  • The feasibility of [P(AA-co-PEGMM)] film as a buccal mucoadhesive patch was previously reported by estimating mucoadhesiveness and release characteristics. To find a rational penetration enhancer of [P(AA-co-PEGMM)] film containing butorphanol tartrate (Bt), penetration of Bt from [P(AA-co-PEGMM)] film which contained various additives was estimated by measuring its flux, Papp and lag tme in in vitro buccal membrane of porcine. EDTA showed almost no increase of Bt permeability, wherease SGC, STDHF and SLS increased the permeability of Bt with the order of SGC > STDHF > SLS. The rational additive concentration of SGC was 4% and its Papp and lag time were $1.93{\times}10^{-4}{\pm}4.21{\times}10^{-6},\;126.60{\pm}21.88min\;(control\;:\;Papp\;0.45{\times}10^{-4};\;lag\;time\;211.01{\pm}16.77\;min)$, respectively.

Study on the In Vitro Maturation and Sperm Penetration Rates of Canine Oocytes

  • The purpose of this study was to investigate the effects of the collection time, co-culture and sperm penetration of canine oocytes on in vitro maturation and fertilization. The oocytes were cultured in TCM-199 media containing hormonal supplements (10% FCS, 10 IU/ml HCG, 10 IU/ml PMSG) at 5% $CO_2$, 95% air, $38^{\circ}C$. The in vitro maturation rate to MII stage of in vitro oocytes recovered from ovaries that collected at follicular, luteal and inactive phases of the reproductive phase for 44~72 hrs were 19.2%, 12.2%, and 6.0%, respectively. Follicular phases oocytes had a significantly higher in vitro maturation rate than oocytes collected at luteal and anestrus stage (p<0.05). The in vitro maturation rates to the MII stage of canine oocytes after 48 hrs of culture with glutathione, pyruvate, or glutathione + pyruvate were 12.5%, 10.7%, and 17.5%, respectively. This was higher than that in both alone or the combination of the two compared to the control group (19.0%). The sperm penetration rates of in vitro matured oocytes by fresh and frozen semen were 29/80 (36.3%) and 18/80 (22.5%), respectively. Although there are limited reports about canine oocytes co-culture and in vitro fertilization, our results on in vitro maturation is comparable to the results from other researches.

  • Generally, the strength of the field shotcrete is heavily dependent on the field mixing and spraying conditions so that it is different from the strength of the shotcrete mixed in laboratories. As a support member, the early strength of shotcrete unlike concrete as structural material is very important to the initial stabilization of the excavation face in tunnels. Therefore, the field methods to efficiently test the early strength of shotcrete have been highly required. This paper aimed to verify the pneumatic pin penetration test and the point load test for measuring the early strength of the field shotcrete. Through a series of uniaxial compression, pin penetration, and point load tests for the range of the early shotcrete strength, two equations to estimate reliably the uniaxial compressive strength by the pin penetration and the point load tests were derived. Field tests in working tunnel were carried out in order to estimate the economic efficiency. As a result, pin penetration method was proved to be the most effective method for testing the early strength of the field shotcrete.

Experiments on the Thermal Stratification in the Branch of NPP

  • The thermal stratification phenomena, frequently occurring in the component of nuclear power plant system such as pressurizer surge line, steam generator inlet nozzle, safety injection system (SIS), and chemical and volume control system (CVCS), can cause through-wall cracks, thermal fatigue, unexpected piping displacement and dislocation, and pipe support damage. The phenomenon is one of the unaccounted load in the design stage. However, the load have been found to be serious as nuclear power plant operation experience accumulates. In particular, the thermal stratification by the turbulent penetration or valve leak in the SIS and SCS pipe line can lead these safety systems to failure by the thermal fatigue. Therefore in this study an 1/10 scaledowned experimental rig had been designed and installed. And a series of experimental works had been executed to measure the temperature distribution (thermal stratification) in these systems by the turbulent penetration, valve leak, and heat transfer through valve. The results provide very valuable informations such as turbulent penetration depth, the possibility of thermal stratification by the heat transfer through valve, etc. Also the results are expected to be useful to understand the thermal stratification in these systems, establish the thermal strati­fication criteria and validate the calculation results by CFD Codes such as Fluent, Phenix, CFX.

Formulation and Skin Permeation Characteristics of Ketoprofen Patches (케토프로펜 패취제의 제제설계 및 피부 투과 특성)

  • Ketoprofen (KP) was formulated as a transdermal patch using the percutaneous penetration enhancers sorbitan monmmleate(SMO), polyvinylpyrrolidone(PVP). The control patch without penetration enhancers showed a KP flux of 8.9$\pm$0.75$\mu\textrm{g}$/$\textrm{cm}^2$/h The flux was increased in proportion to the concentration of SMO added. Furthermore, lag times were decreased upon addition of SMO. Conversely; the skin flux of KP was decreased in proportion to the concentration of PVP added. Pharmacokinetic parameters including $C_{max}$, $T_{max}$, and AUC were increased when SMO was added. However, $C_{mas}$ significantly decreased by the addition of PVP. $T_{max}$ was not significantly different in 2%, 4%, and 8% PVP patches. Patches containing 4% PVP showed the highest AUC value (19.158$\mu\textrm{g}$.h/ml). We found that the effectiveness of the two percutaneous penetration enhancers for topical KP patches was similar, with the addition of appropriate amounts of HPC modifying both skin flux and lag time of KP in the patches. In conclusion, it is possible to manufacture KP patches exhibiting high AUC, high skin flux, and short lag time using percutaneous penetration enhancers of SMO and PVP.

