A Study on the Effect of Herbal-acupuncture with Acanthopanacis cortex solution on Arthritis (오가피약침(五加皮藥鍼)이 류마토이드 관절염(關節炎)에 미치는 실험적(實驗的) 연구(硏究))

  • Objective : The purpose of this study is to observe the effect of Herbal-cupuncture with Acanthopanacis cortex Herbal-Acupuncture Solution (AC-HAS) on rheumatoid arthritis. Methods : The author performed several experimental items : it is paw thickness, weight of spleen, expression of $CD4^+,\;CD8^+,\;B220^+$ in the spleen, production of IL-6, $TNF-{\alpha}$, and examination of histology. Results : The obstain results are summarized s follows. 1. Treatment group were significantly inhibited the incidence of arthritis, hind paw edema and index of arthritis of CIA mice. 2. Treatment group were significantly decreased splenetic weight 3. The number of $CD4^+,\;CD8^+,\;B220^+$ activated cells were significantly inhibited in treatment group. 4. Treatment group were significantly decreased the amout of IL-4, $TNF-{\alpha}$ of CIA mice.. 5. Treatment group were expressed form of new bone, synoviumin and new margine in histology imperison to control group. Conclusions : Taking all these observations into account, AC-HAS injection is considered to be effective in treating rheumatoid arthritis and put to practical use in future rheumatoid arthritis clinic.

Anti-nociceptive and anti-inflammatory effects of Geranii Herba (현지초(玄之草)의 진통(鎭痛) 및 항염증(抗炎症) 효과)

  • Objectives : The present study investigated the anti-nociceptive and anti-inflammatory properties of the water extract of Geranii Herba (The stem and leaves of Geranium thunbergii Sieb. et Zucc.) in the animal models of pain and inflammation. Methods : We evaluated the anti-nociceptive and anti-inflammatory activities of Geranii Herba extract (GHE) using the writhing test, tail-flick test, carrageenan-induced paw edema and xylene-induced ear edema models. Two dose of GHE (100 and 1000 mg/kg) was administrated orally to the mice. Control group received normal saline and ibuprofen (50 mg/kg) was used as a positive control drug. Results : GHE 1000 mg/kg treated group showed an increased tail-flick response time in the tail-flick test and inhibitory effect on writhing syndrome induced by acetic acid. Treatment with GHE at the same dose inhibited ear edema induced by xylene and foot edema induced by carrageenan toxicity. Conclusion : The results demonstrate that GHE has anti-nociceptive and anti-inflammatory effects in the various models of nociception and inflammation.

Anti-inflammatory and anti-noceceptive action of the crude extracts of Costus specious on rodents

  • The effect of alcoholic extracts of Costus specious (Family: Zingiberaceae) was evaluated in experimental models of pain and inflammation. Oral administration of 100, 200 and 300 mg/kg of C. specious extracts were used for the above study. Crude extracts of C. specious (300 mg/kg dose) showed maximum time needed for the response against thermal stimuli ($7.242\;{\pm}\;0.532\;s$) which is comparable to diclofenac sodium ($8.471\;{\pm}\;0.25\;s$) in the hot plate test. The MPH (Maximum Possible Analgesia) has been found to be 14.285 for 300 mg/kg dose of the crude extract while the MPH for diclofenac was 15.857 after 60 min of administration in the hot tail-flick method. The crude extract at 300 and 200 mg/kg doses showed significant reduction in acetic acid induced writhings in mice with a maximum effect of 59.661% reduction at 300 mg/kg dose which is comparable to standard diclofenac sodium (73.4%). Alcoholic extract of C. specious showed significant inhibition in serotonin and egg albumin induced hind paw oedema in rats at 100, 200 and 300 mg/kg of the crude extracts respectively (Serotonin induced edema 44.22; 53.75; 58.51%; egg albumin induced edema - 41.317; 53.892; 59.880% inhibition after 4 h respectively). The antiinflammatory effects showed by the extract were comparable to that of standard indomethacin 5 mg/kg (Serotonin induced edema 77.56%; egg albumin induced edema 77.844% inhibition after 4 h). These results suggest that the extract possesses both the anti-inflammatory and analgesic activity on mice and rat model.

