peony root (PREs) and its active constituents, gallic acid and methyl gallate, exhibited a significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and had an inhibitory effect on lipid peroxidation, as measured by the level of malondialdehyde (MDA) formation. The PREs did not have any pro-oxidant effect. They strongly inhibited the hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, as assessed by single cell gel electrophoresis. Furthermore, the oral administration of 50% PRE (50% ethanol extract of peony root), gallic acid and methyl gallate potently inhibited the formation of micronucleated reticulocytes (MNRET) in the mouse peripheral blood induced by a $KBrO_3$ treatment in vivo. Therefore, PREs containing gallic acid and methyl gallate may be a useful antigenotoxic antioxidant by scavenging free radicals, inhibiting lipid peroxidation and protecting against oxidative DNA damage without exhibiting any pro-oxidant effect.

작약 품종의 가공방법에 따른 엑스 함량 비교 (Comparison on the Extract Content by Different Processing Method in Peony (Paeonia lactiflora Pall.) Root)

  • 한국산 재배작약의 품질평가 기준 확립을 위한 연구의 일환으로 국내 재매작약의 생육년수, 품종, 건조방법에 따른 작약의 성분적 품질평가의 한 요인이 되는 엑스 함량을 검토하고 아울러 작약의 활성효과 중 항염증 작용 및 위액분비 억제 작용에 관여하는 것으로 보고된 엑스의 pH 변화 양상을 검토한 결과를 요약하면 다음과 같다. 1. 생육년수의 차이에 따른 엑스 함량 및 pH변화를 검토한 결과 유피근, 거피근 모두 생육년수에 따른 통계적 차이를 나타내지 않았다. 한편 $2{\sim}4$년생 작약의 유피근 함유엑스 함량이 거피근보다 $3.7%{\sim}9.2%$ 높은 양상을 나타내므로 작약근 껍질부위에도 엑스 추출 시 용출되는 물질이 다량 존재함을 알 수 있다. 2. 4년생 의성작약, 영천작약 및 좀작약을 각각 유피근과 거피근으로 가공하고 상온에서 음건한 후 각 품종별 엑스함량 및 엑스의 pH변화를 조사한 결과 엑스 함량의 경우 유피근, 거피근 모두 품종 간 차이를 나타내었고, 영천작약이 유피근, 거피근 각각 36%, 30% 수준으로 의성 작약 및 좀작약 보다 높은 엑스 함량을 나타내었으며, 엑스의 pH는 품종 간 차이가 인정 되지 않았다. 3. 4년생 의성작약을 대상으로 건조방법에 따른 엑스 함량 및 엑스의 pH 변화를 검토한 결과 유피근, 거피근 모두 건조방법 간에 뚜렷한 차이를 나타내었다. 엑스 함량은 유피근의 경우 상온 음건이, 거피근은 $80^{\circ}C$ 수증건법 이 가장 높았고, 동결건조법은 유피근, 거피근 모두 가장 낮은 함량을 나타내었다. 엑스의 pH변화는 유피근, 거피근 모두 냉동건조법이 pH 5.1 수준으로 가장 높았고, $80^{\circ}C$ 수증건법이 유피근, 거피근 모두 pH 3.7 수준으로 가장 낮은 양상을 나타내었다.

경북지역(慶北地域)에서 재배중(栽培中)인 작약(芍藥)의 Paeoniflorin 함량분석(含量分析) (Analysis of the Content of Paeoniflorin in Peony Roots Cultivated on Kyeongbuk Area)

  • 주(主) 재배품종인 의성종과 영천종의 유효성분을 HPLC 및 TLC로 분석한 결과 주(主)성분인 paeoniflorin 함량은 의성종이 영천종보다 높으며 albiflorin과 oxypaeoniflorin의 함량을 두 재배종에서 비교하면 의성종은 albiflorin 함량이 많고 영천종은 oxypaeoniflorin의 함량이 많았다. 생산지에 따른 paeoniflorin의 함량차이는 2.15% 에서 4.08% 의 범위였고 의성과 거창의 것은 비슷하였으며 영천의 것이 가장 낮았다. 18종의 재배종들은 품종에 따라 다양한 paeoniflorin 함량 차이를 보이며 작약성분분석의 HPLC 크로마토그랍을 albiflorin, oxypaeoniflorin 및 paeoniflorin 피크를 기준으로 비교해본 결과 크게 3가지 패턴으로 분류할수 있었다. 이 세가지 유형은 의성종 패턴 9종, 영천종 패턴 4종 풍기표준종패턴 5종 이었다. 수확시기에 따른 작약의 paeoniflorin의 함량변화를 조사한 결과 3월과 9월에 수확한 것이 낮았으며 5월과 11월에 수확한 작약의 paeoniflorin 함량이 높았다.