Effect of the Addition of β-Hydroxybutyrate to Chemically Defined Maturation Medium on the Nuclear Maturation, Sperm Penetration and Embryonic Development of Porcine Oocytes In vitro

  • We investigated the effects of various concentrations of ${\beta}$-hydroxybutyrate (BHB, 0, 0.1, 1 and 10 mM), a ketone body, added to chemically-defined maturation medium with or without energy substrates (glucose, pyruvate and lactate) on nuclear maturation rates up to the metaphase stage of the second meiotic division (M-II stage). In addition, we also assessed the influence of BHB on glutathione content, sperm penetration rate and embryonic development up to the blastocyst stage of oocytes matured under the presence of these energy substrates. Nuclear maturation rates up to the M-II stage of oocytes matured with BHB in each concentration group did not show a significant increase compared with the control (0 mM) groups in both the presence and absence of energy substrates. Although glutathione contents were not significantly different in each BHB concentration group, the sperm penetration rate in the 1 mM BHB group was significantly higher (p<0.05) and the embryonic development rate of oocytes up to the blastocyst stage was significantly lower (p<0.05) than the respective values of the control groups. These results suggest that BHB added to a chemically-defined maturation medium may stimulate sperm penetration while inhibiting embryonic development of porcine oocytes.

Fabrication of Li2TiO3 Pebbles by Lithium Solution Penetration Method (리튬용액 침투방법에 의한 Li2TiO3 페블 제조)

  • To fabricate spherical lithium titanate ($Li_2TiO_3$) pebbles which are used for a breeder material in fusion reactor, titanium oxide ($TiO_2$) granules were used as a starting material. The granules were pre-sintered, and then aqueous lithium nitrate solution infiltrated into the granules at vacuum condition. The granules were crystallized to $Li_2TiO_3$ after sintering under the control of process parameters. In this study, the concentration of lithium in the solution, as well as the number of penetration times and sintering temperature affected the final crystallite phase and the microstructure of the pebbles. In particular, the sphericity and size of the pebbles were effectively controlled by a technical rolling process. The useful spherical $Li_2TiO_3$ pebbles which have 10~20% porosity and 60~120 N compressive strength were obtained through the sintering at $1000{\sim}1100^{\circ}C$ in the multi-times infiltration process with 50 wt% solution. The physical properties of pebbles such as density, porosity and strength, can be controlled by a selection of $TiO_2$ powders and control of processing parameters. It can be thought that the lithium penetration method is a useful method for the fabrication of mass product of spherical $Li_2TiO_3$ pebbles.

  • The purpose of this study was to evaluate the adaptability to tooth structure of composite resin and glass ionomer cement according to filling methods. In this study. two class V cavities were prepared on the buccal and lingual surface of each tooth of forty extracted human premolars. and they were randomly assigned into 4 groups with 10 teeth. The cavities of each group were filled with the CLEARFIL FII self curing resin(Control Group), Z-100 light curing resin (Group 1). $Vitremer^{TM}$ light curing glass ionomer cement(Group 2) and Z-100 light curing resin over the $Vitremer^{TM}$ liner(Group 3). The specimens underwent temperature changed from $5^{\circ}C$ to $55^{\circ}C$ five hundred times. After thermocycling. specimens were immersed in 2% methylene blue solution and stored in 100% relative humidity at $37^{\circ}C$ for 24 hours. And then. the specimens sectioned buccolingually. Degree of dye penetration at tooth-restoration interfaces were examined by Tool maker's microscope(x 200) and Image analyzer. The results were as follows : 1. On the occlusal margin. among the experimental groups. the group 2 showed the highest dye penetration($2.40{\pm}0.68$) and the group 3 showed the lowest dye penetration($1.15{\pm}0.37$). There was significant difference among the experimental groups(p<0.001). 2. On the gingival margin, among the experimental groups, the group 1 showed the highest dye penetration($3.30{\pm}0.57$) and the group 2 showed the lowest dye penetration($1.65{\pm}0.49$). There was significant difference among the experimental groups(p>0.001). 3. About total degree of dye penetration, the group 1 showed the highest dye penetration($2.25{\pm}1.17$) and the group 3 showed the lowest dye penetration ($1.43{\pm}0.55$). There was significant difference among the experimental groups(p<0.001). 4. The sum of dye penetration at occlusal margin was less than gingival margin. There was significant difference between occlusal margin and gingival margin (p<0.001). The results showed that differences were more pronounced at the gingival margin. Composite restorations inserted over the glass-ionomer liner demonstrated significantly less leakage than single restoration that used composite resin or glass-ionomer cement.