Study on the Effect of Chaenomelis Fructus.Herba Chelidonii.Clematis Florida Thunb on Inflammation and Liver Function of Rats with Arthritis (관절염 백서의 염증과 간기능에 대한 목과.백굴채.위영선(木瓜.白屈菜.威靈仙)의 효능연구)

  • The purpose of this study is to observe the effects of Chaenomelis Fructus Herba Chelidonii Clematis Florida Thunb mixed water extract on the inflammatory edema induced by Found's complete adjuvant and on different blood volume, and then to demonstrate its efficiencies as well as its safety. We injected Chaenomelis Fructus Herba Chelidonii Clematis Florida Thunb mixed water extract on the inflammatory edema induced by Found's complete adjuvant once daily at the same time for a week, then observed the paw-edema, the counts of the blood WBC, Serum total protein, Serum total bilirrubin, SGOT and SGPT The result obtained is as follows : Inflammatory edema on the right plantar in both sample groups showed a significant reduce compared with that in the control group. The counts of WBC in both sample groups showed a significant decrease compared with that in the control group. The volume of serum total protein in the Sample A group showed a significant decrease compared with that in the control group. The volume of serum total bilirrubin, GOT and GPT in both sample groups was not changed compared with that in the control group. As a result of this experiment, it is concluded that Chaenomelis Fructus Herba Chelidonii Clematis Florida Thunb mixed water extract showed that therapeutic effect on the anti-inflammation on arthritis; otherwise it showed non-toxic effect on the liver function.

Clinical Responses of Ultrasound on Adjuvant-Induced Arthritis in Rats (Adjuvant로 유발된 관절염의 흰 쥐에서 초음파의 임상반응)

  • The purpose of this study was to assess the effects of ultrasound on adjuvant-induced arthritis in rats. Adjuvant arthritis was induced in 26 female Sprauge-Dawley rats by the subcutaneous injection of a single dose of .1 mL of Complete Freund's Adjuvant (CFA) (1 mg of Mycobacterium Butyricum suspended in .1 $m{\ell}$ paraffin oil) into the right hind paw. After confirming inflammatory edema and arthritis in the paw 2 weeks later, the arthritic rats were divided into 3 groups, i.e., a control group, a pulsed ultrasound group (Group A), and a continuous ultrasound group (Group B) with 8 rats placed in each group. The rats in Group A were treated with pulsed ultrasound at 1 MHz frequency with .5 $W/cm^2$ intensity in 1 : 4 mode for 3 minutes. The rats in Group B were treated with continuous ultrasound at 1 MHz frequency with 2 $W/cm^2$ intensity in the continuous mode for 3 minutes. The ultrasound treatment was done in the left and right ankles for 2 weeks. Clinical, radiographic and histopathologic findings were then evaluated before and after treatment and yielded the following results. 1. No significant difference was present in body weight between the control group and the treated groups. 2. A statistically significant decrease in the edema of the paw was seen in the rats in Group A that was treated with pulsed ultrasound by 26~29 days after the treatment started (p<.05). 3. According to radiological examination, Group A showed the lowest score in arthritis scale which means it showed a tendency to suppress arthritic inflammation of the left and right hind paws. However, no statistically significant difference was present in the score between the control group, Group A and Group B. 4. According to histopathologic findings, the degree of infiltration by inflammatory cells and hypertrophy of the synovium were less in Group A compared with the control group and Group B. The results of the study show that rats that were treated with the pulsed ultrasound effectively suppressed adjuvant arthritis. However, more effort is needed to objectively prove the effectiveness of ultrasound by developing more sensitive testing methods that could quantitatively evaluate the treatment effects of acute rheumatoid arthritis and by trying out different ultrasound treatment methods.