작약엑스정 1회 경구 투여의 안전성 평가에 관한 연구 (A Single Center Study on the Evaluation of Safety after Single Oral Administration of Peony Root Extract Tablets)

  • Objectives This study is designed to evaluated the safety of peony root extract tablet in healthy male volunteers. Methods 12 healthy male volunteers were recruited, and this study was conducted by a single center. The safety was evaluated by collecting laboratory test and vital signs of volunteers. As the registration process, 12 subjects were assigned by serial number. To evaluate safety, vital signs were checked and blood samples were collected 4 times during the screening period, pre & post-administration (after 8 hours) and post-administration (after 7 days). The difference in variables was summarized by the mean±standard deviation. The normality test was carried out using the Shapiro-Wilk test and Kolmogorov-Smirnov test. When normality is fulfilled, a paired t-test is applied and the significance level was p<0.05. And the incidence of all adverse effects and serious adverse effects are shown in percentage. Results In the case of vital sign, body temperature (BT) (℃) was 0.06±0.05 ℃ (p=0.008), and there was a statistically significant difference in before and after administration. However, clinical symptoms were not occurred and BT (℃) of all subjects before and after administration showed values within the normal reference value. There was no significant difference from the control group in all other vital signs and laboratory test data. And no side-effects associated to clinical trial drugs were followed. Conclusions The peony root extract tablet was considered to be safe for healthy male volunteers.

Amendment with Peony Root Bark Improves the Biocontrol Efficacy of Trichoderma harzianum against Rhizoctonia solani

  • We tested Trichoderma harzianum as a biocontrol agent for Rhizoctonia solani AG2-1, using six natural antifungal materials to improve its efficacy. Among the six materials tested, peony (Paeonia suffruticosa) root bark (PRB) showed the strongest antifungal activity against R. solani AG2-1, and was not antagonistic to T. harzianum. Scanning electron microscopy showed that treatment with PRB extract resulted in shortened and deformed R. so/ani AG2-1 hyphal cells. The control of radish damping-off caused by R. so/ani AG2-1 was greatly increased by combined treatments of T. harzianum and PRB, as compared with either of the two treatments alone, with the control effect increased from 42.3-51.5% to 71.4-87.6%. The antifungal compound in PRB, which was isolated in chloroform and identified as paeonol by mass spectrometry, $^1H$ NMR, and $^{13}C$ NMR analyses, inhibited the growth of R. so/ani AG2-1 but not that of T. harzianum. Thus, PRB powder or extract may be used as a safe additive to T. harzianum to improve the control of the soil borne diseases caused by R. so/ani AG2-1.

백작약 추출물이 항산화활성, LDL 산화 억제 및 혈전용해에 미치는 영향 (Preventive Effects of Peony Root Extracts on Oxidative Stress, Thrombosis and Atherosclerosis)

  • Objectives: There is currently increased interest in the identification of natural antioxidant compounds derived from various plants. Peony Root (PR) is used worldwide for the treatment of many types of cardiovascular disease including atherosclerosis and hypertension. It has been used in Korean traditional medicine for the treatment of glycosuria, hypertension and cancer. However, to date, no studies concerning the antioxidant properties of PR have been conducted. Therefore, this study was conducted to evaluate the in vitro scavenging activity, inhibitory effect of LDL oxidation of pro-oxidant reactive species and anti-thrombosis effect in response to treatment with PR using various screening methods including biological and non-biological oxidants. Methods: In this study, the antioxidant activity of extract from PR was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation and the inhibitory effect on collagen-induced platelet aggregation. Results: The PR extracts were found to have a potent scavenging activity of oxidative stress [DPPH, superoxide anion, hydroxyl radical, nitric oxide and peroxynitrite, etc.] as well as an inhibitory effect on LDL oxidation and on platelet aggregation. Conclusions: The PR extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.

Anti-dementia Effects of Gouteng-san and Si-Wu-Tang

  • Recently, a traditional medicine called Gouteng-san, which consists of eleven herbs, was reported to be effective in treating vascular dementia with a double-blind, placebo-controlled study. Gout-eng-san is also used for patients with vascular dementia in combination with Si-Wu-Tang. The effect of Gouteng-san and Si-Wu-Tang on deficit of learning behavior was investigated using step-down passive avoidance task in mice. Hot-water extract of Gouteng-san (1.5 and 6 g/kg, p.o.) significantly prolonged the step-down latency shortened by scopolamine. The extract of Uncaria hook (150 mg/kg, p.o.), one of the component herb of Gouteng-san, significantly prevented the decrease in the latency after scopolamine. Hot-water extract of Si-Wu-Tang (1.5 and 6 g/kg of dried herbs, p.o.) prevented dose-dependently scopola-mine-induced disruption qf learning behavior. Si-Wu-Tang also prevented the ischemia-induced deficit of learning behavior. Both hot water extract of peony and angelica (1.5 g/kg, p.o.), which are component herbs qf Si-Wu-Tang, prevented the scopolamine-induced learning behavior deficit. Scopolamine (10 uM) suppressed long-term potentiation (LTP) of population spike in the CA1 region of the rat hippocampal slices. Peoniflorin (0.1~ 1uM) extracted from paeony root significantly ameliorated scopolamine-induced inhibition of LTR These results suggest that improvement of deficit of learning behavior by Gouteng-san and Si-Wu-Tang is mediated by direct and/or indirect activation of the cholinergic system in the brain.