Effects of Curcuma longa $L_{INNE}$ Pharmacopuncture at $ST_{36}$ on Adjuvant Induced Arthritis in Rats (강황(薑黃)약침이 CFA로 유발된 류마티스성 관절염에 미치는 영향)

  • Objectives : The objective of this study was to investigate the effects of Curcuma longa $L_{INNE}$ pharmacopuncture at $ST_{36}$ on Complete Freund's Adjuvant(CFA)-induced arthritis in rats. Materials and methods : Arthritis was induced by injecting CFA subcutaneously into the left knee joint and paw, and Curcuma longa $L_{INNE}$ pharmacopuncture(CLL-A. $0.0343{\mu}g/kg$; CLL-B. $0.171{\mu}g/kg$; CLL-C. $0.343{\mu}g/kg$) was injected at $ST_{36}$ each other day for 5 times beginning on day 10 after the CFA injection. Paw edema, withdrawal response, hematological, serological and histological observation were assessed. Results : In paw edema volume all 3 groups(CLL-A, CLL-B, CLL-C) showed significant decrease compared to the CFA control group. In withdrawal response to reaction time and withdrawal response to force all 3 groups(CLL-A, CLL-B, CLL-C) showed significant increase compared to the CFA control group. In serum AST, group CLL-C showed significant decrease compared to the CFA control group. In histological observations, in all 3 groups, more normal chondrocytes were observed compared to the CFA control group and safranin O stain showed high positive reaction in the cartilage tissue close to the bone tissue. Conclusions : The results suggest that Curcuma longa $L_{INNE}$ Pharmacopuncture at $ST_{36}$ has a suppressing inflammation effect on Freund's adjuvant arthritis in rats.

The effect of Gihyeolgwanjeolbang-A(CWS-A) on adjuvant-induced arthritis in rats (기혈관절방(氣血關節方)A(CWS-A)가 CFA로 유발(誘發)된 관절염(關節炎)에 미치는 영향(影響))

  • Objective : To investigate effects of herbal preparations formulated(Gihyeolgwanjeolbang-A, CWS-A) on complete Freund's adjuvant (CFA)-induced arthritis in rats. Method : Arthritis was induced by injecting CFA subcutaneously into the left knee joint and paw, and the herbal preparations formulated(CWS-A I, 82.3 mg/kg ; CWS-A II, 164.6 mg/kg) was administered orally (i.g.) for 10 consecutive days beginning on day 10 after the injection. External shape, paw edema, inflammatory cytokines tumor necrosis factor alpha (TNF-alpha) and interleukin-1 beta (IL-1beta), and histological observation were assessed. Result : In swelling of the paw, CWS-A I and CWS-A II group in 15 days and 20 days were significantly reduced compared to controls. In serum ALT, CWS-A I and CWS-A II group were significantly reduced compared to controls. In TNF-${\alpha}$, CWS-A I and CWS-A II group were a tendency reduced compared to controls. In HGB, HCT, MCHC of erythrocytes, CWS-A II group was increased compared to controls. In histological observations, CWS-A II group was observed synoviocytes more than control group and was observed proteoglycans in the deep layers. Conclusion : The data suggest that CWS-A significant anti-arthritic effects that may be mediated by suppressing inflammatory parameters.

Effects of Sweet Bee Venom Pharmacopuncture and Low Level Laser Acupuncture on a Rat Model with Complete Freund's Adjuvant-induced Arthritis (Sweet 봉약침과 저준위 레이저침이 Complete Freund's Adjuvant로 유발된 관절염 백서 모델에 미치는 영향)

  • Purpose : This research was conducted to investigate the effect of sweet bee venom pharmacopuncture and low level laser acupuncture on paw edema, pain index, anti-inflammatory factor, AST, ALT and complete blood cell count of a rat model with Complete Freund's Adjuvant-induced arthritis. Methods : Five experimental groups were formed with each consisting of six rats: normal group, control group, sweet bee venom pharmacopuncture group, lower level laser acupuncture group, and sweet bee venom pharmacopuncture, lower level laser acupuncture group. The experimental model of arthritis was induced by two injections of Freund's adjuvant into the left knee joint of Sprague Dawley(SD) rats. The second injection of Freund's adjuvant was given ten days after the first one. Ten days later, sweet bee venom pharmacopuncture and low level laser acupuncture were administered separately or together by assigned groups at $GB_{34}$ and $GB_{39}$ of rats twice a week for a total of six times. Thereafter, edema rate, pain index, tumor necrosis factor-${\alpha}$, interleukin-6, aspartate aminortansferase, alanine aminotransferase and complete blood cell count were measured. Results : We noticed synergic effects of sweet bee venom pharmacopuncture and low level laser acupuncture according to the results of the paw edema and Von Frey pain index. The sweet bee venom pharmacopuncture(BVA) and sweet bee venom pharmacopuncture+ low level laser acupuncture(BVA+LLA) groups experienced a more significant effect when compared with the control group. Conclusions : These results suggest that Sweet Bee Venom Pharmacopuncture and low level laser acupuncture at GB34 and GB39 have a significant anti-inflammatory effect on Freund's adjuvant arthritis in rats.

Pharmacokinetic Properties and Antiinflammatory Activity of Ketoprofen Lotion (케토프로펜 로오숀의 약물동력학적 특성과 항염증작용)

  • The pharmacokinetic properties and antiinflammatory activity of 3% ketoprofen lotion (ID-lotion), formulated with poloxamer 407, were evaluated using rats. For the pharmacokinetic study, the lotion, at the dose of 4.5 mg/kg, was applied on the dorsal skin of rats and the drug concentration in plasma was determined using an HPLC method. As references, ketoprofen suspended in saline was administered orally, and E-lotion, which is a 3% ketoprofen lotion in the Japanese market was applied transdermally. Following the transdermal application of ID-lotion and E-lotion, $C_{max}$ were 316 $\pm$22.3 ng/ml and 163 $\pm$ 12.2 ng/ml, respectively, at the same Tma of 2 hours postdose, while $C_{max}$ and $T_{max}$ after oral administration of the drug were 1,030$\pm$89.1 ng/ml and 0.25 hours, respectively. Relative bioavailabilities of ID-lotion and I-lotion were 69.3% and 34.2%, respectively. The antiinflammatory activity of the two 3% ketoprofen lotions was evaluated with carrageeneninduced edema method after 50 mg of the lotions was applied on the paw of rats. ID-lotion showed 67.6% inhibition of the edema formation, while I-lotion showed 34.\\\\`r%. The calculated ED5o after transdermal application of ID-lotion was 2.5 mg/kg, while that after oral administration was 7.0 mg/kg. Based on these results, the relative equiponderal availability of ID-lotion was 296% compared to the oral administration of ketoprofen.n.n.n.

The Therapeutic Effect of Natural Honeybee (Apis mellifera) Venom in Adjuvant-induced Arthritic Rat (관절염 유발 랫드에 대한 생봉독의 치료 효과)

  • This study was performed to assess that clinco-therapeutic effect of natural Italian honeybee (Apis mellifera) venom in adjuvant-induced arthritic rat. Ninety Sprague- Dawley rats of male were injected with complete Freund's adjuvant (CFA). Adjuvant arthritis was produced by a single subcutaneous injection of 1 mg Mycobacterium butyricum suspended in 0.1 ml paraffin oil into the right hindpaw. Righting reflex was uniformly lost and considered to be the point of arthritis development on day 14 after CFA injection. Experimental groups were divided into three groups. When arthritis was developed in the rat hind-paw, tested groups were administrated with prednisolone (10 mg/kg, p.o) and honeybee venom (one bee, s.c) at an interval of two days. Control group was subcutaneously injected with 0.1 ml of physiological saline solution in the rat at an interval of two days. Clinical findings, hematological values and histopathological findings were observed during or after the drugs administration. In tested groups, the development of inflammatory edema and polyarthritis on day 14 after treatment was suppressed. No significant differences of hindpaw edema volume and lameness score between prednisolone and honeybee venom groups were observed during or after therapeutic drugs treatment. WBC counts of prednisolone and honeybee venom treatment groups as compared with the control group were getting remarkably decreased during or after the therapeutic drugs administration(p<0.01). Erosions of articular cartilage and inflammatory cell infiltrations during or after the therapeutic drugs treatment was effectively suppressed in natural honey venom.
